Antimicrobial effects of commensal oral species are regulated by environmental factors.

OBJECTIVES: The objectives of this study are to identify oral commensal species which can inhibit the growth of the main periodontopathogens, to determine the antimicrobial substances involved in these inhibitory activities and to evaluate the influence of environmental factors on the magnitude of these inhibitions. METHODS: The spotting technique was used to quantify the capacity of 13 commensal species to inhibit the growth of Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis and Prevotella intermedia. By altering experimental conditions (distance between spots and size of spo

Polymerizing the fibre between bacteria and host cells: the biogenesis of functional amyloid fibres.

Amyloid fibres are proteinaceous aggregates associated with several human diseases, including Alzheimer's, Huntington's and Creutzfeldt Jakob's. Disease-associated amyloid formation is the result of proteins that misfold and aggregate into beta sheet-rich fibre polymers. Cellular toxicity is readily associated with amyloidogenesis, although the molecular mechanism of toxicity remains unknown. Recently, a new class of 'functional' amyloid fibres was discovered that demonstrates that amyloids can be utilized as a productive part of cellular biology. These functional amyloids will provide unique

Colicin typing of Shigella sonnei.

The colicin typing method for differentiating epidemic strains of Shigella sonnei was evaluated. Forty percent of the strains were untypable, but the method divides 60% of the strains into 15 established colicin types. The method has reasonably good reproducibility, but should not be interpreted with the same degree of confidence as some other characteristics such as serotypes. For this reason, known type strains should be included as controls in each analysis to ensure uniformity of results.

Insights into the mode of action of the two-peptide lantibiotic lichenicidin.

Lantibiotics are promising candidates to address the worldwide problem of antibiotic resistance. They belong to a class of natural compounds exhibiting strong activity against clinically relevant Gram-positive bacterial strains, including methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant Enterococci (VRE). Lichenicidin is a class II two-peptide lantibiotic. The presence of the two mature peptides, Bliα and Bliβ, is necessary for full activity against target bacteria. This work aims at clarifying the synergistic activity of both peptides in their interaction with the t

A New Approach to Harness Probiotics Against Common Bacterial Skin Pathogens: Towards Living Antimicrobials.

In this study, the potential of certain lactic acid bacteria-classified as probiotics and known to be antimicrobially active against pathogens or food-poisoning microorganisms-was evaluated with respect to their activity against bacterial skin pathogens. The aim of the study was to develop a plaster/bandage for the application of inhibitory substances produced by these probiotics when applied to diseased skin. For this purpose, two Streptococcus salivarius strains and one Lactobacillus plantarum were tested for production of antimicrobials (bacteriocin-like substances) active against Gram-posi

Discovery of medically significant lantibiotics.

The emergence of drug-resistant pathogens such as staphylococci and enterococci in the hospital setting has long being recognized as a serious clinical problem. Staphylococcus aureus is the causative agent of many nosocomial infections from minor skin abscesses to serious, potentially life threatening diseases such as bone and soft tissue intra-surgical infections, sepsis and invasive endocarditis, while enterococci are responsible for nosocomial bacteraemia, surgical wound infections and endocarditis. The most infamous drug-resistant forms of these include MRSA (methicillin resistant S. aureu

[Treatment of chronic rhinitis in children].

This paper discusses involvement of intestinal dysbacteriosis in nasal inflammation. Altogether 42 children of preschool age with chronic rhinitis were examined. It was found that microbial changes in the nasal cavity were related to the degree of intestinal dysbacteriosis, which was detected in most patients. The drugs that were used in combined therapy to treat microbial changes in the nasal mucosa and intestine produced good clinical and bacteriological effects. The 4 month follow-up showed that the children on the combined therapeutic protocol remained in the remission stage throughout thi

[Transcription map of a recombinant plasmid carrying a gene for the synthesis of pesticine I and a protein for pesticine I immunity in the plague microbe].

Molecules of the plasmids pBR322 and pRD17 have been compared by electron microscopy technique of R-loops visualization. Comparative location of R-loops on the plasmids has been computerized on minicomputer HP9825A due to the program making possible to define the coordinates of the transcription start and direction. The 5.1 kb fragment coding for pesticin I and immunity protein to pesticin I and cloned in pBR322 vector plasmid is flanked by Bam HI-EcoRI sites. Five promoter regions and direction of transcription were localized on the fragment.

[Importance of the vibriocinogenic factor in the ecology of Vibrio cholerae].

The relationship between 433 V. cholerae strains and 571 enterobacterial strains, isolated from different ecosystems, have been studied by the method of "delayed antagonism". The direct correlation between the virulence of V. cholerae and the width of the spectra of their antagonistic activity towards indicator bacteria was detected. The expediency of studies on the role of the vibriocinogenic factor in the survival of V. cholerae in different ecological systems is substantiated.

Importance of colicinogeny for the course of acute bacillary dysentery.

The development of acute bacillary dysentery was followed in 23 patients involved in two outbreaks and in three sporadic, mutually unrelated cases. Repeated cultivations performed at 2-day intervals for 10 days yielded 386 identifiable strains of "opportune intestinal flora". Escherichia coli colicinogenic activity is one of the significant factors of gastrointestinal tract protection. The period of shigella excretion is significantly reduced (p less than 0.02) if an appropriate colicinogenic E. coli strain is present. Analysis of the results suggested a working hypothesis of differentiated ap

Weissella and the two Janus faces of the genus.

The genus Weissella belongs to the lactic acid bacteria group. It occurs naturally in foods and is a component of the human microbiome. A few Weissella species are candidate probiotics due to their potential for survival under the harsh conditions present in the gastrointestinal tract of humans and animals. Various species have also shown potential for treating and preventing periodontal disease, skin pathologies, and atopic dermatitis; some are used as starters for the fermentation of foods due to their production of exopolysaccharides; and others are used as protective cultures due to their

Post-process treatments are effective strategies to reduce Listeria monocytogenes on the surface of leafy greens: A pilot study.

Growth of L. monocytogenes is among the most important factors affecting the risk of human listeriosis. In ready to eat leafy greens, the use of anti-Listeria treatments represents a good alternative to inhibit growth during storage. Several commercially available antimicrobial agents have been suggested as effective intervention strategies. Among them, phage preparations and bacteriocin-producing strains have shown promising results against L. monocytogenes. In this study, we investigate the efficacy of two commercially available surface treatments, the bacteriophage formulation PhageGuard Li
