Bacteria-mediated modification of insecticide toxicity in the yellow fever mosquito, Aedes aegypti.

The incidence of mosquito-borne disease poses a significant threat to human and animal health throughout the world, with effective chemical control interventions limited by widespread insecticide resistance. Recent evidence suggests that gut bacteria of mosquitoes, known to be essential in nutritional homeostasis and pathogen defense, may also play a significant role in facilitating insecticide resistance. This study investigated the extent to which bacteria contribute to the general esterase and cytochrome P450 monooxygenase (P450)-mediated detoxification of the insecticides propoxur and nale…

A novel method using Autographa californica multiple nucleopolyhedrovirus for increasing the sensitivity of insecticide through calcium influx in insect cell line.

Due to an intensive use of chemical insecticides, resistance mechanisms to insecticides together with adverse effects on non-target organisms have been largely reported. Improvement in pest control strategy represents an urgent need to optimize efficiency in the control of pest insects. In this context, a novel method based on the use of insect specific virus applied in combination with chemical insecticide, which could lead to sensitization of the insect target to insecticides is described. Insect virus, the baculovirus Autographa californica multiple nucleopolyhedrovirus (AcMNPV), applied on

Efficacy of mosquito nets treated with a pyrethroid-organophosphorous mixture against Kdr- and Kdr+ malaria vectors (Anopheles gambiae).

In order to prevent the resistance of Anopheles gambiae s.l. to pyrethroids from spreading too quickly and to lengthen the effectiveness of insecticide impregnated mosquito nets, it has recently been suggested to use mixtures of insecticides that have different modes of action. This study presents the results obtained with tulle mosquito nets treated with bifenthrin (a pyrethroid) and chlorpyrifos-methyl (an organophosphorous) both separately and in mixture on two strains of An. gambiae, one sensitive to all insecticides, and the other resistant to pyrethroids. The values of KDt50 and KDt95 an

Insecticidal toxicity of ω-Atypitoxin-Cs1a and its inhibitory effects on insect voltage-gated calcium channels.

BACKGROUND: Excessive use of chemical insecticides raises concerns about insecticide resistance, urging the development of novel insecticides. Peptide neurotoxins from spider venom are an incredibly rich source of ion channel modulators with potent insecticidal activity. A neurotoxin U1-Atypitoxin-Cs1a from the spider Calommata signata was annotated previously. It was of interest to investigate its insecticidal activity and potential molecular targets. RESULTS: Cs1a was heterologously expressed, purified and pharmacologically characterized here. The recombinant neurotoxin inhibited high-volta…

Susceptibility and resistance profiles of field and laboratory strains of Trogoderma granarium Everts to pirimiphos-methyl, alpha-cypermethrin and spinetoram.

The khapra beetle, Trogoderma granarium Everts, 1898, is a significant pest of stored commodities worldwide. Insecticides are heavily relied upon to manage T. granarium. However, the long-term usage of insecticides has led to the development of resistance to insecticides, reducing their effectiveness against T. granarium. This study investigated variations in susceptibility to pirimiphos-methyl, alpha-cypermethrin and spinetoram in a laboratory and seven field strains of T. granarium, using dose-mortality bioassays. Metabolic resistance mechanisms were investigated through synergism studies us

Multidisciplinary Investigations of Sustained Malaria Transmission in the Greater Mekong Subregion.

In the course of malaria elimination in the Greater Mekong Subregion (GMS), malaria epidemiology has experienced drastic spatiotemporal changes with residual transmission concentrated along international borders and the rising predominance of Plasmodium vivax. The emergence of Plasmodium falciparum parasites resistant to artemisinin and partner drugs renders artemisinin-based combination therapies less effective while the potential spread of multidrug-resistant parasites elicits concern. Vector behavioral changes and insecticide resistance have reduced the effectiveness of core vector control …

A preliminary proteomic study of permethrin resistant and susceptible Aedes aegypti (L.).

The mechanism of insecticide resistance is traditionally attributed to detoxification enzymes, target site alteration, decreased penetration of insecticides and behavioural resistance. Other form of mechanisms, such as the role of protein(s) in resistance is unknown. In the present study, the protein profiling of both IMR-PSS strain (permethrin-selected) and IMR-LS strain (laboratory-susceptible) 24 hours post exposure period to permethrin was carried out via 1D-gel electrophoresis and liquid chromatography mass spectrometry (LC-MS/ MS). The bands which appeared in the gel of 1D-electrophoresi…

Multiple Mechanisms Confer Fipronil Resistance in the German Cockroach: Enhanced Detoxification and Rdl Mutation.

Populations of Blattella germanica (L.) (German cockroach) have been documented worldwide to be resistant to a wide variety of insecticides with multiple modes of action. The phenylpyrazole insecticide fipronil has been used extensively to control German cockroach populations, exclusively in baits, yet the highest reported fipronil resistance is 38-fold in a single population. We evaluated five populations of German cockroaches, collected in 2018-2019 in apartments in North Carolina and assayed in 2019, to determine the status of fipronil resistance in the state. Resistance ratios in field-col

Involvement of esterases in increased resistance to pirimiphos-methyl and bifenthrin in Sitophilus oryzae (Linnaeus, 1763).

Sitophilus oryzae (L.) is a cosmopolitan pest of stored grains, frequently controlled using organophosphates and pyrethroids. However, the continuous use of these insecticides may lead to resistance development. Thus, this study evaluated the evolution of S. oryzae resistance after three generations (F3) of artificial selection with pirimiphos-methyl and bifenthrin, and the persistence of acquired resistance after three additional generations without exposure (F6) to insecticides. Two experiments were conducted: the Continuous Exposure Bioassay (CEB), with exposure to sublethal doses, and the

Rising of pyrethroid resistance mutations (kdr) in the dengue vector Aedes aegypti from northeastern Argentina.

Insecticide resistance poses a significant threat to the control of the main dengue vector, Aedes aegypti. Knockdown resistance mutations (kdr) have been associated with pyrethroid resistance and embody the primary source of genetically resistant mosquitoes. This study describes the evolution of two kdr mutations, V1016I and F1534C, in Ae. aegypti from Posadas, Argentina, based on TaqMan genotyping data generated from two sampling times, 2019 and 2024, at the same collection sites. Consistent with global studies, we observed a significant increase in the frequency of these kdr alleles (>14%) a…

Pyrethroid genetic resistance in the dengue vector (Aedes aegypti) in Posadas, Argentina.

Pyrethroids are extensively used to control adult populations of the arboviral vector Aedes aegypti, raising concerns regarding the increasing frequency and distribution of insecticide resistance mutations (kdr: knock-down resistance) in the voltage-gated sodium channel gene (Nav). The widespread use of pyrethroids imposes a threat to the success of mosquito control and the environment. In this study, we investigated the presence of two kdr mutations (V1016I and F1534C) in the Nav gene and their distribution across four neighborhoods in Posadas, Argentina, with different Ae. aegypti abundance …

Mutations in the GSTe2 Promoter in the Emamectin Benzoate-Resistant Strain of Spodoptera frugiperda Contribute to Elevated Expression Levels and Enhanced Metabolic Detoxification Capacity against Emamectin Benzoate.

While emamectin benzoate (EB) remains a key insecticide against Spodoptera frugiperda, significant resistance has developed, compromising its effectiveness. Identifying response genes and mutations is critical for resistance management. Here, genome resequencing and transcriptome analysis of EB-resistant (EB-R) and EB-susceptible (EB-Sus) strains of S. frugiperda identified 177 candidate genes. Subsequently, four glutathione S-transferase (GST) genes, one Cytochrome P450 gene, and one ABC gene linked to insecticide resistance were selected for functional validation. Among these, GSTe2 was high…
