Synthesis and characterization of self-assembled CdHgTe/gelatin nanospheres as stable near infrared fluorescent probes in vivo.

This work presented a kind of novel near infrared emitting CdHgTe/gelatin nanospheres which were synthesized with Cd(NO(3))(2), Hg(NO(3))(2), NaHTe and a thiol stabilizer in gelatin solution. The self-assembled nanospheres were megranate-like and nearly 40 nm in diameter, with CdHgTe QDs uniformly embedded in gelatin matrix. They exhibited strong fluorescence ranging from 580 to 800 nm that could be tuned by molar ratios of Hg(2+) and gelatin. The full widths at half-maximum of the emission spectra were in the range of 60-80 nm. Compared with bare CdHgTe QDs, the photostability of this compact

Brightly Fluorescent Self-Assembled Supra-J-Aggregate Nanoparticles for Bioanalysis and Imaging.

J-aggregates of molecular dyes have the potential to serve as bright fluorophores with spectrally narrow emission for bioanalysis and imaging; however, this potential is largely untapped due to challenges in developing an architecture that stabilizes the J-aggregates, minimizes nonspecific binding interactions, and facilitates bioconjugation. Here, we develop and deploy colloidal supra-nanoparticle assemblies of J-aggregates as fluorescent labels. The assemblies, denoted as (Supra-J)-Dex, form via the spontaneous layer-by-layer assembly of a polycation, a cyanine dye (TDBC), and a dextran amph

Cancer Cell Targeting Via Selective Transferrin Receptor Labeling Using Protein-Derived Carbon Dots.

Carbon dot (CD) nanoparticles offer tremendous advantages as fluorescent probes in bioimaging and biosensing; however, they lack specific affinity for biomolecules, limiting their practical applications in selective targeting. Nanoparticles with intrinsic affinity for a target have applications in imaging, cytometry, therapeutics, etc. Toward that end, we report the transferrin receptor (CD71) targeting CDs, synthesized for the first time. The formation of these particles is truly groundbreaking, as direct tuning of nanoparticle affinity was achieved by simple and careful precursor selection o

Hole-scavenging in photo-driven N2 reduction catalyzed by a CdS-nitrogenase MoFe protein biohybrid system.

The light-driven reduction of dinitrogen (N2) to ammonia (NH3) catalyzed by a cadmium sulfide (CdS) nanocrystal‑nitrogenase MoFe protein biohybrid is dependent on a range of different factors, including an appropriate hole-scavenging sacrificial electron donor (SED). Here, the impact of different SEDs on the overall rate of N2 reduction catalyzed by a CdS quantum dot (QD)-MoFe protein system was determined. The selection of SED was guided by several goals: (i) molecules with standard reduction potentials sufficient to reduce the oxidized CdS QD, (ii) molecules that do not absorb the excitation

Aggregation of Mouse Serum Amyloid A Protein Was Promoted by Amyloid-Enhancing Factors with the More Genetically Homologous Serum Amyloid A.

Amyloid A (AA) amyloidosis is a condition in which amyloid fibrils characterized by a linear morphology and a cross-β structure accumulate and are deposited extracellularly in organs, resulting in chronic inflammatory diseases and infections. The incidence of AA amyloidosis is high in humans and several animal species. Serum amyloid A (SAA) is one of the most important precursor amyloid proteins and plays a vital step in AA amyloidosis. Amyloid enhancing factor (AEF) serves as a seed for fibril formation and shortens the onset of AA amyloidosis sharply. In this study, we examined whether AEFs

Microgravity Modulates Effects of Chemotherapeutic Drugs on Cancer Cell Migration.

Microgravity or the condition of apparent weightlessness causes bone, muscular and immune system dysfunctions in astronauts following spaceflights. These organ and system-level dysfunctions correlate with changes induced at the single cell level both by simulated microgravity on earth as well as microgravity conditions in outer space (as in the international space station). Reported changes in single bone cells, muscle cells and white blood cells include structural/morphological abnormalities, changes in gene expression, protein expression, metabolic pathways and signaling pathways, suggesting

Ultrastructural differences in pretangles between Alzheimer disease and corticobasal degeneration revealed by comparative light and electron microscopy.

Pretangles are defined under the light microscope as diffuse and granular tau immunoreactivity in neurons in tissue from patients with Alzheimer disease (AD) or corticobasal degeneration (CBD) and are considered to be a premature stage before neurofibrillary tangle formation. However, the ultrastructure of pretangles remains to be described. To clarify the similarities and differences between pretangles from patients with AD and CBD (AD-pretangles and CBD-pretangles, respectively), we examined cortical pretangles in tissue from patients with each of diseases. For direct light and electron micr

LPS unmasking of Shigella flexneri reveals preferential localisation of tagged outer membrane protease IcsP to septa and new poles.

The Shigella flexneri outer membrane (OM) protease IcsP (SopA) is a member of the enterobacterial Omptin family of proteases which cleaves the polarly localised OM protein IcsA that is essential for Shigella virulence. Unlike IcsA however, the specific localisation of IcsP on the cell surface is unknown. To determine the distribution of IcsP, a haemagglutinin (HA) epitope was inserted into the non-essential IcsP OM loop 5 using Splicing by Overlap Extension (SOE) PCR, and IcsP(HA) was characterised. Quantum Dot (QD) immunofluorescence (IF) surface labelling of IcsP(HA) was then undertaken. Qua

YY1 expression predicts favourable outcome in follicular lymphoma.

AIMS: Follicular lymphoma is the second most common type of non-Hodgkin's lymphoma worldwide. The majority of patients diagnosed as having follicular lymphoma have an indolent form of the disease, but a subset of patients have aggressive disease with a shorter survival interval. Optimal treatment stratification requires a distinction between these two groups, though there are presently few prognostic biomarkers available. The transcription factor YY1 has been shown to play an important role in cancer biology. The authors have previously reported a correlation of Yin Yang 1 (YY1) mRNA levels wi

Nitric oxide-enhanced blood-brain barrier penetration and mitochondria-targeted antioxidant carbon dots for Alzheimer's disease.

Alzheimer's disease (AD) involves a complex pathogenesis in which the β-amyloid (Aβ) cascade and oxidative stress hypotheses interact through mitochondrial dysfunction, forming a vicious cycle that underscores the need for multi-targeted therapeutic strategies. In this study, multifunctional carbon dots (EMA-CDs) were rationally designed and synthesised via a simple one-pot hydrothermal approach to integrate antioxidant activity, nitric oxide (NO) release capability, and mitochondrial targeting. EMA-CDs exhibited potent reactive oxygen species (ROS) scavenging abilities, efficiently eliminatin

Dandelion-Derived Carbon Dots with pH-Responsive Charge and Dual ROS Regulation for Anti-Infection and Accelerated Wound Repair.

Bacterial infections associated with biofilm formation hinder wound healing via enhanced antibiotic resistance and impaired tissue repair. Herein, we fabricated dandelion herb-derived carbon dots (DH-CDs) through a green solvothermal-extraction method, developing a biocompatible antimicrobial agent with synergistic wound healing potential. Physicochemical characterization showed that DH-CDs are quasi-spherical (4.87 nm), partially graphitized nanoparticles with abundant surface functional groups and pH-responsive charge-switching capability. In vitro studies demonstrated that DH-CDs exhibit br

Nitrogen-doped carbon nanodots as a biocompatible FRET donor for caspase-3 detection and chemotherapy response evaluation in osteosarcoma cell lysates.

Osteosarcoma is a highly aggressive bone malignancy requiring precise tools for early diagnosis and effective evaluation of chemotherapy efficacy. Here, we report an "off-on" fluorescent nanosensor based on nitrogen-doped carbon nanodots (N-CNDs) for the sensitive and selective detection of caspase-3 activity, a key apoptosis executor. The sensor is constructed by conjugating carboxyl-functionalized N-CNDs with a BHQ1-labeled peptide substrate containing the caspase-3-specific DEVD cleavage motif. In the absence of caspase-3, fluorescence is quenched via FRET. Upon caspase-3 activation, the pe

Suppression of Sepsis Cytokine Storm by Escherichia Coli Cell Wall-Derived Carbon Dots.

Sepsis is a life-threatening disease caused by a dysregulated immune response to infection, often involving the translocation of Gram-negative bacteria such as Escherichia coli (E. coli) into the bloodstream, triggering a cytokine storm. Despite its severity, no effective drugs currently exist for sepsis treatment. This study explores whether pathogen-derived carbon dots can mitigate their inherent toxicity while leveraging their structural similarity to pathogens to competitively bind pattern recognition receptors, thereby inhibiting sepsis. Based on this concept, E. coli wall-derived carbon

Luminescent Electro-Spun Nanofibers Crosslinked with Boronic Esters Exhibiting Controlled Release of Carbon Dots for Detection of Wound pHs and Enhanced Antimicrobial.

Timely and accurate assessment of wounds during the healing process is crucial for proper diagnosis and treatment. Conventional wound dressings lack both real-time monitoring capabilities and active therapeutic functionalities, limiting their effectiveness in dynamic wound environments. Herein, we report our proof-of-concept approach exploring the unique emission properties and antimicrobial activities of carbon nanodots (CNDs) for simultaneous detection and treatment of bacteria. This approach centers on the fabrication of well-defined CND-embedded poly(vinyl alcohol) (PVA) e-spun nanofibrous

Combination therapy enhances efficacy and overcomes toxicity of metal-based anti-diabetic agent.

BACKGROUND AND PURPOSE: Metal-based therapeutic agents are limited by the required concentration of metal-based agents. Hereby, we determined if combination with 17β-oestradiol (E2) could reduce such levels and the therapy still be effective in type 2 diabetes mellitus (T2DM). EXPERIMENTAL APPROACH: The metal-based agent (vanadyl acetylacetonate [VAC])- 17β-oestradiol (E2) combination is administered using the membrane-permeable graphene quantum dots (GQD), the vehicle, to form the active GQD-E2-VAC complexes, which was characterized by fluorescence spectra, infrared spectra and X-ray photoel…

Dose- and duration-dependent cytotoxicity and genotoxicity in human hepato carcinoma cells due to CdTe QDs exposure.

Nanotechnology has achieved more commercial attention over recent years, and its application has increased concerns about its discharge in the environment. In this study, we have chosen human hepatic carcinoma (HuH-7) cells because liver tissue has played an important role in human metabolism. Therefore, the objective of this study was to determine DNA damaging and apoptotic potential of cadmium telluride quantum dots (CdTe QDs; average particle size (APS) 10 nm, 1-25 µg/ml) on HuH-7 cells and the basic molecular mechanism of its cellular toxicity. Cytotoxicity of different concentrations of C…
