Dichloromethane-degrading bacteria in the genomic age.

Dichloromethane (DCM) is a volatile toxic halogenated solvent mainly produced and used industrially. DCM-degrading bacteria have long been models of choice for studying bacterial dehalogenation metabolism at the physiological, biochemical and genetic levels, and have also been used in bioremediation processes. DCM-degrading strains isolated in recent years will be discussed in the context of enzymes known to catalyze dehalogenation of DCM. Insights into the modes of adaptation of bacteria to DCM gained by comparative genomic analysis, highlight the importance of horizontal gene transfer in the

Synthetic consortia towards photosynthetically derived acetate for heterotrophic production of 1-butanol.

Synthetic consortia is an emerging field in biotechnology, offering advantages such as division of labor between the individual members and reduced risk of contamination. Consortia combining phototrophic and heterotrophic bacteria are attractive, as phototrophs can utilize CO₂ and light energy to sustain growth. Several studies have explored such systems where sucrose is most commonly used as the provided carbon source. An alternative is acetate, a smaller two-carbon molecule produced as a by-product by many microorganisms, including the model cyanobacterium Synechocystis PCC 6803 (thereafter

Design features of offshore oil production platforms influence their susceptibility to biocorrosion.

Offshore oil-producing platforms are designed for efficient and cost-effective separation of oil from water. However, design features and operating practices may create conditions that promote the proliferation and spread of biocorrosive microorganisms. The microbial communities and their potential for metal corrosion were characterized for three oil production platforms that varied in their oil-water separation processes, fluid recycling practices, and history of microbially influenced corrosion (MIC). Microbial diversity was evaluated by 16S rRNA gene sequencing, and numbers of total bacteri

Atrazine, chlorpyrifos, and iprodione effect on the biodiversity of bacteria, actinomycetes, and fungi in a pilot biopurification system with a green cover.

The use of biopurification systems can mitigate the effects of pesticide contamination on farms. The primary aim of this study was to evaluate the effect of pesticide dissipation on microbial communities in a pilot biopurification system. The pesticide dissipation of atrazine, chlorpyrifos and iprodione (35 mg kg-1 active ingredient [a.i.]) and biological activity were determined for 40 days. The microbial communities (bacteria, actinomycetes and fungi) were analyzed using denaturing gradient gel electrophoresis (DGGE). In general, pesticide dissipation was the highest by day 5 and reached 95%

Dehalococcoides and general bacterial ecology of differentially trichloroethene dechlorinating flow-through columns.

The diversity of Dehalococcoides mccartyi (Dhc) and/or other organohalide respiring or associated microorganisms in parallel, partial, or complete trichloroethene (TCE) dehalogenating systems has not been well described. The composition of Dhc populations and the associated bacterial community that developed over 7.5 years in the top layer (0-10 cm) of eight TCE-fed columns were examined using pyrosequencing. Columns biostimulated with one of three carbon sources, along with non-stimulated controls, developed into complete (ethene production, whey amended), partial (cis-dichloroethene (DCE) an

Residual effect of pre-emergence herbicides on microbial activities in relation to mineralization of C, N and P in the Gangetic alluvial soil of West Bengal, India.

An experiment has been conducted under laboratory conditions to investigate the residual effect of three pre-emergence herbicides (thiobencarb, pendimethalin and pretilachlor) at fivefold field application rates (7.5, 10.0 and 2.5 kg a.i. ha(-1), respectively), on the changes of microbial activities and some biochemical processes in the Gangetic alluvial soil of West Bengal. Application of herbicides in general significantly increased microbial biomass resulting in greater mineralization of C, N and P in soil. The highest stimulation of microbial biomass C was recorded with thiobencarb (24.4%)

A Slow-Release Substrate Stimulates Groundwater Microbial Communities for Long-Term in Situ Cr(VI) Reduction.

Cr(VI) is a widespread environmental contaminant that is highly toxic and soluble. Previous work indicated that a one-time amendment of polylactate hydrogen-release compound (HRC) reduced groundwater Cr(VI) concentrations for >3.5 years at a contaminated aquifer; however, microbial communities responsible for Cr(VI) reduction are poorly understood. In this study, we hypothesized that HRC amendment would significantly change the composition and structure of groundwater microbial communities, and that the abundance of key functional genes involved in HRC degradation and electron acceptor reducti

Mixotrophic metabolism of Chlorella sorokiniana and algal-bacterial consortia under extended dark-light periods and nutrient starvation.

Microalgae harbor a not fully exploited industrial and environmental potential due to their high metabolic plasticity. In this context, a better understanding of the metabolism of microalgae and microalgal-bacterial consortia under stress conditions is essential to optimize any waste-to-value approach for their mass cultivation. This work constitutes a fundamental study of the mixotrophic metabolism under stress conditions of an axenic culture of Chlorella sorokiniana and a microalgal-bacterial consortium using carbon, nitrogen, and phosphorous mass balances. The hydrolysis of glucose into vol

Biodegradation of hexachlorobenzene by a constructed microbial consortium.

A consortium comprised of an engineered Escherichia coli DH5α and a natural pentachlorophenol (PCP) degrader, Sphingobium chlorophenolicum ATCC 39723, was assembled for degradation of hexachlorobenzene (HCB), a persistent organic pollutant. The engineered E. coli strain, harbouring a gene cassette (camA (+) camB (+) camC) that encodes the F87W/Y96F/L244A/V247L mutant of cytochrome P-450cam (CYP101), oxidised HCB to PCP. The resulting PCP was then further completely degraded by ATCC 39723. The results showed that almost 40 % of 4 μM HCB was degraded by the consortium at a rate of 0.033 nmol/mg

Bio-reduction of tetrachloroethen using a H2-based membrane biofilm reactor and community fingerprinting.

Chlorinated ethenes in drinking water could be reductively dechlorinated to non-toxic ethene by using a hydrogen based membrane biofilm reactor (H2-MBfR) under denitrifying conditions as it provides an appropriate environment for dechlorinating bacteria in biofilm communities. This study evaluates the reductive dechlorination of perchloroethene (PCE) to non-toxic ethene (ETH) and comparative community analysis of the biofilm grown on the gas permeable membrane fibers. For these purposes, three H2-MBfRs receiving three different chlorinated ethenes (PCE, TCE and DCE) were operated under differe

Microbial diversity and changes in the distribution of dehalogenase genes during dechlorination with different concentrations of cis-DCE.

A dechlorinating consortium (designated as TES-1 culture) able to convert trichloroethene (TCE) to ethene was established from TCE-contaminated groundwater. This culture had the ability of complete dechlorination of TCE within about one month. From the clone library analysis of 16S rRNA gene, this culture was mainly composed of fermentation bacteria, such as Clostridium spp., and Desulfitobacterium spp. known as facultative dechlorinator. PCR using specific primers for Dehalococcoides spp. and the dehalogenase genes confirmed that the culture contained the Dehalococcoides spp. 16S rRNA gene an

Development and characterization of DehaloR^2, a novel anaerobic microbial consortium performing rapid dechlorination of TCE to ethene.

A novel anaerobic consortium, named DehaloR^2, that performs rapid and complete reductive dechlorination of trichloroethene (TCE) to ethene is described. DehaloR^2 was developed from estuarine sediment from the Back River of the Chesapeake Bay and has been stably maintained in the laboratory for over 2 years. Initial sediment microcosms showed incomplete reduction of TCE to DCE with a ratio of trans- to cis- isomers of 1.67. However, complete reduction to ethene was achieved within 10 days after transfer of the consortium to sediment-free media and was accompanied by a shift to cis-DCE as the

[Biodegradation of tri- and perchloroethylene in sewage waters and soils by a microbial consortium of compost and phototrophic bacteria].

Compost and phototrophic bacteria are found to be able to degrade trichloroethylene (TCE) and perchloroethylene (PCE). With a TCE dose increase by more than 10 mg/kg, the TCE degradation decreases due to the toxic effect of this pollutant on microbial consortium activity. The addition of compost combined with a liquid culture of phototrophic bacteria (PTB) is experimentally proved to effectively decrease the TCE content in soil and water.

PGPR inoculation and growth enhancement of crops cultivated in hydroponic systems.

Plant growth-promoting rhizobacteria (PGPR) are ubiquitous rhizosphere microorganisms that promote plant health through various mechanisms. Although the study of PGPR inoculants in soil has been done for ages, their application in hydroponic systems has received relatively limited attention. This review identifies PGPR inoculants that are commonly used in hydroponics, methods of application, and their effects on plant growth and nutrient use efficiency. Literature shows that PGPR inoculants improve plant performance in controlled hydroponic systems through the production of growth-stimulating

Changes in the Microbiome Profile in Different Parts of the Intestine in Piglets with Diarrhea.

Determining the taxonomic composition of microbial consortia of the piglet intestine is of great importance for pig production. However, knowledge on the variety of the intestinal microbiome in newborn piglets is limited. Piglet diarrhea is a serious gastrointestinal disease with a high morbidity and mortality that causes great economic damage to the pig industry. In this study, we investigated the microbiome of various sections of the piglet intestine and compared the microbiome composition of healthy and diarrheal piglets using high-throughput sequencing of the 16S rRNA gene. The results sho

Role of Oral Microbiota in Carcinogenesis: A Short Review.

A strong and healthy microbiome is responsible for homeostasis between the host and microbiota which is necessary to achieve the normal functioning of the body. Dysbiosis provokes prevalence of pathogenic microbes, leading to alterations in gene expression profiles and metabolic processes. This in turn results in anomalous immune responses of the host. Dysbiosis may be associated with a wide variety of diseases like irritable bowel syndrome, coeliac disease, allergic conditions, bronchitis, asthma, heart diseases and oncogenesis. Presently, the links between oral microbial consortia and their

Impact of Anthropogenic Activities on the Dissemination of ARGs in the Environment-A Review.

Over the past few decades, due to the excessive consumption of drugs in human and veterinary medicine, the antimicrobial resistance (AR) of microorganisms has risen considerably across the world, and this trend is predicted to intensify. Many worrying research results indicate the occurrence of pools of AR, both directly related to human activity and environmental factors. The increase of AR in the natural environment is mainly associated with the anthropogenic activity. The dissemination of AR is significantly stimulated by the operation of municipal facilities, such as wastewater treatment p

Modeling Healthy and Dysbiotic Vaginal Microenvironments in a Human Vagina-on-a-Chip.

Women's health, and particularly diseases of the female reproductive tract (FRT), have not received the attention they deserve, even though an unhealthy reproductive system may lead to life-threatening diseases, infertility, or adverse outcomes during pregnancy. One barrier in the field is that there has been a dearth of preclinical, experimental models that faithfully mimic the physiology and pathophysiology of the FRT. Current in vitro and animal models do not fully recapitulate the hormonal changes, microaerobic conditions, and interactions with the vaginal microbiome. The advent of Organ-o

Soil microorganisms facilitated the electrode-driven trichloroethene dechlorination to ethene by Dehalococcoides species in a bioelectrochemical system.

Bioelectrochemical dechlorination using organohalide-respiring bacteria (ORBs) is a promising technique for remediating contaminated groundwater. Generally, a longer enrichment period is required for selecting the ORB consortia to achieve bioelectrochemical dechlorination. However, the full dechloriantion is difficult to be achieved due to the absence of functional species (e.g. Dehalococcoides) in previously used enrich cultures. To overcome these challenges, bioelectrochemical dechlorination using a culture enriched with the pre-augmented Dehalococcoides was performed for the first time in t
