Leafroller-induced phenylacetonitrile and acetic acid attract adult Lobesia botrana in European vineyards.

We recently identified unique caterpillar-induced plant volatile compounds emitted from apple leaves infested with the larvae of various leafroller species. In subsequent field tests, binary blends of phenylacetonitrile+acetic acid and 2-phenylethanol+acetic acid were found to be attractive to a range of tortricid leafroller species (Tortricidae: Tortricinae) in both the Southern and Northern Hemispheres. In this work, the caterpillar-induced plant volatiles from the apple-leafroller system were tested in two vineyards in Spain and Hungary for their attractiveness to the grape frugivore Lobesi

Diet influences the bacterial and free fatty acid profiles of the cuticle of Galleria mellonella larvae.

The evolutionary success of insects is arguably due to their ability to build up a complex, highly-adaptable and very effective defense system against numerous pathogens, including entomopathogenic fungi. This system relies on the humoral immune system and cellular defense reactions. The first line of defense against biological pathogens is a cuticle formed of several layers. The cuticular lipids may contain hydrocarbons, free fatty acids (FFA), alcohols, waxes, glycerides, aldehydes and sterols. Cuticular fatty acids may also play a role in defending against fungal invasion. Our present findi

Use of Synthetic Hybrid Strains To Determine the Role of Replicon 3 in Virulence of the Burkholderia cepacia Complex.

The Burkholderia cepacia complex (Bcc) displays a wealth of metabolic diversity with great biotechnological potential, but the utilization of these bacteria is limited by their opportunistic pathogenicity to humans. The third replicon of the Bcc, megaplasmid pC3 (0.5 to 1.4 Mb, previously chromosome 3), is important for various phenotypes, including virulence, antifungal, and proteolytic activities and the utilization of certain substrates. Approximately half of plasmid pC3 is well conserved throughout sequenced Bcc members, while the other half is not. To better locate the regions responsible

Spatial responses of two herbivore groups to a geometrid larva on mountain birch.

Direct or plant-mediated interactions between herbivores may modify their spatial distribution among and within plants. In this study, we examined the effect of a leaf-chewing geometrid, the autumnal moth (Epirrita autumnata), on two different herbivore groups, leaf rolling Deporaus betulae weevils and Eriocrania spp. leafminers, both feeding on mountain birch (Betula pubescens ssp. czerepanovii). The exact locations of herbivores within tree canopies were mapped during three successive summers. In the first 2 years, some trees were artificially colonized by eggs of the autumnal moth to induce

Oral toxicity to flesh flies of a neurotoxic polypeptide.

An insect selective neurotoxic polypeptide from venom of the scorpion Androctonus australis (AaIT, M(r) 8,000) was shown to cross the midgut of the flesh fly Sarcophaga falculata, using assays of oral toxicity, column chromatography, and microscopic autoradiography of the native and radioiodinated toxin. AaIT induced paralysis of flies within 1-2 h after oral administration, with a lethal dose (LD50) of 10 micrograms/100 mg of body weight. Oral toxicity was about 0.14% of toxicity by injection. Hemolymph collection 70-85 min after feeding flies with [125I]AaIT showed that 5% of ingested radioa

Role of virus-like particles in parasitoid-host interaction of insects.

Insect endoparasitoids are capable of suppressing the immune reaction of their habitual hosts in a specific way. Salt (1968) characterized s … le proposal would be that such hypothetical viruses, which were able to suppress immune reaction in lepidopteran hosts, were incorporated into a parasitoid wasp to become part of the life cycle of the parasitoid.(ABSTRACT TRUNCATED AT 400 WORDS)

Nocturnal arthropod occurrences across Natura 2000 protected areas of Madeira Island (Portugal).

BACKGROUND: Artificial light at night (ALAN) is increasingly recognised as an important anthropogenic pressure affecting nocturnal biodiversity, particularly in oceanic islands with high endemism levels. Arthropods, especially moths (Lepidoptera), are key components of nocturnal ecosystems, yet their distribution and abundance patterns in Madeira Island remain insufficiently documented and their responses to ALAN are poorly understood. This study was implemented under the LIFE Natura@night project - Mitigating light pollution in Macaronesia (LIFE20 NAT/PT/001098; https://www.naturaatnight.spea…

Evaluation of the insecticidal activity of the spider-venom peptides Dc1a and Ta1a against economically important lepidopteran pests.

Lepidopterans are the most economically significant crop pests. They are mainly controlled with chemical insecticides that often suffer from insect resistance and adverse effects on the environment and human health. Insect-specific spider-venom peptides are considered safer, eco-friendly alternatives to chemical insecticides, and two insecticidal spider-venom derived peptides have already been commercialised by Vestaron Corporation, while more candidates are being progressed in global discovery pipelines. This study focusses on two insecticidal spider-venom peptides-Dc1a and Ta1a- and assesses…

Direct costs of blood drawings with pre-analytical errors in tertiary paediatric hospital care.

BACKGROUND: Blood drawings is a common hospital procedure involving laboratory and clinical disciplines that is important for the diagnosis and management of illnesses in children. Blood drawings with pre-analytical error (PAE) can lead to increased costs for hospitals and healthcare organisations. The direct cost of blood drawings after a PAE is not fully understood in paediatric hospital care. AIM: The aim of this study was to estimate the average direct cost of PAE per year and per 10,000 blood drawings in tertiary paediatric care. METHODS: A cost analysis using a bottom-up approach was c

In vitro antitrypanosomal activity of the cyclodepsipeptides, cardinalisamides A-C, from the insect pathogenic fungus Cordyceps cardinalis NBRC 103832.

During the search for new antitrypanosomal drug leads, three new antitrypanosomal compounds, cardinalisamides A-C (1-3), were isolated from cultures of the insect pathogenic fungus Cordyceps cardinalis NBRC 103832. Their structures were elucidated using MS analyses and extensive 2D-heteronuclear NMR. The absolute configurations of 1-3 were addressed by chemical degradation and Marfey's analysis. 1-3 showed in vitro antitrypanosomal activity against Trypanosoma brucei brucei with IC50 values of 8.56, 8.65 and 8.63 μg ml(-1), respectively.

Intoxication of cattle on kikuyu grass following army worm (Spodoptera exempta) invasion.

Clinical features and pathological and mycological findings in a field outbreak of intoxication in dairy cattle grazing kikuyu grass are reported. The outbreak followed invasion of the grass by the army worm (Spodoptera exempta). Clinical signs included drooling of saliva, depression, apparent inco-ordination, sunken eyes, ruminal distension and atony, recumbency, moderate diarrhoea and "sham drinking". Seventy-seven cows (64%) were clinically affected over a period of 12 days. Of these, 37 died. Necropsies performed on 4 affected cattle revealed necrosis of the epithelium of the forestomach,

The rectal sac as a second liver for detoxification in the metamorphosing silkworm.

The insect larval fat body is the primary organ for detoxification of environmental pesticides and metabolic waste. During lepidopteran metamorphosis, the larval fat bodies undergo degradation and reprogramming for development, leaving the process of detoxification largely unknown at this stage. Using integrative approaches in the silkworm, Bombyx mori, we demonstrate that the pupal rectal sac, a greatly enlarged hindgut structure, serves as a central detoxification hub during the non-feeding metamorphic period. We show that it actively sequesters metabolic waste (uric acid) and, remarkably, c…

Hypoxia-induced immortalization of Helicoverpa armigera pupal ovarian cells.

Establishing continuous cell lines from lepidopteran insects is notoriously inefficient, with success rates typically below 5% and often below 1%, severely limiting research and biopesticide development for pests such as Helicoverpa armigera (Lepidoptera: Noctuidae). We present a simple, reproducible method, hypoxia-induced immortalization that dramatically improves this efficiency. By applying a brief hypoxic stimulus (5% O2, 7 days) during primary culture of pupal ovary cells, we achieved an overall success rate of 17.8% across four independent batches, more than 3-fold higher than conventio…

The regulatory role of m6A RNA methylation in insect sexual development: a review.

N6-methyladenosine (m6A) is the most prevalent internal mRNA modification in eukaryotes and serves as a key post-transcriptional regulator i … epitranscriptomics. In insects lacking an Sxl-centric pathway, such as Coleoptera, Hymenoptera, and Lepidoptera, m6A depletion does not disrupt core sex differentiation but instead plays essential roles in related processes, including metamorphosis in Tribolium, caste differentiation in honey bees, and gametogenesis across multiple orders. These findings under …

Taxonomic Reevaluation of Blackwellomyces Isolates Preserved in Korean Agricultural Culture Collection and Description of Two New Species.

Blackwellomyces is a genus of entomopathogenic fungi in the family Cordycipitaceae (Hypocreales, Sordariomycetes, Ascomycota) that primarily infects arthropod larvae, particularly those of Lepidoptera. Despite its ecological significance, this genus remains poorly studied in Korea. During the re-identification of Korean Cordyceps sensu lato isolates preserved in the Korean Agricultural Culture Collection (KACC), 12 strains were found to belong to the genus Blackwellomyces. Accordingly, this study aimed to accurately identify and clarify the taxonomic status of these isolates. The 12 Blackwello…

Cyanide detoxification in an insect herbivore: Molecular identification of β-cyanoalanine synthases from Pieris rapae.

Cyanogenic compounds occur widely in the plant kingdom. Therefore, many herbivores are adapted to the presence of these compounds in their diet by either avoiding cyanide release or by efficient cyanide detoxification mechanisms. The mechanisms of adaptation are not fully understood. Larvae of Pieris rapae (Lepidoptera: Pieridae) are specialist herbivores on glucosinolate-containing plants. They are exposed to cyanide during metabolism of phenylacetonitrile, a product of benzylglucosinolate breakdown catalyzed by plant myrosinases and larval nitrile-specifier protein (NSP) in the gut. Cyanide …

A gene horizontally transferred from bacteria protects arthropods from host plant cyanide poisoning.

Cyanogenic glucosides are among the most widespread defense chemicals of plants. Upon plant tissue disruption, these glucosides are hydrolyzed to a reactive hydroxynitrile that releases toxic hydrogen cyanide (HCN). Yet many mite and lepidopteran species can thrive on plants defended by cyanogenic glucosides. The nature of the enzyme known to detoxify HCN to β-cyanoalanine in arthropods has remained enigmatic. Here we identify this enzyme by transcriptome analysis and functional expression. Phylogenetic analysis showed that the gene is a member of the cysteine synthase family horizontally tran…
