Propagation of Polygonatum macranthum (Maxim.) Koidz. from immature seeds using a new sterilization procedure.

Natural seed germination is difficult to achieve in numerous plant species of wide economic importance. The germination of Polygonatum macranthum seeds takes as long as one and a half years under natural conditions. In addition, propagation by rhizome is also extremely slow in this species. Therefore, the natural propagation of P. macranthum through seeds or rhizome is not efficient. In this study, an efficient in vitro propagation system for P. macranthum from immature seeds with seed coat was developed, using a new surface sterilization protocol that utilized a low concentration of hypochlor…

High resolution microscopy to evaluate the efficiency of surface sterilization of Zea Mays seeds.

Surface sterilization of seeds is a key step in providing microorganisms-free seeds for numerous applications like understanding the role of seed-borne microorganisms in plant development, studying microbial cells-plant interactions by inoculating model microorganisms in a simplified system or selective cultivation of seed endobionts. However applying efficient treatment for surface sterilization of seeds without affecting the plant growth is not an easy task. In this study we aimed to provide an efficient surface sterilization treatment for maize seeds using i) hydrogen peroxide (HP), ii) sod…

Modified method for combined DNA and RNA isolation from peanut and other oil seeds.

Isolation of good quality RNA and DNA from seeds is difficult due to high levels of polysaccharides, polyphenols, and lipids that can degrade or co-precipitate with nucleic acids. Standard RNA extraction methods utilizing guanidinium-phenol-chloroform extraction has not shown to be successful. RNA isolation from plant seeds is a prerequisite for many seed specific gene expression studies and DNA is necessary in marker-assisted selection and other genetic studies. We describe a modified method to isolate both RNA and DNA from the same seed tissue and have been successful with several oil seeds …

The seeds and the worms: Ludwik Fleck and the early history of germ theories.

The Polish microbiologist and philosopher of science, Ludwik Fleck (1896-1961), was a pioneer in constructivist history and philosophy of science. Based on studies in the history of syphilis, Fleck hypothesized that many established scientific facts are linked, in their development, to pre-scientific "proto-ideas." In 1935, Fleck proposed that the history of germ theories could be approached through his thesis on proto-ideas. His proposal, however, remained little more than a vague suggestion and was never developed in further detail. This paper introduces the concept of proto-ideas and discus

Distribution of COX-negative mitochondria in myofibers of rats intoxicated with Senna occidentalis seeds.

We have described that administration of seeds or parts of the seed of Senna occidentalis (coffee senna) for long periods, induces histochemical changes in the skeletal muscles of hens and rats that are characteristic of a mitochondrial myopathy--as decrease of SDH and COX activity, with some COX negative fibers. In this experimental model of mitochondrial myopathy, as in many human mitochondrial diseases, there is a random distribution of COX negative fibers. Some fibers are completely COX negative while others are partially negative and others are completely positive. In the present work we …
