Investigation of chromosome 2q in osteoarthritis of the hand: no significant linkage in a Tasmanian population.

BACKGROUND: Previous studies have suggested a strong genetic component to osteoarthritis (OA), especially that of the hand, and three linkage studies have suggested the existence of susceptibility loci in disparate regions of chromosome 2q. OBJECTIVE: To examine for linkage to 2q in a Tasmanian population of women and men with familial hand OA. METHODS: Hand OA (distal interphalangeal, carpometacarpal, and Heberden's nodes) was assessed by a combination of hand photographs and radiographs. A non-parametric linkage (NPL) analysis was performed on chromosome 2q of 69 members in 22 families wit

Two quantitative trait loci influence whipworm (Trichuris trichiura) infection in a Nepalese population.

BACKGROUND: Whipworm (Trichuris trichiura) infection is a soil-transmitted helminth infection that affects >1 billion people. It is a serious public health problem in many developing countries and can result in deficits in growth and cognitive development. In a follow-up study of significant heritability for whipworm infection, we conducted the first genome scan for quantitative trait loci (QTL) influencing the heritability of susceptibility to this important parasitic disease. METHODS: Whipworm egg counts were determined for 1,253 members of the Jirel population of eastern Nepal. All individ

"Higher order" addiction molecular genetics: convergent data from genome-wide association in humans and mice.

Family, adoption and twin data each support substantial heritability for addictions. Most of this heritable influence is not substance-specific. The overlapping genetic vulnerability for developing dependence on a variety of addictive substances suggests large roles for "higher order" pharamacogenomics in addiction molecular genetics. We and others have now completed genome-wide association (GWA) studies of DNAs from individuals with dependence on a variety of addictive substances versus appropriate controls. Recently reported replicated GWA observations identify a number of genes based on com

Role of selection and inbreeding on the incidence of cutaneous malignant melanoma in Sinclair swine.

This paper reports the quantitative analysis of the historical database of a herd of Sinclair swine affected by cutaneous malignant melanoma. The herd was under partial and non-systematic selection for melanoma susceptibility (animals having at least one tumour during the first 6 weeks of life). Weighted selection differentials for the number of tumours at birth and the number of tumours at 6 weeks were generally positive and between -0.43 and 4.76 tumours for the number of tumours at 6 weeks. Estimates of the heritability for number of tumours at birth and at 6 weeks using 1934 animals were 0

Heritabilities of intraocular pressure in the population of Korea: the Korean National Health and Nutrition Examination Survey 2008-2009.

IMPORTANCE: Intraocular pressure (IOP) is the principal modifiable risk factor for the progression of primary open-angle glaucoma. Studies that have measured the IOP directly in large numbers of matched parent-offspring pairs are limited in Asian populations. OBJECTIVE: To compare IOPs in parents and their offspring in Korea. DESIGN, SETTING, AND PARTICIPANTS: Family-based cohort study examining data from the Korean National Health and Nutrition Examination Survey conducted between 2008 and 2009. Data were obtained from 9700 participants from Korea. MAIN OUTCOMES AND MEASURES: Familial corr

Quantitative trait loci that modify the sootiness of yellow pigmentation in KK-A(y)/a mice.

Compared with C57BL/6J-A(y)/a, KK-A(y)/a mice have yellow fur that is markedly darker. Furthermore, there is a considerable variation in the tone of color with a continuous range in F(2) progeny produced from C57BL/6J females and KK-A(y)/a males. The aims of this study are to reveal the phenotypic differences between the two A(y) congenic strains and to elucidate the genetic factors responsible for the sooty yellow pigmentation in the KK background. On the basis of a chemical analysis, the sootiness in KK-A(y)/a was the result of increased eumelanin (PTCA) and decreased pheomelanin (AHP). A st

Identification of quantitative trait loci governing arthritis severity and humoral responses in the murine model of Lyme disease.

A spectrum of disease severity has been observed in patients with Lyme disease, with approximately 60% of untreated individuals developing arthritis. The murine model of Lyme disease has provided strong evidence that the genetic composition of the host influences the severity of arthritis following infection with Borrelia burgdorferi: infected C3H mice develop severe arthritis while infected C57BL/6N mice develop mild arthritis. Regions of the mouse genome controlling arthritis severity and humoral responses during B. burgdorferi infection were identified in the F2 intercross generation of C3H

White matter integrity as an intermediate phenotype: exploratory genome-wide association analysis in individuals at high risk of bipolar disorder.

White matter integrity, as measured using diffusion tensor imaging (DTI), is reduced in individuals with bipolar disorder (BD), their unaffected relatives and carriers of specific risk-alleles. Fractional anisotropy (FA), an index of white matter integrity, is highly heritable but the genetic architecture of this trait has received little investigation. In this study we performed a genome-wide association study with FA as quantitative phenotype, in unaffected relatives of patients with BD (N=70) and a matched control group (N=80). Amongst our top results were SNPs located in genes involved in

Is really endogenous ghrelin a hunger signal in chickens? Association of GHSR SNPs with increase appetite, growth traits, expression and serum level of GHRL, and GH.

Chicken growth hormone secretagogue receptor (GHSR) is a receptor for ghrelin (GHRL), a peptide hormone produced by chicken proventriculus, which stimulates growth hormone (GH) release and food intake. The purpose of this study was to search for single nucleotide polymorphisms (SNPs) in exon 2 of GHSR gene and to analyze their effect on the appetite, growth traits and expression levels of GHSR, GHRL, and GH genes as well as serum levels of GH and GHRL in Mandara chicken. Two adjacent SNPs, A239G and G244A, were detected in exon 2 of GHSR gene. G244A SNP was non-synonymous mutation and led to r

Genome-wide association study of primary open angle glaucoma risk and quantitative traits.

PURPOSE: Primary open angle glaucoma (POAG) is a characteristic optic neuropathy which progresses to irreversible vision loss. Few genes have been detected that influence POAG susceptibility and other genes are therefore likely to be involved. We analyzed carefully characterized POAG cases in a genome-wide association study (GWAS). METHODS: We performed a GWAS in 387 POAG cases using public control data (WTCCC2). We also investigated the quantitative phenotypes, cup:disc ratio (CDR), central corneal thickness (CCT), and intra-ocular pressure (IOP). Promising single nucleotide polymorphisms (S

Association of the rs3743205 variant of DYX1C1 with dyslexia in Chinese children.

BACKGROUND: Dyslexia is a learning disability that is characterized by difficulties in the acquisition of reading and spelling skills independent of intelligence, motivation or schooling. Studies of western populations have suggested that DYX1C1 is a candidate gene for dyslexia. In view of the different languages used in Caucasian and Chinese populations, it is therefore worthwhile to investigate whether there is an association of DYX1C1 in Chinese children with dyslexia. METHOD AND RESULTS: Eight single nucleotide polymorphisms (SNPs) were genotyped from three hundred and ninety three indivi

A novel Z-score-based method to analyze candidate genes for age-related hearing impairment.

OBJECTIVE: Approximately half of the variance of Age-Related Hearing Impairment (ARHI) is attributable to environmental risk factors, and the other half to genetic factors. None of these genes has ever been identified, but the genes involved in monogenic nonsyndromic hearing impairment are good candidates. Here we define and validate a quantitative trait value for ARHI, correcting for age and gender, to allow the genetic study of ARHI as a quantitative trait. DESIGN: Based on the ISO 7029 standard, we convert audiometric data into a Z-score, an age- and gender-independent value expressing to

Resistance to xenobiotic-induced autoimmunity maps to chromosome 1.

Although evidence indicates that environmental factors play a major role in precipitating systemic autoimmunity in genetically susceptible individuals, little is known about the mechanisms involved. Certain heavy metals, such as mercury, are potent environmental immunostimulants that produce a number of immunopathologic sequelae, including lymphoproliferation, hypergammaglobulinemia, and overt systemic autoimmunity. Predisposition to such metal-induced immunopathology has been shown to be influenced by both MHC and non-MHC genes, as well as susceptibility to spontaneous lupus, in mice and othe

Recent progress in the molecular genetics of sclerosing bone dysplasias.

Bone mineral density is a quantitative trait influenced by both genetic and environmental factors. Epidemiological and twin studies have shown heritability for the variance of bone density of up to 80%. The role of genetic factors in bone homeostasis also is clearly illustrated by the existence of an extended group of genetic conditions associated with an abnormal bone density. These conditions are mainly monogenic in that they are caused by mutations in one gene. Recent developments in molecular genetics and genomics have resulted in a dramatically increased power to identify such disease-cau

Assessing bone mineral density in vivo: digitized fluoroscopy and ultrasound.

The genetic component of osteoporosis in caged laying hens is large, and a method for detecting hens susceptible to fracture could be useful in breeding programs. A radiographic absorptiometry film method was modified by video digitization from an image intensifier and computer analysis and termed digitized fluoroscopy (DF). Humeral and ulnar DF values were measured in 165 hens during lay. Relationships (P < 0.001) were seen between DF assessments from 25 wk onward and postmortem measurements at 70 wk. We conclude that DF can detect poor bones in hens early but is problematic. Quantitative ult

Genetic parameters and QTL mapping for novel metabolic traits in early-lactation Holsteins.

Early lactation in dairy cows is characterized by negative energy balance and compromised immune function that could lead to metabolic or inflammatory diseases. Circulating biochemical blood variables are increasingly used as indicators of metabolic and inflammatory diseases in many species. This study aimed to estimate genetic parameters and identify candidate genes and quantitative trait loci (QTL) associated with serum proteins (total protein, albumin, globulin, and albumin-to-globulin ratio), liver enzymes (gamma-glutamyltransferase [GGT], aspartate-amino-transferase [AST], glutamate dehyd

Inheritance of hormonal stress response and temperament in infant rhesus macaques (Macaca Mulatta): Nonadditive and sex-specific effects.

OBJECTIVE: Early life interindividual variation in hypothalamic-pituitary-adrenal (HPA) reactivity to stress is predictive of later life psychological and physical well-being, including the development of many pathological syndromes that are often sex-biased. A complex and interactive set of environmental and genetic causes for such variation has been implicated by previous studies, though little attention has been paid to nonadditive effects (e.g. dominance, X-linked) or sex-specific genetic effects. METHOD: We used a large pedigreed sample of captive 3-4 months old infant rhesus macaques (N

What does infrared thermography tell us about the evolutionary potential of heat tolerance in endotherms?

Anthropogenic climate change affects wild animal populations through increasing average temperatures and more frequent extreme climatic events. Endotherms have evolved the capacity to regulate their body temperature but little is still known about how they can physiologically adapt to the pace of global warming. Adaptive responses would require that heat-tolerance mechanisms, such as the capacity to withstand high body temperatures and regulate evaporative water loss, exhibit sufficient heritable genetic variation for selection to act upon. Unfortunately, the quantitative genetics of these tra

Quantitative genetic analyses of traits related to calcium and phosphorus metabolism in two different laying hen strains.

BACKGROUND: Phosphorus (P) and calcium (Ca) are essential minerals for laying hens. Phosphorus in plant feeds is mainly stored as phytate and needs to be released by the enzyme phytase. Due to the high requirement of Ca, laying hens exhibit limited endogenous phytate degradation and thus plant-P is available to a limited extent. Mineral P supplemented to laying hen feed reduces phytate degradation further and decreases myo-inositol release in the intestinal tract, which is known to have many functions in poultry metabolism. The focus of this study was the investigation of P and Ca metabolism i

A genetic epidemiological study in British adults and older adults shows a high heritability of the combined indicator of vitamin B12 status (cB12) and connects B12 status with utilization of mitochondrial substrates and energy metabolism.

Vitamin B12 deficiency is common among older adults. However, the most commonly used marker of deficiency, total serum vitamin B12 (B12), is not sensitive enough to diagnose true deficiency in a significant proportion of the population. The combined indicator of B12 status (cB12), formulated as a composite score of various biomarkers of vitamin B12 status (which also accounts for low folate status and age) has been shown to offer a more robust and powerful test to diagnose B12 deficiency. There are no epidemiological studies of cB12 variability in older adults. We carried out a twin study to c
