Expression of autophagy‑associated proteins in rat dental irreversible pulpitis.

Autophagy serves an important role in numerous diseases, as well as in infection and inflammation. Irreversible pulpitis (IP) is one of the … questosome 1 were decreased. In addition, the levels of ATG proteins were specifically increased in odontoblasts and microvascular endothelial cells in pulpitis tissue. Based on these findings, autophagy may serve an important role in IP, and the present study data provide a new insight into the IP pathogenesis and treatment.

Molecular genetics of ameloblast cell lineage.

Late tooth morphogenesis is characterized by a series of events that determine crown morphogenesis and the histodifferentiation of epithelial cells into enamel-secreting ameloblasts and of mesenchymal cells into dentin-secreting odontoblasts. Functional ameloblasts are tall, columnar, polarized cells that synthesize and secrete a number of enamel-specific proteins. After depositing the full thickness of enamel matrix, ameloblasts shrink in size and regulate enamel maturation. Amelogenesis imperfecta (AI) is a heterogeneous group of inherited defects in enamel formation. Clinically, AI presents…

The zinc transporter SLC39A13/ZIP13 is required for connective tissue development; its involvement in BMP/TGF-beta signaling pathways.

BACKGROUND: Zinc (Zn) is an essential trace element and it is abundant in connective tissues, however biological roles of Zn and its transporters in those tissues and cells remain unknown. METHODOLOGY/PRINCIPAL FINDINGS: Here we report that mice deficient in Zn transporter Slc39a13/Zip13 show changes in bone, teeth and connective tissue reminiscent of the clinical spectrum of human Ehlers-Danlos syndrome (EDS). The Slc39a13 knockout (Slc39a13-KO) mice show defects in the maturation of osteoblasts, chondrocytes, odontoblasts, and fibroblasts. In the corresponding tissues and cells, impairment …

MEPE/OF45, a new dentin/bone matrix protein and candidate gene for dentin diseases mapping to chromosome 4q21.

Substantial progress has been made regarding the molecular etiology of the dentin diseases dentinogenesis imperfecta types II and III, and d … the human gene. For the first time, MEPE/OF45 expression was shown in dental tissue, in particular odontoblasts, by reverse-transcription polymerase chain reaction (RT-PCR) amplification and characterization of a partial mouse cDNA. Our data provide the first evidence that MEPE/OF45 is expressed during odontogenesis and should be considered as a candidate gen …

Odontoclastic resorption at the pulpal surface of coronal dentin prior to the shedding of human deciduous teeth.

Histological and histochemical observations of more than 150 extracted human deciduous teeth revealed that, prior to shedding, odontoclastic … resorption neared completion, inflammatory cells started to gradually infiltrate into the pulp, and odontoblasts began to degenerate. After that, multinucleate odontoclasts appeared, and resorption proceeded from the predentin to the dentin. The odontoclastic activity was initially detected only on the pulpal surface at the bottom areas of the crown. It gradua …

Distribution patterns of DNA template activity in the embryonic tooth organ: an acridine orange ultracytochemical study.

The ultracytochemical acridine orange (AO) method has been employed to demonstrate DNA template activity within embryonic incisor tooth organs excised from New Zealand White rabbits during the 25th day of gestation. Survey ultrastructural examination revealed characteristic distribution patterns of AO positive cell populations in the inner and outer enamel epithelium and the adjacent ectomesenchyme of the cervical loop region and in populations of fibroblast-like cells connecting as a band the outer AO positive cell groups. With increasing differentiation of ameloblasts and odontoblasts the nu…

Redefining the void: A novel classification for internal resorption in human teeth.

INTRODUCTION: Internal root resorption is the loss of hard tissue of the tooth from the pulpal surface of the root canal space. This pathologic condition is characterized by damage to the organic sheath, loss of predentin, and odontoblastic layer leading to demineralization of hard tissue of the tooth. CONTEXT: Internal root resorption. AIMS: The aim of this article is to classify internal resorptive lesions to distinguish the extent of resorption at various levels as seen in the tooth. SUBJECTS AND METHODS: Internal resorption is less commonly diagnosed as compared to external resorption.

Development of craniofacial structures in transgenic mice with constitutively active PTH/PTHrP receptor.

Parathyroid hormone (PTH) and parathyroid hormone-related peptide (PTHrP) regulate calcium homeostasis, and PTHrP further regulates growth and development. A transgenic mouse carrying the constitutively active PTH/PTHrP receptor (HKrk-H223R) under the control of the mouse bone and odontoblast-specific alpha1(I) collagen promoter (Col1-caPPR) has been developed to demonstrate the complex actions of this mutant receptor in hard tissue formation. We have further characterized Col1-caPPR mice abnormalities in the craniofacial region as a function of development. Col1-caPPR mice exhibited a delay i

Pulpal evaluation of two adhesive systems in rat teeth.

Formocresol is the agent most frequently used with pulpotomies in primary teeth, but its use is currently under discussion in regard to its possible toxic effects. Some current works suggest the use of dentinal adhesives in cavities with minimal dentinal thickness and even in small pulpal exposures. Our objective was to evaluate pulpal response to the application of two dentinal adhesives Syntac and Prime & Bond NT in pulpal exposures in rat teeth. Sprague Dawley rats were used, male, 150 to 200 gm in weight. The maxillary first molar, left and right, was employed in each case, a total of twel

Factors regulating and modifying dental root resorption.

A comparison is made between the resorption of bone and the resorption of the mineralized tissues of teeth. The structure and function of osteoclasts are described well as the factors that regulate their activity. The cells resorbing the dental mineralized tissues are of the same cell type as osteoclasts. The dental tissues are covered by cementoblasts or odontoblasts which differ from the osteoblasts in that they do not respond to hormones and cytokines that stimulate bone resorption. Root resorption therefore seem to require damage of the cementoblastic layer in combination with necrosis or …

Notum regulates the cusp and root patterns in mouse molar.

Notum is a direct target of Wnt/β-catenin signaling and plays a crucial role as a Wnt inhibitor within a negative feedback loop. In the tooth, Notum is known to be expressed in odontoblasts, and severe dentin defects and irregular tooth roots have been reported in Notum-deficient mice. However, the precise expression pattern of Notum in early tooth development, and the role of Notum in crown and root patterns remain elusive. In the present study, we identified a novel Notum expression in primary enamel knot (EK), secondary EKs, and dental papilla during tooth development. Notum-deficient mice …

A spontaneous compound odontoma in an adult Sprague Dawley rat (Rattus norvegicus).

Reports of compound odontomas in rats are very rare. A 14-month-old adult male Sprague Dawley rat was found to have a hard mass associated w … embedded in ossified stroma. Each proto-tooth had a central mesenchyme pulp surrounded by columnar odontoblasts and dentine matrix. The dentine was often bordered by enamel matrix, which was occasionally bounded by ameloblasts. These histological findings were consistent with a compound odontoma. This is the first report of a spontaneous compound odontoma in …

Extracellular matrix degradation by host matrix metalloproteinases in restorative dentistry and endodontics: An overview.

Matrix metalloproteinases (MMPs) are a group of over 25 secreted and membrane-bound enzymes responsible for pericellular substrate degeneration. In response to injury, they play key roles in morphogenesis, wound healing, tissue repair and remodeling. They have been isolated from dentin, odontoblasts, pulp and periapical tissue. They play a major role in the formation of dentin matrix and secondary and tertiary dentin. These are also responsible for releasing dentinal growth factors. MMP family proteins elicit a dual role in the pathogenesis of inflammation, stimulating protective innate and/or…

Cytotoxicity assay using a human pluripotent stem cell-derived cranial neural crest cell model.

Cleft lip and palate are the most common congenital abnormalities that occur early in pregnancy. The majority of cranial mesenchyme is derived from cranial neural crest cells that differentiate into odontoblasts, cartilage, craniofacial bone, and connective tissue. A subset of these cells differentiates into cranial ganglia. We have previously reported an induction protocol of cranial neural crest cell-like cells from human pluripotent stem cells. This study tested detection of the cytotoxic sensitivities of dental materials, including titanium ions, palladium ions, and hydroxyethyl methacryla…
