Regulation of septin phosphorylation: SEPT12 phosphorylation in sperm septin assembly.

The sperm annulus, a septin-based ring structure, is important for reproductive physiology. It is composed of SEPT12-based septin core complex followed by assembling as octameric filament. In clinical examinations, mutations of Septin12 result in male infertility, immotile sperm, as well as sperm with defective annuli. The dynamic assembly of septin filaments is regulated by several post-translational modifications, including sumoylation, acetylation, and phosphorylation. Here, we briefly review the biological significance and the regulation of SEPT12 phosphorylation in the mammalian sperm phy…

MeCP2: multifaceted roles in gene regulation and neural development.

Methyl-CpG-binding protein 2 (MeCP2) is a classic methylated-DNA-binding protein, dysfunctions of which lead to various neurodevelopmental d … unctions, MeCP2 is remarkably modulated by posttranslational modifications such as phosphorylation, SUMOylation, and acetylation, providing more regulatory dimensions to its functions. The role of MeCP2 in the central nervous system has been studied extensively, from neurons to glia. Future investigations combining molecular, cellular, and physiological metho …

The multifaceted oncoprotein Tax: subcellular localization, posttranslational modifications, and NF-κB activation.

The human T-cell lymphotropic virus type-I (HTLV-I) is the etiologic agent of adult T-cell leukemia/lymphoma (ATL) and of tropical spastic p … ure that supports a major role for Tax posttranslational modifications, chiefly ubiquitination, and SUMOylation, in the NF-κB activity of Tax. These modifications indeed participate in the control of Tax subcellular localization and modulate its protein-protein interaction potential. Tax posttranslational modifications, which highlight the ability of HTLV-I t …

Small changes, big impact: posttranslational modifications and function of huntingtin in Huntington disease.

Huntington disease (HD) is a neurodegenerative disorder caused by an elongated polyglutamine tract in huntingtin (htt). htt normally undergoes different posttranslational modifications (PTMs), including phosphorylation, SUMOylation, ubiquitination, acetylation, proteolytic cleavage, and palmitoylation. In the presence of the HD mutation, some PTMs are significantly altered and can result in changes in the clinical phenotype. A rate-limiting PTM is defined as one that can result in significant effects on the phenotype in animal models. For example, the prevention of proteolysis at D586 as well …

Insights on post-translational modifications in fatty liver and fibrosis progression.

Metabolic dysfunction-associated steatotic liver disease [MASLD] is a pervasive multifactorial health burden. Post-translational modifications [PTMs] of amino acid residues in protein domains demonstrate pivotal roles for imparting dynamic alterations in the cellular micro milieu. The crux of identifying novel druggable targets relies on comprehensively studying the etiology of metabolic disorders. This review article presents how different chemical moieties of various PTMs like phosphorylation, methylation, ubiquitination, glutathionylation, neddylation, acetylation, SUMOylation, lactylation,…

Post-translational modifications regulating microglial inflammation in central nervous system disorders: a review.

BACKGROUND: Microglia, the primary immune cells of the central nervous system, play a pivotal role in orchestrating neuroinflammatory responses and maintaining neural homeostasis. Post-translational modifications (PTMs) are critical regulators of microglial inflammatory activation, phagocytic capacity, and crosstalk with other neural cells. FINDINGS: This review highlights seven PTMs-phosphorylation, acetylation, methylation, ubiquitination, succinylation, SUMOylation, and lactylation-that are closely linked to the modulation of microglial inflammation. We discuss how these modifications shap…

Current Technologies Unraveling the Significance of Post-Translational Modifications (PTMs) as Crucial Players in Neurodegeneration.

Neurodegenerative disorders, such as Parkinson's disease, Alzheimer's disease, and Huntington's disease, are identified and characterized by the progressive loss of neurons and neuronal dysfunction, resulting in cognitive and motor impairment. Recent research has shown the importance of PTMs, such as phosphorylation, acetylation, methylation, ubiquitination, sumoylation, nitration, truncation, O-GlcNAcylation, and hydroxylation, in the progression of neurodegenerative disorders. PTMs can alter protein structure and function, affecting protein stability, localization, interactions, and enzymati…

Upregulation of nuclear factor-related kappa B suggests a disorder of transcriptional regulation in minimal change nephrotic syndrome.

Immune mechanisms underlying the pathophysiology of idiopathic nephrotic syndrome, the most frequent glomerular disease in children, are believed to involve a systemic disorder of T cell function and cell mediated immunity. How these perturbations take place remains unclear. We report here that NFRKB, a member of the chromatin remodeling complex, is upregulated in MCNS relapse, mainly in CD4+T cells and B cells and undergo post-translational modifications including sumoylation. We showed that NFRKB was highly expressed in nuclear compartment during the relapse, while it was restricted to cytop…

Role of Nuclear SUMO-Targeted Ubiquitin Ligase Uls2 (Slx5-Slx8) in Quality Control of Cytosolic Proteins.

The interplay between the posttranslational modifiers SUMO and ubiquitin mediates the degradation of proteins. This crosstalk is conveyed by SUMO-targeted ubiquitin ligases (STUbLs). The best-studied STUbL in Saccharomyces cerevisiae is the heterodimer Slx5-Slx8 (Uls2). Uls2 mainly resides in the nucleus and is involved in stress tolerance, DNA repair and genome stability maintenance. So far, the majority of known Uls2 targets are nuclear proteins. The role of Uls2 in the targeting of cytosolic proteins remains to be elucidated. In this study, we identify Nis1 and Fir1 as two cytosolic protein

Epigenetic regulation of metabolism in Saccharomyces cerevisiae: mechanisms, metabolic crosstalk, and engineering applications.

Epigenetic regulation plays a central role in metabolic gene expression in eukaryotes. Focusing on Saccharomyces cerevisiae as a model, this review systematically examines how histone modifications, including acetylation, methylation, phosphorylation, ubiquitination, and SUMOylation, and ATP‑dependent chromatin remodeling regulate metabolic genes. It also summarizes the crosstalk between metabolic signals and epigenetic states. On this basis, practical progress, advantages, and limitations of applying these mechanisms in metabolic engineering are then discussed, including industrial strain opt…
