INTRODUCTION: Alzheimer's disease (AD) biomarkers are assessed on their ability to detect AD pathophysiology in vivo, with confirmation of AD neuropathology only at autopsy.
METHODS: Positron emission tomography (PET), plasma, and cognitive AD biomarkers were compared to AD neuropathology in Harvard Aging Brain Study participants (10 cognitively unimpaired; 6 mild cognitive impairment). Different PET methods were evaluated, for example, standardized uptake volume ratio (SUVR), distribution volume ratio (DVR), spatial extent (EXT), and partial-volume correction (PVC).
RESULTS: Amyloid beta (A

BACKGROUND AND PURPOSE: Central nervous system (CNS) relapse in diffuse large B-cell lymphoma (DLBCL) represents a clinical challenge, with risk stratification based on the CNS International Prognostic Index (CNS-IPI) potentially leading to over- and undertreatment. This study investigated whether diffusion MRI metrics could detect CNS microstructural changes associated with CNS relapse risk factors.
MATERIALS AND METHODS: In this cross-sectional study 63 DLBCL patients without CNS involvement, and 11 healthy controls, underwent brain MRI including multi-shell diffusion tensor imaging, utilis

INTRODUCTION: Exposure to alcohol (ethanol) is the most common toxic insult during human development and is a teratogenic agent associated with abnormalities ranging from behavioral disorders to fetal alcohol syndrome. Ethanol disrupts gene regulatory programs crucial for neuronal identity, including alternative splicing. A distinctive feature of developing neuronal transcriptomes is the high prevalence of microexon inclusion, which consists of short exonic sequences ranging from 3 to 51 nucleotides. However, the impact of ethanol on microexon inclusion remains poorly understood.

Peripheral nerve regeneration depends on interactions among extracellular matrix cues, neuronal bioenergetics, and therapeutic signal presentation, yet progress is limited by the lack of physiologically relevant quantitative in vitro models to evaluate biomaterials and mitochondria-targeted therapies. Here, we present a nerve injury-on-a-chip (NI-Chip) that integrates dorsal root ganglion explants, aligned nanofibers, compartmentalized microfluidics, and precisely controlled axotomy to enable real-time, quantitative analysis of axonal regeneration dynamics. Extracellular matrix composition sig

INTRODUCTION: Neurons depend on the ubiquitin system to maintain proteostasis, which plays crucial roles in processes of neuronal death and axon regeneration after injuries and diseases. Developing druggable targets within the ubiquitin system that simultaneously support neuroprotection and axon regeneration has been considered an attractive therapeutic strategy.
OBJECTIVES: To systematically identify druggable molecules within the ubiquitin signaling pathway and evaluate their efficacy in neuroprotection.
METHODS: To this end, we screened 181 ubiquitination-related small-molecule compounds

Alzheimer disease neuropathologic change (ADNC) is considered to be the most common cause of cognitive decline and dementia worldwide. ADNC level is determined using the density of neuritic plaques in combination with the topographical distribution of β-amyloid (Aβ) plaques and hyperphosphorylated tau (p-tau)-positive neurofibrillary tangles (NFTs). While cognitive decline correlates with the level of ADNC, there remains a great deal of variation in cognitive outcomes between individuals that is unaccounted for by current neuropathologic evaluation metrics. We leveraged quantitative computer-a

Parkinson's disease (PD) encompasses motor and neuropsychiatric symptoms (NPS) including depression, anxiety, and apathy. Cognitive subtypes-PD with mild cognitive impairment (PD-MCI) and PD with dementia (PDD)-and the related Dementia with Lewy Bodies (DLB) exist within the spectrum of Lewy body disease (LBD). While NPS are a major comorbidity, established imaging biomarkers are not available, nor is the neuronal basis well known. Additionally, prevalence of NPS may be linked to cognitive status. The hippocampus is implicated in the development of NPS and may undergo microstructural changes t

Amyloid-beta (Aβ) aggregation is the key component of neuritic plaques that drives Alzheimer's disease (AD) progression and cognitive decline. Although synaptic dysfunction strongly correlates with cognitive impairment, its underlying mechanisms remain unclear. Recently, the kynurenine pathway (KP) of tryptophan metabolism has emerged as a key contributor to AD pathology, and xanthurenic acid (XA), a naturally occurring end-product of the KP, has been implicated in neuroprotection. In this study, we investigated the neuroprotective effects of intranasally administered XA in an Aβ-induced AD mo

INTRODUCTION: Epidemiological studies highlight a significant occurrence of ischemic strokes in cancer patients, particularly in the elderly, where stroke and systemic cancer are the leading causes of death. This comprehensive study investigates the combined effect of melatonin (MT) and exercise (Ex) against stroke, and a chemotherapeutic drug doxorubicin (Dox)-induced cardiotoxicity (DIC).
METHOD: This study employs two models-middle cerebral artery occlusion (MCAo) and DIC-to evaluate the synergistic effects of MT + Ex. For both models, rats were assigned to the same treatment groups (n = 5

Alzheimer disease (AD) polygenic risk scores (ADPRS) built from cell-type (ct) specific genetic variants can be used to infer cell-type contributions to AD. We derived two ct-ADPRSs using variants near single-nuclei RNA-seq (snRNA) derived cell-type specific genes or on single-nuclei ATAC-seq (snATAC) derived cell-type specific accessible chromatin regions. We generated a multi-omic ct-ADPRS for eight neuron subtypes using both single-nuclei datasets. SnATAC-derived ct-ADPRSs demonstrated considerably lower correlations among cell types (average r = 0.071) than snRNA-derived ct-ADPRSs (average

OBJECTIVE: Drug-resistant epilepsy, a severe and diagnostically challenging neurological condition often linked to focal cortical dysplasia (FCD), represents a significant social and healthcare burden. While the thalamus is notably altered in this disorder, it remains unclear whether these changes result from epilepsy manifestation or participate in its pathophysiology. This study aimed to characterize volumetric and microstructural changes in the thalamus and thalamocortical pathways in epilepsy patients with FCD.
METHODS: We prospectively enrolled 24 patients with drug-resistant epilepsy ca

Amyotrophic lateral sclerosis (ALS) is a progressive motor neurons (MNs) degenerative disease. Despite advancements in understanding ALS pathogenesis, drug development lags far behind. The reduced secretion of phosphoglycerate kinase 1 (Pgk1) by NogoA-overexpressing muscle cells inhibits neurite outgrowth of MNs (NOMNs). However, administration of extracellular Pgk1 (ePgk1) reduces phospho-Cofilin (p-Cofilin), a growth cone collapse marker, and mitigates MN degeneration. This improves NOMNs in NSC34 neural cells and locomotion in SOD1-G93A ALS-mice by suppressing the p-P38-T180/p-MK2-T334/p-Li

Neonatal brain injury (NBI) encompasses a variety of neurological acquired conditions affecting newborns. These conditions include hypoxia-ischemia, hyperoxic, periventricular leukomalacia, intrauterine infection, as well as perinatal cerebral hemorrhage. Each year, thousands of babies are born with signs of brain injury. It is estimated that two-thirds of these newborn infants with brain injury would either die or survive with mild to severe neurologic sequelae, largely due to the absence of no widely accepted treatment methods. Erythropoietin (Epo) is a humoral intermediary associated with t

APOEε4 is the major genetic risk factor for sporadic Alzheimer's disease (AD). Although APOEε4 is known to promote Aβ pathology, recent data also support an effect of APOE polymorphism on phosphorylated Tau (pTau) pathology. To elucidate these potential effects, the pTau interactome was analyzed across APOE genotypes in the frontal cortex of 10 advanced AD cases (n = 5 APOEε3/ε3 and n = 5 APOEε4/ε4), using a combination of anti-pTau pS396/pS404 (PHF1) immunoprecipitation (IP) and mass spectrometry (MS). This proteomic approach was complemented by an analysis of anti-pTau PHF1 and anti-Aβ 4G8 i

BACKGROUND: Soluble oligomeric forms of Tau protein have emerged as crucial players in the propagation of Tau pathology in Alzheimer's disease (AD). Our objective is to introduce a single-domain antibody (sdAb) named 2C5 as a novel radiotracer for the efficient detection and longitudinal monitoring of oligomeric Tau species in the human brain.
METHODS: The development and production of 2C5 involved llama immunization with the largest human Tau isoform oligomers of different maturation states. Subsequently, 2C5 underwent comprehensive in vitro characterization for affinity and specificity via

Alzheimer's disease (AD) is an age-related neurodegenerative condition and the most common type of dementia, characterised by pathological accumulation of extracellular plaques and intracellular neurofibrillary tangles that mainly consist of amyloid-β (Aβ) and hyperphosphorylated tau aggregates, respectively. Previous studies in mouse models with a targeted knock-out of the microtubule-associated protein tau (Mapt) gene demonstrated that Aβ-driven toxicity is tau-dependent. However, human cellular models with chronic tau lowering remain unexplored. In this study, we generated stable tau-deplet

BACKGROUND: Variants of the gene triggering receptor expressed on myeloid cells-2 (TREM2) increase the risk of Alzheimer's disease (AD) and other neurodegenerative disorders. Signaling by TREM2, an innate immune receptor expressed by microglia, is thought to enhance phagocytosis of amyloid beta (Aβ) and other damaged proteins, promote microglial proliferation, migration, and survival, and regulate inflammatory signaling. Thus, TREM2 activation has potential to alter the progression of AD. AL002 is an investigational, engineered, humanized monoclonal immunoglobulin G1 (IgG1) antibody designed t

INTRODUCTION: Endometriosis is a chronic inflammatory disease that affects ∼10 % of women. A significant fraction of patients experience limited or no efficacy with current therapies. Tissue adjacent to endometriosis lesions often exhibits increased neurite and vascular density, suggesting that disease pathology involves neurotrophic activity and angiogenesis.
OBJECTIVES: We aim to evaluate the potential for key tyrosine-kinase-receptor-coupled neurotrophic molecules to contribute to endometriosis-associated pain in mice.
METHODS: Peritoneal fluid was collected from endometriosis patients un

INTRODUCTION/AIMS: Diagnosis of small-fiber neuropathy (SFN) is hampered by its subjective symptoms and signs. Confirmatory testing is insufficiently available and expensive, so predictive examinations have value. However, few support the 2020 SFN consensus-case-definition requirements or were validated for non-diabetes neuropathies. Thus we developed the Massachusetts General Hospital Neuropathy Exam Tool (MAGNET) and measured diagnostic performance in 160 symptomatic patients evaluated for length-dependent SFN from any cause and 37 healthy volunteers.
METHODS: We compared prevalences of abn
