Understanding the interaction between brain lipid membranes and therapeutic molecules is essential for unravelling the action mechanism of a drug. This study investigates the influence of amitriptyline hydrochloride (AMT), a tricyclic antidepressant, on lipid monolayers and multilamellar vesicles composed of brain sphingomyelin (BSM) and cholesterol (Chol). Using Langmuir monolayer and small-angle X-ray scattering (SAXS) techniques, it reveals that AMT alters the rigidity, packing density, surface potential, and structural parameters of a phospholipid membrane. The presence of cholesterol in t

The interactions with and effects of five chemically distinct, bioactive phenolic compounds on the lipid bilayers of model dipalmitoylphosphatidylcholine (DPPC) liposomes were investigated. Complementary analytical techniques, including differential scanning calorimetry (DSC) and phosphorus and proton nuclear magnetic resonance spectroscopy (NMR), were employed in order to determine the location of the compounds within the bilayer and to correlate location with their effects on bilayer characteristics and liposomal stability. As compared to the phenolic compounds localized in the glycerol regi…

Cholesterol plays an essential role in biological membranes and is crucial for maintaining their stability and functionality. In addition to biological membranes, cholesterol is also used in various synthetic lipid-based structures such as liposomes, proteoliposomes, and nanodiscs. Cholesterol regulates membrane properties by influencing the density of lipids, phase separation into liquid-ordered (Lo) and liquid-disordered (Ld) areas, and stability of protein-membrane interactions. For planar bilayers, cholesterol thickens the membrane, decreases permeability, and brings lipids into well-order

BACKGROUND: Liposomes are useful tools for amplifying signals in biosensing techniques, which are used to detect and analyze biological molecules. The goal of this study is to improve the sensitivity of immunosorbent assays by optimizing the self-assembled structure of lipids in temperature-responsive liposomes that carry lipophilic fluorescent dyes as a detection probe in their bilayer membrane. We hypothesized that increasing the number of bilayer membranes (lamellarity) in liposomes would enhance the fluorescence signal and improve assay sensitivity, as liposomes with larger lamellarity wou

Antimicrobial peptides (AMPs) are promising complements to antibiotics, yet their membrane-level actions remain incompletely understood. In this work, we characterized how the "de novo" cationic AMP P8.1 interacts with anionic lipid bilayers composed of DPPG (gel phase) or DLPG (fluid phase) using zeta potential, tryptophan and Laurdan fluorescence, Raman microscopy, and a carboxyfluorescein (CF) leakage assay. P8.1 bound both lipids electrostatically, reduced zeta potential, and increased large unilamellar vesicles (LUVs) size. Binding kinetics were faster on DPPG multilamellar vesicles, wher…

Biological membranes are exposed to a number of chemical and physical stresses that may alter the structure of the lipid bilayer in such a way that the permeability barrier to hydrophilic molecules and ions is degraded. These stresses include amphiphilic molecules involved in metabolism and signaling, highly charged polyamines, membrane-permeating peptides, and mechanical and osmotic stresses. As annexins are known to bind to lipid headgroups in the presence of calcium and increase the order of the bilayer lipids, this study addressed whether this activity of annexins provides a potential bene

Aggregated α-synuclein in Lewy bodies is one of the hallmarks of Parkinson's disease (PD). Earlier observations of α-synuclein aggregates in neurons grafted into brains of PD patients suggested cell-to-cell transfer of α-synuclein and a prion-like mechanism. This prompted the current investigation of whether α-synuclein passes over model phospholipid bilayers. We generated giant unilamellar vesicles (GUVs) containing a small amount of a lipid-conjugated red emitting dye (rhodamine B) and varied the membrane charge by using different molar ratios of DOPC and DOPS or cardiolipin. We then used co…

Oxidative stress is a key driver of neurodegenerative diseases, yet its impact on the mechanical properties of complex neuronal membranes remains poorly defined. A combined computational and experimental bottom-up approach was employed to study biomimetic membranes, ranging from simple bilayers to complex mixtures mimicking human neuronal composition. The results indicated that oxidation influenced all investigated membrane properties. While a decrease in membrane thickness and area compressibility was observed across all systems, trends for bending rigidity, area per lipid, and diffusion coef

Membrane shape is commonly described by curvature, area, and thickness, which underpin bilayer mechanics and protein-membrane interactions. In molecular dynamics (MD) simulations, these quantities must be inferred from discrete coordinates, and common grid-based smoothing approaches can introduce resolution-dependent artefacts and ambiguities, especially under periodic boundary conditions. Here we present CATpie, a Python framework for a unified geometric analysis of simulated membranes represented as smooth leaflet height fields over a periodic domain (from user-defined leaflet reference atom

Psychiatric disorders, such as schizophrenia, are treated with antipsychotics. One such molecule is chlorpromazine (CPZ) that mainly exerts its therapeutic effect via its antagonist activity on the dopamine D2 receptor. Due to its high lipophilicity, it can partition and accumulate in membranes, which can lead to a delayed full clinical effect, reached weeks after the beginning of treatment. CPZ's insertion in membranes impacts the bilayer's physicochemical properties, which can indirectly impact membrane-embedded receptors, such as the D2 receptor. To further delve into the potential role of

Caveolin is a monotopic integral membrane protein, widely expressed in metazoans and responsible for constructing enigmatic membrane invaginations known as caveolae. Recently, the high-resolution structure of a purified human caveolin assembly, the CAV1-8S complex, revealed a unique organization of 11 protomers arranged in a tightly packed, radially symmetric spiral disc. One face and the outer rim of this disc are hydrophobic, suggesting that the complex incorporates into membranes by displacing hundreds of lipids from one leaflet. The feasibility of this unique molecular architecture and its

Plasma membranes (PMs) exhibit asymmetry between their two leaflets in terms of phospholipid headgroups, unsaturation, and resulting membrane properties, such as packing and fluidity. Lateral heterogeneity, including the formation of lipid domains, is another crucial aspect of PMs with significant biological implications. However, the nature and even the existence of lipid domains in the two leaflets of PMs remain elusive, hindering a complete understanding of the significance of lipid asymmetry. Using coarse-grained molecular dynamics simulations of the asymmetric PM, we find that the outer l

Biological membrane channels mediate information exchange between cells and facilitate molecular recognition. While tuning the shape and function of membrane channels for precision molecular sensing via de-novo routes is complex, an even more significant challenge is interfacing membrane channels with electronic devices for signal readout, which results in low efficiency of information transfer - one of the major barriers to the continued development of high-performance bioelectronic devices. To this end, we integrate membrane spanning DNA nanopores with bioprotonic contacts to create programm

"Pink" or 1/f noise is a natural phenomenon omnipresent in physics, economics, astrophysics, biology, and even music and languages. In electrophysiology, the stochastic activity of a number of biological ion channels and artificial nanopores could be characterized by current noise with a 1/f power spectral density. In the anthrax toxin channel (PA63), it appears as fast voltage-independent current interruptions between conducting and nonconducting states. This behavior hampers potential development of PA63 as an ion-channel biosensor. On the bright side, the PA63 flickering represents a mesmer

This paper reports all-atom computer simulations of five phospholipid membranes (DMPC, DPPC, DMPG, DMPS, and DMPSH) with focus on the thermal equilibrium fluctuations of volume, energy, area, thickness, and chain order. At constant temperature and pressure, volume and energy exhibit strong correlations of their slow fluctuations (defined by averaging over 0.5 ns). These quantities, on the other hand, do not correlate significantly with area, thickness, or chain order. The correlations are mainly reported for the fluid phase, but we also give some results for the ordered (gel) phase of two memb

Bacterial fructans with a high degree of polymerisation cause a very large increase in surface pressure of lipid monolayers at the air-water interface with a broad range of lipids, including phosphatidylethanolamine and several types of phosphatidylcholines. The surface active effect of fructans contrasts strongly with the maximal effects observed for trehalose, sucrose and glucose under comparable conditions (20 and 0.6 mN/m for fructans and the other sugars, respectively). The results demonstrate a profound and specific membrane interaction of the fructans which is probably very different fr

Lipid bilayers and monolayers composed of dimyristoylphosphatidic acid (DMPA) and cholesterol were characterized by differential scanning calorimetry and film balance measurements. Increasing cholesterol content decreases the bilayer phase transition temperature and enthalpy in a manner similar to that observed before for other lipid/cholesterol systems. In monomolecular films at the air-water interface cholesterol exhibits the well known condensing effect in the liquid-expanded phase, while the liquid-condensed phase is less affected. As with the bilayer phase transition, the transition tempe…

Thylakoid membranes (TMs) of oxygenic photosynthetic organisms are flat membrane vesicles, which form highly organised, interconnected membrane networks. In vascular plants, they are differentiated into stacked and unstacked regions, the grana and stroma lamellae, respectively; they are densely packed with protein complexes performing the light reactions of photosynthesis and generating a proton motive force (pmf). The maintenance of pmf and its utilisation for ATP synthesis requires sealing the TMs at their highly curved regions (CRs). These regions are devoid of chlorophyll-containing protei
