Site-specific transdermal delivery of Qingteng Waifu San in a rheumatoid arthritis rabbit model.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by synovial inflammation, joint swelling, pain, cartilage destruction and bone erosion. Qingteng Waifu San is an external sinomenine-containing preparation, but its site-specific transdermal response requires disease-relevant evaluation. OBJECTIVES: To compare Zusanli (ST36)-site and adjacent non-acupoint transdermal application of Qingteng Waifu San in an ovalbumin/complete Freund's adjuvant-induced RA rabbit model and to evaluate formulation, neuropeptide, inflammatory, behavioural and histopathological outc

Sinomenine protects mice against Staphylococcus aureus-induced endometritis through inhibition of P2X7 receptor-mediated ferroptosis and inflammation.

Endometritis is a major cause of infertility that adversely impacts both human reproductive health and livestock productivity. This study investigated the protective effects and underlying mechanisms of sinomenine (SIN) against Staphylococcus aureus (S. aureus)-induced endometritis. Seventy-two adult female mice were randomly divided into six groups: control group, SIN group (100 mg/kg), S. aureus group (1 × 107 CFU per 200 μL PBS), and SIN (25 mg/kg, 50 mg/kg, and 100 mg/kg, respectively) + S. aureus groups. Uterine tissues were collected for analysis. Inflammatory factors, ferroptosis, and P

Sinomenine protects against atherosclerosis in apolipoprotein E-knockout mice by inhibiting of inflammatory pathway.

Atherosclerosis, a multifaceted and persistent inflammatory condition, significantly contributes to the progression of cardiocerebrovascular disorders, such as myocardial infarctions and cerebrovascular accidents. It involves the accumulation of cholesterol, fatty deposits, calcium and cellular debris in the walls of arteries, leading to the formation of plaques. Our aim is to investigate the potential of sinomenine to counteract atherosclerosis in mice lacking Apolipoprotein E (ApoE-/-) Mice. We employed the high-fat diet-induced method to induce atherosclerosis in ApoE-/- mice, and the mice

Pharmacological characterization of naloxegol: In vitro and in vivo studies.

Opioid-induced constipation is the most prevalent adverse effect of opioid drugs. Peripherally acting mu opioid receptor antagonists (PAMORAs), including naloxegol, are indicated for the treatment of opioid-induced constipation. The aim of this study was the in vitro and in vivo pharmacological characterization of naloxegol in comparison with naloxone. In vitro experiments were performed to measure calcium mobilization in cells coexpressing opioid receptors and chimeric G proteins and mu receptor interaction with G protein and β-arrestin 2 using bioluminescence resonance energy transfer. In vi

Topical administration of the kappa opioid receptor agonist nalfurafine suppresses corneal neovascularization and inflammation.

Corneal neovascularization (CNV) causes higher-order aberrations, corneal edema, ocular inflammation, and corneal transplant rejection, thereby decreasing visual acuity. In this study, we investigated the effects of topical administration of the kappa opioid receptor agonist nalfurafine (TRK-820) on CNV. To induce CNV, intrastromal corneal sutures were placed on the corneal stroma of BALB/c mice for 2 weeks. Nalfurafine (0.1 µg/2 μL/eye) was topically administered to the cornea once or twice daily after CNV induction. The CNV score, immune cell infiltration, and mRNA levels of angiogenic and p

Effects of sinomenine on the proliferation, cytokine production, and regulatory T-cell frequency in peripheral blood mononuclear cells of rheumatoid arthritis patients.

Sinomenine (SN) is a plant-derived alkaloid isolated from Caulis Sinomenii. It has been approved by the State Food and Drug Administration of China for treating rheumatoid arthritis (RA) nearly 20 years ago. To investigate the anti-RA mechanism of SN, a lot of scholars reported the immunosuppressive effect of SN on T lymphocytes. We continued to evaluate the suppressive function of SN by using human peripheral blood mononuclear cells (PBMCs) isolated from RA patients. As the positive control, 10 ng/ml of methylprednisolone (MP) showed the antiproliferation effect on mitogen-activated PBMCs of

Simulation and Prediction of the Drug-Drug Interaction Potential of Naloxegol by Physiologically Based Pharmacokinetic Modeling.

Naloxegol, a peripherally acting μ-opioid receptor antagonist for the treatment of opioid-induced constipation, is a substrate for cytochrome P450 (CYP) 3A4/3A5 and the P-glycoprotein (P-gp) transporter. By integrating in silico, preclinical, and clinical pharmacokinetic (PK) findings, minimal and full physiologically based pharmacokinetic (PBPK) models were developed to predict the drug-drug interaction (DDI) potential for naloxegol. The models reasonably predicted the observed changes in naloxegol exposure with ketoconazole (increase of 13.1-fold predicted vs. 12.9-fold observed), diltiazem

Sinomenine hydrochloride protects against polymicrobial sepsis via autophagy.

Sepsis, a systemic inflammatory response to infection, is the major cause of death in intensive care units (ICUs). The mortality rate of sepsis remains high even though the treatment and understanding of sepsis both continue to improve. Sinomenine (SIN) is a natural alkaloid extracted from Chinese medicinal plant Sinomenium acutum, and its hydrochloride salt (Sinomenine hydrochloride, SIN-HCl) is widely used to treat rheumatoid arthritis (RA). However, its role in sepsis remains unclear. In the present study, we investigated the role of SIN-HCl in sepsis induced by cecal ligation and puncture

Pharmacological characterization and therapeutic potential for the treatment of opioid abuse with ATPM-ET, an N-ethyl substituted aminothiazolomorphinan with κ agonist and μ agonist/antagonist activity.

We previously reported that the κ agonists with mixed μ activity could attenuate heroin self-administration with less potential to develop tolerance. The present study further investigated the effects of (-)-3-N-Ethylamino-thiazolo[5,4-b]-N-cyclopropylmethylmorphinan hydrochloride (ATPM-ET), a κ agonist and μ agonist/antagonist, on the acquisition and reinstatement of morphine-induced conditioned place preference (CPP), heroin self-administration and heroin-primed reinstatement of drug-seeking behavior. We found that ATPM-ET produced a longer duration of potent antinociceptive effects with les

A lack of potency for the delta-opioid antagonist naltrindole after microinjection into the rostral ventrolateral medulla of rabbits.

1. Naltrindole is a novel delta-opioid antagonist which is highly potent in vitro. We examined the effects on arterial pressure of naltrindole (0.3-300 pmol) after microinjection into the pressor area of the rostral ventrolateral medulla of chloralose-anaesthetized rabbits. 2. Naltrindole itself had no significant effects on arterial pressure, and only slightly attenuated the hypotensive effect of an exogenous agonist ( [D-Ala2, D-Leu5]-enkephalin). This is in contrast to previous demonstration of a marked pressor effect following another delta-antagonist, ICI 174,864. 3. Thus, naltrindole, te

Nalbuphine and Althesin anaesthesia. A controlled double-blind comparison for cystoscopy.

In a double-blind, randomized study of 52 unpremedicated outpatients undergoing cytoscopy, either nalbuphine 20 mg or saline 2 ml was given intravenously immediately before induction of anaesthesia with Althesin 0.05 ml/kg. Increments of Althesin 0.5 ml were used to maintain anaesthesia. The patients received nalbuphine required less Althesin than those who received saline (p less than 0.001) for a similar duration of surgery, and there was less movement during anaesthesia (p less than 0.001). During surgery hyperventilation was observed in the patients who received saline; those who received

Neuroprotective effect of sinomenine on parkinson's disease through NLRP3/Caspase-1/GSDMD pathway-mediated pyroptosis inhibition.

Mounting evidence indicates that NLRP3 inflammasome-mediated pyroptosis is critically involved in Parkinson’s disease (PD) pathogenesis. Sinomenine (SN) possesses neuroprotective properties and is known to modulate the NLRP3 inflammasome, but its potential to protect against PD via pyroptosis inhibition remains unexplored. This study demonstrates that SN confers significant neuroprotection in both cellular and animal PD models. Key findings show that SN alleviates motor deficits and nigral pathology in vivo, enhances neuronal viability, and suppresses pyroptosis in vitro. Mechanistically, SN p
