Analysis of N(ε) -ethyllysine in human plasma proteins by gas chromatography-negative ion chemical ionization/mass spectrometry as a biomarker for exposure to acetaldehyde and alcohol.

BACKGROUND: N(ε) -ethyllysine (NEL) is a major stable adduct formed by the reaction of acetaldehyde (AA) with lysine residues in proteins. However, its occurrence and levels in biological specimens and its relationship with AA/alcohol exposure-associated disorders have not been fully elucidated. In this study, we have developed a sensitive and specific method to quantitate NEL levels in human plasma proteins. METHODS: The method consists of (1) purification of the protein fraction of interest by Sephadex G-15 to remove low molecular substances, (2) hydrolysis of proteins with Pronase E in the

Microecological characteristics of water bodies/sediments and microbial remediation strategies after 50 years of pollution exposure in ammunition destruction sites in China.

The effects of long-term ammunition pollution on microecological characteristics were analyzed to formulate microbial remediation strategies. Specifically, the response of enzyme systems, N/O stable isotopes, ion networks, and microbial community structure/function levels were analyzed in long-term (50 years) ammunition-contaminated water/sediments from a contamination site, and a compound bacterial agent capable of efficiently degrading trinitrotoluene (TNT) while tolerating many heavy metals was selected to remediate the ammunition-contaminated soil. The basic physical and chemical propertie

Plant N capture from pulses: effects of pulse size, growth rate, and other soil resources.

In arid ecosystems, the ability to rapidly capture nitrogen (N) from brief pulses is expected to influence plant growth, survival, and competitive ability. Theory and data suggest that N capture from pulses should depend on plant growth rate and availability of other limiting resources. Theory also predicts trade-offs in plant stress tolerance and ability to capture N from different size pulses. We injected K15NO3, to simulate small and large N pulses at three different times during the growing season into soil around the co-dominant Great Basin species Sarcobatus vermiculatus, Chrysothamnus n

Tissue content of thiol-containing amino acids predicts methylmercury in aquatic invertebrates.

Aquatic invertebrates vary in methylmercury (MeHg) levels among systems which has been attributed, in part, to environmental conditions, but may also be linked to differences in their biochemical composition. As MeHg is known to bind to thiol-containing amino acids such as cysteine in proteins of fish, our objective was to determine if these amino acids explain MeHg variability among aquatic invertebrate taxa. Benthic macroinvertebrates from diverse functional feeding groups and bulk zooplankton were collected from six acidic lakes in Kejimkujik National Park, Nova Scotia, Canada, and analyzed

Nitrogen-Tuned Cationic Au Sites on TiO2 Enable Selective Methane Oxidation to Methanol.

Selective oxidation of methane to methanol under mild conditions remains a long-standing challenge because of the high inertness of the C-H bond in methane and the susceptibility of methanol to overoxidation. Here, we report a nitrogen-incorporation strategy for constructing TiO2-supported Au catalysts with cationic Au sites for selective methane oxidation using O2 as the oxidant in aqueous CH4/O2/CO media. The optimized Au/N-TiO2 catalyst delivers a methanol productivity of 3956 μmol gcat-1 h-1, corresponding to 177 mol molAu-1 h-1, with 98% selectivity at 180 °C, demonstrating superior catal

Detailing Atmospheric Nitrate Chemistry in the Northeastern US Under a Period of Changes in Emissions.

The successful implementation of the US Clean Air Act has reduced nitrogen oxides (NOx) emissions, with important improvements in air quality and acid deposition. However, predicting the impact of emission reductions on tropospheric chemistry remains challenging, as NOx influences oxidant concentrations and aerosol formation. Stable oxygen isotopes (Δ17O and δ18O) of atmospheric nitrate (pNO3) serve as powerful observational constraints for the understanding of oxidation processes. Here, we quantified Δ17O and δ18O in pNO3 from a unique 10-year aerosol record in the northeastern US to constrai

Identification of sources and bioaccumulation pathways of MeHg in subantarctic penguins: a stable isotopic investigation.

Seabirds are widely used as bioindicators of mercury (Hg) contamination in marine ecosystems and the investigation of their foraging strategies is of key importance to better understand methylmercury (MeHg) exposure pathways and environmental sources within the different ecosystems. Here we report stable isotopic composition for both Hg mass-dependent (e.g. δ202Hg) and mass-independent (e.g. Δ199Hg) fractionation (proxies of Hg sources and transformations), carbon (δ13C, proxy of foraging habitat) and nitrogen (δ15N, proxy of trophic position) in blood of four species of sympatric penguins bre

Extraction of CO2 from air samples for isotopic analysis and limits to ultra high precision delta18O determination in CO2 gas.

The determination of delta18O values in CO2 at a precision level of +/-0.02 per thousand (delta-notation) has always been a challenging, if not impossible, analytical task. Here, we demonstrate that beyond the usually assumed major cause of uncertainty - water contamination - there are other, hitherto underestimated sources of contamination and processes which can alter the oxygen isotope composition of CO2. Active surfaces in the preparation line with which CO2 comes into contact, as well as traces of air in the sample, can alter the apparent delta18O value both temporarily and permanently. W

Quantitative apportionment of multiple water sources and their implications for coastal nutrient dynamics: A mult-tracer framework of river-dominated system.

Quantifying water source contributions and their environmental impacts is essential for managing water quality in river-dominated coastal systems and sustainable socio-economic development, yet remains methodologically challenging. This study integrated stable isotopes (δD, δ¹⁸O) and rare earth elements within an Optimum Multi-Parameter analysis to resolve source contributions (precipitation, river, shelf, and porewater) in the Changjiang (the world's third-largest river), Jiaojiang, and Oujiang estuaries and the adjacent coastal area. Results delineate distinct source roles. The Changjiang Di

Changes in atmospheric nitrate deposition in Germany--an isotopic perspective.

We investigated the isotopic composition of atmospheric NO3(-) deposition at a moderately polluted site in Western Europe over an annual cycle from December 2011 to November 2012. On average, we measured load-weighted δ(15)N values of +0.1 and +3.0‰ in wet and dry deposition, respectively. A comparison to source-specific N emission trends and to isotope data from the 1980s reveals distinct changes in δ(15)N-NO3(-) values: In contrast to the increasing relative importance of isotopically depleted natural NOx sources, we find an increase of isotope values in comparison to historical data. We exp

Mass spectrometry quantification revealed accumulation of C-terminal fragment of apolipoprotein E in the Alzheimer's frontal cortex.

Polymorphic variation in the apolipoprotein E (apoE) gene is the major genetic susceptibility factor for late-onset Alzheimer's disease (AD) and likely contributes to neuropathology through various pathways. It is also recognized that apoE undergoes proteolytic cleavage in the brain and the resultant apoE fragments likely have a variety of bioactive properties that regulate neuronal signaling and may promote neurodegeneration. ApoE fragmentation in the human brain has been intensively studied using different immunochemical methods, but has never been analyzed in a quantitative manner to establ

Study of ammonia metabolism in a patient with ornithine transcarbamylase deficiency using an 15N tracer.

Ammonia metabolism was studied in an 8-year-old girl with ornithine transcarbamylase (OTC) deficiency, using 15N-tracer. Changes in the incorporation of 15N into amino acids and urea were examined after 15NH4Cl administration. The recovery of total 15N in the urine of the patient in 3 days was 28.5% of the administered 15N whereas that of a control was 69.3%. They were mostly urea. The recovery of 15N-urea in the patient was 28.8% of the control in 1 day, 32% in 2 days and 33.3% in 3 days after the administration of 15NH4Cl. A larger amount of 15N was incorporated into glutamine (alpha-amino N

1H, 13C, and 15N backbone and side-chain resonance assignments of the Nostoc sp. H-NOX C139A variant in complex with the soluble Guanylyl Cyclase inhibitor Zinc Protoporphyrin IX.

The H-NOX (Heme-Nitric oxide/Oxygen binding) domain is highly conserved among both eukaryotes and bacteria. In human soluble Guanylyl Cyclase (sGC), the H-NOX domain functions as a sensor for the gaseous signaling molecule Nitric Oxide (NO). Located at the N-terminus of the enzyme, the heme B-binding H-NOX domain regulates the catalytic domain at the C-terminus, which catalyzes the conversion of GTP (guanosine 5'-triphosphate) to cGMP (cyclic Guanosine MonoPhosphate). The present study employs the bacterial homolog Nostoc sp. H-NOX (Ns H-NOX) C139A mutant together with the sGC inhibitor Zinc P

Putative anaerobic transformation pathway of microcystin-RR inferred from 15N labeling and multi-omics in an enriched shrimp pond sediment microbial community.

The degradation mechanism of MC-RR by an anaerobic degrading microbial community (ADMC), enriched from shrimp pond sediment, was investigated using LC-MS/MS, metagenomic, and metatranscriptomic analyses. Three key degradation products of 15N-labeled MC-RR were tentatively identified: Adda-Glu-Mdha-Ala (m/z 618.3216), a deamination product (m/z 600.2965), and Glu-MeAsp-Ala-Arg-CO2 (m/z 466.2233). The pathway was inferred to involve hydrolytic ring-opening at Arg-Adda or Ala-Arg bonds, followed by deamination, decarboxylation, and stepwise degradation into short peptides and amino acids. Metagen

Effects of gastric bypass on the digestibility and postprandial metabolic fate of 15N dietary protein in rats.

Roux-en-Y Gastric Bypass may be associated with an alteration of protein bioavailability in relation to intestinal remodeling. Our study aimed to test this hypothesis by Roux-en-Y Gastric Bypass. Diet-induced obese rats underwent Roux-en-Y Gastric Bypass surgery (RYGB rats) while a Sham-operated control group was used. All rats received a 15N-labeled protein meal 1 or 3 months after surgery and were euthanized 6h later. Protein digestibility, 15N recovered in organs and urea pool, fractional protein synthesis rate, and intestinal morphometry were assessed. Protein digestibility was similar in

1H, 15N and 13C backbone resonance assignment of the N-terminal region of Zika virus NS4B protein in detergent micelles.

Zika virus has raised global concerns due to its link to microcephaly and Guillain-Barré syndrome in adults. One of viral nonstructural proteins-NS4B, an integral membrane protein, plays crucial roles in viral replication by interacting with both viral and host proteins, rendering it an attractive drug target for antiviral development. We purified the N-terminal region of ZIKV NS4B (NS4B NTD) and reconstituted it into detergent micelles. Here, we report the assignments of the backbone resonances of NS4B NTD in detergent micelles. The available assignment is useful for understanding its structu

1H, 13C, 15N backbone resonance assignment for the 1-164 construct of human XRCC4.

DNA double-strand breaks (DSBs) represent the most cytotoxic DNA lesions, as-if mis- or unrepaired-they can cause cell death or lead to genome instability, which in turn can cause cancer. DSBs are repaired by two major pathways termed homologous recombination and non-homologous end-joining (NHEJ). NHEJ is responsible for repairing the vast majority of DSBs arising in human cells. Defects in NHEJ factors are also associated with microcephaly, primordial dwarfism and immune deficiencies. One of the key proteins important for mediating NHEJ is XRCC4. XRCC4 is a dimer, with the dimer interface med

Biomagnification of mercury in aquatic food webs: a worldwide meta-analysis.

The slope of the simple linear regression between log10 transformed mercury (Hg) concentration and stable nitrogen isotope values (δ(15)N), hereafter called trophic magnification slope (TMS), from several trophic levels in a food web can represent the overall degree of Hg biomagnification. We compiled data from 69 studies that determined total Hg (THg) or methyl Hg (MeHg) TMS values in 205 aquatic food webs worldwide. Hg TMS values were compared against physicochemical and biological factors hypothesized to affect Hg biomagnification in aquatic systems. Food webs ranged across 1.7 ± 0.7 (mean
