Association of the PDYN gene with alcohol dependence and the propensity to drink in negative emotional states.

Synthetic κ-opioid receptor (KOR) agonists induce dysphoric and pro-depressive effects and variations in the KOR (OPRK1) and prodynorphin (PDYN) genes have been shown to be associated with alcohol dependence. We genotyped 23 single nucleotide polymorphisms (SNPs) in the PDYN and OPRK1 genes in 816 alcohol-dependent subjects and investigated their association with: (1) negative craving measured by a subscale of the Inventory of Drug Taking Situations; (2) a self-reported history of depression; (3) the intensity of depressive symptoms measured by the Beck Depression Inventory-II. In addition, 13

Drink and be merry? Gender, life satisfaction, and alcohol consumption among college students.

This study examined the impact of alcohol use and alcohol-related problems on several domains of life satisfaction (LS) in a sample of 353 college students. Alcohol use was associated with lower general satisfaction and anticipated future satisfaction among women. Female abstainers reported higher general and anticipated future satisfaction than did female heavy drinkers. Female students' alcohol use was unrelated to their academic, family, dating, or social satisfaction. Drinking among men showed a positive, curvilinear relation to social satisfaction but was unrelated to other domains of LS.

Predictors of problem drinking and alcohol dependence in a population-based sample of female twins.

OBJECTIVE: To identify characteristics associated with problem drinking (PD) and alcohol dependence (AD) in women. METHOD: Subjects were 2,163 white women aged 17-55 from the population-based Virginia Twin Registry. Measures were selected from a clinical interview and questionnaires to reflect five domains associated with alcoholism in prior studies; demographic characteristics, personality, health, and personal and family history of psychopathology. Logistic and linear regression analyses were used to predict PD and DSM-III-R defined AD. RESULTS: Multiple regression models accounted for 19%

Matching Alcoholism Treatments to Client Heterogeneity: Project MATCH posttreatment drinking outcomes.

OBJECTIVE: To assess the benefits of matching alcohol dependent clients to three different treatments with reference to a variety of client attributes. METHODS: Two parallel but independent randomized clinical trials were conducted, one with alcohol dependent clients receiving outpatient therapy (N = 952; 72% male) and one with clients receiving aftercare therapy following inpatient or day hospital treatment (N = 774; 80% male). Clients were randomly assigned to one of three 12-week, manual-guided, individually delivered treatments: Cognitive Behavioral Coping Skills Therapy, Motivational Enh

Multiple and single location drinking among DWI offenders referred for alcoholism evaluation.

Problem-drinker drivers who drank at multiple locations differ substantially from those who drank at only one location. Persons who drank at more than one location prior to their DWI arrest exhibit the most severe alcohol problems. Multilocation drinkers consumed significantly greater amounts of alcohol--for nearly all alcohol measures including consumption per drinking day, consumption averaged over 30 days, and consumption on the day of the DWI arrest--than single location drinkers. The multilocation group experienced more alcohol problems in their lives, had higher Mortimer-Filkins test sco

Ethanol administration in the drinking fluid to pregnant rats as a model for the fetal alcohol syndrome.

Addition of ethanol (ET) to the drinking fluid of pregnant rats has been questioned as an experimental model for the fetal alcohol syndrome (FAS). This model, however, closely simulates human alcohol intake, and in this study we used a modified version of previous protocols to overcome their major defects. A group of female rats was given 10% ET in drinking fluid for one week, 15% for the second week, 20% for the third, and 25% for the fourth, at the end of which they were mated with non-treated males and given 25% ET throughout gestation. Three groups of non-ET treated sex and age-matched rat

Liver function in a group of hospitalised alcoholics and non-hospitalised problem drinking probationers.

Twenty non-hospitalised problem male drinkers and 20 hospitalised problem drinkers were investigated by the Michigan alcohol screening test (MAST) and by liver function tests (AST, ALT). The MAST scores of the non-hospitalised group were above normal and indicative of alcohol addiction, AST and ALT tended to be higher in the hospitalised but not indicative of severe liver damage.

Sex-specific neural activation to stress and alcohol cues in high-risk drinkers: links between orbitofrontal circuits, alcohol craving, and future drinking.

High-risk drinking is known to disrupt stress and reward pathways including orbitofrontal cortex (OFC) circuits, which may increase cue-induced craving and alcohol use disorder (AUD) risk. Although high-risk drinking is more prevalent in men, it is increasing rapidly among women. It remains unclear whether sex differences in reactivity to stress and alcohol cues contribute to these trends. One hundred eighteen adults (56 high-risk drinkers, 62 low-risk drinkers; 52.5% female) completed an fMRI cue provocation task involving alcohol, stress, and neutral cues in a randomized block design with re

Glucose and fructose differently mediate alcohol cocktail drinking in female and male rats: interaction of glucose and alcohol on post-ingestive behavior.

Sweetened alcoholic beverages are thought to contribute to developing Alcohol Use Disorder by increasing palatability. One monosaccharide, glucose, readily enters the brain more than fructose and directly impacts the activity of central neurons. The objective of this study is to determine the impact of glucose versus fructose on alcohol drinking patterns in female and male rats. Rats drank alcohol cocktails (1.25%-10%) containing either glucose or fructose (10%) in 4-hour sessions. We sought to parse orosensory effects from post-ingestive central effects by analyzing drinking microstructure. W…

Discrepancy between self-reported intoxication and psychomotor vigilance task performance after alcohol intake: associations with BrAC, alcohol-metabolizing genotypes, and drinking tendency.

INTRODUCTION: The degrees of subjective intoxication (SI) and vigilance impairment following alcohol consumption vary among individuals. This variability may reflect both metabolic enzyme genotypes and drinking behavior. The purpose of this study was to examine the integrated associations among alcohol-metabolizing enzyme genotypes, drinking tendency, and acute alcohol-induced changes in breath alcohol concentration (BrAC), SI, and vigilance performance. METHODS: Alcohol (0.5 g/kg body weight) was orally administered to 93 healthy adults. BrAC, SI, and the Psychomotor Vigilance Task (PVT) wer

I'm tired and feel like drinking: Viewing alcohol cues after exerting self-control increases approach motivation among individuals lower in alcohol sensitivity.

Exerting self-control depletes capacity for future self-control, which can promote greater alcohol use. However, certain populations may be more susceptible to these effects of depleted self-control capacity. For example, individuals with lower alcohol sensitivity (i.e., requiring more drinks to experience the effects of alcohol) are a high-risk group who are likely to engage in hazardous alcohol use and develop an alcohol use disorder. Those lower in alcohol sensitivity also exhibit heightened motivational reactivity in response to alcohol-related cues, which may be enhanced following exertio

How do residents perceive alcohol availability and its impact on drinking behaviour? A qualitative study.

INTRODUCTION: Reducing alcohol availability has been identified as a potential approach to reduce alcohol use and related harm. In order to regulate alcohol availability at a local level, it is important to involve communities in decision-making and ensure interventions are locally acceptable and appropriate. To do this, we need to improve our understanding of how residents conceptualise alcohol availability and its impact on behaviours. This paper uses data collected in Scotland, a country with particularly high levels of alcohol-related harm, to explore the perspectives of residents, on loca

From binge drinking to future alcohol severity: The role of emotion regulation and emerging psychopathology.

OBJECTIVES: Binge drinking (BD) increases the risk for alcohol use disorders and other psychopathological disorders later in life. Emotion regulation (ER) deficits have been identified as a transdiagnostic risk factor, yet their role in risk trajectories is not well established. This is especially relevant in young BDs initially free of psychopathology, where emerging symptoms may shape risk trajectories toward severe alcohol use. Thus, this study examined whether ER difficulties mediate the link between BD and later alcohol severity, and whether emerging psychopathology moderates these associ

Perceived consequences of disseminating evidence on untreated recovery and moderate drinking after resolving alcohol concerns.

AIMS: There is substantial evidence that many people recover from serious alcohol concerns without treatment and further, return to moderate alcohol consumption. The current study asks what people believe would happen if this evidence was widely disseminated. DESIGN AND METHODS: Participants (N = 3994), sampled to match the demographic characteristics of people in the UK, were recruited through the Prolific website. Participants were asked a series of questions about their current and past alcohol use. All participants were also asked their beliefs as to whether it would be problematic if evi

Environmental and Social Contexts Associated with Impulsivity and Alcohol Use Among Heavy Drinking College Students.

Background: Although impulsivity is strongly linked to alcohol use, few studies have examined contextual factors that may impact impulsivity in real time. Thus, this study aimed to (1) determine which locations, social settings, and drinking contexts were associated with impulsivity and (2) examine whether the association between impulsivity and alcohol use in-the-moment was moderated by context. Methods: Participants were 89 (47 women) heavy drinking, college students who completed a 14-day ecological momentary assessment (EMA) study. Each day, participants completed three random reports, and

Event-level drinking motives and mood as predictors of alcohol-related consequences among young adults.

OBJECTIVE: People who drink heavily experience positive (e.g., made a new friend) and negative (e.g., embarrassed self) alcohol-related consequences. Important antecedents of alcohol-related consequences are drinking motives, particularly those related to affect regulation (coping, enhancement). Although prior research documents associations at the between-person level, little is known about how mood and motives at the start of a drinking event each relate within-person to positive and negative alcohol-related consequences. METHOD: Participants were U.S. young adults (N = 199; Mage = 22.10, S

Social isolation recruits amygdala-medial prefrontal cortex projections to escalate alcohol drinking in male mice.

Social isolation profoundly alters motivation and increases vulnerability to alcohol misuse in humans, yet the underlying neural mechanisms remain unclear. Here we show that isolation escalates alcohol drinking in male mice but suppresses it in females. Whole-cell recordings revealed that neurons in the basolateral amygdala projecting to the medial prefrontal cortex (BLA-mPFC) track alcohol intake in both sexes. Isolation increased BLA-mPFC excitability in males but decreased it in females, mirroring their opposite behavioral adaptations. Given this divergence, we focused subsequent mechanisti…
