Antibody Responses Elicited by Vaccination with Recombinant LaSota Virus Expressing IBV Spike in Chickens with Maternal Antibodies.

Vaccination with a recombinant Newcastle disease virus (NDV) LaSota (LS) strain expressing Arkansas (Ark) -type infectious bronchitis virus (IBV) spike ectodomain (Se) protein and granulocyte-macrophage colony-stimulating factor (GMCSF) (rLS/ArkSe.GMCSF, where rLS stands for recombinant LS) was evaluated in chickens of commercial origin with NDV and IBV maternally derived antibodies (MDAs). Chickens were vaccinated ocularly with rLS/ArkSe.GMCSF at either 2, 8, 15, or 30 days of age (DOA). Control chickens were vaccinated with the rLS virus (not expressing IBV SE or GMCSF) on the same days. Spe

Investigation of suspected Newcastle disease (ND) outbreaks in Egypt uncovers a high virus velogenic ND virus burden in small-scale holdings and the presence of multiple pathogens.

Highly contagious Newcastle disease (ND) is associated with devastating outbreaks with highly variable clinical signs among gallinaceous birds. In this study we aimed to verify clinical ND suspicions in poultry holdings in Egypt suffering from respiratory distress and elevated mortality, comparing two groups of ND-vaccinated poultry holdings in three governorates. Besides testing for Newcastle disease virus (NDV), samples were screened for infectious bronchitis virus (IBV) and avian influenza virus (AIV) by RT-qPCR as well as by non-directed cell-culture approach on LMH-cells. Virulent NDV was

Influence of drinking water quality on immune responses to viral vaccines in layer chickens.

Drinking water quality is a critical factor in poultry production, and suboptimal drinking water can negatively influence immune functions leading to reduced vaccine efficacy. In the field study, we assessed drinking water quality on Alberta layer farms and evaluated its impact on vaccine-induced immune responses. Chemical, physical and microbiological analyses were performed on 26 water samples collected from Alberta layer farms and serological response for poultry viral vaccines were evaluated at the same time. The water hardness, pH, bicarbonates, and dissolved sodium exceeded acceptable li

Caecal microbiota composition of experimental inbred MHC-B lines infected with IBV differs according to genetics and vaccination.

Interactions between the gut microbiota and the immune system may be involved in vaccine and infection responses. In the present study, we studied the interactions between caecal microbiota composition and parameters describing the immune response in six experimental inbred chicken lines harboring different MHC haplotypes. Animals were challenge-infected with the infectious bronchitis virus (IBV), and half of them were previously vaccinated against this pathogen. We explored to what extent the gut microbiota composition and the genetic line could be related to the immune response, evaluated th

Effect of different vaccination strategies on IBV QX population dynamics and clinical outbreaks.

The extreme variability and rapid evolution of Infectious bronchitis virus (IBV) has always represented the key challenge for its control because of the limited cross-protection among different strains. Several experimental trials have proven a broadening of the protection spectrum when animals are vaccinated with multiple genotypes. Nevertheless, the conditions of vaccine administration in field are so different that the generalization of experimental results is, at least, questionable. In the present study a large scale epidemiological-phylodynamic approach was used to reconstruct the demogr

Host Immune Response Modulation in Avian Coronavirus Infection: Tracheal Transcriptome Profiling In Vitro and In Vivo.

Infectious bronchitis virus (IBV) is a highly contagious Gammacoronavirus causing moderate to severe respiratory infection in chickens. Understanding the initial antiviral response in the respiratory mucosa is crucial for controlling viral spread. We aimed to characterize the impact of IBV Delmarva (DMV)/1639 and IBV Massachusetts (Mass) 41 at the primary site of infection, namely, in chicken tracheal epithelial cells (cTECs) in vitro and the trachea in vivo. We hypothesized that some elements of the induced antiviral responses are distinct in both infection models. We inoculated cTECs and inf

The establishment and characteristics of cell-adapted IBV strain H120.

Avian infectious bronchitis virus is an important pathogen in poultry worldwide. Vaccination is the only effective way to prevent and control IBV infection. H120, one of the safest vaccine strains, which has been used worldwide as a primary vaccine, cannot adapt to passaged cells, which severely restricts the quality of the vaccine. Based on the reverse genetics of our previous research work, we constructed the recombinant R-H120-Beaudette-p65(S) strain by replacing the complete spike gene of H120 with the corresponding spike gene from the Beaudette p65 strain. Some biological characteristics,

AI-based monitoring of broiler vocal repertoire dynamics reveals robust developmental and diurnal patterns but limited disease sensitivity.

Broiler vocalizations may contain relevant information about the welfare and health status; however, there is limited information on how disease affects vocal patterns. This study aimed 1) to investigate whether specific vocalization changes emerge in response to two important poultry diseases: intestinal (Eimeria spp.) and respiratory (infectious bronchitis virus, IBV; combined with avian pathogenic Escherichia coli, APEC), and 2) to characterize changes in broiler chicken vocalizations over age (14-41 days of age), and across time of the day (morning: 6:00 AM-12:00 PM; afternoon: 12:00 PM-6:

Co-infections of Enterococcus cecorum and various avian pathogens resulted in varying rates of SPF broilers with an E. cecorum infection.

Typical E. cecorum lesions can be reproduced in SPF broilers after intravenous, aerosol and oral inoculations. The respiratory route is potentially an infection route for pathogenic E. cecorum bacteria.Co-infections tested in this study or dexamethasone do not exacerbate the proportion of E. cecorum lesions.M.s. in combination with IBV or NDV vaccines exacerbates the proportion of positive reisolations.Immunosuppression induced by early CAV infection increases the proportion of positive reisolations.

Dark brooders during rearing affect fearfulness but not production parameters of broiler chickens.

Dark brooders are often suggested as enrichment for broiler chickens because they promote natural motivated behaviors compared to conventional barren environments in broiler production. Although the use of brooders is thought to convey benefits for animal welfare, it is not well described how they might affect parameters related to production and health. The objective of this study was to investigate if providing dark brooders affects production, health, behavior, and welfare compared to a control group without dark brooders. Six pens of each treatment were used, keeping 60 mixed-sex chickens

Age-dependent immune responses and immune protection after avian coronavirus vaccination.

Infectious bronchitis virus (IBV) is an endemic disease of chickens and a major contributor to economic losses for the poultry industry despite vaccination. Recent observations indicated that chicks may have an immature immune system immediately after hatching when vaccinated for IBV. Therefore we hypothesized that early IBV vaccination will generate an immature, poorly protective IBV-specific immune response contributing to immune escape and persistence of IBV. To test this hypothesis the IBV-specific immune response and immune protection were measured in chicks vaccinated at different ages.

Recombinant infectious bronchitis coronavirus Beaudette with the spike protein gene of the pathogenic M41 strain remains attenuated but induces protective immunity.

We have replaced the ectodomain of the spike (S) protein of the Beaudette strain (Beau-R; apathogenic for Gallus domesticus chickens) of avian infectious bronchitis coronavirus (IBV) with that from the pathogenic M41 strain to produce recombinant IBV BeauR-M41(S). We have previously shown that this changed the tropism of the virus in vitro (R. Casais, B. Dove, D. Cavanagh, and P. Britton, J. Virol. 77:9084-9089, 2003). Herein we have assessed the pathogenicity and immunogenicity of BeauR-M41(S). There were no consistent differences in pathogenicity between the recombinant BeauR-M41(S) and its

First report of molecular genotyping, pathotyping, and histopathological characterization of lentogenic genotype II Newcastle disease virus circulating in young ostrich flocks in Egypt.

BACKGROUND AND AIM: Newcastle disease virus (NDV) is one of the most economically important avian pathogens worldwide, yet information regarding its molecular epidemiology and pathological characteristics in ostriches remains limited. This study investigated the prevalence, molecular characteristics, pathotype, phylogenetic relationships, and histopathological changes associated with NDV circulating in young ostrich flocks in Egypt. MATERIALS AND METHODS: A total of 60 tissue samples were collected from diseased, non-vaccinated ostriches aged 3 weeks to 2 months from eight farms across four E

ROS-mitochondria-NLRP3/STING axis mediates the enhanced anti-coronavirus activity of ZIF8 nanoparticles.

Coronaviruses (CoVs) pose serious threats to both animal and human health, underscoring the urgent need for broad-spectrum antiviral strategies that can provide rapid and immune-independent protection beyond vaccination. In this study, we developed zeolitic imidazolate framework-8 (ZIF8) as an innovative zinc-based anti-coronavirus nanoagent and revealed that ZIF8 exhibits markedly enhanced antiviral activity relative to the commonly used zinc oxide (ZnO). Using porcine epidemic diarrhea virus (PEDV) as a model α-coronavirus, mechanistic studies demonstrated that ZIF8 alleviates excessive reac

Targeting viral Fe-S clusters with TEMPOL confers broad-spectrum anti-coronavirus activity.

Coronaviruses (CoVs) can cause severe respiratory and enteric diseases, posing major threats to human health and the livestock industry. However, the host metabolic mechanisms underlying viral replication remain unclear. Using porcine epidemic diarrhea virus (PEDV) as the primary model, this study investigated the role of iron metabolism in CoV replication and its mechanisms. Our results showed that PEDV infection markedly induced intracellular iron accumulation, while exogenous iron supplementation enhanced viral replication and iron chelation suppressed it. Mechanistically, iron enhances vir

Complex pathogen interactions in upper respiratory tract infections in commercial free-range layers: A cross-sectional study.

The global egg industry of free-range chickens is rapidly expanding owing to perceived higher welfare standards. However, commercial free-range layers are known to be prone to a number of infections including upper respiratory tract infections (URTIs) leading to significant economic and welfare concerns. The present study explored the prevalence and diversity of respiratory tract pathogens in URTIs in free-range layers in Australia. Swabs/tissues were collected from conjunctiva, nasal cavity, infraorbital sinus, and upper trachea of birds from 15 farms across Australia with ongoing URTIs as we

Development of a TaqMan-based real-time PCR assay for rapid and specific detection of fowl aviadenovirus serotype 4.

Twelve serotypes of fowl aviadenovirus, namely, FAdV-(1-8a and 8b-11), have been identified, among which FAdV-4 is the aetiologic agent of hepatitis hydropericardium syndrome (HHS) in chickens. Outbreaks of HHS have been documented in many countries, causing significant economic losses. Real-time PCR methods described so far in the literature cross-detect different serotypes of FAdVs. In this study, we aimed to develop a TaqMan-based real-time PCR assay for the specific detection of FAdV-4. A pair of primers targeting the hexon gene and a TaqMan probe were designed. Using different copy number

Establishment and application of a triplex real-time fluorescent quantitative RT-PCR assay for simultaneous detection of H5, H7 subtype AIV and virulent NDV.

To rapidly monitor clinical infections induced by highly pathogenic avian influenza (HPAI) of H5/H7 subtypes and virulent Newcastle disease virus (NDV), mitigate economic losses in intensive poultry industry and reduce zoonotic spillover risks to human population, a one-tube triplex TaqMan real-time RT-qPCR was developed in this study. Three pairs of subtype-specific primers and corresponding fluorophore-labeled TaqMan probes were rationally designed targeting conserved hemagglutinin (HA) coding regions of H5-AIV, H7-AIV and fusion (F) gene of virulent NDV, respectively, referring to published

Occurrence of Antibody-Dependent Enhancement of Avian Infectious Bronchitis in Target Animal Experiments.

Outbreaks of avian infectious bronchitis virus (IBV) often occur in vaccinated flocks. The antibody-dependent enhancement (ADE) has been proposed as a potential mechanism underlying coronavirus vaccine failure. However, this hypothesis has yet to be substantiated in flocks. This study demonstrates ADE occurrence in IBV (gamacoronavirus) in vitro and in vivo. Using the SPF chicken host model, primary infection with an O-glycosylation-modified attenuated strain enhanced pathogenesis upon secondary homologous/heterologous virulent challenge, increasing morbidity/mortality (≥30%), pathological les

Molecular surveillance of coronaviruses in livestock in northern Vietnam.

Coronavirus (CoV) infections continue to pose a significant challenge to global public health and food security. The COVID-19 pandemic has underscored the importance of One Health approaches and improved surveillance systems. This is due to the considerable potential of CoVs to infect a broad spectrum of hosts and cause respiratory diseases. The present study investigated CoVs circulating in livestock in northern Vietnam using molecular detection and phylogenetic analysis. A total of 412 samples collected from livestock were analysed for the presence of CoVs, and we detected infectious bronchi
