Strategies and limitations of the bat immune response to Pseudogymnoascus destructans: the causative agent of white-nose syndrome.

The rapid spread of white-nose syndrome (WNS), an invasive fungal pathogen in bats caused by the psychrophilic fungus Pseudogymnoascus destructans, represents one of the most severe ongoing wildlife disease crises in North America. Since its emergence in 2006, WNS has driven drastic population declines in several ecologically and economically important bat species, including Myotis lucifugus. Once widespread and abundant, M. lucifugus is now designated as vulnerable or endangered in several jurisdictions, such as under Ontario's Species at Risk Act (SARA). Despite this, current gaps in underst

A Step in the Right Direction: Trends over Time in Bariatric Procedures for Patients with Gastroesophageal Reflux Disease.

INTRODUCTION: While laparoscopic sleeve gastrectomy (LSG) has recently emerged as the predominant surgery type for the national bariatric cohort, the literature suggests that laparoscopic Roux-en-Y gastric bypass (LRYGB) may be more effective in normalizing gastroesophageal physiology for the subset of patients with GERD. This study explored practice patterns over time for patients with GERD or hiatal hernia, a related comorbidity, undergoing bariatric surgery. METHODS: Data for LSG and LRYGB were extracted from the Metabolic and Bariatric Surgery Accreditation and Quality Improvement Program

"It's better for me to drink, at least the stress is going away": perspectives on alcohol use during pregnancy among South African women attending drinking establishments.

The Western Cape of South Africa has one of the highest rates of fetal alcohol spectrum disorders (FASD) globally. Reducing alcohol use during pregnancy is a pressing public health priority for this region, but insight into the experiences of women who drink during pregnancy is lacking. Convenience sampling in alcohol-serving venues was used to identify women who were currently pregnant (n = 12) or recently post-partum (n = 12) and reported drinking during the pregnancy period. In-depth qualitative interviews were conducted between April and August 2013. Interviews explored drinking narratives

Feed status and skin injury modulate immunopathology, global gene expression, and survival in channel catfish during virulent Aeromonas hydrophila infection.

INTRODUCTION: Virulent Aeromonas hydrophila is a major pathogen in channel catfish (Ictalurus punctatus), that causes motile Aeromonas septicemia and significant economic losses. We investigated the effect of feeding status and skin integrity on the host immune response, disease survival, and gastrointestinal pathology following a vAh challenge. METHODS: Using a bath immersion model, channel catfish were divided into four treatment groups: fin clipped and fed (FCF), fin clipped but not fed (FCN), not fin clipped but fed (NCF), and not fin clipped nor fed (NCN) alongside non-challenged control

Haploinsufficiency of the E3 ubiquitin ligase C-terminus of heat shock cognate 70 interacting protein (CHIP) produces specific behavioral impairments.

The multifunctional E3 ubiquitin ligase CHIP is an essential interacting partner of HSP70, which together promote the proteasomal degradation of client proteins. Acute CHIP overexpression provides neuroprotection against neurotoxic mitochondrial stress, glucocorticoids, and accumulation of toxic amyloid fragments, as well as genetic mutations in other E3 ligases, which have been shown to result in familial Parkinson's disease. These studies have created a great deal of interest in understanding CHIP activity, expression and modulation. While CHIP knockout mice have the potential to provide ess

High-Frequency Vibration as a Vestibular Stimulus: From Jerk-Rich Physiology to Clinical Applications.

BACKGROUND: High-frequency bone-conducted vibration (BCV) and skull vibration-induced nystagmus (SVIN) are widely used to probe vestibular function, yet their physical nature and physiological targets remain incompletely defined. In particular, the relationship between vibration physics and jerk, the time derivative of acceleration, and their relevance to vestibular activation has not been systematically explored. OBJECTIVE: To develop an engineering-informed framework for high-frequency vestibular stimulation, linking jerk-rich transient stimuli to otolithic biomechanics and vestibular signa

Woodchuck and deer Hepatitis Delta-like agents show distinct innate immune activation and IFN-resistance compared to human Hepatitis D Virus.

Replication of human hepatitis delta virus (HDV) is sensed by the pattern recognition receptors (PRRs) melanoma differentiation antigen 5 (MDA5) and Laboratory of Genetics and Physiology 2 (LGP2), activating interferon (IFN) responses. Both HDV-induced IFNs and exogenously administered IFN-α and -λ suppress HDV RNA spread via cell division (CDMS) but have only minor effects on replication in resting cells. With the discovery of HDV-like agents (DLAs) in diverse animal species an opportunity arose to comparatively study the interplay between these evolutionarily related replicons with respect t

The HCMV tegument protein UL88 degrades MyD88 and reduces innate immune activation.

The human cytomegalovirus (HCMV) encodes the tegument protein UL88, which supports virus spread by mediating the degradation of the innate immune signaling adapter protein, myeloid differentiation primary response 88 (MyD88). MyD88 transduces signals in multiple innate immune pathways, including acting downstream of pattern recognition receptors and IL-1 cytokine family members. MyD88 is rapidly and robustly upregulated following exposure to HCMV, irrespective of viral gene expression and, even after infection, primarily within uninfected cells in a culture. However, UL88 was required to downr

Lost in translation? The potential psychobiotic Lactobacillus rhamnosus (JB-1) fails to modulate stress or cognitive performance in healthy male subjects.

BACKGROUND: Preclinical studies have identified certain probiotics as psychobiotics - live microorganisms with a potential mental health benefit. Lactobacillus rhamnosus (JB-1) has been shown to reduce stress-related behaviour, corticosterone release and alter central expression of GABA receptors in an anxious mouse strain. However, it is unclear if this single putative psychobiotic strain has psychotropic activity in humans. Consequently, we aimed to examine if these promising preclinical findings could be translated to healthy human volunteers. OBJECTIVES: To determine the impact of L. rham

Genetic advances in inflammatory bowel disease.

The translation of basic science advances to tangible benefits in clinical practice remains a fundamental goal of biomedical research. Genetic approaches provide a unique opportunity to enhance bench-to-bedside translational efforts, with inflammatory bowel disease (IBD) being a model genetic disorder in several respects. The association of the NOD2 (CARD15) mutations to Crohn's disease (CD) represents, in complex human disorders, one of the clearest cases of a definitive disease association. The NOD2 gene is located in the IBD1 genetic linkage region on chromosome 16 and functions as an intra

Expression of microRNA‑26b and identification of its target gene EphA2 in pituitary tissues in Yanbian cattle.

microRNAs (miRNAs/miRs) are a class of single‑stranded non‑coding RNA molecules of 19‑24 nucleotides (nt) in length. They are widely expressed in animals, plants, bacteria and viruses. Via specific mRNA complementary pairing of target genes, miRNAs are able to regulate the expression of mRNA levels or inhibit protein translation following transcription. miRNA expression has a time‑ and space specificity, and it is involved in cell proliferation and differentiation, apoptosis, development, tumor metastasis occurrence and other biological processes. miR‑26b is an miRNA of 22 nt and is important

Disrupting Crystal Growth through Molecular Recognition: Designer Therapies for Kidney Stone Prevention.

Aberrant crystallization within the human body can lead to several disease states or adverse outcomes, yet much remains to be understood about the critical stages leading to these events, which can include crystal nucleation and growth, crystal aggregation, and the adhesion of crystals to cells. Kidney stones, which are aggregates of single crystals with physiological origins, are particularly illustrative of pathological crystallization, with 10% of the U.S. population experiencing at least one stone occurrence in their lifetimes. The human record of kidney stones is more than 2000 years old,
