Impact of the DSP-H1684R Genetic Variant on Ion Channels Activity in iPSC-Derived Cardiomyocytes.

BACKGROUND/AIMS: Mutations of desmosomal genes are known to cause arrhythmogenic cardiomyopathy characterized by arrhythmias and sudden cardiac death. Previously, we described a novel genetic variant H1684R in desmoplakin gene (DSP), associated with a progressive cardiac conduction disease (PCCD). In the present study, we aimed to investigate an effect of the DSP-H1684R genetic variant on the activity of ion channels. METHODS: We used cardiomyocytes derived from induced pluripotent stem cells (iPSC cardiomyocytes) from a patient with DSP-H1684R genetic variant and from two healthy donors. Imm…

MicroRNA-1976 regulates degeneration of the sinoatrial node by targeting Cav1.2 and Cav1.3 ion channels.

Sick sinus syndrome (SSS) is primarily a disease of the elderly, and age-dependent decrease in Cav1.2 and Cav1.3 Ca2+ channels within the sinus node has been shown to play an important role in sinoatrial node (SAN) degeneration; however, posttranscriptional mechanisms regulating decrease in Cav1.2 and Cav1.3 Ca2+ channels remain unclear. Some studies have reported that microRNAs (miRNAs) are involved in age-related cardiovascular diseases. Nevertheless, little is known about the roles of miRNAs in age-related SSS. This study investigated whether miR-1976 was involved in the regulation of SAN d

Role of ion channels in the functional response of conjunctival goblet cells to dry eye.

Optimal vision requires an ocular surface with a stable tear film whose many critical tasks include providing >70% of the eye's refractive power. However, for millions, tear film instability produces uncomfortable sight-impairing dry eye. Despite the multitude of etiologies for dry eye, a universal hallmark is hyperosmolarity of the tear film. Presently, knowledge of how the ocular surface responds to hyperosmolarity remains incomplete with little understood about the role of ion channels. This bioelectric analysis focused on conjunctival goblet cells whose release of tear-stabilizing mucin is…

Blocking mechanosensitive ion channels eliminates the effects of applied mechanical loading on chick joint morphogenesis.

Abnormalities in joint shape are increasingly considered a critical risk factor for developing osteoarthritis in life. It has been shown that mechanical forces during prenatal development, particularly those due to fetal movements, play a fundamental role in joint morphogenesis. However, how mechanical stimuli are sensed or transduced in developing joint tissues is unclear. Stretch-activated and voltage-gated calcium ion channels have been shown to be involved in the mechanoregulation of chondrocytes in vitro In this study, we analyse, for the first time, how blocking these ion channels influe…

Gating modifier toxin interactions with ion channels and lipid bilayers: Is the trimolecular complex real?

Spider peptide toxins have attracted attention because of their ability to target voltage-gated ion channels, which are involved in several pathologies including chronic pain and some cardiovascular conditions. A class of these peptides acts by modulating the gating mechanism of voltage-gated ion channels and are thus called gating modifier toxins (GMTs). In addition to their interactions with voltage-gated ion channels, some GMTs have affinity for lipid bilayers. This review discusses the potential importance of the cell membrane on the mode of action of GMTs. We propose that peptide-membrane…

Functional and structural properties of ion channels at the nerve terminal depends on compact myelin.

KEY POINTS: In the present study, we document the role of compact myelin in regulating the structural and functional properties of ion channels at the nerve terminals, using electrophysiology, dynamic Na(+) imaging and immunohistochemistry. The subcellular segregation of Na(+) channel expression and intracellular Na(+) dynamics at the heminode and terminal was lost in the dysmyelinated axon from Long-Evans shaker rats, which lack compact myelin. In Long-Evans shaker rats, loss of the Nav β4 subunit specifically at the heminode reduced resurgent and persistent Na(+) currents, whereas K(+) chann…

Expression of connexin 43, ion channels and Ca2+-handling proteins in rat pulmonary vein cardiomyocytes.

Atrial fibrillation (AF) is the most common cardiac arrhythmia. AF is thought to be triggered by ectopic beats, originating primarily in the … The results suggest that in the absence of arrhythmia, the investigated molecular properties of the ion channels of rat PV cardiomyocytes resemble those of the working myocardium. This indicates that ectopic beats originating in the myocardial sleeves of the PVs occur only under pathological conditions.

Inhibition of acid-sensing ion channels by levo-tetrahydropalmatine in rat dorsal root ganglion neurons.

Levo-tetrahydropalmatine (l-THP), a main bioactive Chinese herbal constituent from the genera Stephania and Corydalis, has been in use in clinical practice for years in China as a traditional analgesic agent. However, the mechanism underlying the analgesic action of l-THP is poorly understood. This study shows that l-THP can exert an inhibitory effect on the functional activity of native acid-sensing ion channels (ASICs), which are believed to mediate pain caused by extracellular acidification. l-THP dose dependently decreased the amplitude of proton-gated currents mediated by ASICs in rat dor…

Expression and response of acid-sensing ion channels in urinary bladder to cyclophosphamide-induced cystitis.

The expression of acid-sensing ion channel (ASIC) isoforms, ASIC1, ASIC2a, and ASIC3, was examined in the urinary bladder after cyclophosphamide (CYP)-induced cystitis of varying duration (4 h, 48 h, and chronic). Immunohistochemical, Western blot, and quantitative PCR approaches were used to evaluate channel expression and effects of CYP-induced cystitis in whole urinary bladder and split-bladder preparations from control (no inflammation) and CYP-treated rats. Quantitative PCR demonstrated significant (P ≤ 0.01) increases in ASIC2a and ASIC3 transcripts with CYP-induced cystitis (48 h and ch

Biochemical characterization of cysteine-rich peptides from Oxyopes sp. venom that block calcium ion channels.

Oxytoxins (OxyTx1 and OxyTx2) are disulfide-rich peptides isolated from the venom of the spider Oxyopes lineatus that block voltage-sensitive calcium ion channels (VSCCs). OxyTx1 was identified previously and isolated from the related spider Oxyopes kitabensis; however, its pharmacology was unknown. OxyTx1 and OxyTx2 contain 69 and 55 amino acid residues with molecular masses of 8058.2 and 6175.2Da, respectively. Oxytoxins contain five disulfide bridges, are amidated at their C-terminus, antagonize P/Q-, N- or L-type VSCCs, and have low amino acid identity to known VSCC blockers from arthropod…

Characterization of excitability and voltage-gated ion channels of neural progenitor cells in rat hippocampus.

We have observed the excitability of rat neural progenitor cells (NPCs) and characterized the profile of the voltage-gated ion channels, with the help of the electrophysiological measurements in NPCs in vitro, in order to clarify the electrophysiological property of NPCs. The membrane potential changes of neural progenitors were detected with fluorescent dye bis-(1,3-dibutylbarbituric acid) trimethine oxonol (DiBAC4(3)) by a confocal laser scanning microscope. The changes of fluorescent intensity of DiBAC4(3) stain after KCl stimulation were slight which indicated that the NPCs were inexcitabl…

Use of Xenopus oocytes to measure ionic selectivity of pore-forming peptides and ion channels.

The Xenopus laevis oocyte is a widely used system for heterologous expression of exogenous ion channel proteins (1, 2). Among other advantages, these easy to obtain, mechanically and electrically stable, large-sized cells enable multiple types of electrophysiological recordings: two-electrode voltage-clamp, single-cell attached or cell-free patch-clamp, and macropatch recordings. The size of an oocyte (1 mm in diameter) also allows the use of additional electrodes (1-3) for injection of diverse materials (Ca2+ chelators, peptides, chemicals, antibodies, proteic-partners, and so on) before or d

A hot spot for the interaction of gating modifier toxins with voltage-dependent ion channels.

The gating modifier toxins are a large family of protein toxins that modify either activation or inactivation of voltage-gated ion channels. omega-Aga-IVA is a gating modifier toxin from spider venom that inhibits voltage-gated Ca(2+) channels by shifting activation to more depolarized voltages. We identified two Glu residues near the COOH-terminal edge of S3 in the alpha(1A) Ca(2+) channel (one in repeat I and the other in repeat IV) that align with Glu residues previously implicated in forming the binding sites for gating modifier toxins on K(+) and Na(+) channels. We found that mutation of …

The relationship between new ion channels and ectopic discharges from a region of nerve injury.

Changes in ectopic discharges from axons in an injured nerve were examined while agents that interact with ion channels were applied to the site of the nerve injury. Tetraethylammonium (TEA) greatly facilitated spontaneous ectopic discharges or evoked ectopic firing in previously silent axons. Tetrodotoxin, an Na+ channel blocker, completely blocked spontaneous discharges. Verapamil, La3+, and Mn2+, agents that interact with Ca2+ channels, blocked spontaneous discharges and depressed the responses evoked by TEA, noradrenaline and high concentration of K+. Increasing Ca2+ levels facilitated ect…

Mechanisms and therapeutic potential of ultrasound neuromodulation: Focus on ion channel dynamics.

Ultrasound neuromodulation has emerged as a promising noninvasive technique for regulating neuronal activity with high spatial and temporal precision. Among its mechanisms, the modulation of mechanosensitive (MS) ion channels plays a key role in converting acoustic energy into cellular responses. This review highlights the biophysical mechanisms by which ultrasound activates specific ion channels (e.g., Piezo, TRP, K2P, MscL) and discusses their therapeutic implications in neurological and psychiatric disorders, including Parkinson's disease and depression. Advances in sonogenetics, nanopartic…

Ion channel dysfunction in acute pancreatitis: mechanisms, heterogeneity, and therapeutic prospects.

Acute pancreatitis (AP) is a life-threatening gastrointestinal disorder with rising global incidence and persistently high mortality, currently lacking disease-modifying therapies and resulting in substantial unmet clinical needs. Ion channel and transporter dysfunction is proposed as a convergent mechanistic link within a subset of etiologies, driving conserved pathological cascades such as intracellular calcium overload, mitochondrial dysfunction, secretory insufficiency, and systemic inflammation. Building upon prior studies of individual ion channels, this review systematically integrates …

Ion channels and cardiac disease: mechanisms and functions of a disintegrin and metalloproteases and their substrates.

Normal physiological activity of the heart depends on the coordinated regulation of various ion channels in cardiomyocytes (CM), and dysfunction of these channels serves as a key pathological basis for cardiac diseases, including arrhythmias and heart failure Mitochondria play a central role in maintaining the cardiac redox balance and calcium (Ca2+) homeostasis, and cardiac mitochondrial dysfunction constitutes a core mechanism underlying ion channel dysregulation and heart disease pathogenesis. A disintegrin and metalloproteinases (ADAMs) are a family of transmembrane proteases that play cri…

Ion channel TRPML2 (MCOLN2) promotes ovarian cancer metastasis and chemoresistance.

BACKGROUND: Ovarian cancer, with metastasis and chemoresistance being the major obstacles to successful treatment, is a leading cause of death among gynecological malignancies. Identifying new molecular targets and understanding the underlying mechanisms of metastasis and chemoresistance are pressing issues in ovarian cancer therapy. Emerging evidence suggests that an endolysosomal Ca2+-permeable channel TRPML2 may contribute to cancer progression. However, its exact role and associated underlying mechanisms in ovarian cancer remain largely unknown. METHODS: TRPML2 expression was studied by t
