METTL3-Mediated m6A Modification of DSC1 Alleviates Inflammation and Cellular Dysfunction in Periodontitis.

BACKGROUND: The inflammation and cellular dysfunction in periodontitis largely depend on N6-methyladenosine (m6A) modification-related epigenetics. However, the role of methyltransferase-like 3 (METTL3)-differentiated m6A modification of desmocollin 1 (DSC1) in periodontitis remains to be investigated. METHODS: Periodontal ligament (PDL) tissues were collected from patients with periodontitis and healthy volunteers, and a periodontitis model was constructed using lipopolysaccharide (LPS) and human periodontal ligament fibroblasts (HPLF). METTL3 and DSC1 levels in the specimens and cells were

Controlled Inflammation Drives Neutrophil-Mediated Precision Drug Delivery in Heterogeneous Tumors.

Tumor heterogeneity remains a formidable obstacle in targeted cancer therapy, often leading to suboptimal treatment outcomes. This study presents an innovative approach that harnesses controlled inflammation to guide neutrophil-mediated drug delivery, effectively overcoming the limitations imposed by tumor heterogeneity. By inducing localized inflammation within tumors using lipopolysaccharide, it significantly amplify the recruitment of drug-laden neutrophils to tumor sites, irrespective of specific tumor markers. This strategy not only enhances targeted drug delivery but also triggers the re

Isolinderalactone attenuates atherosclerosis through inhibiting NF-κB-mediated inflammation in macrophages.

Atherosclerosis is a chronic inflammatory disease affecting medium and large arteries, characterized by lipid ectopic deposition in arterial wall, whose pathogenesis is closely associated with persistent excessive inflammatory responses. Isolinderalactone (ILL), the primary active sesquiterpene lactone component derived from the traditional Chinese medicine Lindera aggregata, has been demonstrated to possess significant anti-inflammatory and anti-proliferative activities. This study aims to investigate the therapeutic potential of ILL in atherosclerosis and its underlying molecular mechanisms.

Translocator protein facilitates neutrophil-mediated mucosal inflammation in inflammatory bowel diseases.

BACKGROUND: Inflammatory bowel diseases (IBD), including ulcerative colitis (UC) and Crohn's disease (CD), are chronic gastrointestinal disorders with an increasing global prevalence and significant healthcare impact. The exact etiology of this condition remains unclear. Neutrophils play a critical role in IBD pathogenesis. Translocator protein (TSPO), a mitochondrial protein linked to immune responses, has demonstrated potential as an inflammatory marker. However, its role in IBD remains underexplored. AIM: To investigate the role of TSPO in IBD pathogenesis, particularly in neutrophils. ME

Impact of inflammation levels on the liver during the COVID-19 pandemic: a mediation analysis.

OBJECTIVE: The level of systemic inflammation plays a critical role in influencing liver function. This study aims to investigate the impact of various composite inflammatory markers on the liver during the COVID-19 pandemic. METHODS: Data were sourced from the updated NHANES database, reflecting information collected during the COVID-19 pandemic between 2021 and 2023. Based on complete blood count (CBC) data, we identified six composite inflammatory markers: neutrophil-to-lymphocyte ratio (NLR), derived neutrophil-to-lymphocyte ratio (dNLR), monocyte-to-lymphocyte ratio (MLR), neutrophil-mon

Association between heavy metal exposure and chronic cough: the mediating roles of inflammation.

BACKGROUND: Right now, epidemiological research examining the connection between blood heavy metal exposure and chronic coughing is still deficient. Therefore, the survey was aimed at the effects of multiple heavy metals on chronic cough. METHOD: In this investigation, 2647 individuals from NHANES were included. This study applied multiple statistical models to systematically explore the associations between single and mixed blood metal with the prevalence of chronic cough, including logistic regression, weighted quantile sum (WQS) regression, bayesian kernel machine regression (BKMR), and th

Specialized Pro-Resolving Mediator loaded Extracellular Vesicles Mitigate Pulmonary Inflammation.

Specialized pro-resolving mediators (SPMs), including lipoxins derived from arachidonic acid and resolvins, protectins, and maresins derived from docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), orchestrate the active resolution of inflammation. These SPMs are biosynthesized through the coordinated interaction of various cells in a process known as transcellular biosynthesis, involving the sequential action of cyclooxygenase-2 (COX-2), 5-lipoxygenase (5-LOX), 12-lipoxygenase (12-LOX), and/or 15-lipoxygenase (15-LOX) enzymes. Additionally, Aspirin-triggered Resolvins are produced by

Association Between Spinal Cord Injury and Cognitive Impairment, and the Mediating Role of Inflammation.

BACKGROUND: Spinal cord injury (SCI), typically resulting from trauma or cranial surgery, often causes motor and sensory deficits and may also impair brain function, leading to cerebral edema, elevated intracranial pressure, and cognitive decline. Growing evidence implicates systemic inflammation as a key mediator of SCI-related cognitive impairment, yet clinical validation remains limited. This study explores the association between SCI and cognitive dysfunction, with a focus on the mediating role of inflammation. METHODS: A retrospective cohort of 157 participants (SCI patients and controls

Loss of TRPV4 decreases NFκB-mediated myometrial inflammation and prevents preterm labor.

Inflammation is a key initiating event in both spontaneous term and preterm labor. However, the link between inflammation and the onset of labor remains incompletely understood. We identified the transient receptor potential vanilloid 4 (TRPV4) channel as a critical regulator of myometrial calcium (Ca2+) entry and contractility. In this study, we aimed to determine if the TRPV4 channel regulates uterine inflammation and its subsequent effects on myometrial contractility in experimental preterm labor. We demonstrated that global loss of TRPV4 protected mice against inflammation-induced preterm

Impaired SREBP1-mediated regulation of lipid metabolism promotes inflammation in chronic endometritis.

Chronic endometritis (CE) is an inflammatory disease of the uterus that is associated with infertility and poor reproductive outcomes. Although most cases of CE are attributed to bacterial infections, antibiotic treatment is sometimes ineffective, and the mechanisms underlying the development and persistence of inflammation in CE are poorly understood. In the present study, we established a novel mouse model of CE that causes fetal death without affecting implantation and demonstrated that dysregulation of lipid metabolism contributes to its pathology. A deficiency in SREBP1, a key regulator o

The MFN2 Q367H variant reveals a novel pathomechanism connected to mtDNA-mediated inflammation.

Pathogenic variants in the mitochondrial protein MFN2 are typically associated with a peripheral neuropathy phenotype, but can also cause a variety of additional pathologies including myopathy. Here, we identified an uncharacterized MFN2 variant, Q367H, in a patient diagnosed with late-onset distal myopathy, but without peripheral neuropathy. Supporting the hypothesis that this variant contributes to the patient's pathology, patient fibroblasts and transdifferentiated myoblasts showed changes consistent with impairment of several MFN2 functions. We also observed mtDNA outside of the mitochondr

Sirtuin 3 is required for the dexmedetomidine-mediated alleviation of inflammation and oxidative stress in nephritis.

INTRODUCTION: Although sirtuin 3 (SIRT3) is known to be involved in dexmedetomidine (DEX)-mediated alleviation of renal ischemia and reperfusion injury, the influence of the association between DEX and SIRT3 on nephritis development remains unclear. In this study, the role of SIRT3 in DEX-mediated amelioration of inflammation and oxidative stress in nephritis as well as the possible underlying mechanism were explored in vivo and in vitro. METHODS: An animal model of glomerulonephritis was generated by injecting mice with interferon-alpha (IFNα)-expressing adenoviruses, and periodic acid-Schif

Granzyme K mediates IL-23-dependent inflammation and keratinocyte proliferation in psoriasis.

Psoriasis is an inflammatory disease with systemic manifestations that most commonly presents as itchy, erythematous, scaly plaques on extensor surfaces. Activation of the IL-23/IL-17 pro-inflammatory signaling pathway is a hallmark of psoriasis and its inhibition is key to clinical management. Granzyme K (GzmK) is an immune cell-secreted serine protease elevated in inflammatory and proliferative skin conditions. In the present study, human psoriasis lesions exhibited elevated GzmK levels compared to non-lesional psoriasis and healthy control skin. In an established murine model of imiquimod (

Exposure to gestational diabetes mellitus increases subclinical inflammation mediated in part by obesity.

Gestational diabetes mellitus (GDM) is a frequent and serious complication of pregnancy, often associated with obesity. Metabolic dysfunction and metainflammation are evident in both obesity and GDM. In this cross-sectional study, we aimed at defining the direct contribution of the immune system in GDM, across the main metabolic tissues, specifically focussing on elucidating the roles of obesity and GDM to the clinical outcome. Using immunoassays and multicolour flow cytometry, cytokine profiles and immune cell frequencies were measured in maternal circulation and central metabolic tissues [pl

Improvements in Insulin Resistance and Glucose Metabolism Related to Breastfeeding Are Not Mediated by Subclinical Inflammation.

Background: Lactation is known to improve insulin resistance, but this phenomenon remains poorly understood. Our goal was to evaluate whether subclinical inflammation could mediate the association between breastfeeding (BF) and improvement in glucose metabolism and markers of insulin resistance (MIRs) in the postpartum. Methods: A total of 95 adult women (≥18 years) with a BMI ≥ 25 kg/m2 from the outpatient clinic of the Federal University of São Paulo were followed from early pregnancy until 60 to 180 days postpartum. The patients were divided based on their BF status: BF and non-BF groups. A

Piezo1-Mediated Mechanotransduction Contributes to Disturbed Flow-Induced Atherosclerotic Endothelial Inflammation.

BACKGROUND: Disturbed flow generates oscillatory shear stress (OSS), which in turn leads to endothelial inflammation and atherosclerosis. Piezo1, a biomechanical force sensor, plays a crucial role in the cardiovascular system. However, the specific role of Piezo1 in atherosclerosis remains to be fully elucidated. METHODS AND RESULTS: We detected the expression of Piezo1 in atherosclerotic mice and endothelial cells from regions with disturbed blood flow. The pharmacological inhibitor Piezo1 inhibitor (GsMTx4) was used to evaluate the impact of Piezo1 on plaque progression and endothelial infl

Ponatinib Drives Cardiotoxicity by S100A8/A9-NLRP3-IL-1β Mediated Inflammation.

BACKGROUND: The tyrosine kinase inhibitor ponatinib is the only treatment option for chronic myelogenous leukemia patients with T315I (gatekeeper) mutation. Pharmacovigilance analysis of Food and Drug Administration and World Health Organization datasets has revealed that ponatinib is the most cardiotoxic agent among all Food and Drug Administration-approved tyrosine kinase inhibitors in a real-world scenario. However, the mechanism of ponatinib-induced cardiotoxicity is unknown. METHODS: The lack of well-optimized mouse models has hampered the in vivo cardio-oncology studies. Here, we show t
