Capillary cerebral amyloid angiopathy is associated with vessel occlusion and cerebral blood flow disturbances.

The role of cerebral amyloid angiopathy (CAA) in the pathogenesis of Alzheimer's disease (AD) is not fully understood. Here, we studied whether CAA is associated with alterations in microvascularisation in transgenic mouse models and in the human brain. APP23 mice at 25-26 months of age exhibited severe CAA in thalamic vessels whereas APP51/16 mice did not. Wild-type littermates were free of CAA. We found CAA-related capillary occlusion within the thalamus of APP23 mice but not in APP51/16 and wild-type mice. Magnetic resonance angiography (MRA) showed blood flow alterations in the thalamic ve

Exosomal microRNA-143-3P and its effect on biological behavior and angiogenesis in breast cancer.

We undertook this study to investigate the effect of exosomal microRNA-143-3p (miR-143-3p) on the biological behavior and angiogenesis in breast cancer. We transfected human breast cancer cells (MDA-MB-231) with miR-143-3p Agomir or Agomir-NC, and we isolated the exosomes (ex) that were secreted and named these miR-143-3p Agomir-ex and Agomir-NC-ex, respectively. We performed real-time polymerase chain reaction (real-time PCR) to detect the level of exosomal miR-143-3p. We treated the MDA-MB-231 cells with exosomal miR-143-3p Agomir-ex and Agomir-NC-ex. We measured the cellular miR-143-3p conc

Impact of time after anterior cruciate ligament injury on vascular and bone quality at the femoral insertion site: a histological cohort study.

BACKGROUND: Anterior cruciate ligament (ACL) injuries are common, yet little is known about the relationship between time from injury to reconstruction and biological characteristics at the femoral insertion site. This study evaluated whether longer time from ACL injury to reconstruction was associated with differences in vascularity and bone microarchitecture at the femoral ACL footprint. METHODS: In this observational cohort study with cross-sectional histological assessment, 60 adult patients (22-53 years) undergoing primary ACL reconstruction and 10 reference patients (55-65 years) underg

Magnetically ordered yet topologically robust phases emerging in concurrent Kitaev spin liquids.

Peroxisome proliferator-activated receptors (PPARs) are members of the nuclear receptor superfamily that regulate lipid, glucose, and amino acid metabolism. More recently, PPARs and corresponding ligands have been shown in skin and other organs to regulate important cellular functions, including cell proliferation and differentiation, as well as inflammatory responses. These new functions identify PPARs and corresponding ligands as potential targets for the treatment of various skin diseases and other disorders. It has been shown that in inflammatory skin disorders, including hyperproliferativ

CEBPD-mediated SGPP2 upregulation via PERK/ER stress in endothelial cells disrupts S1P homeostasis and impairs angiogenesis in chronic endometritis.

BACKGROUND: Chronic endometritis (CE) is a persistent inflammatory condition associated with adverse pregnancy outcomes. Although impaired endometrial angiogenesis is thought to contribute to its pathogenesis, the underlying molecular mechanisms remain incompletely understood. This study aimed to investigate whether sphingolipid metabolism plays a role in the vascular dysfunction of CE patients. METHODS: Endometrial samples from control and CE patients were assessed for angiogenesis using immunohistochemistry. Sequencing data of endometrial tissues indicated dysregulation of sphingolipid meta

Neuroprotective role of pyrroloquinoline quinone in folate deficiency-induced blood-brain barrier disruption.

Healthy neurodevelopment requires adequate folates (vitamin B9), which are critical for key biosynthetic and homeostatic processes in the central nervous system (CNS). In the brain, folate transport is mediated by three major pathways: folate receptor alpha (FRα), proton-coupled folate transporter (PCFT), and reduced folate carrier (RFC). Folate uptake primarily occurs at the blood-cerebrospinal fluid barrier (BCSFB) by concerted actions of FRα and PCFT. Alterations in this transport pathway can result in cerebral folate deficiency (CFD), a rare but devastating pediatric condition associated w

Modulation of PI3K/AKT/mTOR signaling pathway by combined stem cell and phytochemical treatment improves metabolic and reproductive outcomes in PCOS.

BACKGROUND: PCOS is characterized by ovulatory dysfunction and metabolic abnormalities, and growing evidence suggests that dysregulated autophagy in ovarian granulosa cells contributes to its pathogenesis. Mesenchymal stem cells (MSCs) and Medicine Lin (ML) therapy have been proven to improve these dysfunctions in PCOS patients, enhancing fertility and pregnancy outcomes, though their underlying mechanisms remain unclear. This study hypothesized that MSCs and ML function by regulating the autophagy-mediated PI3K/AKT/mTOR signaling pathway. METHODS: A PCOS mouse model was established through hi

Intratracheal administration of mesenchymal stem cells ameliorates hyperoxia-induced bronchopulmonary dysplasia by inhibiting NLRP3 inflammasome activation: the critical role of Aldh1a2.

BACKGROUND: Bronchopulmonary dysplasia (BPD) is a chronic lung disease driven by inflammation and oxidative stress. Mesenchymal stem cells (MSCs) have shown protective effects against hyperoxic lung injury. However, few studies have thoroughly examined the significantly differentially expressed genes (DEGs) in the lungs before and after MSC treatment. In this study, we analyzed the significant DEGs in lung tissues during both in vivo and vitro umbilical cord-derived mesenchymal stem cells (UCMSCs)-mediated repair of hyperoxic lung injury and investigated their potential mechanisms of action.

Targeted trapping of endogenous endothelial progenitor cells for myocardial ischemic injury repair through neutrophil-mediated SPIO nanoparticle-conjugated CD34 antibody delivery and imaging.

Endothelia progenitor cell (EPC)-based revascularization therapies have shown promise for the treatment of myocardial ischemic injury. However, applications and efficacy are limited by the relatively inefficient recruitment of endogenous EPCs to the ischemic area, while implantation of exogenous EPCs carries the risk of tumorigenicity. In this study, we developed a therapeutic protocol that relies on the capacity of neutrophils (NEs) to target lesions and release preloaded EPC-binding molecules for high efficiency capture. Neutrophils were loaded with superparamagnetic iron oxide nanoparticles

Hepatitis B Virus Small Envelope Protein Promotes Hepatocellular Carcinoma Angiogenesis via Endoplasmic Reticulum Stress Signaling To Upregulate the Expression of Vascular Endothelial Growth Factor A.

Hepatocellular carcinoma (HCC) is a hypervascular tumor, and accumulating evidence has indicated that stimulation of angiogenesis by hepatitis B virus (HBV) may contribute to HCC malignancy. The small protein of hepatitis B virus surface antigen (HBsAg), SHBs, is the most abundant HBV protein and has a close clinical association with HCC; however, whether SHBs contributes to HCC angiogenesis remains unknown. This study reports that the forced expression of SHBs in HCC cells promoted xenograft tumor growth and increased the microvessel density (MVD) within the tumors. Consistently, HBsAg was al

A collagen hydrogel loaded with HDAC7-derived peptide promotes the regeneration of infarcted myocardium with functional improvement in a rodent model.

Myocardial infarction (MI) leads to the loss of cardiomyocytes, left ventricle (LV) dilation, and cardiac dysfunction, eventually developing into heart failure. Most of the strategies for MI therapy require biomaterials that can support tissue regeneration. In this study, we hypothesized that the extracellular matrix (ECM)-derived collagen I hydrogel loaded with histone deacetylase 7 (HDAC7)-derived-phosphorylated 7-amino-acid peptide (7Ap) could restrain LV remodeling and improve cardiac function after MI. An MI model was established by ligation of the left anterior descending coronary artery

SssP1, a Streptococcus suis Fimbria-Like Protein Transported by the SecY2/A2 System, Contributes to Bacterial Virulence.

Streptococcus suis is an important Gram-positive pathogen in the swine industry and is an emerging zoonotic pathogen for humans. In our previous work, we found a virulent S. suis strain, CZ130302, belonging to a novel serotype, Chz, to be associated with acute meningitis in piglets. However, its underlying mechanisms of pathogenesis remain poorly understood. In this study, we sequenced and analyzed the complete genomes of three Chz serotype strains, including strain CZ130302 and two avirulent strains, HN136 and AH681. By genome comparison, we found two putative genomic islands (GIs) uniquely e
