Innate immune role of IL-6 in influenza a virus pathogenesis.

Interleukin-6 (IL-6), a pleiotropic cytokine, is induced by infection of influenza A virus (IAV), where it plays a pivotal role in immune defense and the regulation of inflammation. IAV induces IL-6 transcription upon recognition by pattern recognition receptors, which activate downstream signaling cascades, leading to the activation of transcription factors. Activated transcription factors subsequently regulate the expression of IL-6 in innate immune cells, such as macrophages, dendritic cells, and epithelial cells. IL-6 contributes to antiviral immunity by promoting the recruitment of immune

Emerging role of innate immunity in organ transplantation: part I: evolution of innate immunity and oxidative allograft injury.

The innate immune system is an evolutionarily highly conserved first rapid line of host defense against tissue injury and consists of a whole family of mobile and sessile cells such as antigen-presenting cells, innate lymphocytes, neutrophils, and vascular cells-dendritic cells representing the bridge to development of an adaptive immune response. The system depends on molecules collectively known as pattern recognition receptors to survey the extracellular space and the cytoplasm for the presence of exogenous pathogen-associated molecular patterns derived from microorganisms as well as damage

MDA5 generates compact ribonucleoprotein complexes via ATP-dependent double-stranded RNA unwinding.

Long double-stranded RNA (dsRNA) in the cytosol acts as a potent inflammatory molecule recognized by the receptor MDA5, triggering the innate immune response. Mutations affecting MDA5 ATPase activity lead to severe pathological conditions. MDA5 nucleoprotein filament assembly-disassembly dynamics are proposed to regulate dsRNA recognition, though the exact mechanism remains unclear. Here, we employed magnetic tweezers to monitor the assembly and manipulate MDA5 filaments at the single dsRNA level. Following a slow nucleation event, MDA5 assembles cooperatively and directionally into (partial)

Cinnamon and Derivatives as Modulators of Toll-like Receptors.

Toll-like receptors (TLRs) are critical components of the innate immune system, acting as pattern recognition receptors and interacting with oxidative stress. TLR2, TLR4, and TLR7 play distinct yet overlapping roles in immune regulation, contributing to both physiological processes and disease progression. Therefore, modulation of TLR signaling pathways represents a promising therapeutic strategy for a wide range of diseases. Natural antioxidants have attracted considerable interest as modulators of TLRs, with cinnamon-derived phytochemicals, particularly cinnamaldehyde, emerging as promising

Immunity: defense against infections essential for all living organisms.

All organisms need protection against infection. Bacteria are often primarily seen as infectious agents, but they also need protection against bacterial viruses, so-called bacteriophages. To this end, bacteria have developed very complex defense systems, including apoptosis-like mechanisms, restriction enzymes, and even adaptive-type mechanisms involving immunological memory of immune responses through a system called CRISPR-Cas. An earlier dominating view was that adaptive immunity in eukaryotes only exists in jawed vertebrates, as their immune system includes the classical and highly variabl

Human neutrophils produce IL-12, IL-10, PGE2 and LTB4 in response to Paracoccidioides brasiliensis. Involvement of TLR2, mannose receptor and dectin-1.

The functions of phagocytic cells against pathogens are initiated by the interaction between membrane receptors and molecular structures which compose the cell wall of these microorganisms. Thus our study aimed to identify the neutrophil receptors involved in the recognition of different strains of Paracoccidioides brasiliensis and the consequent modulation of immune response through the production of cytokines and inflammatory mediators. Neutrophils did not produce TNF-alfa in response to both strains. However, these cells produce IL-12, mainly in response to Pb 265, with participation of TLR

Viral proteins targeting host protein kinase R to evade an innate immune response: a mini review.

The innate immune system offers a first line of defense by neutralizing foreign pathogens such as bacteria, fungi, and viruses. These pathogens express molecules (RNA and proteins) that have discrete structures, known as the pathogen-associated molecular patterns that are recognized by a highly specialized class of host proteins called pattern recognition receptors to facilitate the host's immune response against infection. The RNA-dependent Protein Kinase R (PKR) is one of the host's pattern recognition receptors that is a key component of an innate immune system. PKR recognizes imperfectly d

Survey of innate immune responses to Burkholderia pseudomallei in human blood identifies a central role for lipopolysaccharide.

B. pseudomallei is a gram-negative bacterium that causes the tropical infection melioidosis. In northeast Thailand, mortality from melioidosis approaches 40%. As exemplified by the lipopolysaccharide-Toll-like receptor 4 interaction, innate immune responses to invading bacteria are precipitated by activation of host pathogen recognition receptors by pathogen associated molecular patterns. Human melioidosis is characterized by up-regulation of pathogen recognition receptors and pro-inflammatory cytokine release. In contrast to many gram-negative pathogens, however, the lipopolysaccharide of B.

Helicobacter pylori induces the production of interleukin-37 to promote broad immunosuppression and enhance colonization.

Helicobacter pylori infects approximately half of the world's population, resulting in lifelong gastric infections. To promote lifelong colonization, H. pylori modulates host immunity via unknown mechanisms. Here we show that H. pylori can harness the pan-immunosuppressive cytokine interleukin-37 (IL-37) to facilitate pathogenesis, enhance colonization and prevent the development of innate, cellular and humoral immunity. We show that H. pylori increased the production of IL-37 in human gastric biopsies, and IL-37 secretion by gastric epithelial cells and human gastric mucosoids. We found that

Molecular and Functional characterization of a novel C-type lectin during WSSV infection in Eriocheir sinensis.

C-type lectins (CTLs) are key pattern recognition receptors in innate immunity; however, their functions during white spot syndrome virus (WSSV) infection remain to be elucidated. Here, we identified a novel CTL gene from Chinese mitten crab (Eriocheir sinensis), designated as EsLec2. EsLec2 encodes a protein with a conserved carbohydrate-recognition domain (CRD) and shows phylogenetic clustering with other crustacean CTLs. RNA interference (RNAi) and recombinant protein (rEsLec2) injection assays revealed that EsLec2 downregulates ie1 and vp28 expression, indicating that it suppresses WSSV re

Protective role of Toll-like Receptor 3-induced type I interferon in murine coronavirus infection of macrophages.

Toll-like Receptors (TLRs) sense viral infections and induce production of type I interferons (IFNs), other cytokines, and chemokines. Viral recognition by TLRs and other pattern recognition receptors (PRRs) has been proven to be cell-type specific. Triggering of TLRs with selected ligands can be beneficial against some viral infections. Macrophages are antigen-presenting cells that express TLRs and have a key role in the innate and adaptive immunity against viruses. Coronaviruses (CoVs) are single-stranded, positive-sense RNA viruses that cause acute and chronic infections and can productivel

Role of Innate Immunity in Allergic Contact Dermatitis: An Update.

Our understanding of allergic contact dermatitis mechanisms has progressed over the past decade. Innate immune cells that are involved in the pathogenesis of allergic contact dermatitis include Langerhans cells, dermal dendritic cells, macrophages, mast cells, innate lymphoid cells (ILCs), neutrophils, eosinophils, and basophils. ILCs can be subcategorized as group 1 (natural killer cells; ILC1) in association with Th1, group 2 (ILC2) in association with Th2, and group 3 (lymphoid tissue-inducer cells; ILC3) in association with Th17. Pattern recognition receptors (PRRs) including toll-like rec

Eosinophilic Esophagitis and Microbiota: State of the Art.

Eosinophilic esophagitis (EoE) is a chronic, food-triggered, immune-mediated disease of the oesophagus, clinically characterized by symptoms referred to oesophagal dysfunction, and histologically defined by an eosinophil productive inflammation of the oesophagal mucosa, among other cell types. The involvement of an adaptive Th2-type response to food antigens in EoE was known since 2000; several cytokines and chemokines promote food-specific responses, during which local production of IgE, but also IgG4 derived from plasma cells in lamina propria of oesophagal mucosa might play an important rol

Murine dendritic cells transcriptional modulation upon Paracoccidioides brasiliensis infection.

Limited information is available regarding the modulation of genes involved in the innate host response to Paracoccidioides brasiliensis, the etiologic agent of paracoccidioidomycosis. Therefore, we sought to characterize, for the first time, the transcriptional profile of murine bone marrow-derived dendritic cells (DCs) at an early stage following their initial interaction with P. brasiliensis. DCs connect innate and adaptive immunity by recognizing invading pathogens and determining the type of effector T-cell that mediates an immune response. Gene expression profiles were analyzed using mic

Radioiodine-induced cGAS-STING signaling and antitumor immunity in thyroid cancer.

Radioiodine (RAI) is a cornerstone of treatment for differentiated thyroid cancer (DTC), yet the immunological consequences of RAI-induced DNA damage remain poorly understood. Here, we examine cGAS-STING signaling as a potential mechanistic link between RAI-induced genotoxic stress and antitumor immunity. RAI delivers prolonged, thyroid-selective radiation to iodine-avid tumors, providing a plausible source of extranuclear DNA capable of engaging innate DNA sensing. We propose that transient RAI-associated cGAS-STING activation in DTC may favor IRF3-type I interferon-dependent immune priming,

IFI208, an interferon-stimulated gene, promotes host antiviral responses through liquid-liquid phase separation of MDA5.

PYHIN (IFI200/HIN-200) family members are involved in host antiviral immune responses by detecting viral nucleic acids through HIN domains. Within this family, the function of IFI208, which lacks a HIN domain, remains largely unknown. In this study, we identify IFI208, a member of the PYHIN family, as an interferon stimulated gene (ISG). Upon exposure to viral DNA and RNA, IFI208 inhibits viral replication by increasing type I interferon (IFN) production. Mechanistically, upon infection, IFI208 is translocated to the cytoplasm through the chromosome region maintenance 1 (XPO1) transporter prot

RIG-I-dependent sensing of PEDV shapes epithelial antiviral immunity in the intestinal mucosa.

Porcine epidemic diarrhea virus (PEDV) is a highly contagious enteric coronavirus that causes lethal diarrhea in neonatal piglets, yet the viral pathogen-associated molecular patterns (PAMPs) that initiate mucosal antiviral immunity remain poorly defined. Here, integrating intestinal single-cell transcriptomics with in vivo and in vitro infection models, we delineate how PEDV is sensed by host innate immunity. We found that RIG-I, rather than MDA5, functions as the dominant cytosolic sensor that detects PEDV and initiates interferon-mediated antiviral defense in the intestinal mucosa. RIP-seq

Brain endothelial cells orchestrate a neuroprotective antiviral state in the CNS in response to peripheral viral pattern sensing.

West Nile virus (WNV) and other neurotrophic arboviruses are human pathogens that can cause life-threatening encephalitis. To uncover determinants of severe disease progression, we employed a murine infection model of WNV and experimentally uncoupled peripheral pathogen sensing from active viral replication. Peripheral sensing of a viral dsRNA mimic in the footpad led to robust induction of antiviral type I interferon (IFN-I)-stimulated genes and establishment of an antiviral state within the brain. Systematic approaches combining cytokine profiling, single-nuclei transcriptomics, immune modul

A tissue-intrinsic mechanism sensitizes HIV-1 particles for TLR-triggered innate immune responses.

In vivo, HIV-1 replicates within tissues, yet the impact of three-dimensional (3D) environments on viral spread remains unclear. Our laboratory previously showed that collagen-rich 3D extracellular matrix (ECM) imposes an Environmental Restriction to cell-free Virus Infectivity (ERVI). Here, we demonstrate that ERVI is mediated by adhesive ECM components assembled into tissue-like scaffolds. Transient interactions with collagen fibers rapidly diminish virion infectivity across diverse primary strains by impairing virus fusogenicity. Notably, collagen-experienced particles also induce a distinc

An inflammasome-driven differentiation program in intestinal stem cells protects against Salmonella infection.

Here we identify a stem cell-intrinsic immune mechanism whereby Lgr5+ ISCs detect intracellular Salmonella enterica and activate an inflammasome-dependent differentiation program. Using fluorescent-labeled S. enterica, single-cell transcriptomics, fate mapping, organoid models, and genetic perturbations, we show that invaded ISCs undergo rapid reprogramming toward antimicrobial peptide-enriched Paneth cells via ap
