Climate-stressed crop mycobiomes as pre-harvest sentinels of human mycotoxin exposure: an AI-enabled One Health framework linking dysbiosis, exposomics, and toxicopathology.

Climate change is altering the conditions under which crop-associated fungi colonize plants and produce mycotoxins. Current control relies on both pre-harvest management and post-harvest testing; however, field-level fungal-community changes are rarely used to guide where and when testing should be intensified. This Review critically compares evidence linking weather and crop stress, fungal-community composition, toxigenic strain activity, contamination during harvest and storage, human biomarkers, and organ-specific effects. On this basis, we propose the Crop Mycobiome-Exposome-Toxicopatholog…

The Effect of Clay- and Yeast-Based Mycotoxin Binder on Performance, Lymphoid Organs, Blood Parameters, Immune Response and Haematology Parameters of Broilers Exposed to Aflatoxin B1.

BACKGROUND: Feeding broilers aflatoxin-contaminated feed can disrupt the bird's performance, immune system and organs such as the liver, kidneys and heart. One possible strategy to minimise the inevitable impact of aflatoxins could be the use of toxic binders in the feed. OBJECTIVES: The present study aimed to investigate the effect of mycotoxin binder magnotox (MAG) in reducing the toxicity of aflatoxin B1 (AFB1) on performance, lymphoid organs, blood parameters, immune response and haematology parameters of broiler chickens. METHODS: A total of 240 1-day-old male broiler chicks of Ross 308

Food Toxicity of Mycotoxin Citrinin and Molecular Mechanisms of Its Potential Toxicity Effects through the Implicated Targets Predicted by Computer-Aided Multidimensional Data Analysis.

The mycotoxin citrinin, which can contaminate food, is a major global concern. Citrinin is regarded as an inevitable pollutant in foods and feed since fungi are widely present in the environment. To identify contentious toxicity and lessen its severity by understanding the targets of citrinin in the human body and the impacted biosynthetic pathways, we analyzed the production of citrinin from Aspergillus flavus and Penicillium notatum and used a thorough bioinformatics analysis to characterize the toxicity and predict genes and protein targets for it. The predicted median fatal dosage (LD50) f

5-hydroxy-1-methylhydantoin (NZ-419), an endogenous antioxidant, attenuates mycotoxin-induced oxidative stress in human kidney proximal tubular epithelial cells.

BACKGROUND: Exposure to foodborne mycotoxins such as ochratoxin A (OTA) and citrinin (CIT) causes severe oxidative stress-mediated nephropathy. This study investigated the protective potential of 5-hydroxy-1-methylhydantoin (NZ-419), an endogenous creatinine metabolite, against OTA- and CIT-induced toxicity in human proximal tubular epithelial (HK-2) cells. METHODS: We evaluated the radical-scavenging activity of NZ-419 using cell-free assays, including electron spin resonance. In HK-2 cells, its effects on mycotoxin-induced cytotoxicity, reactive oxygen species (ROS) generation, mitochondria…

Repackable microfluidic molecularly imprinted solid-phase extraction coupled with mass spectrometry (μMISPE-MS) for rapid analysis of mycotoxin in agri-foods: an example of zearalenone.

Mycotoxins are detectable in 60-80% of food crops, posing significant threats to human health and food security, and causing substantial economic losses. Most mitigation approaches focus on detecting mycotoxins with standard methods based on liquid chromatography coupled with mass spectrometry (LC-MS). Typical MS methods require extensive sample preparation and clean-up due to the matrix effect, followed by time-consuming LC separation, complicating the analysis process and limiting analytical throughput. This study reports the development of a repackable microfluidic molecularly imprinted sol…

Purinergic signalling displays a pro-inflammatory profile in spleen and splenic lymphocytes of Rhamdia quelen fed with a diet contaminated by fungal mycotoxin: Involvement on disease pathogenesis.

The spleen is an important secondary lymphatic organ that plays a key role in the immune and inflammatory responses of teleost fish. The purinergic signalling has been associated to these types of responses under pathological conditions by the regulation of extracellular adenosine triphosphate (ATP) and its metabolite adenosine (Ado), where both exert potent pro-inflammatory and anti-inflammatory profiles, respectively. The exact pathway involved on the immunotoxic effects of aflatoxin B1 (AFB1) in fish fed with diets containing this mycotoxin remains poorly understood. Thus, the aim of this s

Molecular Dynamics and Quantum Chemical Insights into the Molecular Basis of Differential Toxicity of the Mycotoxin Deoxynivalenol (DON) and Its Microbial Derivatives DOM‑1 and 3-epi-DON.

Deoxynivalenol (DON) is a trichothecene mycotoxin from Fusarium species that contaminates cereal grains, posing health risks. DON binds to site A of the 60S ribosomal subunit in eukaryotes, triggering a ribotoxic stress response. Despite structural similarity, its metabolites, deepoxy-deoxynivalenol (DOM-1) and 3-epi-deoxynivalenol (3-epi-DON), are nontoxic, but the molecular basis of this difference is unclear. This study employed molecular docking, molecular dynamics (MD) simulations, and quantum chemical calculations based on Symmetry-Adapted Perturbation Theory (SAPT) to analyze the intera

Multi-Mycotoxin Quantification in Soy Protein Powders, Soymilk, Soybeans (Glycine max) and Other Legumes Using a New Targeted Differential Ion Mobility LCMS Method.

Foods contaminated with fungal mycotoxins pose a serious risk to human health. Soy and other legumes are a vital source of alternative protein, yet the rates of mycotoxin contamination are not understood. We therefore developed a sensitive stable-isotope-dilution-differential-ion-mobility-spectrometry-LC-MS method for the quantification of 43 mycotoxins in soylike matrixes. Extraction by QuEChERS-d-SPE-PSA reduced matrix effects, while Differential-Ion-Mobility-Spectrometry (DMS) decreased background-noise and enabled the selective quantification of acetyl-deoxynivalenol (ACDON) isomers. Prepubertal exposure to an oestrogenic mycotoxin zearalenone induces central precocious puberty in immature female rats through the mechanism of premature activation of hypothalamic kisspeptin-GPR54 signaling.

Sporadic epidemics and several researches in rodents indicated that zearalenone (ZEA) and its metabolites, the prevailing oestrogenic mycotoxins in foodstuffs, were a triggering factor for true precocious puberty development in girls. Nevertheless, the neuroendocrine mechanism through which ZEA mycoestrogens advance puberty onset is not fully understood. To elucidate this issue, hypothalamic kisspeptin-G-protein coupled receptor-54 (GPR54) signaling pathway that regulates the onset of puberty was focused on in the present study. Immature female SD rats were given a daily intragastric administr…

New Strategies for the Prevention and Control of Foodborne Mycotoxins: Glycyrrhizic Acid Enhances Antioxidant Capacity and Modulates the ROS/NF-κB/NLRP3 Pathway to Treat Zearalenone-Induced Lung Injury in Chickens.

Zearalenone is an estrogen-like mycotoxin produced by Fusarium fungi. Growing evidence shows that ZEA causes multisystem damage via oxidative stress. Glycyrrhizic acid (GA), from Glycyrrhiza glabra, has anti-inflammatory/antioxidant properties. The mechanisms of GA and ZEA in chicken pulmonary oxidative stress and inflammation remain elusive. Therefore, we established zearalenone-exposed chicken lung and CP-II cells models. ZEA exposure triggers an inflammatory cascade via the ROS-NF-κB axis, activating NLRP3 inflammasomes and promoting the release of proinflammatory factors. It also increased

Early-life exposure to the mycotoxin zearalenone causes aberrant lipid metabolism requiring mitochondrial fission in Caenorhabditis elegans: Mechanistic insights from in vivo genetic and in silico analyses.

Zearalenone, a mycotoxin commonly detected in food and the environment, is an underestimated global public health concern, especially as climate change may worsen its prevalence and effects. Although zearalenone has been associated with various adverse health outcomes, the exact mechanistic links to obesogenic effects, mitochondrial dysfunction, and related molecular pathways remain unclear. This research explored the impact of early-life zearalenone exposure on lipid accumulation and its potential relationship to mitochondrial dysfunction in the nematode Caenorhabditis elegans. Multiple lipid

Does chronic dietary exposure to the mycotoxin deoxynivalenol affect the porcine hepatic transcriptome when an acute-phase response is initiated through first or second-pass LPS challenge of the liver?

The sensitivity of pigs to deoxynivalenol (DON) might be increased by systemic inflammation (SI), which also has consequences for hepatic integrity. Liver lesions and a dys-regulated gene network might hamper hepatic handling and elimination of DON whereby the way of initiation of hepatic inflammation might play an additional role. First and second-pass exposure of the liver with LPS for triggering a SI was achieved by LPS infusion via pre- or post-hepatic venous route, respectively. Each infusion group was pre-conditioned either with a control diet (0.12 mg DON/kg diet) or with a DON-contamin

The Role of AFB1, OTA, TCNs, and Patulin in Forensic Sciences: Applications in Autopsy, Criminal Investigations, and Public Health Prevention.

Mycotoxins, specifically aflatoxin B1 (AFB1), ochratoxin A (OTA), trichothecenes (TCNs), and patulin, are a group of secondary metabolites that can contaminate food, leading to severe health implications for humans. Their detection and analysis within forensic toxicology are crucial, particularly as they can be implicated in cases of poisoning, foodborne illnesses, or lethal chronic exposure. However, little is known about the application that mycotoxins could have in forensic investigations and especially about the possibility of extracting and quantifying these molecules on tissues or post-m…
