MST-108494, was subjected to a panel of fermentation and media optimization trials, supported by analytical chemical profiling, to detect and enhance production of a rare class of secondary metabolites. Two new 32-membered macrolide compounds, named Novonestmycins A (1) and B (2), were isolated from the soil strain Streptomyces phytohabitans HBERC-20821. Their structures were elucidated by using spectroscopic methods, including 1D, 2D-NMR and MS spectrometry. Isolates of Legionella from 98 patients with Legionnaires' disease hospitalized in Columbus, Ohio, USA between 1991 through 1995 were tested for antimicrobial susceptibility to macrolides and quinolones using the Etest. Most (87%) isolates were Legionella pneumophila serogroup 1. All isolates tested remain susceptible to erythromycin, azithromycin, clarithromycin, ciprofloxacin, ofloxacin, and levofloxacin. In vitro susceptibility testing of Legionella to representative macrolides and quinolones should be considered to detect the emergence of resistant isolates.
