Antifungal susceptibility profile of Candida species and uncommon yeasts from drug abusers with oral candidiasis.

In Iran, there is limited information regarding the species distribution and antifungal susceptibility profiles of yeast isolates from drug addicts suffering from oral candidiasis (OC). In this study, 104 yeast isolates, including 98 Candida species and 6 uncommon yeasts, were collected from 71 drug abusers with OC. The susceptibility profiles of Candida spp. and uncommon yeasts to amphotericin B (AMB), itraconazole (ITC), nystatin (NYC), fluconazole (FLC), and caspofungin (CAS) were evaluated using the CLSI broth microdilution method. The prevalence of OC in the sampled population was found t…

Adapting an Ergosterol Extraction Method with Marine Yeasts for the Quantification of Oceanic Fungal Biomass.

Ergosterol has traditionally been used as a proxy to estimate fungal biomass as it is almost exclusively found in fungal lipid membranes. Ergosterol determination has been mostly used for fungal samples from terrestrial, freshwater, salt marsh- and mangrove-dominated environments or to describe fungal degradation of plant matter. In the open ocean, however, the expected concentrations of ergosterol are orders of magnitude lower than in terrestrial or macrophyte-dominated coastal systems. Consequently, the fungal biomass in the open ocean remains largely unknown. Recent evidence based on micros

Oral bacteria and yeasts in relationship to oral ulcerations in hematopoietic stem cell transplant recipients.

BackgroundOral mucositis is a serious and debilitating side effect of conditioning regimens for hematopoietic stem cell transplant (HSCT). Through HSCT, the homeostasis in the oral cavity is disrupted. The contribution of the oral microflora to mucositis remains to be clarified. The aim of our study was to investigate the relationship between yeasts, bacteria associated with periodontitis, and oral ulcerations in HSCT recipients.MethodsThis prospective observational study included 49 adult HSCT recipients. Twice weekly, oral ulcerations were scored, and oral rinsing samples were obtained. Samp…

Role of alcohols in growth, lipid composition, and membrane fluidity of yeasts, bacteria, and archaea.

Increased membrane fluidity, which causes cofactor leakage and loss of membrane potential, has long been documented as a cause for decreased … e saturated and unbranched lipids were observed in most of the organisms tested, including archaea, yeasts, and bacteria. However, these shifts did not always result in a decrease in membrane fluidity or in greater tolerance of the organism to alcohol exposure. In general, organisms tolerating the highest concentrations of alcohols maintained membrane fl …

The prevalence of Malassezia yeasts in patients with atopic dermatitis, seborrhoeic dermatitis and healthy controls.

Cultures for Malassezia yeasts were taken from both normal-looking skin and lesional skin in 124 patients with atopic dermatitis, 16 patients with seborrhoeic dermatitis and from normal skin of 31 healthy controls. Positive Malassezia growth was found in fewer patients with atopic dermatitis (56%) than in patients with seborrhoeic dermatitis (88%) or in healthy controls (84%, p<0.01). In the patients with atopic dermatitis, fewer positive cultures were found in lesional (28%) than in non-lesional skin (44%, p<0.05), while positive cultures were found in 75% of both lesional and non-lesional sk…

Systemic ketoconazole is an effective treatment of atopic dermatitis with IgE-mediated hypersensitivity to yeasts.

BACKGROUND: IgE-mediated hypersensitivity to yeasts is often seen in atopic dermatitis (AD) patients, especially when dermatitis is located in the head, neck, and shoulder regions. Two studies have shown the efficacy of ketoconazole in the treatment of this type of AD, in contrast to results of topical treatment. The objective was to assess the clinical efficacy of antifungal treatment in AD in a randomized, double-blind, placebo-controlled study with oral ketoconazole and yeast-specific IgE levels and saprophytic yeast growth monitored simultaneously. METHODS: Eighty patients with AD and pos…

Comparative study of the in vitro antifungal activity of bifonazole, naftifine and sertaconazole against yeasts.

The in vitro activity of three antifungal agents was tested and compared against 151 yeast strains, including ten Candida species, Cryptococcus neoformans, Rhodotorula rubra, and Trichosporon cutaneum. Minimum inhibitory concentrations (MICs) were determined by a microdilution technique in Shadomy modified liquid medium. The mean MICs of sertaconazole (0.34 mg/L) were lower than those of naftifine (16.3 mg/L) and bifonazole (13.2 mg/L). These results suggest that sertaconazole is more active against Candida spp than other topical agents such as bifonazole and naftifine.

[Effect of orally administered polyene antimycotics on the intestinal colonization with yeasts: possibilities and limitations].

On the basis of intestinal yeast colonization different consequences for therapeutic and prophylactic administration of polyene antimycotics have to be drawn. Immunocompromised neutropenic patients should orally receive polyene antifungal drugs (nystatin or amphotericin B) for a long time during the period of increased risk for systemic candidosis. The level of daily dosing is dependent on age, physiological status of the gastrointestinal tract, and underlying disease of the patient. In immunocompetent persons the normal commensal yeast flora should not be suppressed by antifungal chemoprophyl

Divergent patterns of pulmonary blastomycosis induced by conidia and yeasts in athymic and euthymic mice.

Athymic nude (nu/nu) mice are uniformly more susceptible than euthymic nu/+ mice to lethal infection with intranasally inoculated Blastomyces dermatitidis, whether infection is initiated by yeasts or conidia. Conidial infection requires a high inoculum size; the disease produced is prolonged and disseminated. Yeasts are infective at a low inoculum size and produce a rapidly fatal pneumonia. Thymus transplantation is more protective for conidia-infected than yeast-infected nude mice, presumably because the disease course is long enough for an effect to become demonstrable. Yeast inocula multipl…

Comprehensive flavor profiling and neural perception of yeast extracts from five yeast sources via sensomics, machine learning, and EEG.

Yeast extract is widely used as a natural flavor enhancer, yet how yeast source shapes its taste, aroma, and neural perception remains insufficiently understood. Here, we compared yeast extracts from five industrially relevant sources (beer yeast, BY; bread yeast, BRY; Candida utilis, CUY; alcohol yeast, AY; wine yeast, WY) using an integrated sensory-chemical-neural framework. Sensory evaluation revealed clear source-dependent flavor signatures (p < 0.05), with CUY showing the strongest umami intensity. Solvent-assisted flavor evaporation (SAFE) coupled with GC-O-MS identified 120 volatile co

Analysis of the hypoxia-induced ADH2 promoter of the respiratory yeast Pichia stipitis reveals a new mechanism for sensing of oxygen limitation in yeast.

We introduced a reporter gene system into Pichia stipitis using the gene for the artificial green fluorescent protein (GFP), variant yEGFP. This system was used to analyse hypoxia-dependent PsADH2 regulation. Reporter gene activity was only found under oxygen limitation on a fermentable carbon source. The promoter was not induced by oxygen limitation in the Crabtree-positive yeast Saccharomyces cerevisiae. Promoter deletions revealed that a region of 15 bp contained the essential site for hypoxic induction. This motif was different from the known hypoxia response elements of S. cerevisiae but

The effect of canine macrophages on the adherence and growth of Blastomyces dermatitidis yeast: evidence of a soluble factor that enhances the growth of B. dermatitidis yeast.

Blastomycosis is a medically important systemic fungal infection of dogs and humans. Phagocytic cells are the first line of cellular defence against B. dermatitidis, and are a prominent feature in the lesions and exudate of canine blastomycosis. The adherence of B. dermatitidis yeast to canine phagocytes, and the effects of such adherence on the growth of B. dermatitidis yeast, has not been previously reported. The results of this study demonstrate that canine complement enhances the adherence of B. dermatitidis yeast to canine macrophages. Initiation of the canine complement cascade by B. der

Advancement of mannosylerythritol lipids production: enhancement and tailor-made production through genetic modification of producing yeasts.

Mannosylerythritol lipids (MELs) are biosurfactants produced by basidiomycetous yeasts. They are attracting attention not only as surfactants with excellent surface activity but also as environmentally friendly and circular materials that are biodegradable and can be produced from renewable resources. This review focuses on the improvement of MELs production through genetic modification of MELs producers and summarizes studies addressing the challenges associated with expanding MELs use, including reducing production costs through increased productivity and expanding structural variety through…

Mitochondrial retrograde control of transcription evolves with respiratory stress, metabolic adaptation, and virulence in budding yeasts.

The pathway involving the paralogous transcription factors Rtg1 and Rtg3 was first described in Saccharomyces cerevisiae as the retrograde r … e regulatory proteins Rtg2 and Mks1 that were likely present in the last common ancestor of budding yeasts. The regulation of the Rtg transcription factors in C. albicans is different than in S. cerevisiae, as both Rtg2 and Mks1 were lost in the majority of Serinales. Among the target genes, of particular interest is the evolution of the alternative oxid …

Improving protein secretion in yeasts: From secretion leader engineering and AI-driven prediction to bioprocess performance.

Yeast is widely used for recombinant protein production because it supports eukaryotic protein expression and high-cell-density cultivation. Efficient secretion depends on secretion leaders, short N-terminal sequences that direct nascent polypeptides into the secretory pathway. Multiple engineering strategies have been developed to enhance secretion efficiency. These include rational modification of physicochemical properties of secretion leaders, hybrid leader design through the combination of distinct signal elements, codon optimization of the leader sequence, and fusion with translational f

Acetate-based biological platforms: Bridging carbon dioxide utilization and high-value bioproduct production in oleaginous yeasts.

Acetate is emerging as a promising two-carbon substrate in the circular bioeconomy, bridging the gap between single-carbon sources and high-value biofuels and bioproducts. This review examines the key pathways for acetate production, including the electrochemical reduction of carbon dioxide, syngas fermentation, and biological acetogenesis. It focuses on acetate metabolism in oleaginous yeasts, such as Yarrowia lipolytica and Rhodotorula toruloides, which efficiently convert acetate-derived acetyl-CoA units into diverse bioproducts such as lipids, fatty alcohols, triacetic acid lactone, and ca…

Comparing the effect of selenium yeasts, betaine, and capsaicin on heat stress response in fattening bulls.

Prolonged high temperatures cause heat stress in fattened cattle, negatively affecting growth and health. This study evaluated dietary supplements in Limousin bulls under mild heat stress (THI = 75.01 ± 4.13). Forty-eight bulls were divided into four groups, receiving either a standard TMR or TMR supplemented with selenium yeast (YSE), betaine (BET), or capsaicin (CAP) for 60 days. Results showed all additives improved daily gains (p ≤ 0.05). Redox markers varied; total antioxidant capacity (TAC) was highest in the control group, while BET had the lowest. The BET also showed the highest GPx ac

Metabolic Characterization of Two Flor Yeasts During Second Fermentation in the Bottle for Sparkling Wine Production.

The global sparkling wine market continues to grow steadily, reaching approximately 24 million hectoliters in 2023, with an annual increase of around 4% despite a general decline in overall alcoholic beverage consumption. This growth highlights the importance of employing diverse yeast strains to improve product variety and quality. Flor yeasts are specialized strains of Saccharomyces cerevisiae that develop a biofilm on the surface of certain wines during biological ageing. They possess unique physiological properties, including high ethanol tolerance and the capacity to adhere, which support…
