The Role of Salicylic Acid in the Expression of RECEPTOR-LIKE PROTEIN 23 and Other Immunity-Related Genes.

The hormone salicylic acid (SA) plays a crucial role in plant immunity by activating responses that arrest pathogen ingress. SA accumulation also penalizes growth, a phenomenon visible in mutants that hyperaccumulate SA, resulting in strong growth inhibition. An important question, therefore, is why healthy plants produce basal levels of this hormone when defense responses are not activated. Here, we show that basal SA levels in unchallenged plants are needed for the expression of a number of immunity-related genes and receptors, such as RECEPTOR-LIKE PROTEIN 23 (RLP23). This was shown by depl…

Salicylic acid fine-tunes cell survival during plant senescence and pathogen infection.

Salicylic acid (SA) is a crucial plant hormone synthesized via two primary pathways: the ISOCHORISMATE SYNTHASE (ICS) pathway and the PHENYLALANINE AMMONIA-LYASE (PAL) pathway. SA biosynthesis and homeostasis are precisely regulated in plants. Furthermore, SA functions in a concentration-dependent manner to enhance resistance to both abiotic stresses and pathogens, as well as to regulate plant senescence. However, the underlying mechanisms of SA action remain to be elucidated. Recent studies have demonstrated that in juvenile plants, age-related cues, autoimmunity, and autophagy contribute to

The TaRNS4-TaPP2C10 module regulates wheat resistance to stripe rust through RNA degradation and MAPK cascades.

RNase is a kind of RNA hydrolase that widely exists in organisms. RNase T2 in plants functions by hydrolyzing and processing RNA in the cell … phorylation of TaMAPK6. Overall, these findings indicate that the TaRNS4-TaPP2C10 module fine-tunes plant immunity in an antagonistic manner and highlight the versatility of TaRNS4, which enhances plant resistance by synergizing RNA degradation and the MAPK cascade.

Seed priming with calcium chloride enhances stress tolerance in rice seedlings.

Calcium is a crucial second messenger in plant cells and contributes to plant resistance against biotic and abiotic stress. Plant defense priming with natural or synthetic compounds leads to quicker and stronger resistance responses. However, whether pretreatment of plant seeds with calcium could improve their resistance to stress remains poorly understood. In this study, we showed that rice seedlings grown from calcium chloride (CaCl2)-pretreated seeds displayed enhanced resistance to the rice blast fungus Magnaporthe oryzae and the rice bacterial pathogen Xanthomonas oryzae pv. Oryzae (Xoo).

The role of disease transmission and conferred immunity in outbreaks: analysis of the 1993 Cryptosporidium outbreak in Milwaukee, Wisconsin.

The authors examined two competing hypotheses regarding the cause of the 1993 Cryptosporidium outbreak in Milwaukee, Wisconsin. The first was that oocyst contamination of the drinking-water influent, coupled with a treatment plant failure, resulted in a point-source outbreak. The second was that the outbreak was the result of transmission processes that amplified the oocyst concentration in the drinking-water effluent. Analysis of the model suggested that 1) transmission directly from person to person contributed 10% (95% confidence interval: 6%, 21%) of the total cases; 2) closing the drinkin

Comparative oxidation proteomics analyses suggest redox regulation of cytosolic translation in rice leaves upon Magnaporthe oryzae infection.

Pathogen attack can increase plant levels of reactive oxygen species (ROS), which act as signaling molecules to activate plant defense mechanisms. Elucidating these processes is crucial for understanding redox signaling pathways in plant defense responses. Using an iodo-tandem mass tag (TMT)-based quantitative proteomics approach, we mapped 3362 oxidized cysteine sites in 2275 proteins in rice leaves. Oxidized proteins were involved in gene expression, peptide biosynthetic processes, stress responses, ROS metabolic processes, and translation pathways. Magnaporthe oryzae infection led to increa

GSNOR-mediated de-nitrosylation in the plant defence response.

A key feature of the plant defence response is the transient engagement of a nitrosative burst, resulting in the synthesis of reactive nitrogen intermediates (RNIs). Specific, highly reactive cysteine (Cys) residues of low pK(a) are a major site of action for these intermediates. The addition of an NO moiety to a Cys thiol to form an S-nitrosothiol (SNO), is termed S-nitrosylation. This redox-based post-translational modification is emerging as a key regulator of protein function in plant immunity. Here we highlight recent advances in our understanding of de-nitrosylation, the mechanism that d…

Plant-expressed HA as a seasonal influenza vaccine candidate.

Influenza is a globally important respiratory pathogen that causes a high degree of morbidity and mortality annually. Although current vaccines are effective against virus infection, new strategies need to be developed to satisfy the global demand for an influenza vaccine. To address this point, we have engineered and produced the full-length hemagglutinin (HA) protein from the A/Wyoming/03/03 (H3N2) strain of influenza in plants. The antigenicity of this plant-produced HA was confirmed by ELISA and single-radial immunodiffusion (SRID) assays. Immunization of mice with plant-produced HA result

High level expression of surface glycoprotein of rabies virus in tobacco leaves and its immunoprotective activity in mice.

A synthetic gene coding for the surface glycoprotein (G protein) of rabies virus was strategically designed to achieve high-level expression in transgenic plants. The native signal peptide was replaced by that of the pathogenesis related protein, PR-S of Nicotiana tabacum. An endoplasmic reticulum retention signal was included at C-terminus of the G protein. Tobacco plants were genetically engineered by nuclear transformation. Selected transgenic lines expressed the chimeric G protein at 0.38% of the total soluble leaf protein. Mice immunized intraperitoneally with the G protein purified from

[Silencing GmATG5 genes accelerated senescence and enhanced disease resistance in soybean].

Autophagy plays an essential role in recycling/re-utilizing nutrients and in adaptions to numerous stresses. However, the roles of autophagy in soybean have not been investigated extensively. In this study, a virus-induced gene silencing approach mediated by bean pod mottle virus (BPMV) was used to silence autophagy-related gene 5 (ATG5) genes in soybean (referred to as GmATG5). Our results showed that ATG8 proteins were massively accumulated in the dark-treated leaves of the GmATG5-silenced plants relative to the vector control plants (BPMV-0), indicating that autophagy pathway is impaired in

In vivo Imaging Enables Understanding of Seamless Plant Defense Responses to Wounding and Pathogen Attack.

Plants are exposed to varied biotic stresses, including sequential or simultaneous attack by insects and pathogens. To overcome these complex stresses, plants must perceive each of the stresses, then integrate and relay the information throughout the plant body and eventually activate local and systemic resistance responses. Previous molecular genetic studies identified jasmonic acid and salicylic acid as key plant hormones of wound and immune responses. These hormones, combined with their antagonistic interaction, play critical roles in the initiation and regulation of defense responses again

Coronavesicle-engineered hydrogel sequential photothermal-immunometabolic therapy reprograms tissue microenvironment for diabetic-infected wounds healing.

Persistent infection and dysregulated tissue microenvironment constitute the primary obstacles hindering the healing of infectious diabetic wounds. Herein, we developed an NIR-responsive hydrogel dressing incorporating polyphenol-engineered plant-derived nanovesicles (termed Coronavesicles) via an in-situ deposition strategy, designed to implement sequential photothermal-immunometabolic therapy for infectious diabetic wounds. The coronavesicles within the hydrogel preferentially adhere to bacterial surfaces to deliver localized photothermal therapy (PTT), wherein subsequent heat-triggered rupt

Catch Me If You Can: How Xylella fastidiosa Thrives in the Xylem.

The outcome of infection can be seen as a race between the pathogen and the plant immune system. In this review, we use the case study of Xylella fastidiosa (Xf) and explain how the bacterium exploits its slow and fastidious growth to remain below detection thresholds, thereby delaying immune activation. Its self-limiting behavior in the sessile state provides a temporal window for transitioning into a more exploratory lifestyle, enabling systemic colonization of the host. As bacterial populations expand, vessel occlusion and immune overactivation at late stages occur, often propagating beyond

Mobile signals in systemic acquired resistance.

Plants possess a unique form of broad-spectrum long-distance immunity termed systemic acquired resistance (SAR). SAR involves the rapid generation of mobile signal(s) in response to localized microbial infection, which transport to the distal tissue and 'prime' them against future infections by related and unrelated pathogens. Several SAR-inducing chemicals that could be classified as the potential mobile signal have been identified. Many of these function in a bifurcate pathway with both branches being equally essential for SAR induction. This review reflects on the potential candidacy of the

TOR coordinates cytokinin and gibberellin signals mediating development and defense.

Plants constantly perceive and process environmental signals and balance between the energetic demands of growth and defense. Growth arrest upon pathogen attack was previously suggested to result from a redirection of the plants' metabolic resources towards the activation of plant defense. The energy sensor Target of Rapamycin (TOR) kinase is a conserved master coordinator of growth and development in all eukaryotes. Although TOR is positioned at the interface between development and defense, little is known about the mechanisms by which TOR may potentially regulate the relationship between th

Fructan oligosaccharide priming alters apoplastic sugar dynamics and improves resistance against Botrytis cinerea in chicory.

Carbohydrates such as fructans can be involved in priming or defence stimulation, and hence potentially provide new strategies for crop protection against biotic stress. Chicory (Cichorium intybus) is a model plant for fructan research and is a crop with many known health benefits. Using the chicory-Botrytis cinerea pathosystem, we tested the effectiveness of fructan-induced immunity, focussing on different plant and microbial fructans. Sugar dynamics were followed after priming and subsequent pathogen infection. Our results indicated that many higher plants might detect extracellular levan ol

Actin depolymerization factor (ADF) moonlighting: Nuclear immune regulation by interacting with WRKY transcription factors and shaping the transcriptome.

Remodeling of the actin cytoskeleton is a critical process for plant immunity, essential for the transport, activation, and stabilization of immune-regulatory molecules and organelles. In this process, actin depolymerization factors (ADFs) function as key players through severing and depolymerizing actin microfilaments. However, recent evidence suggests that ADFs may possess noncanonical immune functions inside the nucleus, in addition to the canonical cytosolic role, a phenomenon not adequately explained by the traditional mechanistic model of ADF-actin dynamics. In this study, we demonstrate…
