Latitudinal Clines in Diapause Onset and Circadian Clock Characteristics in Drosophila littoralis and Drosophila montana.

Drosophila littoralis and Drosophila montana are two widespread species of the virilis group that occur at latitudes between approximately 40°N and 70°N. To survive the winter, they undergo a photoperiodically controlled diapause triggered by the decreasing photoperiod in late summer to autumn. Previous studies have shown that the critical photoperiod at which diapause is triggered depends on latitude. Fly populations living at high latitudes have a longer critical photoperiod than populations living at low latitudes, meaning that they enter diapause earlier. It is not yet fully understood how…

Daily and seasonal variations in the concentration of melatonin in the human pineal gland.

To elucidate whether pineal melatonin secretion is affected by changes in day length, we determined the concentration of melatonin in human pineal glands obtained at autopsy from 66 male subjects, aged 16-84 years over a period of 12 consecutive months. Based on the time of death, a day-night difference in pineal melatonin levels was evident only in the long photoperiod (April-September) with significantly higher melatonin concentrations occurring at night (2200-1000 h). Nighttime values in the long photoperiod were significantly higher than the nighttime values during the short photoperiod (O…

Hormonal modulation of pineal-mediated seasonal events: effects of thyroid or gonadal manipulations.

The ability of various manipulations of the endocrine system to affect pineal-mediated events was examined in the present studies. Male Syrian hamsters were analyzed for pineal-induced gonadal regression and depressions in serum thyroxine and testosterone levels after treatments which altered thyroid or gonadal function. Hamsters were thyroidectomized, received thyroxine implants (5 mg), or were thyroidectomized plus implanted with thyroxine. The animals were exposed to short photoperiods (10L:14D) for 9 wk, and plasma hormone levels and gonadal status were determined at the end of the experim…

Changes in goose hypothalamus under different photocycles: RNA-Seq reveals new pathways and molecules.

Reproduction in avian species is regulated by light, and the hypothalamus is the most important regulator of the response to light signals. The objective of this study was to identify the key molecules and signaling pathways in the hypothalamus that regulate reproduction in birds based on the photoperiod. Yangzhou geese kept in a closed goose barn were selected for transcriptome sequencing. The light program during rearing was as follows: a short photoperiod (light[L]: dark[D]=6.5 h:17.5 h) for 60 days, followed by a prolonged photoperiod (L:D = 11.5 h:12.5 h). Samples were collected on the 38…

Integrated analysis of differential expression profile of miRNA in the uterus of seasonal estrus sheep.

BACKGROUND: Photoperiod is one of the important factors affecting seasonal estrus of sheep. The importance of the uterus to reproduction is self-evident. However, the uterine molecular mechanisms involved in photoperiodic regulation of seasonal estrus events in sheep remain poorly understood. In recent years, the role of uterine microRNA (miRNA) in mammalian reproduction has been continuously revealed, and it is necessary to analyze and discuss their participation in the seasonal estrus of sheep. RESULTS: We compared the miRNA expression profile in uterine tissues of Sunit sheep at 3 differen…

Pineal melatonin is a circadian time-giver for leptin rhythm in Syrian hamsters.

Nocturnal secretion of melatonin from the pineal gland may affect central and peripheral timing, in addition to its well-known involvement in the control of seasonal physiology. The Syrian hamster is a photoperiodic species, which displays gonadal atrophy and increased adiposity when adapted to short (winter-like) photoperiods. Here we investigated whether pineal melatonin secreted at night can impact daily rhythmicity of metabolic hormones and glucose in that seasonal species. For that purpose, daily variations of plasma leptin, cortisol, insulin and glucose were analyzed in pinealectomized h…

Seasonal vs opportunistic breeders, a seasonal morphological and endocrine comparative study of the gonadal cycle in birds.

Avian reproduction is usually seasonal and strongly influenced by environmental factors, primarily photoperiod, which is mainly perceived by deep brain photoreceptors in the hypothalamic region and regulates the activation of the hypothalamic-pituitary-gonadal axis. However, certain species have developed adaptable reproductive strategies that differ from the classical seasonal model. In this study, we analyzed seasonal variations and gonadal asymmetry in three dove species with different reproductive strategies: The White-tipped Dove (Leptotila verreauxi) and the Eared Dove (Zenaida auriculat…

A brain circuit and neuronal mechanism for decoding and adapting to change in daylength.

Changes in daylight amount (photoperiod) drive pronounced alterations in physiology and behaviour1,2. Adaptive responses to seasonal photoperiods are vital to all organisms - dysregulation is associated with disease, from affective disorders3 to metabolic syndromes4. Circadian rhythm circuitry has been implicated5,6 yet little is known about the precise neural and cellular substrates that underlie phase synchronization to photoperiod change. Here we present a previously unknown brain circuit and novel system of axon branch-specific and reversible neurotransmitter deployment that together prove…

Dietary cadmium induces histopathological changes despite a sufficient metallothionein level in the liver and kidneys of the bank vole (Clethrionomys glareolus).

The objective of this study was to correlate hepatic and renal cadmium (Cd) accumulation, Cd-binding capacity of metallothionein (MT) and lipid peroxidation with the tissue injury in the male bank voles raised under short (8 h light/16 h dark) and long (16 h light/8 h dark) photoperiods that affect differently Cd accumulation and MT induction in these rodents. The animals were exposed to dietary Cd (0, 40 and 80 microg/g) for 6 weeks. The accumulation of Cd in the liver and kidneys appeared to be dose-dependent in bank voles from the two photoperiod groups; however, the short-photoperiod anima…

The fish pituitary directly responds to daylength and drives seasonality.

Seasonal timing is important for many critical life history events of organisms, and annual changes in daylength provide a reliable seasonal cue. In birds and mammals, photoperiod-driven seasonality is caused by changes in pituitary thyroid stimulating hormone (TSH), brain deiodinase (DIO) and triiodothyronine (T3), but it is unclear if a similar mechanism exists in fish. We used anadromous Atlantic salmon that migrate downstream and enter the ocean in spring to examine photoperiod signaling in fish. Manipulations of diurnal and circannual photoperiod cycles indicate that pituitary tshβb and b…

Redefining the limits of day length responsiveness in a seasonal mammal.

At temperate latitudes, increases in day length in the spring promote the summer phenotype. In mammals, this long-day response is mediated by decreasing nightly duration of melatonin secretion by the pineal gland. This affects adenylate cyclase signal transduction and clock gene expression in melatonin-responsive cells in the pars tuberalis of the pituitary, which control seasonal prolactin secretion. To define the photoperiodic limits of the mammalian long day response, we transferred short day (8 h light per 24 h) acclimated Soay sheep to various longer photoperiods, simulating those occurri…

Seasonal time measurement during reproduction.

Most species living outside the tropical zone undergo physiological adaptations to seasonal environmental changes and changing day length (photoperiod); this phenomenon is called photoperiodism. It is well known that the circadian clock is involved in the regulation of photoperiodism such as seasonal reproduction, but the mechanism underlying circadian clock regulation of photoperiodism remains unclear. Recent molecular analysis have revealed that, in mammals and birds, the pars tuberalis (PT) of the pituitary gland acts as the relay point from light receptors, which receive information about …

Monosodium glutamate-induced arcuate nucleus damage affects both natural torpor and 2DG-induced torpor-like hypothermia in Siberian hamsters.

Siberian hamsters (Phodopus sungorus) have the ability to express daily torpor and decrease their body temperature to approximately 15 degrees C, providing a significant savings in energy expenditure. Daily torpor in hamsters is cued by winterlike photoperiods and occurs coincident with the annual nadirs in body fat reserves and chronic leptin concentrations. To better understand the neural mechanisms underlying torpor, Siberian hamster pups were postnatally treated with saline or MSG to ablate arcuate nucleus neurons that likely possess leptin receptors. Body temperature was studied telemetri…

Both Low Temperature and Shorter Duration of Food Availability Delay Testicular Regression and Affect the Daily Cycle in Body Temperature in a Songbird.

Photoperiodic control of reproduction in birds is based on two processes, a positive effect leading to gonadal maturation and an inhibitory effect subsequently inducing regression. Nonphotoperiodic cues can modulate photoperiodic control, particularly the inhibitory process. In previous studies of common starlings (Sturnus vulgaris), (1) restriction of food availability to 8 h after dawn had little effect on testicular maturation but dramatically delayed subsequent regression and (2) lower ambient temperature also had little effect during maturation but delayed regression. Could the effects of…

Cyclic AMP-inducible genes respond uniformly to seasonal lighting conditions in the rat pineal gland.

The encoding of photoperiodic information ensues in terms of the daily profile in the expression of cyclic AMP (cAMP)-inducible genes such as the arylalkylamine N-acetyltransferase (AA-NAT) gene that encodes the rate-limiting enzyme in melatonin formation. In the present study, we compared the influence of the photoperiodic history on the cAMP-inducible genes AA-NAT, inducible cyclic AMP early repressor (ICER), fos-related antigen-2 (FRA-2), mitogen-activated protein kinase phosphatase-1 (MKP-1), nerve growth factor inducible gene-A (NGFI-A) and nerve growth factor inducible gene-B (NGFI-B) in…

Pineal and ovarian response to 22- and 24-h days in the ewe.

Melatonin secretion in ewes was entrained by 22-h light-dark cycles whether of long (16L:6D) or short (6L:16D) photoperiod. In photoperiods of 6L:16D, a phase-delay of melatonin secretion was evident, leading to a dark-phase duration shorter than that found in 8L:16D. Early onset of estrus was induced in anestrous ewes kept in 8L:16D, but not 6L:16D, from 22 July compared to controls in natural light. In photoperiods of 16L:6D, the melatonin profile corresponded precisely to the dark phase. Early offset of estrus was induced in estrous ewes kept in both 18L:6D and 16L:6D from 18 December compa…
