Dystonia 16 (DYT16) mutations in PACT cause dysregulated PKR activation and eIF2α signaling leading to a compromised stress response.

Dystonia 16 (DYT16) is caused by mutations in PACT, the protein activator of interferon-induced double-stranded RNA-activated protein kinase (PKR). PKR regulates the integrated stress response (ISR) via phosphorylation of the translation initiation factor eIF2α. This post-translational modification attenuates general protein synthesis while concomitantly triggering enhanced translation of a few specific transcripts leading either to recovery and homeostasis or cellular apoptosis depending on the intensity and duration of stress signals. PKR plays a regulatory role in determining the cellular r…

Aconserved salt-stress co-expression network in japonica rice reveals partially dissociated hub gene connectivity and transcriptional induction.

INTRODUCTION: Soil salinity severely compromises rice (Oryza sativa L.) growth and grain yield by disrupting ionic and osmotic homeostasis. Although transcriptome profiling has advanced understanding of salt-stress responses in rice, the conservation of co-expression networks across genetic backgrounds and the relationship between network connectivity and transcriptional induction remain poorly characterized. METHODS: We performed integrated RNA sequencing and weighted gene co-expression network analysis (WGCNA) on three japonica rice varieties under control (0 mM), moderate (85.6 mM), and se

Diabetes-associated MYT1 and ST18 genes regulate human beta cell insulin secretion and survival via other diabetes risk genes.

AIMS/HYPOTHESIS: Genetic and environmental factors work together to cause islet beta cell failure, leading to type 2 diabetes. How these factors are integrated to regulate beta cells remains largely unclear. Based on our previous findings that the family of myelin transcription factors (MYT TFs; including MYT1, MYT1L and ST18) prevents mouse beta cell failure by repressing the overactivation of stress response, their regulation by obesity-related nutrition signals in human beta cells and their association with type 2 diabetes, we postulate that these factors prevent human beta cell failure und

The protein kinase Ire1 impacts pathogenicity of Candida albicans by regulating homeostatic adaptation to endoplasmic reticulum stress.

The unfolded protein response (UPR), crucial for the maintenance of endoplasmic reticulum (ER) homeostasis, is tied to the regulation of multiple cellular processes in pathogenic fungi. Here, we show that Candida albicans relies on an ER-resident protein, inositol-requiring enzyme 1 (Ire1) for sensing ER stress and activating the UPR. Compromised Ire1 function impacts cellular processes that are dependent on functional secretory homeostasis, as inferred from transcriptional profiling. Concordantly, an Ire1-mutant strain exhibits pleiotropic roles in ER stress response, antifungal tolerance, ce

An oxidative stress - and immunotherapy-related six-gene signature defines immune subtypes and predicts prognosis and immunotherapy response in hepatocellular carcinoma.

BACKGROUND: Oxidative stress and the tumor immune microenvironment jointly shape hepatocellular carcinoma (HCC) progression and response to immunotherapy, yet integrated biomarkers linking these processes are lacking. METHODS: Transcriptomic and clinical data from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) datasets were used to identify oxidative stress- and immunotherapyrelated differentially expressed genes (OSIRDEGs). Functional enrichment, weighted gene co-expression network analysis (WGCNA) and LASSO-Cox regression were used to construct a prognostic signature. Cons

Neuronal translational control in physiology, infection, and inflammation.

Neurons are highly specialized cells whose function depends on dynamic regulation of gene expression. With increasing subcellular compartmentalization, neuronal gene expression becomes less dependent on transcription in the perinuclear region and tends to be increasingly controlled by local translation. The integrated stress response is a central, evolutionarily conserved pathway of translational regulation that maintains neuronal homeostasis through precise control of protein synthesis. Transient cellular stress, such as neuronal infection, adaptively activates this pathway, whereas persisten…

Removing dysfunctional mitochondria from axons independent of mitophagy under pathophysiological conditions.

Chronic mitochondrial dysfunction has been implicated in major neurodegenerative diseases. Long-term cumulative pathological stress leads to axonal accumulation of damaged mitochondria. Therefore, the early removal of defective mitochondria from axons constitutes a critical step of mitochondrial quality control. We recently investigated the axonal mitochondrial response to mild stress in wild-type neurons and chronic mitochondrial defects in amyotrophic lateral sclerosis (ALS)- and Alzheimer disease (AD)-linked neurons. We demonstrated that remobilizing stressed mitochondria is critical for ma

Redox-sensitive m6A epitranscriptomic remodeling in cadmium stress and toxicity.

Environmental stressors that disrupt redox homeostasis pose a significant threat to metabolic balance, tissue integrity, and organismal development. Increasing evidence identifies N6-methyladenosine (m6A), a dynamic and stress-responsive RNA modification, as a central regulator that translates oxidative cues into functional changes in RNA metabolism and cellular behavior. Redox imbalance can recalibrate the activity, localization, and substrate selectivity of m6A writers, erasers, and readers, thereby reshaping transcriptomic programs that control inflammation, antioxidant defense, proteostasi

Reduced White Matter Damage and Lower Neuroinflammatory Potential of Microglia and Macrophages in Hri/Eif2ak1 -/- Mice After Contusive Spinal Cord Injury.

Cellular stressors inhibit general protein synthesis while upregulating stress response transcripts and/or proteins. Phosphorylation of the translation factor eIF2α by one of the several stress-activated kinases is a trigger for such signaling, known as the integrated stress response (ISR). The ISR regulates cell survival and function under stress. Here, germline knockout mice were used to determine contributions by three major ISR kinases, HRI/EIF2AK1, GCN2/EIF2AK4, and PKR//EIF2AK2, to pathogenesis of moderate contusive spinal cord injury (SCI) at the thoracic T9 level. One-day post-injury (…

Teratogenic effects induced by paracetamol, ciprofloxacin, and their mixture on Danio rerio embryos: Oxidative stress implications.

Even though the toxic effects of paracetamol (PCM) and ciprofloxacin (CPX) have been deeply studied in the last decades, the impact of the PCM-CPX mixture may induce in aquatic organisms is poorly known. Thus, the objective of this work was to investigate the teratogenic effects and oxidative stress that PCM, CPX, and their mixture induce in Danio rerio embryos. Moreover, we aimed to determine whether the PCM-CPX mixture induces more severe effects on the embryos than the individual drugs. For this purpose, zebrafish embryos (4 hpf) were exposed to environmentally relevant concentrations of PC

Nitric oxide rectifies acid-base disturbance and modifies thyroid hormone activity during net confinement of air-breathing fish (Anabas testudineus Bloch).

Nitric oxide (NO), a short-lived freely diffusible radical gas that acts as an important biological signal, regulates an impressive spectrum of physiological functions in vertebrates including fishes. The action of NO, however, on thyroid hormone status and its role in the integration of acid-base, osmotic and metabolic balances during stress are not yet delineated in fish. Sodium nitroprusside (SNP), a NO donor, was employed in the present study to investigate the role of NO in the stressed air-breathing fish Anabas testudineus. Short-term SNP treatment (1 mM; 30 min) interacted negatively wi

Dietary Modulation of Inflammatory and Oxidative Pathways in Type 2 Diabetes: Biomarkers and Cardiorenal Outcomes.

Type 2 diabetes mellitus is a relevant cardio-renal-metabolic disorder in which chronic low-grade inflammation and oxidative stress have a crucial function in linking insulin resistance, endothelial dysfunction, β-cell impairment, and progressive organ injury. In this context, nutrition has emerged as a key modifiable determinant of metabolic homeostasis, capable of influencing inflammatory signalling, redox balance, mitochondrial function, and gut microbiota-host interactions. The objective of this review is to critically summarise the mechanistic connections among inflammation, oxidative str

Temporal Transcriptomic and Metabolomic Reprogramming Unveils a Two-Phase Salt Tolerance Mechanism in Apocynum venetum.

Soil salinization poses a major constraint to global agriculture. Apocynum venetum, a salt-tolerant halophyte, provides an effective model for investigating salt-adaptive strategies; however, the temporal dynamics of its tolerance-associated genes and metabolites remain unclear. In this study, integrated transcriptomics, metabolomics (UHPLC-MS), physiological assays, and weighted gene co-expression network analysis (WGCNA) were conducted to characterize early (7-day) and late (18-day) responses to 200 mM NaCl stress. NaCl stress significantly reduced chlorophyll content while increasing Na+ ac

Breed-specific physiological, biochemical, and tissue transcript responses of Harri and Naemi lambs to acute heat stress.

Climate change is an increasing threat to livestock production in arid regions. Understanding how indigenous breeds deploy immediate acute physiological and biochemical adjustments to acute hyperthermia at systemic and transcript-level indices is necessary to isolate initial homeostatic limits. This research compared the short-term thermoregulatory, metabolic, and multi-tissue transcriptomic adjustments of Naemi and Harri lambs exposed to acute heat stress. Forty male lambs (Naemi, Harri) were subjected to a 180-min exposure to either thermoneutral (25.0 °C Ta, 38.0% RH) or severe acute heat-s
