[Radiation-induced hematopoietic disorders].

Hematopoietic stem cells are one of the most sensitive tissues to radiation exposure. Epidemiological studies of atomic bomb survivors in Nagasaki and Hiroshima revealed a radiation dose-response relationship for acute myeloid leukemia, acute lymphoblastic leukemia, and Chronic myelogenous leukemia. In our study of myelodysplastic syndrome (MDS) in Nagasaki, the relative risk was higher in the proximal-exposed group. Additionally, the frequency of chromosome abnormalities associated with poor prognosis was higher in the proximal-exposed group than in the non-exposed group. However, no associat

[Frequency, dynamics, and structure of congenital malformations in populations under long-term exposure to ionizing radiation].

Information on the dynamics, frequency, and structure of congenital malformations (CMs) and chromosomal diseases in two generations of rural populations from the Semipalatinsk region was obtained for the first time. The tests performed in the Semipalatinsk nuclear test site (STS) for many years were demonstrated to deteriorate the genetic health of the populations of the zones of extreme and maximum radiation risks. Mutagenic and teratogenic effects of long-term ionizing irradiation were detected. These were mainly expressed as an increase in the frequencies of strictly registered CMs (SR CMs)

[Ultraviolet radiation in the pathogenesis of lupus erythematosus].

Photosensitivity represents an increased inflammatory reaction to sunlight, which can be observed particularly in the autoimmune disease lupus erythematosus. Cutaneous lupus erythematosus (CLE) can be provoked by ultraviolet (UV) radiation and can cause both acute, nonscarring and chronic, scarring skin changes. In systemic lupus erythematosus, on the other hand, provocation by UV radiation can lead to flare or progression of systemic involvement. The etiology of lupus erythematosus is multifactorial and includes genetic, epigenetic and immunologic mechanisms. In this review, we address the ef

The prevalence of chromosomal aberrations in Argentine air crew members.

The authors analyzed the effects of chronic exposure of Argentine air crew members to low doses of ionizing radiations. Genetic damage induced by either low doses or low rates of ionizing radiation was higher than expected. Seventy-one heparinized blood samples were obtained from technical ground workers (group A; n = 10), pilots of domestic flights (group B; n = 14), pilots of transequatorial flights (group C; n = 17;), pilots of transpolar flights (group D; n = 17) and retired pilots (group E; n = 10) The frequency of dicentric chromosomes was higher in groups B and C compared with groups D

Controversial cytogenetic observations in mammalian somatic cells exposed to radiofrequency radiation.

During the years 1990-2003 a large number of investigations were conducted using rodents, cultured rodent and human cells, and freshly collected human blood lymphocytes to determine the genotoxic potential of exposure to radiofrequency (RF) radiation. The results of most of these studies (58%) did not indicate increased damage to the genetic material (assessed from DNA strand breaks, incidence of chromosomal aberrations, micronuclei and sister chromatid exchanges) in cells exposed to RF radiation compared to sham-exposed and/or unexposed cells. Some investigations (23%) reported an increase in

Mouse genomic associations with in vitro sensitivity to simulated space radiation.

Exposure to ionizing radiation is considered by NASA to be a major health hazard for deep space exploration missions. Ionizing radiation sensitivity is modulated by both genomic and environmental factors. Understanding their contributions is crucial for designing experiments in model organisms, evaluating the risk of deep space (i.e. high-linear energy transfer, or LET, particle) radiation exposure in astronauts, and also selecting therapeutic irradiation regimes for cancer patients. We identified single nucleotide polymorphisms in 15 strains of mice, including 10 collaborative cross model str

The Birth of the Illegitimate Linear No-Threshold Model: An Invalid Paradigm for Estimating Risk Following Low-dose Radiation Exposure.

This paper examines the birthing process of the linear no-threshold model with respect to genetic effects and carcinogenesis. This model was conceived >70 years ago but still remains a foundational element within much of the scientific thought regarding exposure to low-dose ionizing radiation. This model is used today to provide risk estimates for cancer resulting from any exposure to ionizing radiation down to zero dose, risk estimates that are only theoretical and, as yet, have never been conclusively demonstrated by empirical evidence. We are literally bathed every second of every day in lo

Only the strong survive: therapeutic selective pressure drives medulloblastoma leptomeningeal metastasis.

Medulloblastoma (MB) is the most common malignant tumor in the central nervous system in childhood and regularly metastasizes to the leptomeninges following radiation therapy. Using patient-derived medulloblastoma models and genetically engineered mouse models, Nör et al. observed enhanced inflammation and infiltration of myeloid cells within the brain following irradiation. The authors identified inflammatory cytokines and the resulting breakdown of blood-brain barriers as the main culprits of MB leptomeningeal metastasis. This study demonstrated that targeting inflammation through the use of

Multidisciplinary Management of Angiosarcoma - A Review.

Angiosarcomas (AS) are a diverse group of soft tissue sarcomas, arising from blood and lymphatic vessels. They frequently present in the elderly, and in patients with previous radiation or lymphedema. A wide range of genetic derangements contribute to their development, and AS histology is often high-grade in keeping with aggressive disease biology. The clinical presentation, while often innocuous, is marked by its infiltrative and aggressive nature, with a proclivity for metastatic spread, and outcomes are often poor. Surgery is performed for localized, resectable cases. A multidisciplinary a

[Anthropological aspects of skin color].

The highly variable spectrum of colors of human skin depends primarily on differences in the activity of the melanocytes and the melanin content, besides other pigments such as carotenoids. In case of a homogenous early-man-color, the following moments may be relevant for the genetic differentiation: primary factors (latitude, climate, radiation), secondary (vegetation, morbific agents) and tertiary (nutritional status, diseases) factors. They modify selection, resulting from population movement, adaptation to new habitats, and temporary isolation. Regarding the protective function of the inte

Frequency of reciprocal translocations and dicentrics induced in human blood lymphocytes by X-irradiation as determined by fluorescence in situ hybridization.

This study was designed to test the scoring efficiency of reciprocal translocations and dicentrics induced by X-irradiation in vitro using the fluorescence in situ hybridization (FISH) technique. An excess was found in the frequencies of reciprocal translocations relative to those of dicentrics by measurement with FISH at the first cell division after irradiation (translocation:dicentric approximately 60:40). However, when the same metaphases were also evaluated sequentially by a conventional staining method, the ratio of about 50:50 was restored. This was due in part to misclassification of c

Rapid translocation frequency analysis in humans decades after exposure to ionizing radiation.

This paper presents an analysis of the utility of fluorescence in situ hybridization (FISH) with whole-chromosome probes for measurement of the genomic frequency of translocations found in the peripheral blood of individuals exposed to ionizing radiation. First, we derive the equation: Fp = 2.05fp(1-fp)FG, relating the translocation frequency, Fp, measured using FISH to the genomic translocation frequency, FG, where fp is the fraction of the genome covered by the composite probe. We demonstrate the validity of this equation by showing that: (a) translocation detection efficiency predicted by t

Case Report: Solid variant of papillary thyroid carcinoma in a young adult with Turner syndrome with chronic thyroiditis.

Turner syndrome is associated with an increased risk of developing several neoplasms. In particular, a clinical feature of Turner syndrome with chronic thyroiditis implies a relationship with thyroid malignancies. We report a very rare case of a solid variant of papillary thyroid carcinoma that was identified during a follow-up of chronic thyroiditis in a 22-year-old woman with Turner syndrome. The patient had no notable history of radiation exposure. No genetic mutations relating to the occurrence of the solid variant of papillary thyroid carcinoma, including RET/PTC rearrangements and mutati

Patterns of second malignant neoplasms in children.

A search of the records of 10 pediatric oncology centers revealed 102 children with more than one malignant neoplasm. In this group of 102 patients, all pediatric cancers were seen as initial lesions, but Wilms' tumor and retinoblastoma were over-represented and leukemia and brain tumors underrepresented. Survival variation as well as tumor susceptibility may be responsible for this disproportion. Osteosarcomas and chondrosarcomas were the most frequent second malignant neoplasms (SMN). Embryonal tumors were rare as SMN and adult-type tumors (carcinomas) appeared at earlier than expected ages,

Neonatal sunburn and melanoma in mice.

Retrospective epidemiological data have indicated that cutaneous malignant melanoma may arise as a consequence of intense, intermittent exposure of the skin to ultraviolet radiation, particularly in children, rather than from the cumulative lifetime exposure that is associated with other forms of skin cancer. Here we use a genetically engineered mouse model to show that a single dose of burning ultraviolet radiation to neonates, but not adults, is necessary and sufficient to induce tumours with high penetrance which are reminiscent of human melanoma. Our results provide experimental support fo

Improved outcomes and treatment guidance in pediatric therapy-related AML: AML-BFM study group recommendations.

Therapy-related acute myeloid leukemia (AML; tAML) is one of the most feared therapy-emergent complications. This study aims to determine the clinical and pathological significance and define therapeutic implications and poor prognosticators in pediatric tAML. We analyzed a total of 119 pediatric patients (aged 2-20 years) who were centrally diagnosed with tAML within the Acute Myeloid Leukemia Berlin-Frankfurt-Münster (AML-BFM) study group between 1993 and 2019. Compared with de novo AML, tAML was associated with decreased white blood count and involvement of the central nervous system. Laten

Personal and Familial Malignancy History in Patients with Malignant Peripheral Nerve Sheath Tumors with a Focus on Sporadic Tumors.

BACKGROUND: Malignant peripheral nerve sheath tumors (MPNSTs) occur sporadically, in patients with neurofibromatosis 1, and in patients with prior radiation therapy. The incidence of unrelated prior malignancies and family history of malignancy in patients with MPNSTs has not been previously described. METHODS: A retrospective search for cases of MPNSTs at our institution for the years 1994-2019 was performed. The electronic medical record was reviewed for documentation of personal and family history of malignancies. RESULTS: The study included 331 patients. Of patients, 301 had documentatio

[Practical aspects of mutagenesis--today and later].

The struggle of medicine and biology with the phenomenon mutation is discussed from a philosophical (C. BRESCH: "Zwischenstufe Leben") and a practical point of view. In the beginning age of the intellectual evolution mutations have become a pruely negative relict of the declining phase of the biological evolution. Effective means of medical genetics suitable in reducing the number of manifest mutants are: prenatal genetic diagnoses in as many pregnancies as possible; protection from exogenous mutagenic sources such as chemical mutagens and consequent protection from ionizing radiations, especi
