Synthesis of 4-amino-4,5-dideoxy-L-lyxofuranose derivatives and their evaluation as fucosidase inhibitors.

The nitrone 4 (4,5-dideoxy-4-hydroxylamino-3,4-O-isopropylidene-L-lyxofuranose) was synthesised from D-ribose and used as key intermediate for the preparation of fucosidase inhibitors. We describe two transformations of 4. Hydrolysis with aqueous sulfur dioxide gave the known potent nanomolar inhibitor 4-amino-4,5-dideoxy-L-lyxofuranose (3). 1,3-Dipolar cycloaddition with enol ethers led to the related 1,2,5,6-tetradeoxy-2,5-imino-L-altroheptonic ester 2a, acid 2b and the corresponding heptitol 2c. The new iminosugars have been evaluated for their inhibitory activity against α-L-fucosidase fro

Favism: effect of divicine on rat erythrocyte sulfhydryl status, hexose monophosphate shunt activity, morphology, and membrane skeletal proteins.

Favism is an acute anemic crisis that can occur in susceptible individuals who ingest fava beans. The fava bean pyrimidine aglycone divicine has been identified as a hemotoxic constituent; however, its mechanism of toxicity remains unknown. We have shown recently that divicine can induce a favic-like response in rats and that divicine is directly toxic to rat red cells. In the present study, we have examined the effect of hemotoxic concentrations of divicine on rat erythrocyte sulfhydryl status, hexose monophosphate (HMP) shunt activity, morphology, and membrane skeletal proteins. In vitro exp

Chronic UVB irradiation induces superoxide dismutase activity in human epidermis in vivo.

In order to study the effects of repeated UVB exposures on the epidermal antioxidant defence system, we obtained epidermis samples from male volunteers who were exposed to chronic UVB irradiation. Chronic UVB irradiation was shown to be accompanied by induction of epidermal superoxide dismutase (SOD) activity in vivo, while the activities of the other antioxidant enzymes were not significantly changed. The repeated exposure of the epidermis to UVB irradiation was not accompanied by accumulation of products of lipid peroxidation reactions. As superoxide dismutase is of major importance in scave

Enterococcus faecalis biofilm rewires neutrophil metabolism to suppress antimicrobial activity.

Enterococcus faecalis is an opportunistic pathogen that persists in biofilm-associated infections despite robust neutrophil recruitment. How E. faecalis evades neutrophils remains poorly understood. Here, we investigated how E. faecalis biofilms alter effector functions in human neutrophils and a murine wound infection model. We found that E. faecalis biofilm suppresses neutrophil extracellular trap (NET) formation, bacterial engulfment, and neutrophil-mediated control of biofilm growth. This suppression is partly driven by production of lactic acid through the lactate dehydrogenases (LDHs) of

Integrated Transcriptome and Metabolome Analysis Reveals the Mechanism of Sweetness Formation in Vegetable Soybean Seeds.

To understand the mechanism of sweetness formation in vegetable soybean seeds, an integrative transcriptomic and metabolomic analysis was conducted using high sweetness (HS) and low sweetness (LS) varieties selected from 287 resources based on electronic tongue evaluation. The HS variety exhibited significantly higher levels of soluble sugars (58.91 mg/g) and free amino acids (54.15 mg/g). Transcriptomic results indicated that DEGs correlated with glycolysis/gluconeogenesis, starch and sucrose metabolism, and biosynthesis of amino acids pathways were significantly up-regulated in the HS variet

Virus-induced deconjugation of ISG15 dysregulates innate immunity and cellular metabolism.

Interferon-stimulated gene 15 (ISG15) encodes a ubiquitin-like protein that regulates diverse cellular responses, including antiviral immunity, through its conjugation to proteins in a process known as ISGylation. Several pathogens, including SARS-CoV-2, subvert ISGylation by encoding deISGylating enzymes. However, the direct targets and physiological consequences of coronaviral deISGylation remain poorly defined. Here, we genetically ablated the deISGylating activity of the SARS-CoV-2-encoded papain-like protease (PLpro) and found that loss of deISGylation boosted innate immune activation, at

Multi-omics profiling uncovers metabolic regulation in cumulus cells during oocyte maturation.

Bidirectional signalling between the oocyte and surrounding cumulus cells is essential for coordinating follicular development and acquiring oocyte competence. The metabolic networks governing bovine cumulus cell function, however, remain poorly characterized. To address this gap, we performed integrated metabolomic and proteomic analyses of cumulus cells isolated from cumulus-oocyte complexes at two critical maturation stages: (1) meiotically arrested germinal vesicle stage (0 h) and (2) metaphase II stage (24 h). This temporal profiling reveals dynamic metabolic reprogramming events supporti

Mapping the Binding Landscape of Allosteric Inhibitor G6PDi-1 on Human G6PD.

Targeting the oxidative pentose phosphate pathway by inhibiting glucose-6-phosphate dehydrogenase (G6PD) is a promising anticancer strategy, yet clinically useful inhibitors remain unavailable. A key limitation is the lack of molecular insight into how allosteric and noncompetitive inhibitors perturb enzymatic activity, limiting rational optimization. Here, we investigated the mechanism of G6PDi-1, a potent, reversible, noncompetitive G6PD inhibitor, by combining biochemical assays with molecular dynamics simulations, Markov state model, and MM/PBSA binding energy calculations. Experimentally,

Sex differences in Drosophila intestinal metabolism contribute to sexually dimorphic infection outcome and alter gut pathogen virulence.

Sexual dimorphism in infection outcomes is widespread, yet its underlying mechanisms remain incompletely understood. Utilizing Pseudomonas entomophila intestinal infection in Drosophila, we demonstrate that sex differences in intestinal redox processes contribute to female-biased susceptibility to infection. Female inability to overcome pathogen-induced oxidative stress results in defecation blockage, pathogen persistence, and host death. Male flies exhibit increased carbohydrate metabolism and pentose phosphate pathway activity-a key antioxidant defense system. This allows males to withstand

The pivotal role of endoplasmic reticulum in cancer glucose metabolism.

The endoplasmic reticulum (ER) supports essential biosynthetic and quality control functions. These processes rely on sustained energy supply and precise redox control within the ER lumen. While ATP can be imported from mitochondria, pyridine nucleotides are impermeable to the ER membrane, necessitating compartment-specific mechanisms to regulate NAD(H) and NADP(H) pools. Here, we demonstrate that the ER-confined pentose phosphate pathway (ER-PPP), driven by hexose-6-phosphate dehydrogenase (H6PD), processes large amounts of glucose equivalents to preserve the local redox homeostasis in triple

Prostate cancer exploits BRD9-driven metabolic reprogramming to shape the aggressive phenotype.

The aggressive phenotype of prostate cancer (PCa) requires adaptation to androgen deprivation (AD) to progress into castration-resistant PCa (CRPC), including adaptation to AD-induced oxidative stress. However, our understanding of the oncogenes that maintain the redox balance during CRPC progression is limited. Here, we identified Bromodomain-containing protein 9 (BRD9) as a metabolic checkpoint for reprogramming cell metabolism to support tumor growth and impart a castration-resistant phenotype under metabolic and oxidative stress. Following oxidation, BRD9 recruited the nuclear transcriptio

Comparative Physiological and Transcriptomic Analyses Revealed Key Metabolic Pathways Involved in Ethanol Fermentation in Nanguo Pears during Ripening.

Nanguo pears produce a distinctive "alcoholic taste" during ripening, while Jingbai pears do not. To better understand the regulatory mechanism of "alcohol taste" formation in Nanguo pears, differences in ethanol fermentation-related metabolisms between the two cultivars were investigated. Compared to Jingbai pears, Nanguo pears had higher activities of pyruvate decarboxylase (PDC) and alcohol dehydrogenase (ADH) and contents of pyruvate, acetaldehyde, and ethanol during ripening. Nanguo pears generally showed higher starch degradation and higher levels of acetyl-CoA and citric acid but lower

G6PD deficiency triggers dopamine loss and the initiation of Parkinson's disease pathogenesis.

Loss of dopaminergic neurons in Parkinson's disease (PD) is preceded by loss of synaptic dopamine (DA) and accumulation of proteinaceous aggregates. Linking these deficits is critical to restoring DA signaling in PD. Using murine and human pluripotent stem cell (hPSC) models of PD coupled with human postmortem tissue, we show that accumulation of α-syn micro-aggregates impairs metabolic flux through the pentose phosphate pathway (PPP). This leads to decreased nicotinamide adenine dinucleotide phosphate (NADP/H) and glutathione (GSH) levels, resulting in DA oxidation and decreased total DA leve

G6PD deficiency in brain induces schizophrenia-like behaviors and synaptic dysfunction.

Schizophrenia is a severe mental disorder characterized by aberrant brain development, influenced by genetic and environmental factors, with an incompletely understood etiology. Glucose-6-phosphate dehydrogenase (G6PD), a critical enzyme in the pentose phosphate pathway (PPP), facilitates NADPH production for biosynthesis and redox homeostasis. Recent studies suggests that PPP inhibition and consequent oxidative stress contribute to schizophrenia pathogenesis. While clinical investigations have proposed a link between G6PD deficiency and schizophrenia, the underlying biological mechanisms rema

Study on the Characteristics of Traditional Chinese Medicine Syndromes in Patients with Erosive Gastritis Based on Metabolomics.

According to traditional Chinese medicine theory, tongue coatings reflect changes in the body. The goal of this study was to identify a metabolite or a set of metabolites capable of classifying characteristics of traditional Chinese medicine syndromes in erosive gastritis. In this study, we collected tongue coatings of patients with erosive gastritis with damp-heat syndrome (DHS), liver depression and qi stagnation syndrome (LDQSS), and healthy volunteers. Then, we analyzed the differences in metabolic characteristics between the two groups based on metabolomics. We identified 14 potential bio

Diabetes Primes Neutrophils for Neutrophil Extracellular Trap Formation through Trained Immunity.

Neutrophils are primed for neutrophil extracellular trap (NET) formation during diabetes, and excessive NET formation from primed neutrophils compromises wound healing in patients with diabetes. Here, we demonstrate that trained immunity mediates diabetes-induced NET priming in neutrophils. Under diabetic conditions, neutrophils exhibit robust metabolic reprogramming comprising enhanced glycolysis via the pentose phosphate pathway and fatty acid oxidation, which result in the accumulation of acetyl-coenzyme A. Adenosine 5'-triphosphate-citrate lyase-mediated accumulation of acetyl-coenzyme A a

Mechanosensing regulates pDC activation in the skin through NRF2 activation.

Plasmacytoid DCs (pDCs) infiltrate the skin, chronically produce type I interferon (IFN-I), and promote skin lesions and fibrosis in autoimmune patients. However, what controls their activation in the skin is unknown. Here, we report that increased stiffness inhibits the production of IFN-I by pDCs. Mechanistically, mechanosensing activates stress pathways including NRF2, which induces the pentose phosphate pathway and reduces pyruvate levels, a product necessary for pDC responses. Modulating NRF2 activity in vivo controlled the pDC response, leading to resolution or chronic induction of IFN-I
