The industrial use of certain insects, such as the black soldier fly (BSF, Hermetia illucens), has become a burgeoning way of converting residual biomass into a valuable source of biomolecules for the feed and food industry. Here, the integration of a valuable step as an upcycling technology using biological waste from the 5th instar BSF larvae processing as a source of bioactive chitosan was explored. The goal is to pave the way for sustainable chitosan production from insect-based resources in addition to proteins, lipids, and fertilizers. The macronutrient profile of the larval waste was ev

The neotropical insect Hermetia illucens has become a cosmopolite species, and it is considered a highly promising insect in circular and sustainable economic processes. Being able to feed on a wide variety of organic substrates, it represents a source of lipids and proteins for many uses and produces recyclable waste. We investigated the characteristics and differences in the poorly-known locomotory behaviour of larvae of different instars, paying particular attention to the unique characteristics of the prepupal stage, key to farming and industrial processes. Moreover, we attempted to develo

Tropomyosin is a pan-allergen in crustaceans known to cross-react with homologous proteins from edible insects. However, epitope-level cross-reactivity in edible insects remains poorly defined. This study aimed to identify IgE-binding epitopes of tropomyosin and to evaluate their cross-reactivity between shrimp and the edible two-spotted cricket. Here, sera from shrimp-allergic patients were screened for IgE-binding to proteins from Pacific white shrimp (Penaeus vannamei) and two-spotted cricket (Gryllus bimaculatus). Candidate tropomyosin epitopes from the two-spotted cricket were predicted.

On the background of the previous findings concerning the passage of a low molecular basic protein from cobra snake venom through the gut of a blowfly (Primor, N., Teitelbaum, Z. and Zlotkin, E. (1980) Biochim, Biophys, Acta 627, 71--81) several additional proteins were tested in the present study. With the aid of toxicity tests, radioiodinated proteins, gel filtration chromatography, and the usage of Sarcophaga falculata flies as test animals, the following information was obtained: 1. Out of the five species (representing four orders) of insects tested, only flies demonstrated the lethality

BACKGROUND: The PIWI-interacting RNA (piRNA) pathway is a conserved transposon silencing mechanism essential for genomic integrity and gametogenesis. While Piwi proteins are known to play essential roles in insect germline development, their functional characteristics remain largely unexplored in agricultural pests. Here, we investigated BdPiwi in Bactrocera dorsalis, a highly invasive pest whose exceptional reproductive capacity underpins its global pest status.
RESULTS: Phylogenetic analysis revealed that BdPiwi is evolutionarily conserved among insects and contains the canonical PAZ and PI

Many plant species have evolved surfaces that reduce insect attachment. Among such plants are deceptive trap flowers of Ceropegia. Their gliding zones consist of convex epidermal cells, each with a bristle-like central protuberance and a single small liquid droplet on its tip. So far, the molecular and physical mechanisms controlling the function of these droplets are unknown. We analyzed the droplets of Ceropegia sandersonii flowers by microscopic approaches, studied how they behave when getting in contact with the feet of fly pollinators, and analyzed their chemical composition. The droplets

Wheat (Triticum aestivum L.) is one of the most important staple crops all over the world. Its productivity is adversely affected by aphid infestation. Plant volatiles play a critical role in plant communication, inducing direct and indirect defenses against insect pests. However, little is known about the priming mechanism of key volatiles in wheat. To determine whether and how plant volatile induced defense priming in wheat against the grain aphid Sitobion avenae, a combination of insect bioassays, phytohormone and defense metabolite quantification, and transcriptome analyses were performed

Embryo microinjection techniques are essential for many molecular and genetic studies of insect species. They provide a means to introduce exogenous DNA fragments encoding genes of interest as well as favorable traits into the insect germline in a stable and heritable manner. The resulting transgenic strains can be studied for phenotypic changes resulting from the expression of the integrated DNA to answer basic questions or used in practical applications. Although the technology is straightforward, it requires of the investigator patience and practice to achieve a level of skill that maximize

Hepatitis E virus (HEV) is associated with acute hepatitis disease. Numerous truncated HEV capsid proteins have been successfully expressed using different expression systems. Among these, p495, a protein truncated at its N- and C-termini by 111 and 54 amino acids (aa), respectively (HEV ORF2 aa 112-606) can self-assemble into T = 1 virus-like particles (VLPs) when expressed by insect cells. A shorter p239 (aa 368-606) protein is a particulate antigen that we have previously used in our commercialized HEV vaccine, Hecolin. Here, we sought to express p495 in its soluble form (named Ep495) in E.

Mesothelin is a glycosylphosphatidylinositol-anchored glycoprotein that is highly expressed on the cell surface of malignant mesothelioma. Monoclonal antibodies against mesothelin are being evaluated for the treatment of mesothelioma. Immunocytokines represent a novel class of armed antibodies. To provide an alternative approach to current mesothelin-targeted antibody therapies, we have developed a novel immunocytokine based on interleukin-12 (IL12) and the SS1 Fv specific for mesothelin. IL12 possesses potent anti-tumor activity in a wide variety of solid tumors. The newly-developed recombina

Two benzenoid esters, methyl salicylate (MeSA) and methyl benzoate (MeBA), were detected from insect-damaged rice plants. By correlating metabolite production with gene expression analysis, five candidate genes encoding putative carboxyl methyltransferases were identified. Enzymatic assays with Escherichia coli-expressed recombinant proteins demonstrated that only one of the five candidates, OsBSMT1, has salicylic acid (SA) methyltransferase (SAMT) and benzoic acid (BA) methyltransferase (BAMT) activities for producing MeSA and MeBA, respectively. Whereas OsBSMT1 is phylogenetically relatively

Yersinia pestis diverged from Yersinia pseudotuberculosis</= 20 000 years ago, during which time it evolved to be transmitted by fleas. In comparing the ability of these closely related species to infect the rat flea Xenopsylla cheopis, we found that Y. pseudotuberculosis, unlike Y. pestis, is orally toxic to fleas. Fleas showed signs of acute toxicity, including diarrhoea, immediately after feeding on blood containing Y. pseudotuberculosis in response to protein toxin(s) produced by the bacteria. Adherence of Y. pseudotuberculosis to the midgut and large intracellular vacuoles in midgut epith

The preservation along evolution of specific core motifs in proteins of diverse functions and taxonomic origins pinpoints a possible developmental advantage at the structural level. Such a preservation was observed in a group of membrane potential modulators including plant gamma-thionins, scorpion toxins, insect and scorpion defensins, bee venom apamin and MCD peptide, snake sarafotoxins, and human endothelins. These substances are short polypeptides of various lengths and nonhomologous sequences that affect organisms of distant phyla. Despite the structural differences, comparative analysis

The catalytic subunit of human DNA polymerase delta has been overexpressed in insect cells by a recombinant baculovirus. The recombinant protein has a Mr = approximately 125,000 and is recognized by polyclonal antisera against N-terminal and C-terminal peptides of the catalytic subunit of human DNA polymerase delta. The recombinant protein was purified to near homogeneity (approximately 1200-fold) from insect cells by chromatography on DEAE-cellulose, phosphocellulose, heparin-agarose, and single-stranded DNA-cellulose. The purified protein had both DNA polymerase and 3'-5' exonuclease activit

BACKGROUND AND AIMS: The labellar 'hairs' of some Cymbidium spp. are said to be thin-walled and to contain 'plasma', oil and sugars and it has long been speculated that they may function as food-hairs. However, the present authors' preliminary studies showed that certain atypical papillae may have a different role and, by reflecting light, function as a speculum. The purpose of the paper is to test this hypothesis.
METHODS: Light microscopy, scanning electron microscopy, transmission electron microscopy, histochemistry and ultraviolet photography were used to investigate the structure, food c

To obtain an insight into the salivary transcriptome and proteome (sialome) of the adult female mosquito Culex quinquefasciatus, a cDNA library was randomly sequenced, and aminoterminal information for selected proteins and peptides was obtained. cDNA sequence clusters coding for secreted proteins were further analyzed. The transcriptome revealed messages coding for several proteins of known families previously reported in the salivary glands of other blood-feeding insects as well as immune-related products such as C-type lectin, gambicin, and members of the prophenol oxidase cascade. Addition

We report the complete mitochondrial DNA genome of the soybean stem fly (SSF) Melanagromyza sojae from Brazil Santa Catarina state based on Illumina MiSeq sequence data. The estimated mitogenome is 15 475 base pairs (bp) (KT597923), with 13 protein-coding genes (PCGs), two ribosomal RNA (rRNA) genes, and 22 tRNAs, and an estimated 579 bp AT-rich control region. Similar to other insects, the SSF mitogenome is A-T bias with 40.9% A, 36.7% T, 13.6% C, and 8.8% G. Molecular characterization of SSF mitogenome will facilitate the development of effective molecular markers, and robust and rapid ident

BACKGROUND: The endosymbiont Wolbachia pipientis infects a broad range of arthropod and filarial nematode hosts. These diverse associations form an attractive model for understanding host:symbiont coevolution. Wolbachia's ubiquity and ability to dramatically alter host reproductive biology also form the foundation of research strategies aimed at controlling insect pests and vector-borne disease. The Wolbachia strains that infect nematodes are phylogenetically distinct, strictly vertically transmitted, and required by their hosts for growth and reproduction. Insects in contrast form more fluid

Over the past decades, many molecular players have been uncovered to control distinct steps of olfactory circuit assembly in Drosophila. Among these, multi-member gene families encoding cell-surface proteins are of interest as they can act as neuron-specific recognition tags in combinations and contribute to circuit assembly in complex brains. Recently, a multi-protein interactome has been described between Beat and Side families of IgSF proteins. Here, we use newly generated gene trap transgenic driver lines to probe the spatial expression pattern of beat/side genes in olfactory receptor neur
