Salmonid fish: model organisms to study cardiovascular morphogenesis in conjoined twins?

BACKGROUND: There is a gap in knowledge regarding the cardiovascular system in fish conjoined twins, and regarding the cardiovascular morphogenesis of conjoined twins in general. We examined the cardiovascular system in a pair of fully developed ventrally conjoined salmonid twins (45.5 g body weight), and the arrangement of the blood vessels during early development in ventrally conjoined yolk sac larvae salmonid twins (<0.5 g body weight). RESULTS: In the fully developed twins, one twin was normal, while the other was small and severely malformed. The mouth of the small twin was blocked, inh…

Ketoconazole inhibits Malassezia furfur morphogenesis in vitro under filamentation optimized conditions.

Malassezia furfur, a constituent of the normal human skin flora, is an etiological agent of pityriasis versicolor, which represents one of t … on both M. furfur yeast and mycelial cells, ketoconazole also has a dramatic impact on suppressing morphogenesis. Since mycelia typified the pathogenic form of Malassezia infection, the capacity of ketoconazole to block morphogenesis may represent an additional important effect of the antifungal.

Nordihydroguaiaretic acid (NDGA) inhibits replication and viral morphogenesis of dengue virus.

Dengue is the most common mosquito borne viral disease in humans. The infection with any of the 4 dengue virus serotypes (DENV) can either b … reduction in number of lipid droplets (LDs), the neutral lipid storage organelles involved in DENV morphogenesis that are known to increase in number during DENV infection. Furthermore, NDGA treatment resulted in dissociation of the C protein from LDs. Overall our results suggest that NDGA inhibits DENV infection by targeting genome replication and viral assem …

Evidence of interlobular repulsion during branching morphogenesis in mouse salivary glands.

We developed a culture method for detecting repulsion among epithelial lobules during branching morphogenesis in mouse submandibular glands. Three epithelia were placed at each vertex of an imaginary triangle apart but near enough to meet with one another if each of them expands radially during the culture period. No repulsion was observed following cultivation with neuregulin 1 and lysophosphatidic acid; the epithelia actively branched and nearly contacted one another in the triangle's center. In contrast, strong repulsion was observed among the epithelia cultured with fibroblast growth facto…

A transcriptionally silent RXRalpha supports early embryonic morphogenesis and heart development.

Retinoic acid (RA) receptors (RARs) alpha, beta, and gamma heterodimerized with rexinoid receptors (RXRs) alpha, beta, and gamma mediate the RA signal. To analyze the contribution of the transcriptional activity of RXRalpha, the main RXR during embryogenesis, we have engineered a mouse line harboring a transcriptionally silent RXRalpha mutant that lacks the activation functions AF1 and AF2. All homozygous mutants (Rxra(afo)) display the ocular defects previously observed in compound Rar-null and Rxra/Rar-null mutants, thus demonstrating that a transcriptionally active RXRalpha is required duri

Planar cell polarity signalling couples cell division and morphogenesis during neurulation.

Environmental and genetic aberrations lead to neural tube closure defects (NTDs) in 1 out of every 1,000 births. Mouse and frog models for t … accumulations and NTDs. Remarkably, blocking cell division leads to rescue of trilobite neural tube morphogenesis despite persistent defects in convergence and extension. These results reveal a function for PCP signalling in coupling cell division and morphogenesis at neurulation and indicate a previously unrecognized mechanism that might underlie NTDs.

Role for retinoid signaling in left-right asymmetric digestive organ morphogenesis.

The looping events that establish left-right asymmetries in the vertebrate gut tube are poorly understood. Retinoic acid signaling is known to impact left-right development in multiple embryonic contexts, although its role in asymmetric digestive organ morphogenesis is unknown. Here, we show that the genes for retinaldehyde dehydrogenase (RALDH2) and a retinoic acid hydroxylase (CYP26A1) are expressed in complementary patterns in the Xenopus gut during looping. A late-stage chemical genetic assessment reveals that agonists and antagonists of retinoid signaling generate abnormal gut looping top…

Abnormal Purkinje cell morphogenesis in human renal agenesis. A Golgi study.

The prenatal histogenesis of the human cerebellum in renal agenesis has been studied in four fetuses (32-, 33-, 33-, and 36-week-old) using the rapid Golgi method. A fundamental anomaly involving the developing Purkinje cell has been found. The normal histogenesis of this neuron is aborted about the 32nd-33rd week of gestation. Subsequently, the neuron undergoes a progressive transformation acquiring an abnormal dendritic morphology in only four weeks. The perikaryal basal dendrites are retained rather than disappear, and they continue to grow spreading horizontally through the ganglionic plat

Spatial Relations between Coccoliths and Their Confining Membrane During Crystal Morphogenesis.

Coccoliths are multicrystalline calcite structures formed by microalgae within an intracellular vesicle. The morphology of each crystal is complex, and recent studies postulate that coccolith morphogenesis is regulated by the bounding membrane of its vesicle. However, the limited information about the native-state organization within the cell makes it difficult to understand which structural aspects of the membrane are responsible for morphogenesis. Here, we examined the vesicular environment during the formation of Calcidiscus leptoporus coccoliths, using advanced cryo-electron microscopy and…

Intestine organotypic culture recapitulates human embryonic villus morphogenesis at week 8.

The human small intestine depends on villi to generate the large absorptive surface area required for nutrient uptake and barrier integrity. Existing human in vitro models fail to reproduce early villus morphogenesis, as they lack developmental signaling and epithelial-mesenchymal interactions that drive villus initiation. Such models are crucial for understanding the mechanisms of human villus formation to study intestinal development and disease. This study aimed to establish a human organotypic intestine model that recapitulates early villus development and to compare its features with huma…

Transcriptome Dynamics of Terminal Buds During Flower Bud Morphogenesis in Blueberry.

Background/Objectives: Flower bud morphogenesis is a critical developmental phase during which blueberry transitions from vegetative to reproductive growth, yet the transcriptome dynamics and regulatory networks within buds during this process have not been systematically characterized. Methods: Terminal buds of the rabbiteye blueberry 'Brightwell' were sampled at six time points spanning from summer shoot cessation to bud swelling and dormancy entry. RNA sequencing, trend clustering, and Pearson correlation network analyses were performed to identify potentially important genes and regulatory…

Temporal dynamics of BMP/Nodal ratio drive tissue-specific gastrulation morphogenesis.

Anteroposterior elongation of the vertebrate body plan is driven by convergence and extension (C&E) gastrulation movements in both the mesoderm and neuroectoderm, but how or whether molecular regulation of C&E differs between tissues remains an open question. Using a zebrafish explant model of anteroposterior axis extension, we show that C&E of the neuroectoderm and mesoderm can be uncoupled ex vivo, and that morphogenesis of individual tissues results from distinct morphogen signaling dynamics. Using precise temporal manipulation of BMP and Nodal signaling, we identify a critical developmenta…

FOXF1 Regulates Alveolar Epithelial Morphogenesis through Transcriptional Activation of Mesenchymal WNT5A.

Mutations in the FOXF1 (forkhead box F1) gene, encoding the mesenchymal FOX (forkhead box) transcription factor, are linked to alveolar capillary dysplasia with misalignment of pulmonary veins (ACDMPV), a severe congenital disorder associated with the loss of alveolar capillaries and lung hypoplasia. Although proangiogenic functions of FOXF1 have been extensively studied, the role of FOXF1 in mesenchymal-epithelial signaling during lung development remains uncharacterized. Herein, we used murine lung organoids to demonstrate that the S52F FOXF1 mutation (found in patients with ACDMPV) stimulat
