Proteome-based systems biology analysis of the diabetic mouse aorta reveals major changes in fatty acid biosynthesis as potential hallmark in diabetes mellitus-associated vascular disease.

BACKGROUND: Macrovascular complications of diabetes mellitus are a major risk factor for cardiovascular morbidity and mortality. Currently, studies only partially described the molecular pathophysiology of diabetes mellitus-associated effects on vasculature. However, better understanding of systemic effects is essential in unraveling key molecular events in the vascular tissue responsible for disease onset and progression. METHODS AND RESULTS: Our overall aim was to get an all-encompassing view of diabetes mellitus-induced key molecular changes in the vasculature. An integrative proteomic and

Ubiquitin proteasome system and glaucoma: A survey of genetics and molecular biology studies supporting a link with pathogenic and therapeutic relevance.

Glaucoma represents a group of progressive neurodegenerative diseases characterized by the loss of retinal ganglion cells (RGCs) and their axons with subsequent visual field impairment. The disease develops through largely uncharacterized molecular mechanisms, that are likely to occur in different localized cell types, either in the anterior (e.g., trabecular meshwork cells) or posterior (e.g., Muller glia, retinal ganglion cells) segments of the eye. Genomic and preclinical studies suggest that glaucoma pathogenesis may develop through altered ubiquitin (Ub) signaling. Ubiquitin conjugation,

Identification of joint gene players implicated in the pathogenesis of HTLV-1 and BLV through a comprehensive system biology analysis.

BACKGROUND: Human T-cell lymphotropic virus type 1 (HTLV-1) and bovine leukemia virus (BLV) are oncogenic viruses that induce adult T cell leukemia/lymphoma (ATLL) and enzootic bovine leukosis (EBL), respectively. HTLV-1 principally infects CD4+ T cells comprising regulatory T cells (Tregs), T helper 1 (Th1), and T helper 2 (Th2), while BLV infects B lymphocytes. Both viruses may impel cell proliferation and malignancy. METHODS: To survey the transcriptomic variations due to HTLV-1 and BLV infection and further hematologic malignancies, differential expression genes (DEGs) were explored betwe

Pristionchus pacificus: A Genetic Model System for the Study of Evolutionary Developmental Biology and the Evolution of Complex Life-History Traits.

INTRODUCTIONPristionchus pacificus is a nematode that has been established as a model system for evolutionary developmental biology. Initially, P. pacificus was used as a convenient nematode with which to compare the processes of vulva and gonad development as well as sex determination to Caenorhabditis elegans, one of the best-studied animal models. P. pacificus shares many features with C. elegans, including a short generation time, its ability to be easily cultured in the laboratory, and self-fertilization as a mode of reproduction. These features allowed forward and reverse genetic tools t

How close are modern broilers to their physiological ceiling? Have genetic, nutritional, and precision technologies reached biological limits? A systems biology perspective through physiological ceiling theory.

This review examines the hypothesis that modern broiler chickens may be approaching a multidimensional physiological limit to further productivity improvement and evaluates the extent to which current evidence supports this proposition. Over the past century, genetic selection, nutritional optimization, health management, and technological innovations have dramatically enhanced growth rate, feed conversion efficiency, and carcass yield. However, these gains have increasingly been accompanied by physiological trade-offs, including skeletal abnormalities, muscle myopathies, metabolic disorders,

A systems biology approach and in vitro experiment indicated Rapamycin targets key cancer and cell cycle-related genes and miRNAs in triple-negative breast cancer cells.

An anticancer drug known as Rapamycin acts by inhibiting the mammalian target of the Rapamycin pathway. This agent has recently been investigated for its potential therapeutic benefits in sensitizing drug-resistant breast cancer (BC) treatment. The molecular mechanism underlying these effects, however, is still a mystery. Using a systems biology method and in vitro experiment, this study sought to discover essential genes and microRNAs (miRNAs) targeted by Rapamycin in triple-negative BC (TNBC) cells to aid prospective new medications with less adverse effects in BC treatment. We developed the…

From Spore Biology to System Performance: Strain-Resolved Mechanisms, Biosafety and Translational Readiness of Bacillus Probiotics in Aquaculture.

Bacillus-based probiotics are widely promoted as antibiotic-sparing interventions in aquaculture, yet translation from controlled trials to commercial production remains constrained by strain heterogeneity, inconsistent dosing, incomplete biosafety assessment and frequent inference of mechanisms from non-specific biomarkers. This scoping review reorganizes the evidence reported for 67 empirical studies published during 2015-2025 into a strain-host-route-system framework. Across finfish and aquatic invertebrates, Bacillus supplementation was commonly associated with improved growth, feed utiliz

Bioregulatory periodontal medicine: could the molecular networks, inflammation and system biology modify the auto-regulatory capacity of the patients?

BACKGROUND: The traditional healthcare model, focused on symptom management, has shown limitations in addressing chronic diseases. In response, Bioregulatory Systems Medicine (BrSM) has emerged, an approach that promotes the body's self-regulation through multi-component drug and phytotherapeutic treatments. Its potential application in dentistry, particularly in periodontitis, has spurred the study of various natural molecules with the potential to prevent or reduce bone resorption. This qualitative systematic review analyzes the available evidence from animal and human models in the field of

Surgical Intensity and Survival After Surgery for Extradural Spinal Metastases: Mechanical Instability, Tumor Biology, and Systemic Reserve.

Background/Objectives: Surgery for extradural spinal metastases must balance neurologic and mechanical goals against limited survival. We evaluated whether a predefined three-tier surgical-intensity classification aligned with established measures of operative burden and whether intensity was associated with 90-day mortality or overall survival. Methods: This retrospective cohort included 141 adults with extradural spinal metastases operated on between June 2020 and December 2025. Surgical intensity was classified as low, moderate, or high. SMII was calculated for all patients as a secondary c

Microplastic trafficking in maternal-fetal system: a systematic review and quantitative profiling linking particle characteristics to developmental biology.

Microplastics (MPs) have been detected across multiple maternal and neonatal biological matrices, suggesting widespread exposure during critical developmental stages. However, the mechanisms shaping their distribution and accumulation within the maternal-fetal system remain poorly understood. This systematic review and quantitative synthesis provides a comprehensive quantitative profiling of MP burden across six maternal and neonatal biological matrices placenta, breast milk, endometrium, amniotic fluid, meconium, and feces. Our analysis reveals three critical insights: (1) Tissue‑specific MP

Identification of Potential Biomarkers Related to the Progression and Prognosis of Parkinson's Disease and Melanoma via Combined System Biology Approaches.

Parkinson's disease (PD) and melanoma are considered high risk in affecting an individual's health. The association between PD and melanoma has been reported with consistent results by various epidemiological studies. The identification of differentially expressed genes (DEGs) and pathways between the two diseases can support the findings of the epidemiological studies. Transcriptomics studies play a vital role in investigating DEGs with better specificity and sensitivity. Hence, we have performed transcriptomic data analysis to discover the gene expression profiles and significant pathways an
