Headache under simulated microgravity is related to endocrine, fluid distribution, and tight junction changes.

Head-down-tilted bed rest (HDTBR) induces headaches similar to headaches during space flights. The objective of this investigation was to study hematological, endocrinological, fluid changes and tight junctions in HDTBR-induced headaches as a proxy for space headache. The randomized crossover HDTBR design by the European Space Agency included 12 healthy, nonheadache male subjects. Before, during, and after confined HDTBR periods, epinephrine (urine), cortisol (saliva), hematological, endothelium markers, and fluid distribution parameters were measured. Headaches were assessed with a validated

Consequences of cardiovascular adaptation to spaceflight: implications for the use of pharmacological countermeasures.

There is little evidence obtained from space flight to support the notion that occurrence of cardiac dysrhythmias, impaired cardiac and vascular function, and manifestation of asymptomatic cardiovascular disease represent serious risks during space flight. Therefore, the development of orthostatic hypotension and instability immediately after return from spaceflight probably reflect the most significant operational risks associated with the cardiovascular system of astronauts. Significant reductions in stroke volume and lower reserve for increasing peripheral vascular resistance contribute to

A Validated Transcriptomic NMJ Remodeling Score Reveals Synaptic Dysfunction Independent of Muscle Atrophy after Immobilization in a Microgravity Analog.

Muscle weakness after immobilization often exceeds that explained by loss of muscle mass alone, suggesting a role for neuromuscular synaptic changes. To quantify these adaptations, we developed a composite transcriptomic Neuromuscular Junction (NMJ) Remodeling Score and evaluated its behavior relative to classical atrophy pathways during short-term unloading. We analyzed vastus lateralis RNA sequencing data from adults undergoing 10 days of unilateral lower-limb suspension followed by a 21-day recovery, generating NMJ and atrophy scores for 15 and 10 genes, respectively. Transcriptome-wide tes

0.33g mitigates muscle atrophy while 0.67g preserves muscle function and myofiber type composition in mice during spaceflight.

As human space exploration advances, understanding how different gravity levels affect skeletal muscle is critical for long-term health. Among the major organ systems, skeletal muscle is particularly sensitive to gravitational unloading, yet the gravity threshold required to maintain homeostasis remains unclear. Using the Multiple Artificial-gravity Research System aboard the International Space Station, mice were exposed to graded gravity levels, microgravity, 0.33g, 0.67g, and 1g, and their muscles were analyzed postflight. In the gravity-sensitive soleus, the cross-sectional area was preser

Effect of Exercise on Coronary Hemodynamics Under the Earth's Gravity and Microgravity Conditions: A CFD Study.

Exposure to microgravity affects the cardiovascular system and fluid distribution in the vasculature system of the human body depending on the short term or long term exposure. Literature suggests that in the initial few days in the microgravity environment, a short term exposure, the cardiac output and plasma volume are altered. Gradually, after about a week of long exposure to the microgravity environment, these parameters return to the same levels as those under the Earth's gravity. In this computational fluid dynamics (CFD) study, we investigate the effect of microgravity environment on co

Microgravity-Induced Ocular Changes Assessed by Optical Coherence Tomography.

INTRODUCTION: Long-duration space missions pose a risk to visual health due to cephalad fluid shifts in microgravity; this can affect ocular structures, causing decreased astronaut performance in addition to long-term side effects. Understanding these changes is essential for developing effective countermeasures against spaceflight-associated neuro-ocular syndrome. We reviewed how head-down tilt (HDT)-a terrestrial analog of microgravity-affects ocular parameters measured by optical coherence tomography (OCT) and intraocular pressure (IOP). METHODS: Seven databases were searched through May 2

Bio-Monitor Detects Reduced Obstructive Sleep Apnea and Susceptibility to Arrhythmia in Spaceflight.

BACKGROUND: Attenuation of sleep disordered breathing in astronauts entering microgravity was previously reported during shuttle missions. However, the impacts of microgravity on obstructive sleep apnea (OSA) have not previously been quantified. CASE REPORT: An astronaut with OSA participated in the noninvasive assessment of multiple physiological variables using the Bio-Monitor biometric shirt before, twice during, and after a 6-mo mission to the International Space Station. Cardiorespiratory variables, including electrocardiogram, respiratory rate and tidal volume, oxygen saturation, and ac

Otoacoustic Estimate of Astronauts' Intracranial Pressure Changes During Spaceflight.

PURPOSE: To investigate the potential correlation between prolonged exposure to microgravity on the International Space Station and increased intracranial fluid pressure, which is considered a risk factor for the astronauts' vision, and to explore the feasibility of using distortion product otoacoustic emissions as a non-invasive in-flight monitor for intracranial pressure changes. METHODS: Distortion product otoacoustic emission phase measurements were taken from both ears of five astronauts pre-flight, in-flight, and post-flight. These measurements served as indirect indicators of intracran

The reciprocal regulation between mitochondrial-associated membranes and Notch signaling in skeletal muscle atrophy.

Skeletal muscle atrophy and the inhibition of muscle regeneration are known to occur as a natural consequence of aging, yet the underlying mechanisms that lead to these processes in atrophic myofibers remain largely unclear. Our research has revealed that the maintenance of proper mitochondrial-associated endoplasmic reticulum membranes (MAM) is vital for preventing skeletal muscle atrophy in microgravity environments. We discovered that the deletion of the mitochondrial fusion protein Mitofusin2 (MFN2), which serves as a tether for MAM, in human induced pluripotent stem (iPS) cells or the red

Daily generation of a footward fluid shift attenuates ocular changes associated with head-down tilt bed rest.

Astronauts have presented with a constellation of visual changes referred to as spaceflight-associated neuro-ocular syndrome (SANS). However, neither have early markers of microgravity-induced optic remodeling been fully identified nor have countermeasures been developed. To identify early markers of SANS, we studied 10 subjects with optical coherence tomography and ultrasonography when upright and supine and again after 24 h of 6° head-down tilt (HDT) bed rest. Upon acute transition from the upright to the supine position, choroid area (2.24 ± 0.53 to 2.28 ± 0.52 mm2, P = 0.001) and volume (9

Tracheal intubation in microgravity: a simulation study comparing direct laryngoscopy and videolaryngoscopy†.

BACKGROUND: The risk of severe medical and surgical events during long-duration spaceflight is significant. In space, many environmental and psychological factors may make tracheal intubation more difficult than on Earth. We hypothesised that, in microgravity, tracheal intubation may be facilitated by the use of a videolaryngoscope compared with direct laryngoscopy. METHODS: In a non-randomised, controlled, cross-over simulation study, we compared intubation performance of novice operators and experts, using either a direct laryngoscope or a videolaryngoscope, in weightlessness and in normogr…

Neuroplasticity changes during space flight.

Neuroplasticity refers to the ability of neurons to alter some functional property in response to alterations in input. Most of the inputs received by the brain and thus the neurons are coming from the overall sensory system. The lack of gravity during space flight or even the reduction of gravity during the planned Mars missions are and will change these inputs. The often observed "loop swimming" of some aquatic species is under discussion to be based on sensory input changes as well as the observed motion sickness of astronauts and cosmonauts. Several reports are published regarding these ch

Cytogenetic studies of blood lymphocytes from cosmonauts after long-term space flights on Mir station.

Long-term space missions may increase risks of unfavorable consequences for cosmonauts as a result of radiation effects. This paper presents results of a study of cytogenetic damage in cosmonauts' peripheral blood lymphocytes induced by space radiation. Cultivation of lymphocytes and analysis of chromosomal aberrations were made according to generally accepted methods. It is shown that the yields of dicentrics and centric rings scored after long-term space flights are considerably higher than those scored prior to the flights. An attempt was made to assess individual doses received by cosmonau

[Studies on the solvent structure in protein crystals grown in microgravity].

Structures of Space- and ground-grown protein crystals were determined and compared for hen egg-white lysozyme and acidic phospholipase A2 from snake venom. The results show that microgravity might improve the structure of weakly bound ordered water molecules, though it could not change the protein conformation and the structure of solvent molecules bound stronger to protein molecules. The results also imply that the degree of quality improvement of microgravity-grown protein crystals might be related to the solvent content in protein crystals. These findings might, in a respect, reveal the me

Pharmacokinetic consequences of spaceflight.

Spaceflight induces a wide range of physiological and biochemical changes, including disruption of gastrointestinal (GI) function, fluid and electrolyte balance, circulatory dynamics, and organ blood flow, as well as hormonal and metabolic perturbations. Any of these changes can influence the pharmacokinetics and pharmacodynamics of in-flight medication. That spaceflight may alter bioavailability was proposed when drugs prescribed to alleviate space motion sickness (SMS) had little therapeutic effect. Characterization of the pharmacokinetic and/or pharmacodynamic behavior of operationally crit

[Low-gravitational changes in the bone system].

This review surveys reported data and observations made by the present authors concerning the effect of real and simulated hypogravity on bones of man and various animals. Bone biomechanics and mechanisms of osteodystrophic changes in hypogravity are discussed. Based on published data about bone metabolism regulation, the concept of bone formation and degradation in normal gravity and hypogravity is advanced. The conclusion is made that hypogravity-induced changes obey regulation at different levels, with local mechanisms being predominant. It is emphasized that a detailed investigation of eve

Hormonal responses of metoclopramide-treated subjects experiencing nausea or emesis during parabolic flight.

Metoclopramide was orally administered (10 or 20 mg) to 22 subjects, 75 min before parabolic flight. Serum levels of ACTH, EPI, NE, and vasopressin (AVP) were unaltered by metoclopramide. AVP and ACTH (1.2 and 36 pg.ml-1) were elevated 77 and 3.8-fold (92.3 and 135 pg.ml-1) following emesis, after 40 parabolas (68.7 and 140 pg.ml-1) and landing (8.7 and 79 pg.ml-1). Seven subjects displaying no nausea and no emesis demonstrated smaller elevations (8.2 and 2.2-fold). Of 15 vomiting subjects, 7 reported no nausea and had lower elevations of AVP with faster recoveries. These findings are consiste

Motion sickness and space sickness: clinical and experimental findings.

Vertigo in motion sickness forms a clear clinical picture. We can confirm that complex symptoms are due to abnormal stimulations of labyrinth receptors subsequent to simultaneous multi-plane angular accelerations. This labyrinth stimulation originates from Coriolis' complementary accelerations. The seriousness of kinetosis symptoms can vary according to different causes; level of labyrinth stimulation, subject's constitution, psychological and physical conditions and whether or not the subject is accustomed to kinetic stimulations. Space sickness shows similar symptoms and similar psychologica
