Beta-lactamase production, beta-lactam sensitivity and resistance to synergy with clavulanate of 737 Bacteroides fragilis group organisms from thirty-three US centres.

Beta-Lactamase production and agar dilution sensitivities to amoxycillin, amoxycillin/clavulanate, ticarcillin, ticarcillin/clavulanate, cefoxitin, imipenem and metronidazole were determined for 737 Bacteroides fragilis group strains isolated between 1986 and 1988 from 33 US centres. The strains comprised 441 B. fragilis, 114 B. thetaiotaomicron, 35 B. ovatus, 58 B. distasonis, 58 B. vulgatus, 26 B. uniformis and five B. caccae. Overall, addition of clavulanate lowered the geometric mean MICs of of amoxycillin from 46.7 to 0.6 mg/l, and of ticarcillin from 37.2 to 1.3 mg/l. Addition of clavula…

Infective Endocarditis Secondary to Bacteroides Thetaiotaomicron in a Patient With Uterine Smooth Muscle Tumor of Uncertain Malignant Potential: A Case Report and Review of the Literature.

Bacteroides species are significant clinical pathogens with an associated mortality of more than 19% and are found in most anaerobic infections. Our report documents for the first time a case of infective endocarditis (IE) secondary to Bacteroides thetaiotaomicron (BT). We discuss the case of a 65-year-old female with a medical history of smooth muscle tumor of uncertain malignant potential (STUMP) who presented to the ED with lower quadrant pain. In the hospital, she was found to be in septic shock. A transthoracic echocardiogram showed large vegetation on the aortic valve with severe aortic …

Glutamine synthetase, glutamate synthase and glutamate dehydrogenase in Rhizobium japonicum strains grown in cultures and in bacteroids from root nodules of Glycine max.

The growth yields of three strains of Rhizobium japonicum (CB 1809, CC 723, CC 705) in culture solutions containing L-glutamate were about twice those grown with ammonium. The activities of glutamine synthetase (GS; EC 6.3.1.2) and glutamate dehydrogenase (GDH; EC 1.4.1.4) were dependent on the nitrogen source in the medium and also varied with growth. Both NADPH-and NADH-dependent glutamate synthase (GOGAT; EC 1.4.1.13) and NADPH-dependent GDH were found in strains grown with either glutamate or ammonium but NADH-linked GDH was only detected in glutamate-grown cells. Glutamine synthetase was

Bacteroides coprocola protects dopaminergic neurons in rotenone-induced Parkinson's disease mouse model by modulating gut microbiota dysbiosis and inhibiting the NLRP3 signaling pathway.

BACKGROUND: Parkinson's disease (PD) is a prevalent neurodegenerative disease and its pathogenesis is still unclear. Emerging evidence supports the gut-origin hypothesis, highlighting gut microbiota dysbiosis as a contributing factor in PD pathogenesis. Our previous clinical study showed that Bacteroides coprocola (B. coprocola), a gut bacterium producing short-chain fatty acids (SCFAs), was significantly reduced in PD patients. This study was aimed to investigate the potential of B. coprocola in ameliorating PD pathology and explore the underlying mechanisms in a rotenone-induced PD mouse mod…

Differential toll-like receptor 2 activation by Akkermansia muciniphila and Bacteroides thetaiotaomicron mediates the beneficial effects of Fu brick tea polysaccharide against colitis.

Fu Brick Tea Polysaccharide (FBTP) ameliorates dextran sulfate sodium (DSS)-induced colitis in mice. However, the key intestinal bacterial strains and downstream molecular mechanisms mediating these protective effects remain unclear. In this study, FBTP ameliorated colitis and concurrent liver injury in a microbiota-dependent manner, primarily by enriching Akkermansia muciniphila (A. muciniphila) and depleting Bacteroides thetaiotaomicron (B. thetaiotaomicron). The essential role of the microbiota was confirmed through fecal microbiota transplantation. Mechanistically, A. muciniphila synergist…

Bacteroides acidifaciens alleviates high-fat diet-induced obesity-related osteoporosis by regulating gut microbiota and bile acid metabolism via the gut-bone axis.

Obesity disrupts bone formation-resorption balance. Despite the established role of Bacteroides acidifaciens (B. acidifaciens) in modulating metabolic homeostasis, reducing inflammation, and improving lipid metabolism, its impact on obesity-associated osteoporosis is still not well understood. In this study, we delved into the potential protective influence of B. acidifaciens on high-fat diet (HFD) induced bone loss. The results showed that B. acidifaciens sharply improved weight gain, glucose and lipid metabolism in 16 weeks HFD mice. Both In vitro and in vivo experiments have conclusively de…

Improving fatty liver hemorrhagic syndrome in laying hens through gut microbiota and oxylipin metabolism by Bacteroides fragilis: A potential involvement of arachidonic acid.

Bacteroides fragilis (B. fragilis), a crucial commensal bacterium within the gut, has shown connections with hepatic lipid metabolism and inflammation regulation. Nonetheless, the role of B. fragilis in the progression of fatty liver hemorrhagic syndrome (FLHS) remains unknown. This study aims to explore the ameliorative effects of B. fragilis on FLHS in laying hens, as well as its underlying mechanisms. This is the first study to employ a chicken FLHS model, combining microbiomics and oxylipin metabolomics to investigate the mechanism of action of intestinal symbiotic bacteria. Exp. 1: 40 lay…

Microbial enterotypes beyond genus level: Bacteroides species as a predictive biomarker for weight change upon controlled intervention with arabinoxylan oligosaccharides in overweight subjects.

Recent studies indicate that microbial enterotypes may influence the beneficial effects of wholegrain enriched diets including bodyweight regulation. In a 4-week intervention trial, overweight subjects were randomized to consume either arabinoxylan-oligosaccharides (AXOS) (10.4 g/d) from wheat bran or polyunsaturated fatty acids (PUFA) (3.6 g/d). In the present study, we have stratified the subjects participating in the intervention (n = 29) according to the baseline Prevotella-to-Bacteroides (P/B) ratios through a post-hoc analysis and applied a linear mixed model analysis to identify the inf…

The bone-resorbing activities in tissue culture of lipopolysaccharides from the bacteria Actinobacillus actinomycetemcomitans, Bacteroides gingivalis and Capnocytophaga ochracea isolated from human mouths.

The activities of lipopolysaccharides (LPS) were assessed by measuring the calcium release from mouse calvaria in vitro and compared to that of LPS from Salmonella typhimurium. Stimulation of bone resorption was maximal at an LPS concentration of 10 micrograms/ml and at this dose all oral LPS preparations showed similar levels of activity and less than that of LPS from S. typhimurium. Only S. typhimurium LPS and B. gingivalis LPS retained bone-resorbing activity at 0.1 microgram/ml. No bone-resorbing activity was observed against killed bone and histochemical observations of stable acid phosph

Fabrication of Colon-Targeted Delivery System of Astaxanthin Based on Bacteroides-Dependent Biodegradation Strategy and Its Role in Ameliorating DSS-Induced Colitis in Mice.

Astaxanthin (Ax) is a potent antioxidant, yet its poor water solubility and instability limit its application. While alginate-Ca encapsulation protects Ax during digestion, its release in the colon is often inefficient. This study aims to optimize colon-targeted delivery by integrating inulin and fucoidan, which respond selectively to Bacteroides-mediated fermentation. A novel delivery system was developed using Ax-containing particles formulated in an alginate-inulin-fucoidan matrix (Ax-Mix), with most particles ranging from 2 to 20 μm. In vitro results showed that the incorporation of inulin…

Bacteroides Fragilis-Derived Outer Membrane Vesicles Deliver MiR-5119 and Alleviate Colitis by Targeting PD-L1 to Inhibit GSDMD-Mediated Neutrophil Extracellular Trap Formation.

Inflammatory bowel disease (IBD) results from a breakdown in the symbiotic relationship between the intestinal commensal microflora and the mucosal immune system. Non-toxigenic Bacteroides fragilis, a common human colon symbiote, has been shown to alleviate colitis. However, the underlying mechanisms of this alleviation remain incompletely understood. Herein, it is demonstrated that promoting the secretion of B. fragilis outer membrane vesicles (BfOMVs+) enhances its ability to alleviate dextran sodium sulfate (DSS)-induced colitis, while inhibiting B. fragilis OMV secretion (BfOMVs-) reduces …
