Small-molecule degraders for oncogenic KRASG12C and pan-KRAS mutations.

KRAS, a frequently mutated oncogene, has been challenging to target therapeutically. Although covalent inhibitors like sotorasib against KRASG12C have been developed, their efficacy is often limited by acquired resistance. Targeted protein degradation offers a potential solution but has largely relied on large PROTAC molecules. Here, we report DJX-A-KM, a small-molecule degrader of KRASG12C, designed by incorporating an acrylamide warhead into the MRTX849 scaffold. It induces potent and sustained degradation of KRASG12C in cells and in vivo. Mechanistic investigation reveal that degradation is

Flawed translation triggers oncogenic B-T cell communication.

B-T cell cross talk is safeguarded by central and peripheral tolerance and cell-intrinsic checkpoints. In this issue of JEM, Lin et al. (https://doi.org/10.1084/jem.20251968) show that altered translation in early germinal center B cells triggers a vicious cycle of aberrant B-T cell interactions culminating in lymphomagenesis.

Pancancer Analysis and the Oncogenic Role of UBTF in Breast Invasive Carcinoma.

Upstream binding transcription factor (UBTF) is a nuclear transcription factor implicated in ribosome biogenesis, yet its pancancer relevance and immunological associations remain incompletely understood. We integrated datasets from The Cancer Genome Atlas (TCGA), Genotype-Tissue Expression (GTEx), Human Protein Atlas (HPA), Cancer Cell Line Encyclopedia (CCLE), and cBioPortal databases to characterize UBTF expression, genomic alterations, and prognostic value across 33 cancer types. Immune microenvironment analyses were performed using ESTIMATE and multiple deconvolution algorithms. CRISPR-Ca

Oncogene activation mechanism determines the limits of targeted protein degradation.

Protein degrader drugs such as PROTACs are being advanced as therapeutics targeted against oncogenic proteins. During tumorigenesis, oncogenic proteins can become constitutively activated via mechanisms including gene amplification, which increases protein production, and point mutations, which can extend protein half-life. Few experimental studies have addressed how disease-associated changes in target protein homeostasis influence PROTAC activity. We developed orthogonal methods to increase production or enhance stability of β-catenin, an important oncoprotein and target for degrader therape

Phosphorylation Protects Oncogenic RAS from LZTR1-Mediated Degradation.

Oncogenic KRAS and NRAS mutations are common in hematologic malignancies, but their signaling in this context remains less well characterized than in carcinomas. Using multi-omics screens in multiple myeloma, we sought to identify regulators of RAS activity. We found that the phosphatase PP1C dephosphorylated conserved RAS residue T148, permitting LZTR1-dependent proteasomal degradation. LZTR1 was ineffective against KRAS A146 gain-of-function mutations, which lie adjacent to T148 and are enriched in hematologic cancers, such as diffuse large B cell lymphoma and acute myeloid leukemia. FANCM is required for the PAX3::FOXO1-driven oncogenic program in rhabdomyosarcoma.

Many cancers are driven by mutationally altered transcription factors (TFs) that rewire cells to an oncogenic state. Cells must activate spe … ains as essential for this dependency, linking FANCM-mediated replication fork binding to sustained oncogenesis.

Redox guardian or oncogenic driver? The dual roles of PRDX1 in tumorigenesis.

Peroxiredoxin 1 (PRDX1) is a thiol-dependent peroxidase with paradoxical roles in cancer. In response to oxidative stress and hyperoxidation of its peroxidatic cysteine, PRDX1 undergoes reversible oligomerization, switching from a peroxidase to a molecular chaperone, thereby dictating context-dependent tumor effects. In early malignancy, PRDX1 suppresses transformation by limiting oxidative DNA damage and inhibiting oncogenic signaling (c-Myc, PTEN/AKT). However, PRDX1 overexpression in advanced cancers promotes survival, proliferation, and chemoresistance via MAPK modulation, DNA repair enhan

The oncogenic potential of YAP requires Y357 phosphorylation in cholangiocytes but not in hepatocytes.

PURPOSE: The transcriptional cofactor yes-associated protein (YAP) is activated in primary liver cancers (PLCs). YAP is canonically inhibited through serine 127 (S127) phosphorylation via the Hippo pathway but activated by phosphorylation at tyrosine 357 (Y357) by SRC family kinases. Due to liver plasticity, PLCs can originate from different cell types. However, it is unknown whether YAP activation favors tumorigenesis from specific cells of origin or promotes lineage commitment during tumorigenesis. This study investigates how YAP Y357 phosphorylation influences tumor phenotype and cell of or

RIT1 Drives Oncogenic Transformation and Is an Actionable Target in Lung Adenocarcinoma.

UNLABELLED: RIT1 is a small GTPase of the RAS family, and RIT1 mutations have been identified in lung cancer, leukemia, and the developmental disorder Noonan syndrome. Mutations in RIT1 lead to increased protein levels due to impaired proteolysis, resulting in dysregulation of RAS/MAPK signaling and other pathways. In this study, we documented the diversity of RIT1 mutations in human lung cancer and showed that physiologic expression of RIT1 M90I is sufficient to drive autochthonous lung tumor development in vivo in mouse models. Evaluation of complementary methods to either inhibit RIT1 direc

Extrachromosomal DNA-Driven Oncogene Spatial Heterogeneity and Evolution in Glioblastoma.

UNLABELLED: Oncogenes amplified on extrachromosomal DNA (ecDNA) contribute to treatment resistance and poor survival across cancers. Currently, the spatiotemporal evolution of ecDNA remains poorly understood. In this study, we integrate computational modeling with samples from 94 treatment-naive human glioblastomas (GBM) to investigate the spatiotemporal evolution of ecDNA. We observe oncogene-specific patterns of ecDNA spatial heterogeneity, emerging from random ecDNA segregation and differing fitness advantages. Unlike PDGFRA-ecDNAs, EGFR-ecDNAs often accumulate prior to clonal expansions, c…

Microplastic Contamination: A Rising Environmental Crisis With Potential Oncogenic Implications.

Microplastics have emerged as pervasive environmental contaminants detected in air, water, food, and even human tissues such as blood and placenta. These particles are now recognized as potential health hazards to humans. Due to their high surface area and ability to adsorb toxic substances, microplastics can act as vectors for heavy metals and persistent organic pollutants, many of which are established carcinogens. Evidence from animal models links microplastic exposure to oxidative stress, inflammation, immune disruption, and tissue damage, mechanisms implicated in cancer pathogenesis. Alth

TAK1 is a key regulator of oncogenic signaling and differentiation blockade in rhabdomyosarcoma.

Rhabdomyosarcoma (RMS) is a malignant soft tissue sarcoma with a skeletal muscle phenotype, accounting for approximately 50% of all pediatric soft tissue sarcomas and 8% of all childhood cancers. Although RMS cells express myogenic regulatory factors, they fail to undergo terminal differentiation into mature muscle cells. Transforming growth factor β-activated kinase 1 (TAK1) is a major signaling protein that activates multiple intracellular pathways in response to growth factors, cytokines, and microbial products. Emerging evidence suggests that TAK1 is also an important regulator of self-ren

NUP98-BPTF promotes oncogenic transformation through PIM1 upregulation.

INTRODUCTION: Nucleoporin 98 (NUP98) fusion proteins are recurrently found in leukemia and are associated with unfavorable clinical outcomes. They are distributed to the nucleus and contribute to leukemogenesis via aberrant transcriptional regulation. We previously identified NUP98-BPTF (NB) fusion in patients with T-cell acute lymphoblastic leukemia (T-ALL) using next-generation sequencing. The FG-repeat of NUP98 and the PHD finger and bromodomain of bromodomain PHD finger transcription factor (BPTF) are retained in the fusion. Like other NUP98 fusion proteins, NB is considered to regulate ge

The Effect of Atorvastatin on Oncogenic miRNAs in Hematological Malignancies: A Central Study.

The efficacy of statins as anti-cancer drugs has been demonstrated in several malignancies but has been poorly investigated in hematological malignancies (HM). By studying its effect on oncogenic miRNAs, we investigated the effect of statin therapy on HM patients. The data were used to identify enriched pathways that were altered due to statin treatment. The main aim of this study was to identify significantly differentially expressed miRNAs and involved regulatory pathways post-atorvastatin treatment in HM patients. A panel of 95 plasma circulating miRNAs involved in tumorigenesis, apoptosis,

BCLAF1 is Expressed as a Potential Anti-oncogene in Bile Duct Cancer.

Validating the role of BCLAF1 in the development of Bile Duct Cancer. Differential expression of BCLAF1 in Bile Duct Cancer and normal tissues was analyzed bioinformatically, and immuno-infiltration analysis was performed by R. We also derived the correlation between the expression of BCLAF1 and HIF-1α by bioinformatics analysis and validated it by Western Blotting, qRT-PCR and scratch assays before and after hypoxia. Through bioinformatics analysis, we found that BCLAF1 mRNA was significantly higher in the tumor tissues of Bile Duct Cancer. The high expression of BCLAF1 implied a more advance

Exploring the oncogenic role of RGS19 in bladder cancer progression and prognosis.

This study investigates the role of autophagy-related genes (ARGs) in bladder cancer (BLCA), focusing on the regulator of G protein signaling 19 (RGS19). Using data from The Cancer Genome Atlas (TCGA) and the Human Autophagy Database (HADb), we identified RGS19 as significantly upregulated and linked to poor prognosis in BLCA. Kaplan-Meier survival analysis confirmed its association with increased mortality and. In vitro, RGS19 knockdown in BLCA cell lines inhibited proliferation, migration, and invasion, while inducing apoptosis and autophagy. Transmission electron microscopy showed autophagi

Oncogenic alterations in KIR3DL1 in cutaneous acral CD8+ lymphoproliferative disorder.

BACKGROUND: Primary cutaneous acral CD8+ T-cell lymphoproliferative disorder (TLPD) is a rare and indolent lymphoma entity. Although TLPD was first identified many years ago, the molecular pathogenesis is still not fully understood. OBJECTIVES: In order to better understand the molecular pathogenesis of cutaneous acral CD8+ TLPD and to identify further discriminatory markers to differentiate this lymphoma subtype from other CD8+ cutaneous lymphomas, we analysed five cases of cutaneous acral CD8+ TLPD for putative molecular alterations. METHODS: Somatic alterations were assessed using whole-e

Systematic analysis identifies REST as an oncogenic and immunological biomarker in glioma.

The repressor element 1 silencing transcription factor (REST) has been proposed to function as a transcription factor to silence gene transcription by binding to repressor element 1 (RE1), a highly conserved DNA motif. The functions of REST in various tumors have been studied, but its role and correlation with immune cell infiltration remains uncertain in gliomas. REST expression was analyzed in datasets of The Cancer Genome Atlas (TCGA) and the Genotype-Tissue Expression (GTEx) and validated by the Gene Expression Omnibus and Human Protein Atlas databases. The clinical prognosis of REST was e

Oncogenic drivers dictate immune control of acute myeloid leukemia.

Acute myeloid leukemia (AML) is a genetically heterogeneous, aggressive hematological malignancy induced by distinct oncogenic driver mutations. The effect of specific AML oncogenes on immune activation or suppression is unclear. Here, we examine immune responses in genetically distinct models of AML and demonstrate that specific AML oncogenes dictate immunogenicity, the quality of immune response and immune escape through immunoediting. Specifically, expression of NrasG12D alone is sufficient to drive a potent anti-leukemia response through increased MHC Class II expression that can be overco…
