Adolescent exposure to THC in female rats disrupts developmental changes in the prefrontal cortex.

Current concepts suggest that exposure to THC during adolescence may act as a risk factor for the development of psychiatric disorders later in life. However, the molecular underpinnings of this vulnerability are still poorly understood. To analyze this, we investigated whether and how THC exposure in female rats interferes with different maturational events occurring in the prefrontal cortex during adolescence through biochemical, pharmacological and electrophysiological means. We found that the endocannabinoid system undergoes maturational processes during adolescence and that THC exposure d

Glucocorticoid inhibits growth factor-induced differentiation of hippocampal progenitor HiB5 cells.

In the present study, we investigated the effect of glucocorticoid on neuronal differentiation of hippocampal progenitor HiB5 cells. Dexamethasone (DEX), a synthetic glucocorticoid, inhibited platelet-derived growth factor (PDGF)-induced differentiation of HiB5 cells. The inhibitory effect of DEX was antagonized by RU486, a glucocorticoid receptor (GR) antagonist, indicating the GR-mediated processes. Nestin mRNA level was decreased and midsize neurofilament (NF-M) mRNA level was increased as a function of neuronal differentiation. DEX significantly blocked PDGF-induced down-regulation of nest

Improvements in MPTP-induced object retrieval deficits and behavioral deficits after fetal nigral grafting in monkeys.

Improvements in MPTP-induced deficits were only found in subjects that received fetal substantia nigra transplants into the caudate nucleus. … led that cells of the fetal substantia nigra grafted into the caudate nucleus survived and extended neurites into the host striatum. Indeed, grafted dopamine neurons were often associated with appreciable innervation of the caudate nucleus and appeared to be well incorporated into the host brain. DENN/MADD/IG20 alternative splicing changes and cell death in Alzheimer's disease.

The potential effects of alternative splicing of death-domain expressing genes and neuronal death have not been determined in Alzheimer's disease (AD). We analyzed DENN/MADD/IG20 (DMI), the complex of four splice variants. IG20 is known to be involved in cell death and the DENN/MADD splice variant (DM-SV) in cell survival in non-neural systems. DENN/MADD (DM) and DENN/MADD splice variant 2 were also included. Using SH-SY5Y human neuroblastoma cultures exposed to high concentrations of oligomeric Aβ peptides (oAβ) as a model for neuronal death, there was initially an increased ratio of DM-SV to

Variability of beta-amyloid protein deposited lesions in Down's syndrome brains.

An immunohistochemical study was carried out on the brains of 7 adult Down's syndrome cases (ages 31 to 62) using antibodies to beta-protein, beta-amyloid protein precursor and tau-protein. Variable forms of beta-protein deposited lesions (including senile plaques and cerebrovascular amyloidosis) were observed in extensive areas of the neocortex of all cases and coexistence of both beta-protein amyloid fibrils and beta-amyloid protein precursors was also seen in some of these lesions. Moreover, 3 cases at an advanced stage showed a few plaque-like lesions with beta-protein immunoreactivity in

Microglial depletion prevents extracellular matrix changes and striatal volume reduction in a model of Huntington's disease.

Huntington's disease is associated with a reactive microglial response and consequent inflammation. To address the role of these cells in disease pathogenesis, we depleted microglia from R6/2 mice, a rapidly progressing model of Huntington's disease marked by behavioural impairment, mutant huntingtin (mHTT) accumulation, and early death, through colony-stimulating factor 1 receptor inhibition (CSF1Ri) with pexidartinib (PLX3397) for the duration of disease. Although we observed an interferon gene signature in addition to downregulated neuritogenic and synaptic gene pathways with disease, overt

Alpha-synuclein accumulates in Purkinje cells in Lewy body disease but not in multiple system atrophy.

Alpha-synuclein has an important role in the pathogenesis of Parkinson disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy (MSA), comprising a new disease concept, that of alpha-synucleinopathies. Cerebellar degeneration with Purkinje cell depletion is present in the majority of MSA cases. By contrast, cerebellar pathology has not been demonstrated unequivocally in either PD or DLB. Recent immunohistochemical studies using anti-alpha-synuclein antibodies have shown that LB-type degeneration in PD and DLB is more widespread than previously recognized. To determine whether

Topology-aware segmentation for tubular structure in 3D microscopy.

High-resolution 3D microscopy has become a foundational tool in biomedical research by reconstructing vascular and neural networks from subcellular to organ scales, thereby facilitating quantitative analysis of tissue microenvironments, disease progression, and early pathological changes. However, accurately segmenting tubular structure like microvascular and neurite in 3D microscopy remains challenging due to their densely packed, span a wide range of calibers, and branch extremely frequently. These characteristics hinder existing deep learning-based segmentation methods from simultaneously e

Biological plausibility of single-shell NODDI-DTI in non-demented older adults: associations with plasma biomarkers and follow-up cognition.

BACKGROUND: White matter (WM) degeneration is increasingly recognized as a major feature of aging and neurodegenerative disorders, such as Alzheimer's disease (AD). NODDI-DTI is a technique that allows the estimation of neurite density index (NDI) and orientation dispersion index (ODI) in white matter from single-shell diffusion tensor imaging. However, it should be interpreted as a tensor-derived approximation and biological support for these metrics is needed. This study aimed to provide multimodal evidence supporting the biological plausibility of NODDI-DTI estimates. METHODS: Demographic,

A sustained-release sinomenine hydrogel targets microglial ferroptosis via Nrf2-hmox1 activation to alleviate diabetic peripheral neuropathy.

BACKGROUND: Sinomenine (SIN), a bioactive alkaloid from Sinomenium acutum, possesses anti-inflammatory properties, but its efficacy against diabetic peripheral neuropathy (DPN) is limited by poor bioavailability and undefined mechanisms. PURPOSE: This study aimed to evaluate the therapeutic effects and mechanism of a SIN-loaded hydrogel (Gel-SIN-M) in DPN. METHODS: Gel-SIN-M was engineered via crosslinking for controlled SIN delivery. DPN mice received perineural Gel-SIN-M injections. Pain thresholds (von Frey) and motor coordination (gait analysis) were quantified. Intraepidermal nerve fibe

Single-Cell Proteomics Reveals Proteome Remodeling and Cellular Heterogeneity During NGF-Induced PC12 Neuronal Differentiation.

Single-cell proteomics enables direct measurement of cellular heterogeneity during dynamic biological processes, but its application to fragile and highly adherent neuronal models remains challenging. Here, we developed and applied an optimized single-cell proteomics workflow to characterize proteome remodeling during nerve growth factor (NGF)-induced differentiation of PC12 cells. To enable reliable single-cell analysis, we implemented gentle dissociation, antiaggregation strategies, and thermal inkjet-based cell dispensing, achieving high accuracy in single-cell isolation. Inclusion of n-dod

Natural Products and Neuroregeneration: Rethinking Discovery Beyond Bioavailability Through Pseudo-Natural Product Design.

Natural product (NP)-based neuroregeneration research has generated extensive preclinical evidence over the past five decades. Pharmacological activity across core processes of central nervous system (CNS) repair including neurogenesis, axonal regeneration, and neuroplasticity have been documented. Despite consistent observations of neurite outgrowth, neuroprotection, and partial functional recovery in cellular and animal models, translation into durable clinical therapies has remained limited. Neurotrophins such as nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF) similar

Frequency of mixed neuropathologies in individuals with down syndrome with and without Alzheimer's dementia.

Individuals with Down syndrome (DS) develop Alzheimer's disease neuropathological change (ADNC) by the age of 40 years, and most develop dementia by their early 50s. The frequency of co-pathologies in clinically and neuropathologically characterized adults with DS has not been systematically characterized. We characterized the frequency of ADNC and common co-pathologies, including cerebral amyloid angiopathy (CAA), Lewy pathology (LP), limbic predominant age-related TDP-43 encephalopathy neuropathological change (LATE-NC), hippocampal sclerosis (HS), and other cerebrovascular and macroscopic f

Correlative ultrastructural mapping of Lewy pathology reveals regional diversity in Parkinson's and dementia with Lewy bodies.

Lewy body diseases, including Parkinson's disease (PD) and dementia with Lewy bodies (DLB), are defined by neuronal accumulation of misfolded α-synuclein (α-Syn), yet the ultrastructural diversity of these inclusions across brain regions and disease contexts remains unclear. Here, we applied large-scale correlative light and electron microscopy (CLEM) to map α-Syn pathology across cortical regions (entorhinal cortex, ENT; anterior cingulate cortex, AC; hippocampal CA2 region) and substantia nigra (SN) in clinically and pathologically confirmed PD and DLB donors. We identified pronounced region
