Molecular genetics of the early development of hindbrain serotonergic neurons.

The serotonergic (5HT) system plays a key role in modulating behaviors, such as appetite and anxiety and has been implicated in many human disorders of mood and mind. Recent studies have begun to identify the signaling molecules and transcriptional cascades governing 5HT neuron development in the hindbrain. Already at early stages, local differences in requirements of 5HT neuron development have become apparent. These studies point toward cryptic heterogeneity amongst 5HT neurons and suggest that 5HT neuron determination and differentiation may be more flexible and less absolute biologic proce

Role of cold-inducible RNA-binding protein in hypothalamic regulation of feeding behavior during fasting and cold exposure.

Appetite regulation is a complex process that is critical for maintaining energy balance and is governed by intricate molecular and cellular mechanisms in the hypothalamus. RNA-binding proteins play vital roles in the post-transcriptional regulation of mRNA and influence feeding behavior and energy metabolism. This study explored the role of cold-inducible RNA-binding protein (Cirbp) in hypothalamic neurons under metabolic stress conditions, such as fasting and cold exposure. Next-generation sequencing (NGS) of the hypothalami from fasted mice identified 67 differentially expressed RNA-binding

Respiratory and emotional reactivity to ethanol odor in human neonates is dependent upon maternal drinking patterns during pregnancy.

BACKGROUND: Beyond the well-known deleterious effects of ethanol defining Fetal Alcohol Spectrum Disorders (FASD), the notion of fetal alcohol programming has gained scientific support. This phenomenon implies early neural plasticity relative to learning mechanisms comprising ethanol´s sensory cues and physiological effects of the drug; among others, its reinforcing properties and its depressant effects upon respiration. In this study, as a function of differential ethanol exposure during gestation, we analyzed neonatal physiological and behavioral responsiveness recruited by the odor of the d

Efficacy of Home-Based Remotely Supervised Transcranial Direct Current Stimulation for Managing Neuropsychiatric Symptoms in Older Adults With Alzheimer's Disease and Related Dementias.

BACKGROUND: Neuropsychiatric symptoms (NPS) are prevalent among persons with Alzheimer's disease and related dementias (ADRD). However, there are limited safe and effective nonpharmacological treatments for controlling NPS. Transcranial direct current stimulation (tDCS) is a promising noninvasive and safe treatment. MATERIALS AND METHODS: This study investigated the effects of remotely supervised tDCS in managing NPS in older adults with mild to moderate ADRD. Forty older adults diagnosed with early-stage ADRD were randomly assigned in a 1:1 ratio to receive home-based active tDCS (n = 20) or

From Mistrust to Malice: Examining the Influence of Adverse Childhood Experiences on Reactive and Appetitive Aggression in Male Forensic Psychiatric Patients with a History of Drug Abuse Through the Lens of Psychodynamic Personality Structures.

Adverse childhood experiences (ACEs) represent one of the most critical factors contributing to the manifestation of psychiatric disorders later in life. Furthermore, such experiences are often associated with deficits in interpersonal relationships, manifesting as mistrust and violent behaviors, and are indicative of a fragmented personality. This study aimed to analyze the correlative relationships between personality deficits influenced by ACEs and the expression of reactive and appetitive aggression using self-report questionnaires in 53 male forensic psychiatric patients with a drug depen

Prefronto-cerebellar neuromodulation affects appetite in obesity.

Human neuroimaging studies have consistently reported changes in cerebellar function and integrity in association with obesity. To date, however, the nature of this link has not been studied directly. Emerging evidence suggests a role for the cerebellum in higher cognitive functions through reciprocal connections with the prefrontal cortex. The purpose of this exploratory study was to examine appetite changes associated with noninvasive prefronto-cerebellar neuromodulation in obesity. Totally, 12 subjects with class I obesity (mean body mass index 32.9 kg/m2) underwent a randomized, single-bli

Relatively strong automatic appetitive action-tendencies in male carriers of the OPRM1 G-allele.

This study investigated whether automatic approach action tendencies for alcohol-related stimuli were associated with variation in the mu-opioid receptor gene (OPRM1), previously related to rewarding effects of alcohol and craving. An adapted approach avoidance task was used, in which participants pulled or pushed a joystick in reaction to the format of a picture shown on the computer screen (e.g. pull landscape pictures and push portrait pictures). Picture size on the screen changed upon joystick movement, so that upon a pull movement picture size increased (creating a sense of approach) and

Acute and chronic administration of melanin-concentrating hormone enhances food intake and body weight in Wistar and Sprague-Dawley rats.

AIM: Although melanin-concentrating hormone (MCH) is believed to be an important regulator of feeding behavior, both its acute and chronic effects on food intake as well as its interaction with other brain peptides involved in the control of appetite remain unclear. Therefore, the acute effects of MCH on food intake and the chronic effect of MCH on food intake and the gene expression of various hypothalamic peptides involved in the control of appetite were studied in rats. METHODS AND RESULTS: Either the acute or the continuous intraventricular infusion of MCH for 12 days stimulated feeding i

Endogenous opioid peptides in intrapartum uterine blood.

Endogenous opioid peptides have consistently been reported to exert significant influence on several parameters of reproductive physiology, including tolerance to pain, release of prolactin, pleasurable behavior patterns, and appetite satiety. Immunoreactive endorphins measured in several hundred samples of postpartum uterine blood had a mean value of 190 pg/ml and a range of 60 to 520 pg/ml. Individual values in 100 mothers who were delivered at the Northwest Hospital failed to correlate with doses of postpartum pain medications, infant feeding patterns, and recorded estimates of maternal bon

Heightened play following play observation in the absence of 22-kHz calls in juvenile rats.

Emotional contagion is often studied through aversive-state transmission, leaving positive affect sharing less understood. Here, we investigated whether observing social play facilitates a subsequent play opportunity in juvenile male rats. Observer dyads were socially isolated for 30, 120, or 270 min before observing either highly playful demonstrator dyads that had been socially isolated for 24 h, or less playful demonstrator dyads that were socially housed. During observation, visual, auditory, and olfactory cues were available, but physical contact was prevented. When observers were reunite

Prenatal Exposure to Low Alcohol and Alcohol Motivation in the Offspring.

Prenatal alcohol exposure (PAE), even at low doses, has been linked to long-term alterations in alcohol-related behaviors, particularly increased alcohol consumption and preference in offspring. This chapter examines how low-dose PAE affects the motivational effects of alcohol, potentially influencing the balance between its rewarding and aversive properties. While high-dose PAE has been extensively studied for its teratogenic effects, the implications of lower, more common exposures remain less understood. Preclinical studies using rodent models suggest that even moderate PAE (1-3 g/kg/day) e

Apathy and Impulse Control Disorders: Yin & Yang of Dopamine Dependent Behaviors.

Neuropsychiatric symptoms are common non-motor symptoms in Parkinson's disease (PD). Apathy and impulse control disorders (ICD) are two opposite motivational expressions of a continuous behavioural spectrum involving hypo- and hyperdopaminergia. Both syndromes share pathological (decreased vs increased) dopamine receptor stimulation states. Apathy belongs to the spectrum of hypodopaminergic symptoms together with anhedonia, anxiety and depression. Apathy is a key symptom of PD which worsens with disease progression. Animal models, imaging and pharmacological studies concur in pointing out dopa

Generalization of courtship learning in Drosophila is mediated by cis-vaccenyl acetate.

Reproductive behavior in Drosophila has both stereotyped and plastic components that are driven by age- and sex-specific chemical cues. Males who unsuccessfully court virgin females subsequently avoid females that are of the same age as the trainer. In contrast, males trained with mature mated females associate volatile appetitive and aversive pheromonal cues and learn to suppress courtship of all females. Here we show that the volatile aversive pheromone that leads to generalized learning with mated females is (Z)-11-octadecenyl acetate (cis-vaccenyl acetate, cVA). cVA is a major component of

Investigating fecal microbiota transplants from individuals with anorexia nervosa in antibiotic-treated mice using a cross-over study design.

Anorexia nervosa (AN) is a complex and serious mental disorder, which may affect individuals of all ages and sex, but primarily affecting young women. The disease is characterized by a disturbed body image, restrictive eating behavior, and a lack of acknowledgment of low body weight. The underlying causes of AN remain largely unknown, and current treatment options are limited to psychotherapy and nutritional support. This paper investigates the impact of Fecal Microbiota Transplants (FMT) from patients with AN on food intake, body weight, behavior, and gut microbiota into antibiotic-treated mi

Behavioral and Neural Responses to Vitamin C Solution in Vitamin C-deficient Osteogenic Disorder Shionogi/Shi Jcl-od/od Rats.

To investigate the appetite for vitamin C (VC), we conducted behavioral and neural experiments using osteogenic disorder Shionogi/Shi Jcl-od/od (od/od) rats, which lack the ability to synthesize VC, and their wild-type controls osteogenic disorder Shionogi/Shi Jcl- +/+ (+/+) rats. In the behavioral study, rats were deprived of VC for 25 days and then received two-bottle preference tests with a choice between water and 10 mM VC. The preference for 10 mM VC solution of od/od rats was significantly greater than that of +/+ rats. In the neural study, the relative magnitudes of the whole chorda tym

Acute post-orchiectomy pain does not reduce alpha rams' interest in feed resources.

Sheep pain is an animal welfare issue monitored based on behavioral responses, including appetite. Dominant (alpha) males have priority for accessing limited feed resources, however, the effects of pain on feed interest in members of a group with defined social hierarchy are unknown. Our objective was to investigate effects of acute post-orchiectomy pain on alpha rams' interest in accessing a limited feed resource. Eighteen rams were randomly housed in pens of 3 rams. After acclimation, the first 5-d (consecutive) battery of a behavior test was performed. In this test, 180 g of the regular die
