External quality assessment for molecular detection of sand fly-borne phleboviruses circulating in the Mediterranean Basin.

BACKGROUND: Sand fly-borne phleboviruses (SbPV) are globally distributed and pose potential public health risks. Despite increased detection in recent decades, detailed knowledge of their ecology, characteristics and clinical relevance remains limited. Many cases of SbPV infection likely go unreported or misdiagnosed due to limited awareness and the lack of standardized screening. The External Quality Assessment (EQA) reported herein was organized within the framework of the European Union CLIMOS (EU Climate Monitoring and Decision Support Framework for Sand Fly-borne Diseases Detection and Mi…

A novel sand cat swarm optimization algorithm-based SVM for diagnosis imaging genomics in Alzheimer's disease.

In recent years, brain imaging genomics has advanced significantly in revealing underlying pathological mechanisms of Alzheimer's disease (AD) and providing early diagnosis. In this paper, we present a framework for diagnosing AD that integrates magnetic resonance imaging (fMRI) genetic preprocessing, feature selection, and a support vector machine (SVM) model. In particular, a novel sand cat swarm optimization (SCSO) algorithm, named SS-SCSO, which integrates the spiral search strategy and alert mechanism from the sparrow search algorithm, is proposed to optimize the SVM parameters. The optim…

Genomic analysis of two phlebotomine sand fly vectors of Leishmania from the New and Old World.

Phlebotomine sand flies are of global significance as important vectors of human disease, transmitting bacterial, viral, and protozoan pathogens, including the kinetoplastid parasites of the genus Leishmania, the causative agents of devastating diseases collectively termed leishmaniasis. More than 40 pathogenic Leishmania species are transmitted to humans by approximately 35 sand fly species in 98 countries with hundreds of millions of people at risk around the world. No approved efficacious vaccine exists for leishmaniasis and available therapeutic drugs are either toxic and/or expensive, or …

Efficient rural sewage treatment with manganese sand-pyrite soil infiltration systems: Performance, mechanisms, and emissions reduction.

The application of soil infiltration systems (SISs) in rural domestic sewage (RDS) is limited due to suboptimal denitrification resulting from factors such as low C/N (<5). This study introduced filler-enhanced SISs and investigated parameter impacts on pollutant removal efficiency and greenhouse gas (GHG) emission reduction. The results showed that Mn sand-pyrite SISs, with hydraulic load ratios of 0.003 m3/m2·h and dry-wet ratios of 3:1, achieved excellent removal efficiency of COD (92.7 %), NH4+-N (95.8 %), and TN (76.4 %). Moreover, N2O and CH4 emission flux were 0.046 and 0.019 mg/m2·d, r…

AN OUTBREAK OF FELINE INFECTIOUS PERITONITIS IN THREE RELATED SAND CATS (FELIS MARGARITA) IN HUMAN CARE.

Feline infectious peritonitis (FIP) is a systemic disease in felid species caused by infection with mutated forms of feline coronavirus (FCoV), and outbreaks can devastate exotic felid populations in human care. Feline infectious peritonitis was diagnosed in three of four related juvenile sand cats (Felis margarita) from a single institution over a 6-wk period. Case 1 was a 7-mon-old male found deceased with no premonitory signs. Case 2, an 8-mon-old male (littermate to Case 1), and Case 3, a 6-mon-old male (from a different litter with identical parentage), were evaluated for lethargy and ano…

Freeze-thaw cycles induce diverse bacteria release behaviors from quartz sand columns with different water saturations.

Bacteria present in natural environment especially those in cold regions would experience freeze-thaw (FT) process during day-night and season turns. However, knowledge about the influence of FT on bacteria release behaviors in porous media was limited. In present study, the bacteria release behaviors from quartz sand columns without and with 1 and 3 FT treatment cycles under three water saturations (θ=100%, 90%, and 60%) were investigated. We found that for all three water saturated columns without FT treatment, negligible bacteria released from columns via background salt solution elution, w…

Particle size reduction along the digestive tract of fat sand rats (Psammomys obesus) fed four chenopods.

It is generally accepted that microbial digestion contributes little to digesta particle size reduction in herbivores, and that faecal particle size reflects mainly chewing efficiency, and may vary with diet. Nevertheless, a decrease in mean particle size (MPS) along the gastrointestinal tract (GIT) has been reported, especially in hindgut fermenters. However, to what degree the very fine particle fraction (non-food origin, especially microbes) affects MPS is unclear. Fat sand rats (Psammomys obesus, diurnal herbivores, n = 23, 175 ± sd 24 g) consumed one of four chenopods (natural dietary ite…

DNA barcoding and fauna of phlebotomine sand flies (Diptera: Psychodidae: Phlebotominae) from Los Tuxtlas, Veracruz, Mexico.

Mexico has great diversity of phlebotomine sand flies related to cases of leishmaniasis, yet few studies have dressed the molecular taxonomy of these sand fly species. The use of the cytochrome oxidase subunit 1 (COI) gene, as a DNA Barcode has facilitated the molecular identification of sand flies species worldwide. We use the DNA barcode as a useful tool for the identification of phlebotomine sand flies of the natural reserve Los Tuxtlas from Veracruz, México. A fragment of 536 bp of the COI gene was obtained from 36 individuals belonging to eight species of five genera (Dampfomyia, Lutzomyi…

[Soil Analysis of Potential Breeding Sites of Sand Flies (Diptera: Psychodidae) in Aichoun Locality, Central Morocco].

The epidemiology of leishmaniasis is related with habitat and behavior of the sand fly vector. Each species of sand flies (Diptera: Psychodidae) has a specific characteristic of their sites for the development of their immatures. Information on natural breeding sites of phlebotomine sand flies is limited, due to the difficulty of isolation of immature from the soil. This study, realized from May 2014 to September 2014, aimed to determine the effect of properties of soil on behavior and abundance of sand flies in breeding habitat in Aichoun locality. Sand flies were collected using sticky paper…

Pretreatment using Opuntia cochenillifera followed by household slow sand filters: technological alternatives for supplying isolated communities.

Household slow sand filter (HSSF) performance in continuous and intermittent flows was evaluated when influent water was treated with a natural coagulant extracted from Opuntia cochenillifera. The water under study, used as influent, had a turbidity of 111 ± 17.3 NTU. When clarifying the water with O. cochenillifera, the best condition obtained was 30  mg L-1 in natural pH (without correction), generating clarified water with turbidity satisfactory to filters operation (7.83 ± 2.32 NTU). The results indicated a better performance of continuous flow HSSF in turbidity removal (79.2% ± 8.39%) and…

Field study of the improved rapid sand fly exposure test in areas endemic for canine leishmaniasis.

BACKGROUND: Canine leishmaniasis (CanL) is a severe chronic disease caused by Leishmania infantum and transmitted by sand flies of which the main vector in the Western part of the Mediterranean basin is Phlebotomus perniciosus. Previously, an immunochromatographic test (ICT) was proposed to allow rapid evaluation of dog exposure to P. perniciosus. In the present study, we optimized the prototype and evaluated the detection accuracy of the ICT in field conditions. Possible cross-reactions with other hematophagous arthropods were also assessed. METHODOLOGY/PRINCIPAL FINDINGS: The ICT was optimi…

A randomized, blinded, controlled trial to assess sand fly mortality of fluralaner administered orally in dogs.

BACKGROUND: Leishmania infantum is the parasite responsible for the disease in humans known as zoonotic visceral leishmaniasis (ZVL). Dogs are considered the main domestic reservoir of ZVL and sand flies are the proven vectors. The use of systemic insecticides in dogs has been studied as an alternative strategy to control ZVL in endemic areas. One systemic insecticide in dogs, fluralaner, has a proven anti-sand fly effect in membrane-fed studies. However, the efficacy and duration on sand flies directly feeding from dogs treated with fluralaner remains unknown. METHODS: Direct feeding bioassa…

Feeding preferences of Lutzomyia longipalpis (Diptera: Psychodidae), the sand fly vector, for Leishmania infantum (Kinetoplastida: Trypanosomatidae).

Leishmania infantum, the causative agent of visceral leishmaniasis (VL) in Brazil, is spread mostly by the bite of the sand fly Lutzomyia longipalpis (Lutz & Neiva). We trapped sand flies in endemic neighborhoods near Natal, Brazil, where cases of human and dog VL were documented. Amplification of species-specific cytochrome b (Cyt b) genes by polymerase chain reaction revealed that sand flies from rural and periurban areas harbored blood from different sources. The most common source ofbloodmeal was human, but blood from dog, chicken, and armadillo was also present. We tested the preference f…
