Contribution to the quality and safety assessment of traditional herbal remedy Tinospora crispa stem using phytochemical analyses and CYP3A4 inhibition assay in HepaRG cell model.

Legal use of herbal health products must comply with quality and safety standards required by health regulatory authorities in order to protect consumers from risks associated with misidentification, falsification and undesirable toxic effects. Some plant-derived compounds can act as CYP450 inhibitors, potentially leading to herb-drug interactions and, in some cases, direct hepatotoxicity, particularly when CYP450-mediated bioactivation is involved. There is a critical need to develop reliable and efficient screening assays to identify potential CYP450 inhibitors in such complex mixtures and h

Using 13C to Quantify Phloem Transport on Tall Plants in the Field.

Phloem transport dynamics play a crucial role in understanding the movement of photoassimilates within plants, particularly in tall species, where tracing carbon allocation and monitoring transport velocities present unique challenges. The difference in time lags between a labeling pulse of 13CO2 of the foliage and the appearance of labeled C in the respiration at different locations along the stem of a tall plant is used to estimate at which velocities the isotope tracer, i.e., the labeled carbohydrates, are transported in the phloem sap. Here, we describe a method for pulse labeling tall pla

Identification of the anti-epileptic effect of speciogynine through molecular network-aided metabolite profiling of M. speciosa alkaloids using zebrafish.

Mitragyna speciosa is a native plant to Southeast Asian countries, known its medicinal use as an analgesic and stimulant. Apart from the indole-type alkaloids, which have been shown to be responsible for its pharmacological properties, there is still limited information on other classes of compounds present in various parts of the plant. In an effort to fill this knowledge gap, the present study applied tandem liquid chromatography-mass spectrometry (LC-MS/MS)-based molecular networking to obtain more information on the chemodiversity of M. speciosa. Analysis of the generated molecular map obt

Mind the bubbles: achieving stable measurements of maximum hydraulic conductivity through woody plant samples.

The maximum specific hydraulic conductivity (k(max)) of a plant sample is a measure of the ability of a plants' vascular system to transport water and dissolved nutrients under optimum conditions. Precise measurements of k(max) are needed in comparative studies of hydraulic conductivity, as well as for measuring the formation and repair of xylem embolisms. Unstable measurements of k(max) are a common problem when measuring woody plant samples and it is commonly observed that k(max) declines from initially high values, especially when positive water pressure is used to flush out embolisms. This

Ecology of Australia: the effects of nutrient-poor soils and intense fires.

Australia, the flattest, driest, and geologically oldest vegetated continent, has a uniquely high proportion of nutrient-poor soils. We develop a "Nutrient-Poverty/Intense-Fire Theory," which postulates that most anomalous features of organisms and ecosystems of Australia are the evolutionary consequences of adaptations to nutrient poverty, compounded by intense fire that tends to occur as a result of nutrient poverty. The fundamental tenet of the theory is that plants growing in environments with plentiful light and periodic adequate moisture, but on soils poor in phosphorus, zinc, and other

Transcriptome analysis revealed key genes and pathways related to cadmium tolerance and accumulation in coix (Coix lacryma-jobi L.).

Coix (Coix lacryma-jobi L.) is an essential medicinal and edible plant with great economic value. Nevertheless, little is known about the molecular mechanisms underlying coix response to cadmium (Cd) stress. Coix germplasm, YY03-03, was exposed to 0 (control), 15, and 30 mg kg-1 Cd. We observed that YY03-03 exhibited low Cd absorption and transportation capacity. Further, various enrichment and translocation factors decreased under Cd stress. Moreover, under Cd stress levels of 15 and 30 mg kg-¹, transpiration rate, stomatal conductance, and photosynthetic rate significantly decreased (by 54.0

Research on quality homogenization of preparations from multi-part botanical materials based on pseudotargeted metabolomics and machine learning: a case study of Cocculus orbiculatus (L.) DC.

The use of multiple medicinal parts from a single herbal source for disease treatment is a common practice in traditional Chinese medicine. However, variations in the ratio of these parts can lead to inconsistencies in product quality. Therefore, ensuring uniform quality by controlling the ratio of different plant parts is a key challenge for Traditional Chinese Medicine (TCM) products. In this study, UHPLC-Q-TOF-MS was employed to acquire MS data of Cocculus orbiculatus (L) DC. (C. orbiculatus) quality control samples under different collision energies. Ion pairs were screened using the MRM-I

Metabolic profiling, in-situ spatial distribution, and biosynthetic pathway of functional metabolites in Dendrobium nobile stem revealed by combining UPLC-QTOF-MS with MALDI-TOF-MSI.

The stem of Dendrobium nobile Lindl. (Orchidaceae), called "Shihu" in traditional Chinese medicine, is a well-known medicinal and edible plant material in China. It is used as an antipyretic, analgesic, and tonic to nourish the stomach and Yin (i.e., to improve the production of body fluids). These therapeutic properties are attributed to its alkaloids, sesquiterpenoids, bibenzyls, fluorenones, and phenanthrenes. However, a comprehensive understanding of these metabolites and their spatial distribution in stems is lacking. In this study, ultra-performance liquid chromatography/quadrupole time-

ATP-citrate lyase B (ACLB) negatively affects cell death and resistance to Verticillium wilt.

BACKGROUND: ATP-citrate lyase (ACL) plays a pivotal role in histone acetylation and aerobic glycolysis. In plant, ACL is a heteromeric enzyme composed of ACLA (45 kD) and ACLB (65 kD). So far, the function of ACL genes in cotton still remains unknown. RESULTS: Here, we identified three ACLA homologous sequences and two ACLB homologous in each genome/sub-genome of cotton species. Silencing ACLB in cotton led to cell death at newly-grown leaves and stem apexes. Simultaneously, in ACLB-silenced plants, transcription factors related to senescence including SGR, WRKY23 and Osl57 were observed to b

Pharmacognostic standardization of Turnera aphrodisiaca Ward.

Turnera aphrodisiaca Ward (Turneraceae) has been used traditionally as an aphrodisiac, stimulant, nerve tonic, and laxative and in kidney, menstrual, and pregnancy disorders. Despite a long tradition of use in the treatment of various ailments, no systematic phytochemical and pharmacological work has ever been carried out on T. aphrodisiaca. The authors suggest that the major stumbling block in systematic exploration of the plant is non-availability of authentic plant material. In the present investigation, various pharmacognostic standards for the plant have been generated so that authentic T

Proteomics of isolated sieve tubes from Nicotiana tabacum: sieve element-specific proteins reveal differentiation of the endomembrane system.

Symplasmicly connected cells called sieve elements form a network of tubes in the phloem of vascular plants. Sieve elements have essential functions as they provide routes for photoassimilate distribution, the exchange of developmental signals, and the coordination of defense responses. Nonetheless, they are the least understood main type of plant cells. They are extremely sensitive, possess a reduced endomembrane system without Golgi apparatus, and lack nuclei and translation machineries, so that transcriptomics and similar techniques cannot be applied. Moreover, the analysis of phloem exudat
