Benzylamine-Based Organoselenium Compounds as Glutathione Peroxidase Mimics: Antioxidant and Antibacterial Agents.

Benzylamine-based organoselenium compounds were designed and synthesized to explore their antioxidant and antibacterial properties. The synthetic strategy involves nucleophilic substitution of benzyl bromides with in situ generated sodium selenolate, obtained from sodium borohydride reduction of the corresponding diselenides, affording the desired products. The multifunctional antioxidant properties, including radical-trapping and glutathione peroxidase-like antioxidant activities were evaluated using 2,2-diphenyl-1-picrylhydrazyl and thiophenol assay, respectively. These antioxidants exhibite

Immobilized horseradish peroxidase hybrid biocatalysts for degradation of pharmaceuticals and other emerging micropollutants.

With the increasing detection of emerging pollutants (EPs), such as pharmaceuticals, personal care products, and pesticides, in water bodies, there is a growing research priority to develop efficient methods for their degradation. In the present study, the degradation of 21 EPs from different classes was investigated using horseradish peroxidase (HRP) covalently immobilized onto two photocatalytic supports, TiO2 and ZnO. The hybrid biocatalysts (TiO2-HRP and ZnO-HRP) were used to degrade EPs with and without the presence of a redox mediator, 1-hydroxybenzotriazole (HOBT). The results revealed

Molecular characterization and functional analysis of thioredoxin peroxidase-2 from Babesia duncani.

Human babesiosis caused by Babesia duncani is an emerging zoonotic disease that poses a growing threat to public health. Since the first reported case in 1991, knowledge of this pathogen has remained limited. Recent completion of the B. duncani genome sequence has accelerated research aimed at elucidating its molecular biology. During its intraerythrocytic life cycle, B. duncani is exposed to oxidative stress generated by redox reactions. In this study, we identified and characterized an antioxidant enzyme from B. duncani-thioredoxin peroxidase-2 (BdTPx-2), a member of the peroxiredoxin family

Peroxynitrite detoxification by Trypanosoma cruzi heme peroxidase supports parasite survival in macrophages.

Trypanosoma cruzi, the causative agent of Chagas disease, must overcome a host's nitro-oxidative burst to establish the infection. Among the reactive species generated by immune cells, peroxynitrite stands out as a highly cytotoxic molecule for the parasite. Yet, the parasite's defense mechanisms against peroxynitrite remain incompletely understood. In this work, we demonstrate that the T. cruzi heme-containing ascorbate peroxidase cytochrome c peroxidase (APx-CcP), an antioxidant enzyme previously shown to increase parasite infectivity, acts as a key peroxynitrite-detoxifying enzyme. Using di

Colorimetric Detection of Acid Phosphatase Using Nanoparticle CuNCs as Peroxidase Mimicking Activity.

Acid phosphatase (ACP) serves as a critical biomarker for disease diagnosis, yet conventional detection methods face limitations in cost and complexity. Copper nanoclusters (CuNCs), leveraging Earth-abundance, low expense, and noble metal-like catalytic properties, offer an ideal nanozyme platform. This study harnesses the intrinsic peroxidase mimicking activity of CuNCs to construct a visual colorimetric sensor for ACP detection. CuNCs oxidize the substrate 3,3',5,5'-tetramethylbenzidine (TMB) to generate a blue chromogen (λmax = 654 nm). ACP introduction triggers reduction of this chromogen,

Tetracyclines at subinhibitory concentrations are lethal for NADH peroxidase-deficient mutants of Enterococcus faecium.

OBJECTIVES: Tigecycline is a bacteriostatic antibiotic member of the glycylcycline family that inhibits protein synthesis. Tigecycline is a last-line treatment for infections caused by MDR pathogens like vancomycin-resistant Enterococcus faecium (VR-Efm). We recently explored oxidative stress defences in E. faecium and we here aimed to assess their role in antibiotic resistance. METHODS: Antibiotic susceptibility was evaluated in mutants deficient in primary oxidative stress defences by monitoring bacterial survival after a 24 h treatment. Hydrogen peroxide (H2O2) levels were quantified to li

Molecular identification of a thioredoxin peroxidase in Babesia gibsoni with potential against oxidative stress.

Babesia gibsoni is the infectious agent of canine babesiosis, a vector-borne infection that poses a global threat to the canine health. As B. gibsoni is an erythrocytic intracellular parasite, the completion of its genome and transcriptome sequencing and analysis facilitates the elucidation of the mechanism of B. gibsoni residue in the erythrocyte. The main function of red blood cells (RBCs) is oxygen delivery; thus, B. gibsoni may be exposed to high levels of oxidative stress. To date, no report is available on the mechanism by which B. gibsoni survives oxidative stress inside the RBCs. In th

Comparative Assessment of Lipid Peroxidase in Oral Cancer and Oral Potentially Malignant Disorders.

Introduction Oral potentially malignant disorders (OPMD) are abnormally altered tissues that could potentially develop into oral cancer. From the literature, it is understandable that not all OPMDs develop into oral cancer. Hence, it is essential to identify the high-risk lesions that are more likely to develop into oral cancer. Lipid peroxidase (LPO) is a byproduct of phospholipid metabolism, and its levels are an oxidative stress marker that can probably help us predict the onset of cancer in OPMDS. This study aimed to assess the levels of LPO in OPMD, oral cancer, and normal patients. Mater

Light-Driven Self-Cascade Peroxidase-like Nanozymes without Exogenous H2O2.

The peroxidase (POD)-like nanozyme typically requires the addition of exogenous H2O2. To address the limitation, previous work mainly adopted a cascade strategy for H2O2 production. Herein, we propose a new light-driven self-cascade strategy to construct POD-like nanozymes without exogenous H2O2. The model nanozyme resorcinol-formaldehyde resin-Fe3+ (RF-Fe3+) is synthesized with the hydroxyl-rich photocatalytic material RF as the carrier to in situ chelate metal oxides, which can simultaneously achieve the functions of in situ H2O2 generation under irradiation and substrate oxidation via POD-l

Amorphous copper(II)-cyanoimidazole frameworks as peroxidase mimics for hydrogen sulfide assay.

Metal-organic frameworks with hierarchical porosities and exposed active sites are promising for ideal enzyme mimics. In this work, we developed a simple and feasible air oxidation strategy to prepare amorphous Cu(II)-cyanoimidazole frameworks (aCu(II)-CIFs) using CuI as the metal source in dimethylsulfoxide. Benefiting from coordination unsaturation and hierarchical porosities, aCu(II)-CIFs exhibit inherent peroxidase-mimic activity for rapid colorimetric reaction of 3,3',5,5'-tetramethylbenzidine (TMB). aCu(II)-CIFs were utilized to develop a colorimetric platform for specific H2S assay in t

Destroying glutathione peroxidase improves the oxidative stress resistance and pathogenicity of Listeria monocytogenes.

INTRODUCTION: Glutathione peroxidase is abundant in eukaryotes as an important antioxidant enzyme. However, prokaryotic glutathione peroxidase has not been thoroughly studied. Listeria monocytogenes is a facultative intracellular pathogen that is capable of causing listeriosis in animals as well as humans. Despite the fact that L. monocytogenes encodes a putative glutathione peroxidase, GSH-Px (encoded by lmo0983)), the functions of the enzyme are still unknown. Here we revealed the unusual roles of L. monocytogenes GSH-Px in bacterial antioxidants and pathogenicity. METHODS: L. monocytogenes

Catalase-peroxidase StKatG is a bacterial manganese oxidase from endophytic Salinicola tamaricis.

Manganese (Mn) oxides in iron/manganese plaques are widely distributed in the rhizosphere of wetland plants and contribute significantly to elemental cycling and pollutant removal. Mn oxides are primarily produced by bacterial processes using Mn oxidases. However, the molecular mechanism underlying the formation of rhizosphere Mn oxides is still largely unknown. This study identified a manganese-oxidizing enzyme, the catalase-peroxidase StKatG, from an endophytic bacterium Salinicola tamaricis from the wetland plant. The gene encoding StKatG was cloned and overexpressed in Escherichia coli. Th

Diagnosis of Pediatric Non-Esophageal Eosinophilic Gastrointestinal Disorders by Eosinophil Peroxidase Immunohistochemistry.

BACKGROUND: Diagnosis of non-esophageal eosinophilic gastrointestinal disorders requires quantification of tissue eosinophils. Our objective was to evaluate eosinophil peroxidase (EPX) immunohistochemistry (IHC) as a method for histologic diagnosis of eosinophilic gastritis (EG) and eosinophilic duodenitis (EoD). METHODS: We performed a retrospective analysis of biopsies from pediatric EG/EoD cases and controls. Subjects with EG or EoD had ≥30 eosinophils per high power field (eos/hpf) in ≥5 hpf in the stomach and/or ≥3 hpf in the duodenum, respectively. Controls had no histopathologic diagno

Rhizobium leguminosarum Glutathione Peroxidase Is Essential for Oxidative Stress Resistance and Efficient Nodulation.

Glutathione (GSH) plays a key role in regulating the cellular Redox Homeostasis, and appears to be essential for initiation and development of root nodules. Glutathione peroxidase (Gpx) catalyzes the reduction of H2O2 and organic hydroperoxides by oxidation of GSH to oxidized GSH (GSSG), which in turn is reduced by glutathione reductase (GR). However, it has not been determined whether the Rhizobium leguminosarum Gpx or GR is required during symbiotic interactions with pea. To characterize the role of glutathione-dependent enzymes in the symbiotic process, single and double mutants were made i

Bioinspired Spiky Peroxidase-Mimics for Localized Bacterial Capture and Synergistic Catalytic Sterilization.

Besides the pandemic caused by the coronavirus outbreak, many other pathogenic microbes also pose a devastating threat to human health, for instance, pathogenic bacteria. Due to the lack of broad-spectrum antibiotics, it is urgent to develop nonantibiotic strategies to fight bacteria. Herein, inspired by the localized "capture and killing" action of bacteriophages, a virus-like peroxidase-mimic (V-POD-M) is synthesized for efficient bacterial capture (mesoporous spiky structures) and synergistic catalytic sterilization (metal-organic-framework-derived catalytic core). Anti-thyroid peroxidase antibody and subclinical hypothyroidism in relation to hypertension and thyroid cysts.

Hypertension frequently occurs in subclinical hypothyroidism (SCH). By bolstering thyroid inflammation, anti-peroxidase antibody (TPO-Ab) causes autoimmune thyroiditis, which is one of the most common causes of SCH. Since the absence of thyroid cysts is associated with TPO-Ab (+) based on the indication of latent thyroid damage, we explored the potential mechanism underlying the association among TPO-Ab, SCH, hypertension, and thyroid cysts. A cross-sectional study of 1,483 Japanese aged 40-74 years was conducted. Thyroid cysts were defined as those having a maximum diameter of ≥ 2.0 mm, conta
