Application of Noninvasive Brain Stimulation for Sleep Quality Enhancement and Cognitive Improvement.

Sleep is a fundamental process for maintaining our physical and mental health by adjusting brain homeostasis and repairing axons to refresh memories. Due to its essentiality, sleep disorders and insufficiency can cause both physiological and behavioral risks. This report diverges from traditional medical treatments and focuses mainly on physiotherapy-based neuroregulatory techniques for sleep treatment. Noninvasive brain stimulation (NiBS) techniques have been developed to enhance patients' sleep and memory, including transcranial electrical stimulation (tES), transcranial magnetic stimulation

Sleep Abnormalities and Risk of Alzheimer's Disease.

PURPOSE OF REVIEW: This review aimed at investigating sleep abnormalities as risk factors for Alzheimer's disease (AD), with a focus on their potential utility in early disease detection and risk modification. RECENT FINDINGS: Impaired sleep quality, circadian misalignment, and disruptions in sleep architecture are significantly associated with an elevated risk of AD. Moreover, excessive or insufficient sleep, reductions in slow-wave and REM sleep, and fragmented rest-activity rhythms have been linked to early alterations in amyloid-β and tau biomarkers, even in cognitively unimpaired individ

Chemosensory event-related potentials during sleep--a pilot study.

Aim of the present pilot study was to investigate whether cortically generated chemosensory event-related potentials (ERPs) can be recorded during sleep. Chemosensory function during sleep was assessed in 14 healthy female volunteers. An overnight polysomnography was performed to assess nocturnal sleep and to classify sleep stages. Chemosensory ERPs were recorded using air-dilution olfactometry. H2S (4ppm) was used for olfactory and CO2 (40%, v/v) for trigeminal stimulation. Chemosensory ERPs could be recorded during sleep for both olfactory and trigeminal stimuli in some but not all subjects.

You Snooze, You Win? An Ecological Dynamics Framework Approach to Understanding the Relationships Between Sleep and Sensorimotor Performance in Sport.

Sleep has a widespread impact across different domains of performance, including sensorimotor function. From an ecological dynamics perspective, sensorimotor function involves the continuous and dynamic coupling between perception and action. Sport performance relies on sensorimotor function as successful movement behaviors require accurate and efficient coupling between perceptions and actions. Compromised sleep impairs different aspects of sensorimotor performance, including perceptual attunement and motor execution. Changes in sensorimotor performance can be related to specific features of

Sleep variables are unaltered by zolantidine in rats: are histamine H2-receptors not involved in sleep regulation?

The effects of the H1-receptor antagonist diphenhydramine and the brain-penetrating H2-receptor antagonist zolantidine were studied in rats implanted for chronic sleep recordings. Diphenhydramine (1.0-4.0 mg/kg) significantly increased slow wave sleep and decreased wakefulness. Zolantidine (0.25-8.0 mg/kg) had no significant effects on any of the sleep parameters examined. One possibility is that zolantidine did not enter the brain in sufficient concentration to produce significant changes on sleep and wakefulness. Another possibility is that blockade of H2-receptor involved parts of the brain

Obstructive sleep apnea syndrome (OSAS): Pathophysiology in Estonians.

The aim of the study was to clarify the roles of age, obesity, smoking, alcohol, pathoanatomy and -physiology in Estonian's OSAS. For this 164 randomly chosen such patients were selected in different regions of Estonia. They underwent naso-oropharyngeal examination, physical examination of craniofacial abnormalities, and polysomnography. They also completed a self-reported questionnaire about smoking, alcohol use, excessive daytime sleepiness, hypertension, cardiac disorders, headaches, concentration disorders, and recurrent upper-airway diseases. The patients (129 men; 35 women) aged between

Developmental and/or Epileptic Encephalopathy with Spike-Wave Activation in Sleep: From Thalamocortical Mechanisms to Precision Therapy.

Developmental and/or epileptic encephalopathy with spike-wave activation in sleep (D/EE-SWAS), previously described as continuous spike-wave during slow-wave sleep (CSWS) or electrical status epilepticus in sleep (ESES), is a childhood-onset disorder characterized by cognitive, language, behavioral, and/or motor regression or stagnation associated with marked activation of epileptiform discharges during non-rapid eye movement sleep. Three themes are central to the evolving understanding of D/EE-SWAS. First, it is a network disorder in which thalamocortical dysfunction, impaired sleep-dependent

Cannabinoid receptor activation disrupts the internal structure of hippocampal sharp wave-ripple complexes.

Cannabinoid agonists impair hippocampus-dependent learning and memory. Using mouse hippocampal slice preparations, we examined the effect of anandamide, an endogenous cannabinoid, on sharp wave-ripple (SW-R) complexes, which are believed to mediate memory consolidation during slow-wave sleep or behavioral immobility. Anandamide reduced the frequency of SW-Rs recorded from the CA3 region, and this effect was abolished by AM251, a cannabinoid CB1-receptor antagonist. We further addressed the action of anandamide using a functional multineuron calcium imaging technique. Anandamide reduced the fir

Effects of locus coeruleus lesions upon sleeping and waking in the rabbit.

The effects of radiofrequency lesions in the region of the locus coeruleus (LC) upon sleep--waking states and behaviors were investigated in chronically implanted New Zealand White rabbits. Polygraphic recordings were taken prior to and at 5- and 14-day intervals following lesioning. In animals exhibiting absence of paradoxical sleep and the presence of bizarre motor behavior, additional recordings were taken 30 days postlesion. Prelesion sleep-wakefulness pattern data were comparable to those previously observed in intact rabbits, including the recently reported absence of sustained PS-relate

Parasomnias. Things that go thump in the night.

BACKGROUND: General practitioners are well versed with patients presenting to the surgery with sleep symptoms, however, the approach to evaluating these symptoms is often haphazard. Insomnia is the commonest presenting complaint. Sleep apnoea, although readily treatable, carries with it significant morbidity and mortality. OBJECTIVE: This article aims to highlight those disorders that occur infrequently during the night but which interrupt sleep--the parasomnias. DISCUSSION: Knowledge of the spectrum of parasomnias and their symptoms usually allows a reasonably accurate clinical diagnosis an

[Alcohol and sleep disorders].

Despite an initial sedative effect, alcohol disrupts sleep persistently and should not be used as a sleeping aid. Nocturnal withdrawal symptoms may lead to an increased duration of wakefulness, and to tachycardia and sweating in the second half of the night. It is not known by which mechanism alcohol affects sleep; however, effects do not appear to depend on the stimulation of benzodiazepine receptors or the antagonism at adenosine receptors. Alcohol can exacerbate primary sleep disturbances such as sleep apnea and nocturnal myoclonus, and thereby contribute to excessive daytime sleepiness. Th

[Thyroid gland and sleep].

A set of data suggests that the thyroid gland plays a role in the bi-directional interaction between the electrophysiological and the endocrine components of sleep, e.g. the nonREM-REM-cycle and the patterns of nocturnal hormone secretion, respectively. In detail thyroid-stimulating hormone (TSH) and thyroxin (T4) show circadian rhythms. A specific relationship was observed between TSH and REM sleep. Blunted TSH levels were found in healthy elderly subjects and, probably due to overactivity of corticotropin-releasing hormone in patients with depression in comparison to young normal controls. P

Sleep deprivation in prion protein deficient mice sleep deprivation in prion protein deficient mice and control mice: genotype dependent regional rebound.

We have previously reported a larger and more prolonged increase of slow wave activity (SWA) in NREM sleep after sleep deprivation (SD) in prion protein deficient mice (PrP) compared to wild-type mice. Regional differences in the SWA increase were investigated by comparing the effect of 6 h SD on a frontal and occipital derivation in PrP deficient mice and wild-type mice. The larger increase of SWA after SD in PrP deficient mice was restricted to the occipital derivation. The difference appeared after the waking-NREM sleep transitions, making it unlikely that PrP is involved in the mechanisms

Neurophysiological aspects of the completely locked-in syndrome in patients with advanced amyotrophic lateral sclerosis.

OBJECTIVE: Amyotrophic lateral sclerosis (ALS) patients in completely locked-in syndrome (CLIS) are incapable of expressing themselves, and their state of consciousness and awareness is difficult to evaluate. Due to the complete paralysis included paralysis of eye muscles, any assessment of the perceptual and psychophysiological state can only be implemented in passive experimental paradigms with neurophysiological recordings. METHODS: Four patients in CLIS were investigated in several experiments including resting state, visual stimulation (eyes open vs eyes closed), auditory stimulation (mo

The Hypothalamic Pituitary Thyroid Axis and Sleep.

Sleep has a bidirectional relationship with the hypothalamic-pituitary-thyroid (HPT) axis, and both these homeostatic processes are inter-dependent for robust physiological functioning. The quality and quantity of sleep influence the circadian pattern of TSH and thyroid hormone secretion. Short term sleep restriction significantly reduces the amplitude of nocturnal TSH secretion and may modulate active thyroid hormone secretion, likely through an increased sympathetic tone. Conversely, TSH and active thyroid hormone affect the quantity and architecture of sleep. For instance, low TSH values ar

Effects of antipsychotic treatment on polysomnographic measures in schizophrenia: a replication and extension.

OBJECTIVE: The authors sought to replicate and extend previous observations of improvement in some EEG sleep measures during the course of antipsychotic treatment in schizophrenia patients. METHOD: Fourteen medication-free patients with schizophrenia underwent 2 nights of sleep EEG monitoring before and after 3-4 weeks of treatment with clinically determined doses of haloperidol or thiothixene. RESULTS: Measures of sleep continuity improved consistently. REM latency increased, although five of 14 patients continued to exhibit short REM latencies (less than 60 minutes). Stage 3 sleep increase

[A multicenter trial of a device for treating obstructive sleep apnea by continuous positive pressure].

The efficacy and tolerance of a nasal CPAP device marketed in France (Pression +, Sefam) for the treatment of obstructive sleep apnoea syndromes have been evaluated in a co-operative trial including 12 patients. This study confirmed the efficacy of nasal CPAP on sleep parameters: total sleep time was increased; light non-REM sleep was diminished; slow-wave sleep and REM sleep were augmented; sleep apnoeas were eliminated completely or almost completely; oxygen saturation was markedly improved. At one month follow-up, most clinical features were improved; daytime blood gases showed little chang

Longitudinal Alterations in Sleep EEG Biomarkers of Memory Consolidation in Middle-Aged and Older Adults.

The precise coordination of slow oscillations (SO) and sleep spindles during non-rapid eye movement (NREM) sleep supports memory consolidation and may serve as a sensitive marker of cognitive aging. However, longitudinal changes in their oscillatory dynamics in midlife and older age remain poorly understood. Using polysomnography with high-density EEG at two timepoints over ~2.5 years, we examined changes in local NREM slow wave (SW), sleep spindle (occurring in the 11-16 Hz sigma range), and SO-sigma coupling strength in cognitively unimpaired middle-aged to older adults at risk for Alzheimer

Transmission and integration of biologically meaningful olfactory information after bilateral transection of the lateral olfactory tract in the rat.

The contribution of the olfactory inputs conveyed by the lateral olfactory tract (LOT) to the central olfactory areas in the onset of behavioral and electrophysiological responses i.e. multiunit activity of mitral cells was studied in rats which were submitted to a bilateral transection of LOT. The characteristic emotional reactions elicited in sham-operated rats by biologically meaningful odorants--odors of predator or of conspecific--were no longer observed after such lesion. At the olfactory bulb level, the differential habituation of mitral cell electrical responses according to the biolog
