Cortical markers of excitation/inhibition balance are associated with sensory responsivity from infancy in longitudinal cohorts enriched for autism and ADHD.

Autism and ADHD are characterised by atypical sensory responsivity, and this may be driven by alterations in the balance of cortical excitation to inhibition (E/I). Studies early in development are required to establish when sensory responsivity differences emerge and whether they predict later neurodevelopmental condition outcomes. We utilised data from a prospective longitudinal cohort of infants with and without a family history (FH) of autism and/or ADHD (N = 151; 55% male, 83% white). We extracted electroencephalography (EEG) metrics of E/I balance at 5, 10 and 14 months; the aperiodic ex

Young Adults with Anxiety Disorders Show Reduced Inhibition in the Dorsolateral Prefrontal Cortex at Higher Trait Anxiety Levels: A TMS-EEG Study.

BACKGROUND: The neuropathology of anxiety disorders, including specific phobias, social phobias, and generalized anxiety disorders (GAD), has been believed to be rooted in a reduced inhibition of limbic areas by the dorsolateral prefrontal cortex (DLPFC). Trait anxiety has been linked to insufficient recruitment of DLPFC mechanisms for attentional control. Despite limited research on individuals with anxiety disorders, our study utilized transcranial magnetic stimulation to assess DLPFC cortical activity and emotional states using the N100 as an indicator of GABA-B-mediated cortical inhibition

Suppression of reward-induced dopamine release in the nucleus accumbens in animal models of depression: Differential responses to drug treatment.

Anhedonia, the loss of interest or pleasure in previously enjoyable activities, is a core symptom of major depressive disorder, suggesting that the brain reward system may be dysfunctional in this condition. Neurochemical changes in the mesolimbic dopamine (DA) system are not fully understood in animal models of depression. We investigated reward (30% sucrose intake)-induced DA release in the nucleus accumbens (NAc) and the effect of chronic treatment with the antidepressant escitalopram (5mg/kg, intraperitoneally twice daily for 3 weeks) in two animal models of depression. Exposure to chronic

Pre- but not post-menopausal female CBA/CaJ mice show less prepulse inhibition than male mice of the same age.

Prepulse inhibition (PPI) of the acoustic startle reflex (ASR) has been reported to be weaker in females than males for both humans and rats. Although there are exceptions, on balance these data suggest that PPI is sensitive to sex-specific neurosteroids; in contrast, most studies with mice have not replicated this effect. We compared PPI for noise decrement prepulses (quiet gaps) in female CBA/CaJ mice at 3-8 months (pre-menopausal: n = 55) and 17-25 months of age (post-menopausal, n = 33) with similarly aged groups of males (n = 48, 35). Both PPI and ASR levels were significantly reduced in

Spatially local inhibition and synaptic plasticity together enable dynamic, context-dependent integration of parallel sensory pathways.

Retinal ganglion cells have traditionally been grouped into cells that are sensitive to luminance but not spatial structure and cells with responses that are enhanced by spatial structure. Neither category describes mouse Off-transient alpha cells, which respond strongly to spatially homogeneous inputs and are suppressed by spatial structure. We identified two circuit mechanisms that together can explain this unusual spatial selectivity. First, the inhibition that controls responses of these cells is tuned to finer spatial structure than excitation, causing the balance of excitation and inhibi

Convergence and Segregation of Excitatory and Inhibitory Afferents in the Paraventricular Thalamic Nucleus.

The paraventricular thalamic nucleus (PVT) integrates subcortical signals related to arousal, stress, addiction, and anxiety with top-down cortical influences. Increases or decreases in PVT activity exert profound, long-lasting effects on behavior related to motivation, addiction, and homeostasis. Yet the sources of its subcortical excitatory and inhibitory afferents, their distribution within the PVT, and their integration with layer-specific cortical inputs remain unclear. Using transgenic male and female mice selective for GABAergic and glutamatergic neurons, or for different cortical layer

Motor Function and Physiology in Youth With Neurofibromatosis Type 1.

BACKGROUND: Neurofibromatosis type 1 (NF1) is a genetic neurocutaneous disorder commonly associated with motor and cognitive symptoms that greatly impact quality of life. Transcranial magnetic stimulation (TMS) can quantify motor cortex physiology, reflecting the basis for impaired motor function as well as, possibly, clues for mechanisms of effective treatment. We hypothesized that children with NF1 have impaired motor function and altered motor cortex physiology compared to typically developing (TD) control children and children with attention-deficit/hyperactivity disorder (ADHD). METHODS:

Multi-modal investigation of transcranial ultrasound-induced neuroplasticity of the human motor cortex.

INTRODUCTION: There is currently a gap in accessibility to neuromodulation tools that can approximate the efficacy and spatial resolution of invasive methods. Low intensity transcranial focused ultrasound stimulation (TUS) is an emerging technology for non-invasive brain stimulation (NIBS) that can penetrate cortical and deep brain structures with more focal stimulation compared to existing NIBS modalities. Theta burst TUS (tbTUS, TUS delivered in a theta burst pattern) is a novel repetitive TUS protocol that can induce durable changes in motor cortex excitability, thereby holding promise as a

Multimodal assessment of the GABA system in patients with fragile-X syndrome and neurofibromatosis of type 1.

Fragile-X syndrome (FXS) and Neurofibromatosis of type 1 (NF-1) are two monogenic disorders sharing neurobehavioral symptoms and pathophysiological mechanisms. Namely, preclinical models of both conditions show overactivity of the mTOR signaling pathway as well as GABAergic alterations. However, despite its potential clinical relevance for these disorders, the GABAergic system has not been systematically studied in humans. In the present study, we used an extensive transcranial magnetic stimulation (TMS) assessment battery in combination with magnetic resonance spectroscopy (MRS) to provide a

Predicting brain organization with a computational model: 50-year perspective on lateral inhibition and oscillatory gating by dendrodendritic synapses.

The first compartmental computer models of brain neurons using the Rall method predicted novel and unexpected dendrodendritic interactions between mitral and granule cells in the olfactory bulb. We review the models from a 50-year perspective on the work that has challenged, supported, and extended the original proposal that these interactions mediate both lateral inhibition and oscillatory activity, essential steps in the neural basis of olfactory processing and perception. We highlight strategies behind the neurophysiological experiments and the Rall methods that enhance the ability of detai

Cortical neurophysiology of primary isolated dystonia and non-dystonic adults: A meta-analysis.

Transcranial magnetic stimulation (TMS) is a non-invasive method to assess neurophysiology of the primary motor cortex in humans. Dystonia is a poorly understood neurological movement disorder, often presenting in an idiopathic, isolated form across different parts of the body. The neurophysiological profile of isolated dystonia compared to healthy adults remains unclear. We conducted a systematic review with meta-analysis of neurophysiologic TMS measures in people with isolated dystonia to provide a synthesized understanding of cortical neurophysiology associated with isolated dystonia. We pe

Enhancing excitatory projections from the ventral subiculum to the nucleus accumbens shell contribute to the MK-801-induced impairment of prepulse inhibition.

Prepulse inhibition (PPI), a measure of sensorimotor gating, has been shown to be disrupted in several animal models of neuropsychiatric disorders, such as schizophrenia. The neural circuits involving the hippocampus and nucleus accumbens (NAC) have been studied in rats to uncover the neurochemical and neuroanatomical substrates that regulate PPI. Majority of the studies of the hippocampus on PPI to date have been focused on CA1, CA2, and dentate gyrus (DG) area. Little is known about the role of the subiculum, which maintains the hippocampal formation intact, on the sensorimotor gating. In th

Pain After Spinal Cord Injury Is Associated With Abnormal Presynaptic Inhibition in the Posterior Nucleus of the Thalamus.

UNLABELLED: Pain after spinal cord injury (SCI-Pain) is one of the most debilitating sequelae of spinal cord injury, characterized as relentless, excruciating pain that is largely refractory to treatments. Although it is generally agreed that SCI-Pain results from maladaptive plasticity in the pain processing pathway that includes the spinothalamic tract and somatosensory thalamus, the specific mechanisms underlying the development and maintenance of such pain are yet unclear. However, accumulating evidence suggests that SCI-Pain may be causally related to abnormal thalamic disinhibition, lead

Signatures of Somatic Inhibition and Dendritic Excitation in Auditory Brainstem Field Potentials.

Extracellular voltage recordings (Ve ; field potentials) provide an accessible view of in vivo neural activity, but proper interpretation of field potentials is a long-standing challenge. Computational modeling can aid in identifying neural generators of field potentials. In the auditory brainstem of cats, spatial patterns of sound-evoked Ve can resemble, strikingly, Ve generated by current dipoles. Previously, we developed a biophysically-based model of a binaural brainstem nucleus, the medial superior olive (MSO), that accounts qualitatively for observed dipole-like Ve patterns in sustained

Simultaneously Excitatory and Inhibitory Effects of Transcranial Alternating Current Stimulation Revealed Using Selective Pulse-Train Stimulation in the Rat Motor Cortex.

Transcranial alternating current stimulation (tACS) uses sinusoidal, subthreshold, electric fields to modulate cortical processing. Cortical processing depends on a fine balance between excitation and inhibition and tACS acts on both excitatory and inhibitory cortical neurons. Given this, it is not clear whether tACS should increase or decrease cortical excitability. We investigated this using transcranial current stimulation of the rat (all males) motor cortex consisting of a continuous subthreshold sine wave with short bursts of suprathreshold pulse-trains inserted at different phases to pro

Cortical inhibitory function in cervical dystonia.

OBJECTIVE: To assess the specificity of cortical inhibitory deficits in cervical dystonia patients. METHODS: A systematic test battery was developed to assess spatial and temporal aspects of cortical inhibition, in motor and somatosensory systems of the hand. We tested 17 cervical dystonia (CD) patients and 19 controls assessing somatosensory spatial inhibition (grating orientation test, interdigital feedforward subliminal inhibition), somatosensory temporal inhibition (temporal discrimination threshold, feedforward subliminal inhibition), motor spatial inhibition (surround inhibition), and m

Impaired synaptic plasticity in RASopathies: a mini-review.

Synaptic plasticity in the form of long-term potentiation (LTP) and long-term depression (LTD) is considered to be the neurophysiological correlate of learning and memory. Impairments are discussed to be one of the underlying pathophysiological mechanisms of developmental disorders. In so-called RASopathies [e.g., neurofibromatosis 1 (NF1)], neurocognitive impairments are frequent and are affected by components of the RAS pathway which lead to impairments in synaptic plasticity. Transcranial magnetic stimulation (TMS) provides a non-invasive method to investigate synaptic plasticity in humans.

Human occipital cortices differentially exert saccadic suppression: Intracranial recording in children.

By repeating saccades unconsciously, humans explore the surrounding world every day. Saccades inevitably move external visual images across the retina at high velocity; nonetheless, healthy humans don't perceive transient blurring of the visual scene during saccades. This perceptual stability is referred to as saccadic suppression. Functional suppression is believed to take place transiently in the visual systems, but it remains unknown how commonly or differentially the human occipital lobe activities are suppressed at the large-scale cortical network level. We determined the spatial-temporal

Analgesic effects of dyspnoea: "Air hunger" does not inhibit the spinal nociception reflex in humans.

Dyspnoea has distinct sensory modalities, including air hunger and the sensation of excessive breathing "work/effort". Both have analgesic properties. In the case of work/effort, spinal mechanisms have been documented (inhibitory effect on the spinal nociceptive flexor reflex, RIII). This mechanism involves C-fibres. As C-fibres are unlikely to play a major role in air hunger, we hypothesised that inducing this type of dyspnoea would not result in RIII inhibition. Eight healthy volunteers were exposed to a hypercapnic hyperoxic gas mixture (5% CO2 and 95% O2) and asked to voluntarily fight the

Covert movements trigger repetition suppression of electroencephalography in sensorimotor cortex.

'Repetition suppression' (RS) denotes the decrease of neural responses to repeated external sensory stimuli. We showed that RS can be also triggered by internal processes alone. When individuals perform repetitive covert movements, that is, motor imagery or quasi-movements, both of which are associated with pericentral cortical activity without muscle activations, there was a significant recovery of electroencephalographic oscillations over sensorimotor cortices back to resting baseline level. After 58 s of task performance only 20% of α and 5% of β suppressions remained (overt movements: 34%
