Baclofen affects the semicircular canals but not the otoliths in humans.

CONCLUSION: This study showed that GABAB agonist baclofen (10 mg) affects the semicircular canals (SCCs), both centrally and peripherally, but does not influence the otolithic function. OBJECTIVES: The aim of the study was to identify the effects of baclofen on the complete vestibular system, i.e. semicircular canals, saccules and utricles. METHODS: The study had a double-blind, placebo-controlled, repeated measures design and was conducted on healthy male volunteers. With electronystagmography (ENG), the SCC function was evaluated, whereas utricular function was determined by means of unila

[Validation of the French version of the situational characteristics questionnaire in the measurement of space and motion discomfort].

INTRODUCTION: Space and motion discomfort (SMD) refers to various symptoms that occur in environments with unreliable visual and kinesthetic information that do not permit adequate spatial orientation. Some studies have demonstrated that there is a stable and predictable relationship between vestibular dysfunction and anxiety disorders. Further, vestibular dysfunction can predispose or trigger the development of panic disorder with or without agoraphobia (PD/A) or reinforce phobic avoidance. It therefore seems clinically useful to develop and validate instruments for evaluating SMD in various

Cerebral hypoperfusion precedes nausea during centrifugation.

INTRODUCTION: Nausea and motion sickness are important operational concerns for aviators and astronauts. Understanding the underlying mechanisms associated with motion sickness may lead to new treatments. The goal of this work was to determine if changes in cerebral blood flow precede the development of nausea in subjects susceptible to motion sickness. METHODS: Cerebral flow velocity in the middle cerebral artery (transcranial Doppler), BP, and end-tidal CO2 were measured while subjects were rotated on a centrifuge (250 degrees x s(-1)). Following 5 min of rotation, subjects were translated

Predicting motion sickness during parabolic flight.

BACKGROUND: There are large individual differences in susceptibility to motion sickness. Attempts to predict who will become motion sick have had limited success. In the present study, we examined gender differences in resting levels of salivary amylase and total protein, cardiac interbeat intervals (R-R intervals), and a sympathovagal index and evaluated their potential to correctly classify individuals into two motion sickness severity groups. METHODS: Sixteen subjects (10 men and 6 women) flew four sets of 10 parabolas aboard NASA's KC-135 aircraft. Saliva samples for amylase and total pro

Neuro-cognitive performance is enhanced during short periods of microgravity.

There is increasing interest in the effects of microgravity on cognitive processing, particularly as it relates to the potential for human space travel. While findings to date are quite inconsistent, studies reporting a decrement in cognitive performance have generally not been able to distinguish between the direct influence of microgravity, and any associated influence of stress. Furthermore, the currently available findings are primarily based on behavioral observations, and there is a need to better understand the underlying neurophysiological responses. The current study aimed to determin

[Cutaneous absorption of chemicals].

Chemicals have become indispensible for the maintenance of health in animals and man. The route of administration of each medicament is decided by factors such as site of desired action, chemistry of the active ingredient, age and species of the patient, and frequency of administration (or desired duration of activity). In situations where the oral and hypodermic routes, which are used most frequently, are inadequate or unsatisfactory, dermal application can provide a valuable alternative method to achieve systemic activity. Examples of formulations currently available for dermal application c

The relation of motion sickness to the spatial-temporal properties of velocity storage.

Tilting the head in roll to or from the upright while rotating at a constant velocity (roll while rotating, RWR) alters the position of the semicircular canals relative to the axis of rotation. This produces vertical and horizontal nystagmus, disorientation, vertigo, and nausea. With recurrent exposure, subjects habituate and can make more head movements before experiencing overpowering motion sickness. We questioned whether promethazine lessened the vertigo or delayed the habituation, whether habituation of the vertigo was related to the central vestibular time constant, i.e., to the time con

[Some problems of space medicine].

The paper discusses the problems to be resolved by space medicine and the main stages in the development of this branch of science, beginning with the vertical launches of rockets and ending with the flights of orbital stations. On the basis of ground-based simulation experiments and real space flights it presents a classification of the major symptomocomplexes that may occur inflight. The paper describes the main stages of adaptation to weightlessness and physiological changes in the weightless state. The paper also outlines further pathways in the development of space medicine.

Multiple field tests on landing day: Early mobility may improve postural recovery following spaceflight.

Adaptation to microgravity causes astronauts to experience sensorimotor disturbances during return to Earth leading to functional difficulties. Recently, the Field Test (FT) study involving an incrementally demanding sensorimotor functional test battery has allowed for an unprecedented view into early decrements and recovery from multiple tests conducted on the landing day following 6-months International Space Station missions. Although the protocol was challenging and temporarily increased motion sickness symptoms, there were anecdotal reports that performing these tasks within the first few

Converging indicators for assessing individual differences in adaptation to extreme environments.

It is well known that microgravity results in various physiological alterations, for example, head-ward fluid shifts which can impede physiological adaptation. Other factors that may affect crew operational efficiency include disruption of sleep-wake cycles, high workload, isolation, confinement, stress, and fatigue. From an operational perspective, it is difficult to predict which individuals will be most or least affected in this unique environment given that most astronauts are first-time flyers. During future lunar and Mars missions space crews will include both men and women of multi-nati

Cerebral blood velocity and other cardiovascular responses to 2 days of head-down tilt.

Spaceflight induces a cephalad redistribution of fluid volume and blood flow within the human body, and space motion sickness, which is a problem during the first few days of spaceflight, could be related to these changes in fluid status and in blood flow of the cerebrum and vestibular system. To evaluate possible changes in cerebral blood flow during simulated weightlessness, we measured blood velocity in the middle cerebral artery (MCA) along with retinal vascular diameters, intraocular pressure, impedance cardiography, and sphygmomanometry on nine men (26.2 +/- 6.6 yr) morning and evening f…

Effects of microgravity on osteoblast growth.

Studies from space flights over the past two decades have demonstrated that basic physiological changes occur in humans during space flight. These changes include cephalic fluid shifts, loss of fluid and electrolytes, loss of muscle mass, space motion sickness, anemia, reduced immune response, and loss of calcium and mineralized bone. The cause of most of these manifestations is not known and until recently, the general approach was to investigate general systemic changes, not basic cellular responses to microgravity. Recently analyzed data from the 1973-1974 Skylabs disclose that there is a r…

Motion sickness: a modulatory role for the central cholinergic nervous system.

The present review has extended the general theory of motion sickness proposed by Wood and Graybiel [135, 136] by identifying specific neurophysiological mechanisms that are involved in motion sickness and by interpreting the actions of both scopolamine and amphetamine as effective anti-motion sickness drugs within this neurophysiological context. The neurochemical and neurophysiological effects of scopolamine have been reviewed in relationship to central cholinergic pathways. Cholinergic pathways have been associated with both the perception and expression of normal and excessive levels of mo

[The role of individual reaction s of thermal and water-electrolyte metabolism to suited immersion].

Role of individual reactions of thermal and water-electrolyte metabolism and their relation to the mechanisms of hormonal control were studied during suited immersion (SI). The investigation consisted of 177 man/exposures from 3 hrs up to 3 days of 46 male volunteers. Parameters of mass, heat and water-electrolyte metabolism, and hormonal control were determined on a background of monitoring consumption of special ratios. Based on the complex evaluation of tolerance of weight-free SI for 1 to 3 days, parameters of thermal and hydroionic homeostasis were found to be the predictor variables. Ext

Effect of the character of autonomic response and the emotional and personality features of a human being on reactions of the vestibular system.

A moderate degree (R = 0.37-0.45) of correlation between the values of vestibular reactivity, autonomic response, and emotional and personality characteristics was revealed. A conclusion was made that the following features of the background functional organization of the vestibular system, autonomic and emotional and personality characteristics are space sickness predictors: (1) a high coefficient of enhancement of the vestibuloocular reflex (VOR) and its asymmetry with an excess of the physiological norm limits, (2) the presence of depressive manifestations, (3) the impulsive and autistic pe
