Although paternal post-partum depression (PPD) significantly impacts the mental wellbeing of fathers and their families, there are many barriers to fathers being assessed and supported for PPD. Given mothers are positioned both close to fathers and healthcare services, we employed mixed-methods to investigate mothers' potential role in facilitating fathers' recognition of and help-seeking for PPD. In Study 1, 189 mothers in Australia (M age = 35.0, 78.9% Australian-European) completed an online survey assessing mothers' levels of mental health literacy regarding paternal PPD. Results showed th

Advanced paternal age affects embryonic development and offspring phenotypes. We previously demonstrated that resveratrol prevents age-associated declines in the mitochondrial DNA copy number (mt-cn) and telomere length (TL) in embryos. The present study was performed to investigate the effects of paternal resveratrol intake on the mt-cn and TL in pups and embryos produced either via in vitro fertilization (IVF) or IVF using sperm preincubated in epididymal fluid (EF). C57BL/6N male mice were administered drinking water containing either the vehicle (ethanol, 1/2500) or 0.1 mM resveratrol. Whe

High-glycemic foods such as white rice (WR) are major dietary contributors to metabolic disorders, yet paternal transmission of their metabolic effects remains unknown. This study investigated whether paternal WR feeding influences metabolic phenotypes in Drosophila (fruit fly) offspring. Fly eggs were reared on either a control, 50% WR, or 50% brown rice (BR) during development, and adult males were subsequently maintained on respective diets prior to mating with virgin females maintained on a normal diet. The resulting F1 and F2 offspring were maintained on either a normal diet or a high-sug

Growing evidence challenges the long-standing assumption that the paternal germline is insulated from environmental perturbation and identifies preconception paternal exposure as an underrecognized pathway of developmental neurotoxicity. Across mammalian models, paternal exposure to substances of abuse, including cocaine, nicotine, ethanol, morphine, and cannabinoids, induces reproducible, often sex-specific alterations in offspring brain function and behavior, including changes in cognition, stress responsivity, reward processing, and affective behaviors, accompanied by transcriptional and ep

Folate-dependent one-carbon metabolism supplies methyl donors required for chromatin modification, yet how metabolic conditions shape epigenome establishment in the male germline remains poorly understood. Here, using a folate-deficient mouse model, we identify meiotic prophase I as a metabolically sensitive window in the male germline. By integrating in vivo germline analysis with bulk and single-cell transcriptomic and epigenomic profiling, we show that folate deficiency disturbs transcriptional programs in pachytene spermatocytes and preferentially perturbs CpG island (CGI)-associated promo

Paternal pre-conceptual exposure to pathogenic infection can alter offspring phenotypes via changes to sperm epigenetics, including small non-coding RNAs (ncRNAs). This phenomenon occurs even in the absence of infection when pathogen mimetics trigger paternal immune activation (PIA). While this implicates a shared component of the immune response, the specific mechanisms responsible remain unknown. Cytokines are a key component of innate immunity and are highly upregulated following exposure to both pathogens and their mimetics. Here we investigate whether paternal administration of the pro-in

Childhood obesity can induce long-lasting metabolic programming, with consequences that may extend to subsequent generations. However, whether paternal early-life obesity programs hypothalamic neuropeptide pathways controlling feeding behavior in offspring remains poorly understood. We investigated the effects of paternal obesity induced by lactational overnutrition on hypothalamic neuropeptide expression, circulating leptin, and food intake in adult offspring. On postnatal day (PND) 0, male Wistar rats were assigned to normal litters (NL; 10 pups/dam) or small litters (SL; 3 pups/dam) to indu

In recent decades, obesity and diabetes have reached pandemic levels, with obesity now recognised as a major health risk factor. Evidence shows that metabolic diseases are more pronounced in the offspring of malnourished parents, suggesting that predisposition can be inherited via epigenetic information in gametes. This has sparked growing interest in small regulatory RNAs in sperm as carriers of epigenetic inheritance. However, the functional annotation of dysregulated sperm microRNAs (miRNAs) in obesity and diabetes remains limited. This work addresses this gap by analysing publicly availabl

BACKGROUND: Depression occurring during the perinatal period (PND) could affect both future mother and father. PND may lead to several adverse physical and mental health outcomes for the whole family. Several psychopathological determinants have been identified, even though few studies investigated the role of paternal mental health in the onset of maternal perinatal depression (MPND). Hence, a retrospective cohort study was carried out in order to investigate the relationship between paternal mental health and the occurrence of antenatal maternal depression as well as identifying potential so

BACKGROUND: As fathers become more involved in pregnancy and early parenting, more research attention has been given to their emotional connection with their unborn child. The transition to fatherhood is a critical developmental phase involving psychological and relational changes that shape paternal identity and bonding with the fetus. However, the factors influencing prenatal father-infant bonding remain unclear. This study examines whether emotional abuse and neglect experienced in childhood impact paternal bonding during pregnancy, and whether antenatal depressive symptoms mediate this rel

BACKGROUND: Epidemiological studies have reported an association between advanced paternal age at conception and an increased risk of neurodevelopmental disorders in offspring, such as autism spectrum disorder. Evidence suggests that DNA methylation alterations in spermatozoa of older men may be transmitted to the feto-placental unit and associated with offspring brain development and behavioral differences later in childhood.
OBJECTIVE: We aimed to assess the association of advanced paternal age with DNA methylation alterations in the human placenta and compare the results to previous findin

Anti-TNF drugs are effective in the treatment of inflammatory arthritis. However, concerns about the impact of anti-TNF therapy on fertility, particularly in male patients, have been insufficiently explored. This study aimed to investigate the effects of anti-TNFs on male fertility and examine the pregnancy outcomes of their partners. This study was conducted on male patients with inflammatory arthritis undergoing anti-TNF therapy. Male patients whose female partners became pregnant during the anti-TNF therapy were evaluated regarding their partners' pregnancy outcomes. The characteristics of

Cocaine use remains a major public health concern, with rising global prevalence and a well-established profile of neurotoxicity and addictive potential. While the central nervous system has been the primary focus of cocaine research, emerging evidence indicates that cocaine also disrupts male reproductive physiology. In the testis, cocaine alters the endocrine microenvironment, induces cell-specific damage, and disrupts spermatogenesis. Cocaine also interferes with epigenetic programming in germ cells and mature sperm, potentially leading to heritable epimutations. Epidemiology data reveal th

Post-traumatic stress (PTS) is a debilitating mental health condition that is highly prevalent in Veteran populations owing to their increased exposure to combat-related trauma. PTS is associated with numerous comorbid conditions including major depressive disorder, anxiety, and chronic pain. Although the causal mechanisms are currently unknown, paternal trauma has been linked to an increased risk for pathology in their offspring. Therefore, using a preclinical model of combat trauma, we examined the relationship between paternal PTS and offspring socio-emotional functioning, pain perception,

Nanoplastics (NPs), as emerging contaminants, have been shown to cause testicular disorders in mammals. However, whether paternal inheritance effects on offspring health are involved in NP-induced reproductive toxicity remains unclear. In this study, we developed a mouse model where male mice were administered 200 nm polyethylene nanoparticles (PE-NPs) at a concentration of 2 mg/L through daily gavage for 35 days to evaluate the intergenerational effects of PE-NPs in an exclusive male-lineage transmission paradigm. We observed that paternal exposure to PE-NPs significantly affected growth phen

There is growing evidence that poor paternal diet at the time of conception increase the risk of offspring developing a range of non-communicable metabolic diseases, such as obesity, diabetes and cardiovascular disease, in adulthood. We hypothesise that a paternal low protein-high carbohydrate diet perturbs offspring tissue lipid abundance through both sperm and seminal plasma-mediated mechanisms. To test our hypothesis, we fed male C57BL/6 mice either a control normal protein diet (NPD; 18% protein) or an isocaloric low protein diet (LPD; 9% protein) for a minimum of 8 weeks. We generated off

BACKGROUND: Gene-environment interactions contribute to metabolic disorders such as diabetes and dyslipidemia. In addition to affecting metabolic homeostasis directly, drugs and environmental chemicals can cause persistent alterations in metabolic portfolios across generations in a sex-specific manner. Here, we use inorganic arsenic (iAs) as a prototype drug and chemical to dissect such sex differences.
METHODS: After weaning, C57BL/6 WT male mice were treated with 250 ppb iAs in drinking water (iAsF0) or normal water (conF0) for 6 weeks and then bred with 15-week-old, non-exposed females for
