GABA(A) positive modulator and NMDA antagonist-like discriminative stimulus effects of isoflurane vapor in mice.

RATIONALE: Several neurotransmitter systems have been hypothesized to be involved in the in vivo effects of volatile anesthetics. Drug discrimination may represent a novel procedure to explore the neurochemical systems underlying the sub-anesthetic behavioral effects of these compounds. OBJECTIVES: The purpose of the present study was to examine the contribution of GABA(A) and NMDA receptors to the discriminative stimulus effects of a behaviorally active sub-anesthetic concentration of isoflurane vapor. METHODS: Sixteen B6SJLF1/J mice were trained to discriminate 10 min of exposure to 6,000 …

Effects of isoflurane on glutamate and taurine releases, brain swelling and injury during transient ischemia and reperfusion.

The volatile anesthetic agent isoflurane was thought to provide neuroprotection against ischemic damage; however, this effect remains controversial. Using the middle cerebral artery occlusion model and intracerebral microdialysis, the authors monitored the variations of glutamate and taurine concentrations in the extra-cellular space in male rats anesthetized with pentobarbital or isoflurane. Brain injury and edema were evaluated 24 h after ischemia. Isoflurane prevented the ischemia-induced efflux of glutamate and reduced the release of taurine. No difference in the size of the brain lesions …

Combination of isoflurane and caspase inhibition reduces cerebral injury in rats subjected to focal cerebral ischemia.

BACKGROUND: Recent data indicate that the neuroprotective efficacy of isoflurane is not sustained. Delayed neuronal death, mediated in part by apoptosis, contributes to the gradual increase in the size of the infarction. These data suggest that isoflurane may not be able to inhibit delayed neuronal death. The prevention of apoptosis by a caspase inhibitor might provide neuroprotection in addition to that provided by isoflurane. The current study was conducted to determine whether isoflurane-mediated neuroprotection can be made more durable with the administration of z-VAD-fmk, a nonspecific ca…

Isoflurane, but not halothane, improves indices of diastolic performance in dogs with rapid ventricular, pacing-induced cardiomyopathy.

BACKGROUND: The left ventricular (LV) mechanical effects of isoflurane and halothane were examined in dogs with rapid LV pacing-induced cardiomyopathy. These experiments tested the hypothesis that isoflurane and halothane differentially enhance indices of diastolic performance in dogs with moderate LV dysfunction. METHODS: Eight dogs were chronically instrumented for measurement of LV and aortic pressures, subendocardial segment length, and cardiac output. Contractility was quantified by preload recruitable stroke work (Mw). Diastolic function was evaluated with a time constant of isovolumic …

Isoflurane-chlorodifluoroethene interaction in human liver microsomes. Role of cytochrome P4502B6 in potentiation of haloethene metabolism.

Short-chain saturated halocarbons, including isoflurane and the chlorofluorocarbon substitute HCFC-123, can strongly potentiate the cytochrome P450-dependent oxidation of gaseous haloethenes, such as 2-chloro-1,1-difluoroethene (CDE) and vinyl chloride, in vivo and in vitro. P450 isozyme specificity in this effect is suggested by the fact that the interaction is pronounced in microsomes from rats treated with phenobarbital, but does not occur in microsomes of isoniazid- or beta-naphthoflavone-treated animals. We examined the effect of isoflurane on CDE defluorination in liver microsomes from 1…

Effects of subanesthetic concentrations of isoflurane and their interactions with epinephrine on acquisition and retention of the rabbit nictitating membrane response.

BACKGROUND: Evidence concerning the concentrations of volatile anesthetics that prevent learning and recall is limited. Epinephrine is believed to enable learning during anesthesia. We investigated the effects of isoflurane and its interaction with epinephrine on learning and subsequent retention of the rabbit's classically conditioned nictitating membrane response. METHODS: In experiment 1, a tone (conditioned stimulus, CS) preceded paraorbital shock (unconditioned stimulus, US) during 60-min daily sessions of 60 presentations of these paired stimuli for 6 days of acquisition training under …

Differences in cardiodepressant effects of halothane and isoflurane after inotropic stimulation in guinea-pig papillary muscles.

The effects of halothane and isoflurane on action potentials and force of contraction were studied in guinea-pig isolated papillary muscles in order to investigate the cardiodepressant action of the anesthetics in the presence of clinically relevant inotropic drugs. In control conditions, equipotent concentrations of the two volatile anesthetics depressed force of contraction to a similar degree. Halothane (2%) slightly shortened the action potential duration, whereas isoflurane (2.8%) did not influence the shape of the action potential. The muscles were then treated with three different inotr…

The effects of convective cooling and rewarming on systemic and central nervous system physiology in isoflurane-anesthetized dogs.

Recent studies have demonstrated that small (i.e., 2-5 degrees C) reductions in temperature may protect the brain and spinal cord from ischemic injury. The present study evaluated the physiologic response of anesthetized animals to convective-based cooling and warming. Six shaved, isoflurane-anesthetized (1.50% end-expired; 1 MAC), pancuronium-paralyzed dogs were subjected to temperature manipulation. The flow of cool (13-14 degrees C) or warm (39-41 degrees C) air was uniformly applied to the the dorsal and lateral surfaces of the dog using an inflatable blanket with perforations in the inter…

Sex-specific Effects of Acute Stress on Emergence from Isoflurane Anesthesia via Brainstem Circuitry in Mice.

BACKGROUND: Preoperative stress is a well documented phenomenon in surgical patients and leads to clinically significant variability in anesthetic requirements. This variability poses important challenges for safe anesthesia practice, as stress may alter the function of neural circuits targeted by general anesthetics. However, the specific mechanisms linking stress to changes in anesthetic efficacy remain unclear. This study aimed to investigate how acute stress affects the efficacy of isoflurane anesthesia and to uncover the associated neural circuitry and receptor mechanisms. METHODS: Using…
