Zinc oxide nanoparticles (ZnO-NPs) have gained attention for their anticancer and antimicrobial activity. Our study highlights a novel anti-virulence strategy against multidrug-resistant pathogens by showing that ZnO-NPs suppress bacterial virulence and quorum-sensing genes (rmpA, fnbA, cna, and LuxS) at sub-MIC levels. In this study, we synthesized ZnO-NPs using the chemical co-precipitation process, we confirmed their characteristics with the techniques TEM, XRD, UV-Vis spectroscopy, and measuring their zeta potential. ZnO-NPs are almost spherical, 30 nanometers in size, with a notable UV ab

BACKGROUND: Cancer cells, such as non-small cell lung cancer (NSCLC) cells, exhibit remarkable phenotypic plasticity and undergo epigenetic reprogramming, characteristics that enable them to evade targeted therapies. However, how NSCLC cells use epigenetic regulatory mechanisms (including endogenous DNA base damage) to drive transcriptome reprogramming and develop resistance to EGFR-tyrosine kinase inhibitor (EGFR-TKI) therapy remains unclear. METHODS: We employed an integrated multi-omics approach in erlotinib-sensitive and resistant NSCLC cells, including genome-wide mapping of abasic sites

BACKGROUND: Vibrio vulnificus is a Gram-negative pathogen that infects humans through foodborne or wound infections. Victims of V. vulnificus infections face significant health risks, including cellulitis and septicemia, which have rapid disease progression and high mortality rates. Diguanylate cyclase is responsible for producing the secondary messenger cyclic di-GMP. It plays a crucial role in regulating various bacterial physiological processes, such as motility, toxicity, and pathogenicity, through transcriptional regulation and affecting cyclic di-GMP levels. However, the DGC-mediated pat

Zika virus (ZIKV) infection during pregnancy causes congenital defects such as fetal microcephaly. Monoclonal antibodies (MAbs) against the nonstructural protein 1 (NS1) have the potential to suppress ZIKV pathogenicity without enhancement of disease, but the pathways through which they confer protection remain obscure. Here, we report two types of NS1-targeted human MAbs that inhibit ZIKV infection through distinct mechanisms. MAbs 3G2 and 4B8 show a better efficacy than MAb 4F10 in suppressing ZIKV infection in C57BL/6 neonatal mice. Unlike MAb 4F10 that mainly triggers antibody-dependent ce

Xanthomonas oryzae is a serious pathogen causing bacterial leaf blight (BLB) disease in rice, markedly reducing its yield. In this study, the rice chorismate mutase (OsCM) gene was overexpressed in a bacterial leaf blight-susceptible rice line to investigate the functional role of OsCM in response to bacterial leaf blight stress. We reported that overexpression of OsCM altered the downstream pathway of aromatic amino acids, mitigating pathogen stress by altering stress-responsive genes and hormonal accumulation. Phenotypic evaluation showed that the lesion length in the transgenic line was sig

Xeroderma pigmentosum complementation group A (XPA), is defective in xeroderma pigmentosum patients, causing pre-disposition to skin cancer and neurological abnormalities, which is not well understood. Here, we analyzed the XPA-deficient cells transcriptional profile under oxidative stress. The imbalance in of ubiquitin-proteasome system (UPS) gene expression was observed in XPA-deficient cells and the involvement of nuclear factor erythroid 2-related factor-2 (NFE2L2) was indicated. Co-immunoprecipitation assays showed the interaction between XPA, apurinic-apyrimidinic endonuclease 1 (APE1) a

Members of the Multiple antibiotic resistance Regulator (MarR) family of DNA binding proteins regulate transcription of a wide array of genes required for virulence and pathogenicity of bacteria. The present study reports the molecular characterization of HosA (Homologue of SlyA), a MarR protein, with respect to its target gene, DNA recognition motif, and nature of its ligand. Through a comparative genomics approach, we demonstrate that hosA is in synteny with nonoxidative hydroxyarylic acid decarboxylase (HAD) operon and is present exclusively within the mutS-rpoS polymorphic region in nine d

Cronobacter is a group of important foodborne pathogens associated with neonatal meningitis, septicemia, and necrotizing enterocolitis. Among Cronobacter species, Cronobacter sakazakii is the most common species in terms of isolation frequency. However, the molecular basis involved in virulence differences among C. sakazakii isolates is still unknown. In this study, based on the determination of virulence differences of C. sakazakii G362 (virulent isolate) and L3101 (attenuated isolate) through intraperitoneal injection, histopathologic analysis (small intestine, kidney, and liver) further con

We characterized key components and major targets of the c-di-GMP signaling pathways in the foodborne pathogen Listeria monocytogenes, identified a new c-di-GMP-inducible exopolysaccharide responsible for motility inhibition, cell aggregation, and enhanced tolerance to disinfectants and desiccation, and provided first insights into the role of c-di-GMP signaling in listerial virulence. Genome-wide genetic and biochemical analyses of c-di-GMP signaling pathways revealed that L. monocytogenes has three GGDEF domain proteins, DgcA (Lmo1911), DgcB (Lmo1912) and DgcC (Lmo2174), that possess diguany

The results of the examination of association of polymorphisms of DNA repair genes and chromosomal aberrations in lung cancer patients are discussed. A significant positive association between the hOGG1 G/G genotypes, XPD G/G genotype and lung cancer was found. The hOGG1 C/C genotypes were significantly negatively associated with lung cancer. The patient chromosomal aberration frequencies were significantly higher than in control. Carriers of all APE1 and XPD genotypes, XRCC1 G/G genotype, ADPRT T/T genotype, hOGG1 C/C and Ser/Cys genotypes had statistically significant differences in the leve

1. The aim of the study was to determine if H(2)S is involved in the development of hypoxia-induced pulmonary hypertension in broilers, a condition frequently observed in a variety of cardiac and pulmonary diseases. 2. Two-week-old broilers were reared under normoxic conditions or exposed to normobaric hypoxia (6 h/day) with tissue levels of H(2)S adjusted by administering sodium hydrosulfide (NaHS, 10 µmol/kg body weight/day). Mean pulmonary arterial pressure, right ventricular mass, plasma and tissue H(2)S levels, the expression of cystathionine-β-synthase (CSE) and vascular remodeling were

The 'stage albinism line of winter wheat' FA85 exhibits a severe block in chlorophyll (Chl) biosynthesis with prolonged low-temperature treatment. The correlations between leaf color and low temperature provide more comprehensive understanding of low temperature as an environmental signal that regulate the metabolic changes in the entire Chl-synthesizing pathway. In this study, we investigated differences in Chl biosynthesis between leaves of Aibian1 and FA85 by measuring their Chl precursors and heme content, transcripts for key genes of Chl biosynthesis and key enzyme activities. The physiological behavior of Helicobacter pylori in different growth conditions was investigated to approach its growth standardization. H. pylori free-living and biofilm modes of growth were assessed in four different liquid culture media (Brucella broth, brain heart infusion broth and Ham's F-12 supplemented with 2% fetal calf serum and Ham's F-12 without serum). Free-living growth was monitored during 72 h in each medium and characterized for bacterial density, culturability, viability and morphology. The biofilm formation in the same media was evaluated for biomass production, colony form

Crohn's disease (CD) and celiac disease (CelD) are chronic intestinal inflammatory diseases, involving genetic and environmental factors in their pathogenesis. The two diseases can co-occur within families, and studies suggest that CelD patients have a higher risk to develop CD than the general population. These observations suggest that CD and CelD may share common genetic risk loci. Two such shared loci, IL18RAP and PTPN2, have already been identified independently in these two diseases. The aim of our study was to explicitly identify shared risk loci for these diseases by combining results

Reactive oxygen species (ROS) play an important role as mediators of pulmonary damage in mineral dust-induced diseases. Studies carried out to date have largely focused on silica-induced production of ROS by lung phagocytes. In this study we investigated the hypothesis that crystalline silica Min-U-Sil 5 can induce elevations in intracellular ROS in human bronchial epithelial cells BEAS-2B, via an indirect mechanism that involves ROS-inducing intracellular factors, through a reduction of antiglycation (glyoxalase enzymes) and antioxidant (paraoxonase 1 and glutathione-S-transferases) enzymatic

Two subspecies of Trypanosoma brucei s.l. co-exist within the animal populations of Eastern Africa; T. b. brucei a parasite which only infects livestock and wildlife and T. b. rhodesiense a zoonotic parasite which infects domestic livestock, wildlife, and which in humans, results in the disease known as Human African Trypanosomiasis (HAT) or sleeping sickness. In order to assess the risk posed to humans from HAT it is necessary to identify animals harbouring potentially human infective parasites. The multiplex PCR method described here permits differentiation of human and non-human infective p

Glyoxalases are involved in a ubiquitous detoxification pathway. In pursuit of a better understanding of the biological function of the enzyme, the recombinant glyoxalase I (LdGLOI) protein has been characterized from Leishmania donovani, the most important pathogenic Leishmania species that is responsible for visceral leishmaniasis. A 24kDa protein was heterologously expressed in Escherichia coli. LdGLOI showed a marked substrate specificity for trypanothione hemithioacetal over glutathione hemithioacetal. Antiserum against recombinant LdGLOI protein could detect a band of anticipated size ap
