Reduced susceptibility to host-defense cationic peptides and daptomycin coemerge in methicillin-resistant Staphylococcus aureus from daptomycin-naive bacteremic patients.

BACKGROUND: We hypothesized that, for methicillin-resistant Staphylococcus aureus (MRSA), in vitro daptomycin susceptibility could be influenced by exposures to endogenous host defense peptides (HDPs) prior to clinical exposure to daptomycin. METHODS: Two endovascular HDPs were used: thrombin-induced platelet microbicidal protein (tPMP) and human neutrophil defensin-1 (hNP-1) from neutrophils. Forty-seven unique MRSA isolates obtained from bacteremic patients in multicenter prospective clinical trials were studied. Clinical characteristics, microbiologic parameters, prior vancomycin therapy,

Detection of CTX-M-15-producing and carbapenem-resistant Acinetobacter baumannii strains from urine from an Indian hospital.

This one-year study describes isolation of 6 carbapenem-resistant Acinetobacter baumannii strains from 920 urine samples from urinary tract infection patients attending an indian hospital. A search for bla(CTX-M), bla(TEM), armA, rmtA, rmtB and integrons was performed. four strains were identified as extended-spectrum-β-lactamase (ESBL)-producers. Two bla(CTX-M-15 )and 2bla(TEM-150 )and 3 Class 1 integrons were amplified. Ceftazidime and cefotaxime resistance markers were found to be located on the plasmid DNA of the clonally related bla (CTX-M-15)-positive strains and were transmissible throu

Decreased linezolid uptake in an in vitro-selected linezolid-resistant Staphylococcus epidermidis mutant.

OBJECTIVES: The aim of this study was to characterize the mechanism of resistance to linezolid in an in vitro-selected linezolid-resistant Staphylococcus epidermidis mutant. METHODS: A linezolid-resistant strain of S. epidermidis was selected by serial passages with increasing concentrations of linezolid. The MICs of linezolid, ciprofloxacin, tetracycline, rifampicin, vancomycin, gentamicin, tobramycin, chloramphenicol and oxacillin were determined. The 23S rRNA gene was amplified and sequenced, to search for mutations conferring linezolid resistance. The MIC of linezolid was also determined

Functional analysis of a PcsB-deficient mutant of group B streptococcus.

Group B streptococcus (GBS) is the major cause of bacterial sepsis and meningitis in neonates and poses a significant threat to parturient women. Recently, we identified in GBS the polypeptide PcsB, which is a protein required for cell separation of GBS, and which is also involved in the antibiotic sensitivity of these bacteria. In the present study, the introduction of the pcsB-carrying plasmid pATpcsB into the PcsB-deficient GBS mutant Sep1 restored the phenotype and the antibiotic susceptibility of this strain to that of the GBS wild-type. Although Northern blots revealed a four- to five-fo

Molecular and biochemical characterization of Ambler class A extended-spectrum beta-lactamase CGA-1 from Chryseobacterium gleum.

Antibiotic susceptibility testing by disk diffusion of a Chryseobacterium gleum isolate, strain CIP 103039, showed a typical synergy image between clavulanic acid and expanded-spectrum cephalosporins. Shotgun cloning gave a recombinant plasmid in Escherichia coli that produced a beta-lactamase, CGA-1, with a pI value of 8.9 that conferred resistance to most penicillins (except ureidopenicillins) and narrow-spectrum cephalosporins and an intermediate susceptibility to expanded-spectrum cephalosporins and aztreonam. The CGA-1 amino acid sequence shared only 60% amino acid identity with CME-1 and

Pilot study of the genetic diversity of the pneumococcal nasopharyngeal flora among children attending day care centers.

A pilot study was conducted to determine the genetic diversity of multiple colonies of pneumococci recovered from 37 nasopharyngeal (NP) samples of children. A total of 239 pneumococcal isolates (typically, six to eight colonies per sample) were typed by pulsed-field gel electrophoresis (PFGE). In most NP samples (89%) the multiple colonies shared common PFGE types and serotypes. However, four samples were heterogeneous (samples A through D): each contained two strains with different PFGE types, antibiotypes, and serotypes. Samples A and B each contained one strain of a vaccine capsular type a

Persistence, replacement, and microevolution of Cryptococcus neoformans strains in recurrent meningitis in AIDS patients.

Six separate human immunodeficiency virus-positive patients with cryptococcal meningitis were each found to have been infected with a unique strain of Cryptococcus neoformans on the basis of genomic DNA finger-printing analysis with the microsatellite sequence-containing oligonucleotide probe (GGAT)4 and by random amplification of polymorphic DNA. Two patients (A and B) experienced a recurrent episode of infection. Between 12 and 16 single-colony isolates recovered from primary isolation media (> 50% of C. neoformans colonies recovered) from cerebrospinal fluid specimens were fingerprinted fro

Role of pili and the phase-variable PilC protein in natural competence for transformation of Neisseria gonorrhoeae.

The Gram-negative bacterial pathogen Neisseria gonorrhoeae is naturally competent for transformation with species-related DNA. We show here that two phase-variable pilus-associated proteins, the major pilus subunit (pilin, or PilE) and PilC, a factor known to function in the assembly and adherence of gonococcal pili, are essential for transformation competence. The PilE and PilC proteins are necessary for the conversion of linearized plasmid DNA carrying the Neisseria-specific DNA uptake signal into a DNase-resistant form. The biogenesis of typical pilus fibers is neither essential nor suffici

Relationship of staphylococcal conjugative plasmids to large gentamicin-resistance nonconjugative plasmids in clinical isolates of Staphylococcus epidermidis.

Self-mobilization of large plasmids was not observed in 15 of 15 strains of gentamicin-resistant (Gmr) Staphylococcus epidermidis clinical isolates that were taken from bacteremic newborns in a neonatal intensive care unit. Alternatively, nine Gmr Staphylococcus aureus clinical isolates, which were isolated along with the S. epidermidis strains, were all shown previously to contain Gmr conjugative plasmids. All of the nonconjugative strains had plasmids that were similar in size to the S. aureus conjugative plasmids. Transfer of seven of these nonconjugative plasmids by protoplast transformati

Fitness cost of ceftazidime-avibactam resistance acquisition in carbapenem-resistant hypervirulent Klebsiella pneumoniae.

BACKGROUND: Antibiotic-resistant Klebsiella pneumoniae (KP) poses a serious global public health threat. However, research on the resistance mechanisms and accompanying phenotypic changes in carbapenem-resistant hypervirulent KP (CR-hvKP) under antibiotic treatment remains limited. This study aims to investigate the resistance mechanisms of CR-hvKP to ceftazidime-avibactam (CAZ-AVI) and its concomitant phenotypic shifts, employing an in vitro induction assay. METHODS: Six ceftazidime-avibactam (CAZ-AVI)-susceptible CR-hvKP clinical isolates were subjected to in-vitro resistance induction. We

Widely dispersed clonal expansion of multi-fungicide-resistant Aspergillus fumigatus limits genomic epidemiology prospects.

Invasive aspergillosis, caused by Aspergillus fumigatus, represents a critical public health concern, particularly due to increasing resistance to triazole antifungals linked to TR34/TR46 cyp51A haplotypes. In our genomic epidemiology study of 157 A. fumigatus isolates from Dutch environmental hotspots and two clinical centers, we identified near-identical genomes in several environmental and patient isolates, indicating a probable link. However, the geographic and temporal data alone are not sufficient to explain direct transmission pathways. Furthermore, a comparison with more than 1,200 glo

Dominance of high-risk clones ST2 and ST571 and the diversity of resistance islands in clinical Acinetobacter baumannii isolates from Hanoi, Vietnam.

Multidrug-resistant Acinetobacter baumannii poses a significant threat to hospital environments worldwide, including Vietnam. In this study, we conducted whole-genome sequencing on 30 clinical A. baumannii isolates from Hanoi to explore their genomic diversity, antibiotic resistance determinants, virulence factors and mobile genetic elements. Phylogenetic analyses, utilizing both SNP-based and multilocus sequence typing-based approaches, revealed that the isolates clustered into various sequence types (STs). Among these, ST2 and ST571 emerged as the dominant high-risk clones. The ST2 isolates

Pseudomonas fragi or not? Beyond MALDI-TOF in the frontier of Pseudomonas diversity.

Advanced platforms, such as whole genome sequencing (WGS), should be employed to enhance and refine microbial identification compared to phenotypic methods, including miniaturized biochemical tests and MALDI-TOF. The application of WGS has led to the reclassification of clinical bacterial pathogens previously misidentified by phenotypic techniques. In this study, eight clinical isolates initially identified as Pseudomonas fragi by VITEK MS were subjected to WGS and bioinformatics analysis. The results revealed one strain as Pseudomonas lundensis, while average nucleotide identity and phylogene

Aspergillus fumigatus sensu stricto genetic diversity from cystic fibrosis patients.

We aimed to access the genetic diversity of Apergillus fumigatus strains obtained from cystic fibrosis (CF) patients from southern Brazil. A. fumigatus sensu stricto isolates from respiratory clinical specimens were genotyped by microsatellite markers and azole resistance was evaluated by azole-agar screening. Twenty-seven isolates from twenty-seven patients showed a high genetic diversity, with the differentiation of 25 different genotypes (25 unique and one common to two isolates). All isolates were susceptible to the azoles tested. We believe that prospectively monitoring A. fumigatus genet

Genetic characterization of vancomycin-resistant Enterococcus faecium isolates from neutropenic patients in Tunisia: spread of the pandemic CC17 clone associated with high genetic diversity in Tn1546-like structures.

AIMS: In Tunisia, limited research has focused on characterizing clinical vancomycin-resistant Enterococcus faecium (VREfm). This study aimed to bridge this knowledge gap by molecular characterization of antimicrobial resistance, determining the genetic elements mediating vancomycin-resistance, and whole-genome sequencing of one representative VREfm isolate. METHODS AND RESULTS: Over 6 years (2011-2016), a total of eighty VREfm isolates responsible for infection or colonization were identified from hospitalized patients, with the incidence rate increasing from 2% in 2011 to 27% in 2016. All o
