In Parkinson's disease (PD), the thalamus plays an important role in pathogenesis and disease symptoms; however, the morphological changes in thalamic subnuclei have not been clearly investigated. And there are still many challenges in individual PD diagnosis, especially clinical condition evaluations. Structural magnetic resonance imaging (MRI) was performed on 131 PD patients and 69 healthy controls (HC), and the volumes of 25 thalamic subnuclei were evaluated by FreeSurfer and a newly developed thalamus segment algorithm. Then, the individual PD diagnosis and clinical condition prediction w

The thalamus is composed of multiple nuclei densely connected with the cortex in an organized manner, forming parallel thalamocortical networks critical to sensory, motor, and cognitive functioning. Thalamocortical circuit dysfunction has been implicated in multiple neurodevelopmental disorders, including schizophrenia, which also often exhibit sex differences in prevalence, clinical characteristics, and neuropathology. However, very little is known about developmental and sex effects on thalamocortical networks in youth. The present study characterized the effects of age, sex and psychosis sy

Enhanced bidirectional connectivity was also observed within the frontoparietal network, between the pregenual cingulate gyrus and superior frontal gyrus, and between lateral thalamic nuclei (LTN) and frontal pole. …

The rodent orbitofrontal cortex is involved in a variety of cognitive and behavioral functions that require thalamic input to be successfully expressed. Although the thalamic nucleus submedius (Sm) is a major source of afferents to the orbitofrontal cortex, thalamocortical projection from the Sm has not been fully elucidated. In the present study, we first divided the rat Sm into dorsal and ventral parts according to the distribution of vesicular glutamate transporter 2-immunoreactive varicosities, which were somatosensory afferents from the brain stem. Subsequently we investigated dendritic a

Functional magnetic resonance imaging (fMRI) of the auditory and visual sensory systems of the human brain is an active area of investigation in the study of human health and disease. The medial geniculate nucleus (MGN) and lateral geniculate nucleus (LGN) are key thalamic nuclei involved in the processing and relay of auditory and visual information, respectively, and are the subject of blood-oxygen-level-dependent (BOLD) fMRI studies of neural activation and functional connectivity in human participants. However, localization of BOLD fMRI signal originating from neural activity in MGN and LG…

The thalamus plays a critical role in the coordination of information as it passes from region to region within the brain. A disruption of t … ptoms. The thalamus is a starting point for several parallel, overlapping networks that extend from thalamic nuclei to the cortex. Evidence is emerging that changes in the thalamic nodes of these networks are echoed by changes at other points along the chain; this suggests that schizophrenia might be a disease of disrupted thalamocortical neural networks. This mo …

Mast cells accessing the brain parenchyma through the blood-brain barrier in healthy animals are limited to pre-cortical sensory relays - the olfactory bulb and the thalamus. We have demonstrated that unilateral repetitive stimulation of the abdominal wall generates asymmetry in midline thalamic mast cell (TMC) distribution in cyclophosphamide-injected rats, consisting of contralateral side-prevalence with respect to the abdominal wall stimulation. TMC asymmetry 1) was generated in strict relation with cystitis, and was absent in disease-free and mesna-treated animals, 2) was restricted to the

Background: Stroke induces both focal and remote brain alterations, including early degeneration of deep grey nuclei (DGN). This study evaluates whether volumetric and diffusion changes in DGN during the acute-subacute phase after cortical middle cerebral artery (MCA) stroke can predict 3-month functional outcome. Methods: In this retrospective study, consecutive patients ≥ 18 years with first acute-subacute cortical MCA stroke (< 21 days) who underwent 3T MRI including DWI and T1-MP-RAGE imaging between 2018 and 2020 were included. Demographic variables, cardiovascular risk factors, time from

OBJECTIVE: Nonhypothesis-based MRI-analysis techniques including deformation-based morphometry and automated tissue segmentation have suggested that preterm infants at term-equivalent age have reduced tissue volume in the basal ganglia and thalami, which is most apparent among infants with supratentorial lesions. The aim of our study was to test this hypothesis by direct measurement of thalamic and lentiform nuclei volumes in preterm infants at term-equivalent age and term-born controls using manual volumetry.
DESIGN/METHODS: Forty preterm infants at term-equivalent age (median gestational ag

Cocaine dependence is associated with deficits in cognitive control. Previous studies demonstrated that chronic cocaine use affects the activity and functional connectivity of the thalamus, a subcortical structure critical for cognitive functioning. However, the thalamus contains nuclei heterogeneous in functions, and it is not known how thalamic subregions contribute to cognitive dysfunctions in cocaine dependence. To address this issue, we used multivariate pattern analysis (MVPA) to examine how functional connectivity of the thalamus distinguishes 100 cocaine-dependent participants (CD) fro

A hypothetical mechanism of the basal ganglia involvement in visual hallucinations is proposed. Hallucinations as conscious visual patterns are the result of selection of signals circulating in several interconnected loops each of whi …

Functional brain neuroimaging essentially relies on two basic principles: functional segregation and functional interaction. Recent studies of the functional segregation of the human brain during sleep, using positron emission tomography and statistical parametric mapping, indicate that human brain function is organized in a very specific way in each state of vigilance. During slow wave sleep, the most deactivated areas are the upper brainstem, thalamic nuclei and basal forebrain; deactivation of the basal ganglia is also seen. In the cortex, the least active areas are the associative cortices…

The Medio-Dorsal Nuclei (MDN) including the thalamic magnocellular and parvocellular thalamic regions has been implicated in verbal memory function. In a 77 year old lady, with a prior history of a clinically silent infarct of the left MDN, we observed the acute onset of spontaneous confabulations when an isolated new infarct occurred in the right MDN. The patient and five age-matched healthy subjects underwent Magnetic Resonance Imaging (MRI) and Diffusion Tensor Imaging (DTI). The thalamic lesions were localized by overlapping Morel Thalamic Atlas with structural MRI data. DTI was used to as

In a patient with a unilateral embolic infarct in the left posterior thalamo-subthalamic paramedian artery territory, neuropathological studies showed involvement of the intralaminar, dorsomedial, and internal part of the ventral posterior nuclei of the thalamus, of the rostral part of the mesencephalic reticular formation, and of the posterior commissure. The patient showed upgaze palsy for voluntary saccades, smooth pursuit and vestibulo-ocular movements, sustained downgaze, right-sided motor hemineglect and facio-brachial hypaesthesia, motor transcortical aphasia and anterograde amnesia. The neuropathology of 8 cases of S. Paulo south coast epidemic encephalitis (Rocio flavivirus), a new arbo B virus encephalitis, is described. The topographic pattern of the lesions appears to be almost specific. The gray matter is predominantly affected. Interstitial mononuclear infiltration, microglial proliferation and perivascular lymphocytic cuffing were seen. Neuronophagia was seldom seen except during the acute phases of the disease. Throughout the neuraxis, the gray matter was affected to a greater degree than white matter. The more damaged structures, in descending order, were as foll
