Juvenile hormone enhances viral activity in bumble bees.

Bees are susceptible to viral infections that can compromise individual and colony health. As viruses depend on host cellular resources for replication, we investigated the influence of a key insect regulator, Juvenile hormone (JH), which enhances reproduction and metabolic rate on viral activity. We hypothesized that elevated JH levels would promote viral replication in a tissue-specific manner. To test this hypothesis, we used the bumble bee, Bombus terrestris, and the Israeli acute paralysis virus (IAPV). In our first experiment, we compared IAPV replication in pre- and post-diapause queens

Effects of temperature and relative humidity on germination, vegetative growth, sporulation, and pathogenicity of native Beauveria bassiana isolates and formulations against adult desert locust (Schistocerca gregaria Forskål).

Entomopathogenic fungi such as Beauveria bassiana are promising biocontrol agents against the desert locust, Schistocerca gregaria, but their performance is strongly influenced by temperature and relative humidity (RH). In this study, native B. bassiana isolates recovered from soils in Isiolo and Laikipia Counties, Kenya, were molecularly identified and evaluated under factorial combinations of temperature (20, 25, 30, and 35 °C) and RH (50, 75, and 90%) to determine their effects on conidial germination, vegetative growth, sporulation, and pathogenicity against adult S. gregaria. Three native

Microcrystalline tyrosine as a novel depot-forming agent in venom immunotherapy: a pre-clinical evaluation in bee-venom allergic mice.

BACKGROUND: Venom immunotherapy (VIT) with aqueous venom extracts is a standard treatment for severe insect venom allergies. In other fields of allergen immunotherapy (AIT), depot adjuvants have been used for decades, with benefits for both safety and efficacy. Biodegradable microcrystalline tyrosine (MCT), is well-established in AIT, and has proven both safe and effective through sustained release and adjuvancy. The objective of the current study was to evaluate MCT as a depot-forming agent in VIT using a murine model of bee venom allergy. MATERIALS AND METHODS: Mice were sensitised with bee

Glycosomal Phosphoenolpyruvate Carboxykinase CRISPR/Cas9-Deletion and Its Role in Trypanosoma cruzi Metacyclogenesis and Infectivity in Mammalian Host.

Trypanosoma cruzi, the causative agent of Chagas disease, possesses glycosomes-unique organelles that house key metabolic enzymes, several of which are promising therapeutic targets. Among them, phosphoenolpyruvate carboxykinase (PEPCK) plays a central role in succinic fermentation, the main pathway for NAD+ regeneration within the organelle. Using CRISPR/Cas9 editing, the PEPCK gene was disrupted in T. cruzi, producing single-allele knockout epimastigotes (TcPEPCK-sKO) with reduced PEPCK expression and enzyme activity. In a high glucose environment, PEPCK disruption impaired glucose consumpti

Landscape modeling as a surveillance strategy for Trypanosoma cruzi vectors in Brazil.

BACKGROUND: The transmission of Trypanosoma cruzi in nature is closely linked to the ecology of its vectors, the triatomine insects. Although the geographic distribution of these species is relatively well-known, understanding the characteristics that define their ecological niche and how human interventions alter these conditions remains limited. Rapid and intense environmental changes, driven by activities such as agricultural expansion, raise important questions about how vector ecology is being transformed and the new implications for Chagas disease transmission risk. METHODS: This study

MicroRNA Lse-miR-2944c-3p regulates female reproduction in Lasioderma serricorne by targeting the heat shock protein LsHSP70-3.

BACKGROUND: The reproductive plasticity of female insects is governed by the complex structure and function of their ovaries. However, the molecular mechanisms underlying ovarian development in Lasioderma serricorne, a globally significant pest of stored products, remain largely unexplored. The role of microRNAs (miRNAs) in regulating genes associated with reproduction remains poorly characterized. RESULTS: This study investigated the regulatory mechanisms involving the LsHSP70-3 gene, which encodes a heat shock protein, and its associated miRNA in the reproductive processes of female L. serr

Gut microbial community structure of the adult citrus root weevil Diaprepes abbreviatus.

Diaprepes abbreviatus is an agricultural pest known to affect around 270 plant species across the Caribbean and the United States, posing significant challenges to pest management. Chemical control dominates management, but environmental and health concerns motivate microbiome-informed alternatives. However, limited information exists on the gut anatomy, physicochemical environment, and microbial composition of D. abbreviatus. In this study, we provide the first comprehensive characterization of the gut morphology, pH, and microbiota of adult D. abbreviatus in both females and males collected

Bioaccumulation and transferreing for impacts on Cd and Pb by aphid consumption of the broad bean, Vicia faba L, in soil heavy metal pollution.

Heavy metal pollution threatens human and ecological health. Heavy metals can exist in the soil for a long time and migrate to organisms along the food chain. However, only a few studies have investigated the effects of a single stress on broad beans. Here, we aimed to characterize Cd and Pb bioaccumulation, at varying concentrations, in the broad bean, Vicia faba L. We also determined how the bioaccumulated metals are impacted by aphids that consume the plant. No significant difference was noted in the germination rates of broad beans at the early stage of planting (after 8 days), but eventua

New insight into environmental oxidation of phosmet insecticide initiated by HO˙ radicals in gas and water - a theoretical study.

Phosmet is an organophosphorus insecticide widely used in agriculture to control a range of insects; recently, it was banned by the European Union in 2022 due to its harmful effects. However, its environmental degradation and fate have not yet been evident. Thus, phosmet oxidation by HO˙ radicals was theoretically studied in this work using the DFT approach at the M06-2X/6-311++G(3df,3pd)//M06-2X/6-31+G(d,p) level of theory. Three different mechanisms were considered, including formal hydrogen transfer (FHT), radical adduct formation (RAF), and single electron transfer (SET). The mechanisms, k

Association between the potential distribution of Lutzomyia longipalpis and Nyssomyia whitmani and leishmaniasis incidence in Piauí State, Brazil.

BACKGROUND: Leishmaniases are vector borne diseases caused by Leishmania spp. parasites transmitted by female sandflies (Diptera: Psychodidae) whose geographic distribution is influenced by environmental factors. Among the main tools for studying the distribution of vector species, modeling techniques are used to analyze the influence of climatic and environmental factors on the distribution of these insects and their association with human cases of the disease. METHODOLOGY/PRINCIPAL FINDINGS: Here, we used a multiscale ecological niche modeling approach to assess the environmental suitabilit

Toxic assessment, growth disrupting and neurotoxic effects of red seaweeds' botanicals against the dengue vector mosquito Aedes aegypti L.

Application of synthetic pesticides over decades to control insects, pests, and disease vectors has resulted in negative impacts on environment and health. The current study assessed the toxicological effects of 12 botanicals obtained from 4 different red seaweeds against the dengue vector mosquito Aedes aegypti L. (Diptera: Culicidae). Four species of red seaweeds, namely Laurencia karachiana, Gracilaria foliifera, Jania rubens, Asparagopsis taxiformis, were collected from Karachi coast and extracted with hexane, dichloromethane and methanol. The efficiency of these extracts was determined by

Functional Characterization of Odorant Binding Protein 27 (RproOBP27) From Rhodnius prolixus Antennae.

Olfactory proteins mediate a wide range of essential behaviors for insect survival. Odorant binding proteins (OBPs) are small soluble olfactory proteins involved in the transport of odor molecules (=odorants) through the sensillum lymph to odorant receptors, which are housed on the dendritic membrane of olfactory sensory neurons also known as olfactory receptor neurons. Thus, a better understanding of the role(s) of OBPs from Rhodnius prolixus, one of the main vectors of Chagas disease, may ultimately lead to new strategies for vector management. Here we aimed at functionally characterize OBPs

High level efficacy of lufenuron against sea lice (Lepeophtheirus salmonis) linked to rapid impact on moulting processes.

Drug resistance in the salmon louse Lepeophtheirus salmonis is a global issue for Atlantic salmon aquaculture. Multiple resistance has been described across most available compound classes with the exception of the benzoylureas. To target this gap in effective management of L. salmonis and other species of sea lice (e.g. Caligus spp.), Elanco Animal Health is developing an in-feed treatment containing lufenuron (a benzoylurea) to be administered prior to seawater transfer of salmon smolts and to provide long-term protection of salmon against sea lice infestations. Benzoylureas disrupt chitin s

Incongruent nuclear and mitochondrial genetic structure of new world screwworm fly populations due to positive selection of mutations associated with dimethyl- and diethyl-organophosphates resistance.

Livestock production is an important economic activity in Brazil, which has been suffering significant losses due to the impact of parasites. The New World screwworm (NWS) fly, Cochliomyia hominivorax, is an ectoparasite and one of the most important myiasis-causing flies endemic to the Americas. The geographic distribution of NWS has been reduced after the implementation of the Sterile Insect Technique (SIT), being eradicated in North America and part of Central America. In South America, C. hominivorax is controlled by chemical insecticides, although indiscriminate use can cause selection of

Effect of dietary carotenoids on vitamin A status and skin pigmentation in false tomato frogs (Dyscophus guineti).

Several species in captivity develop nutritional diseases including vitamin A deficiency; cases of this disease have been documented in amphibians, which may be linked to an insectivorous diet lacking in vitamin A or carotenoids. Adults and young of Dyscophus guineti were fed three diets over 9 weeks to evaluate effects on carotenoids and vitamin A status and skin pigmentation. Feeder crickets were either supplemented with soy oil (control, CON), soy oil enriched with β-carotene (BC) or mixed carotenoids (MIX) by direct injection of known dosages. Vitamin A from feeder crickets (measured as re

Reciprocal regulation of NF-kB (Relish) and Subolesin in the tick vector, Ixodes scapularis.

BACKGROUND: Tick Subolesin and its ortholog in insects and vertebrates, Akirin, have been suggested to play a role in the immune response through regulation of nuclear factor-kappa B (NF-kB)-dependent and independent gene expression via interaction with intermediate proteins that interact with NF-kB and other regulatory proteins, bind DNA or remodel chromatin to regulate gene expression. The objective of this study was to characterize the structure and regulation of subolesin in Ixodes scapularis. I. scapularis is a vector of emerging pathogens such as Borrelia burgdorferi, Anaplasma phagocyto

Loss of function of FATTY ACID DESATURASE7 in tomato enhances basal aphid resistance in a salicylate-dependent manner.

We report here that disruption of function of the ω-3 FATTY ACID DESATURASE7 (FAD7) enhances plant defenses against aphids. The suppressor of prosystemin-mediated responses2 (spr2) mutation in tomato (Solanum lycopersicum), which eliminates the function of FAD7, reduces the settling behavior, survival, and fecundity of the potato aphid (Macrosiphum euphorbiae). Likewise, the antisense suppression of LeFAD7 expression in wild-type tomato plants reduces aphid infestations. Aphid resistance in the spr2 mutant is associated with enhanced levels of salicylic acid (SA) and mRNA encoding the pathogen

The greening of pesticide-environment interactions: some personal observations.

BACKGROUND: Pesticide-environment interactions are bidirectional. The environment alters pesticides by metabolism and photodegradation, and pesticides in turn change the environment through nontarget or secondary effects. OBJECTIVES: Approximately 900 currently used commercial pesticides of widely diverse structures act by nearly a hundred mechanisms to control insects, weeds, and fungi, usually with minimal disruption of nature's equilibrium. Here I consider some aspects of the discovery, development, and use of ecofriendly or green pesticides (i.e., pesticides that are safe, effective, and
