Biomimetic Macrophage Cell Membrane-Based Nanoparticles for Effective Treatment of Glioblastoma Through Boron Neutron Capture Therapy Combined With Immunotherapy.

Glioblastoma multiforme (GBM) remains largely incurable due to the blood-brain barrier (BBB) and immunosuppressive microenvironment. While boron neutron capture therapy (BNCT) selectively eradicates tumor cells via 10B(n, α)-7Li reactions, its clinical potential in GBM is unrealized because of the suboptimal pharmacokinetics of conventional boron agents and their failure to achieve therapeutic intracranial concentrations. Here, we design macrophage membrane-coated PLGA nanoparticles (M@PLGA-10BN) that exploit innate biological transport mechanisms to overcome dual therapeutic barriers. The mac

Identification of a key water molecule involved in the macrophage migration inhibitory factor-catalyzed tautomerization of para-hydroxyphenylpyruvate using neutron crystallography.

Neutron crystallography was used to determine a 2.5-Å resolution all-atom structure of macrophage migration inhibitory factor (MIF) interacting with 3-(4-hydroxyphenyl)-pyruvate (HPP). MIF is a pro-inflammatory, pro-tumorigenic protein that may be an attractive therapeutic target. MIF catalyzes the interconversion of the keto and enol forms of HPP by a tautomerase reaction. Although HPP is evidently not a physiological substrate of MIF, many compounds that inhibit this activity in enzymatic assays have been found also to inhibit physiological activities of MIF. Therefore, the MIF-catalyzed HPP

Incorporating boron distribution variations in microdosimetric kinetic model-based relative biological effectiveness calculations for boron neutron capture therapy.

This study introduces the MKM_B model, an approach derived from the MKM model, designed to evaluate the biological effectiveness of Boron Neutron Capture Therapy (BNCT) in the face of challenges from varying microscopic boron distributions. The model introduces a boron compensation factor, allowing for the assessment of compound Biological Effectiveness (CBE) values for different boron distributions. Utilizing the TOPAS simulation platform, the lineal energy spectrum of particles in BNCT was simulated, and the sensitivity of the MKM_B model to parameter variations and the influence of cell siz

Synthesis of Gd-DTPA Carborane-Containing Compound and Its Immobilization on Iron Oxide Nanoparticles for Potential Application in Neutron Capture Therapy.

Cancer is one of the leading causes of global mortality, and its incidence is increasing annually. Neutron capture therapy (NCT) is a unique anticancer modality capable of selectively eliminating tumor cells within normal tissues. The development of accelerator-based, clinically mountable neutron sources has stimulated a worldwide search for new, more effective compounds for NCT. We synthesized magnetic iron oxide nanoparticles (NPs) that concurrently incorporate boron and gadolinium, potentially enhancing the effectiveness of NCT. These magnetic nanoparticles underwent sequential modification

Profile analysis of adverse events after boron neutron capture therapy for head and neck cancer: a sub-analysis of the JHN002 study.

The purpose of this study was to outline the course and profile of adverse events specific to boron neutron capture therapy (BNCT) for head and neck cancer. This was a sub-analysis of the phase II JHN002 trial. Patients received 400 mg/kg borofalan(10B), followed by neutron irradiation. The course of adverse events after BNCT was documented in the JHN002 Look Up study. Patients were grouped into face/front (FF), face/lateral (FL) and neck (N) beam groups according to the point of skin incidence of the epithermal neutron beam axis, and the profile of adverse events dependent on beam incidence p

Proton transfer and drug binding details revealed in neutron diffraction studies of wild-type and drug resistant HIV-1 protease.

HIV-1 protease is an essential therapeutic target for the design and development of antiviral inhibitors to treat AIDS. We used room temperature neutron crystallography to accurately determine hydrogen atom positions in several protease complexes with clinical drugs, amprenavir and darunavir. Hydrogen bonding interactions were carefully mapped to provide an unprecedented picture of drug binding to the protease target. We demonstrate that hydrogen atom positions within the enzyme catalytic site can be altered by introducing drug resistant mutations and by protonating surface residues that trigg

Bone Marrow Endothelial Cells Influence Function and Phenotype of Hematopoietic Stem and Progenitor Cells after Mixed Neutron/Gamma Radiation.

The bone marrow (BM) microenvironment plays a crucial role in the maintenance and regeneration of hematopoietic stem (HSC) and progenitor cells (HSPC). In particular, the vascular niche is responsible for regulating HSC maintenance, differentiation, and migration of cells in and out of the BM. Damage to this niche upon exposure to ionizing radiation, whether accidental or as a result of therapy, can contribute to delays in HSC recovery and/or function. The ability of BM derived-endothelial cells (BMEC) to alter and/or protect HSPC after exposure to ionizing radiation was investigated. Our data

Experimental yield and evaluation of proton induced reactions for neutron production and synthesis of beryllium-7 using lithium compounds as target material.

We had investigated the nuclear processes occur during nuclear reactions of protons with lithium containing targets. Experimental measurements were undertaken to obtain the thick-target yield of 7Be and infer upon the neutron production. Three candidate target materials having the chemical compositions LiF, LiBO2, and Li2B4O7 were used. The 7Be yield was probed using its 477 keV day γ-line while the neutron yield was probed using 63Cu(n,γ)64Cu. The 7Be and neutron yields were found different among the different compositions. Discussion on the details of Li (p,n) reactions and decay of exit cha

Determination of relative krypton fission product yields from 14 MeV neutron induced fission of 238U at the National Ignition Facility.

Precisely-known fission yield distributions are needed to determine a fissioning isotope and the incident neutron energy in nuclear security applications. 14 MeV neutrons from DT fusion at the National Ignition Facility induce fission in depleted uranium contained in the target assembly hohlraum. The fission yields of Kr isotopes (85m, 87, 88, and 89) are measured relative to the cumulative yield of 88Kr and compared to previously tabulated values. The results from this experiment and England and Rider are in agreement, except for the 85mKr/88Kr ratio, which may be the result of incorrect nucl…

Growth, shrinking, and breaking of pluronic micelles in the presence of drugs and/or beta-cyclodextrin, a study by small-angle neutron scattering and fluorescence spectroscopy.

The associative structures between F127 Pluronic micelles and four drugs, namely, lidocaine (LD), pentobarbital sodium salt (PB), sodium naproxen (NP), and sodium salicylate (SAL), were studied by small-angle neutron scattering (SANS). Different outcomes for the micellar aggregates are observed, which are dependent on the chemical nature of the drug and the presence of charge or otherwise: the micelles grow with LD, are hardly modified with PB, and decrease in size with both NP and SAL. The partition coefficient, determined by fluorescence spectroscopy, is directly correlated to the amount of

[Prophylaxis of early radiation injuries to intact tissues following exposure to combined photon-neutron radiotherapy for malignant head and neck tumors].

Our data are presented on comparison of early- and late-response and complications in 320 patients with head and neck tumors after combined photon-neutron radiotherapy (CPNT) alone or in conjunction with cisplatin. Laser therapy via the LGN-222 quartz installation alternating with irradiation sessions was used to prevent and palliate radiation injuries. As a result, the incidence of acute (grade I-II) reactions was significantly lowered and severe fast-neutron-induced complications were avoided. Grade I (RTOG) skin and mucous membrane injuries were rarer in the main group receiving laser thera

Effect of monomeric and polymeric co-solutes on cetyltrimethylammonium bromide wormlike micelles: rheology, Cryo-TEM and small-angle neutron scattering.

The effect of hydrophobic and hydrophilic co-solutes on the rheological properties of wormlike micelles of cetyltrimethylammonium bromide (CTAB) and sodium salicylate (NaSal) or sodium bromide (NaBr) was investigated. Monomeric (ethanol, 2-propanol, benzene and benzylic alcohol) and polymeric species (poly(ethylene oxide), poly(vinyl alcohol) and poly(propylene oxide), respectively PEO, PVA and PPO) of varying molecular weight were studied in order to assess the effect of co-solute 'length' on the interactions with the wormlike micelles. Rheological properties were characterised by the plateau

Deranged Ca, Al and Mg content in the tissues of patients with chronic renal failure, as measured by non-destructive neutron activation analysis.

Ca, Al and Mg content in the bone, aorta, skin and hair of 55 persons was measured by non-destructive neutron activation analysis. The Ca content in the bone of nondialyzed and dialyzed patients was found to decrease. Al and Mg were relatively higher than Ca in the patients compared to the controls. The present study suggests that fluctuation in the content of Al and Mg may have some relation to renal osteodystrophy such as osteomalacia as well as similar and related disorders. Al and Mg content in the aorta, and that of Ca in the hair of uremic patients, were higher than the controls. The mea

Whole-body elemental composition in patients with renal failure and after transplantation studied using total-body neutron-activation analysis.

Whole body elemental composition in seventy-five patients with either renal failure or a kidney transplant was measured by neutron-activation analysis and whole-body counting. Comparison was made with expected normal values and with a control group; between undialysed, dialysed and transplanted patients; and between those receiving dialysis treatment for various periods. A frequent finding was low body calcium, reflecting the prevalence of renal osteodystrophy. Sodium and chlorine were increased in undialysed patients, a consequence of their renal disease. Patients with transplanted kidneys ha

Application of solid phase extractants TEVA and TK200 for the separation of medically relevant 199Au from thermal neutron irradiated Pt target.

This study describes the separation of no-carrier-added (n.c.a.) 199Au from thermal neutron irradiated natural platinum targets using two extraction chromatographic resins: TEVA containing a quaternary amine (Aliquat 336) and TK200 impregnated with trioctylphosphine oxide (TOPO, Cyanex 921). The behavior of Au, Pt, and Ir in HCl media (0.1-10 mol·L-1 HCl) is examined through batch distribution coefficient (Kd) measurements and dynamic column experiments. For TEVA, the Kd of Au remains high across the entire HCl range (2-3·104 mL·g-1), whereas Pt shows a maximum Kd of 3.3·104 mL·g-1 at 1 mol·L-

The Relation between Colloid Size and Nanoscale Water Transport Pathways in Waterborne Acrylic Coatings as Seen by Small-Angle Neutron Scattering.

Waterborne and water-reduced coatings are increasing in relevance in many sectors as an alternative to solventborne coatings. In this work, the internal structure of waterborne polymers as a function of colloid particle size is unveiled and directly related to macroscopic water absorption. To this aim, a set of acrylic waterborne films was prepared from dispersions of different colloidal particle sizes (100, 150, and 200 nm) with the same surfactant coverage. Macroscopic water absorption and water affinity were studied by Dynamic vapor sorption (DVS) and immersion tests. Small-Angle Neutron Sc

A Porphyrinic Covalent Organic Polymer Nanoplatform for Carborane Delivery and Multifunctional Imaging-Guided Boron Neutron Capture Therapy of Breast Cancer.

INTRODUCTION: The advancement of boron neutron capture therapy (BNCT) is critically dependent on the development of efficient and traceable boron delivery agents. METHODS: We designed and synthesized a porphyrin-based covalent organic polymer (T-COP) loaded with ortho-carborane (o-CB) and surface-functionalized with DSPE-PEG2000, successfully constructing a novel nanoplatform (T-COP@o-CB NPs). This platform enables real-time imaging-guided boron neutron capture therapy (BNCT), providing a new strategy for the treatment of breast cancer. RESULTS: The resulting nanocomposite exhibits a high bo

Sensitivity analysis of neutron and photon equivalent doses in patient organs during 3D conformal radiotherapy using Monte Carlo simulations.

This study evaluates neutron and photon equivalent dose sensitivity in patient organs during prostate three-dimensional conformal radiotherapy (3DCRT) using Monte Carlo simulations. A validated Varian Clinac 2100C LINAC model and a hybrid XCAT adult male phantom were used with an 18 MV photon beam. Component-wise sensitivity analysis showed that removal of the flattening filter caused the largest increase in neutron doses, with ratios of 1.49 in the testes, 1.39 in the bladder, and 1.38 in the colon, indicating a dominant shielding role. The primary collimator also showed significant shielding

Whole-brain spatial metabolomics reveals metabolic gradient shifts in a murine glioma model following boron neutron capture therapy.

BACKGROUND: Glioblastoma (GBM) is the most aggressive and lethal primary brain tumor, characterized by poor prognosis and resistance to current therapies. Boron neutron capture therapy (BNCT) is a tumor-selective particle radiation therapy in which high-linear energy transfer (LET) α-particles and recoil 7Li nuclei are generated through the capture reaction between 10B and thermal neutrons. Although BNCT has been shown to prolong survival in GBM, its effects on global metabolomic alterations of the whole brain, particularly in relation to the tumor microenvironment (TME), remain incompletely u…
