Loss of Cajal bodies in motor neurons from patients with novel mutations in VRK1.

Distal hereditary motor neuropathies (dHMNs) are a heterogeneous group of diseases, resembling Charcot-Marie-Tooth syndromes, but characterized by an exclusive involvement of the motor part of the peripheral nervous system. Here, we describe two new compound heterozygous mutations in VRK1, the vaccinia-related kinase 1 gene, in two siblings from a Lebanese family, affected with dHMN associated with upper motor neurons (MNs) signs. The mutations lead to severely reduced levels of VRK1 by impairing its stability, and to a shift of nuclear VRK1 to cytoplasm. Depletion of VRK1 from the nucleus alt

Mammalian birth versus arousal from hibernation: thyroid hormones, common regulators of metabolic transition?

Mammalian birth and arousal from hibernation are both endogenously regulated transitional events, characterized by an increase in metabolic rate (MR) and onset of thermogenesis. Thyroid hormones (THs) are known to be key regulators of metabolic and thermogenic activity. To explore the similarities and differences in the role of THs during mammalian birth as opposed to arousal from hibernation, a comprehensive review is given of the levels and kinetics of serum thyrotropin-releasing hormone (TRH), thyroid stimulating hormone (TSH), thyroxine (T4), triiodothyronine (T3), and reverse triiodothyro

[Invisible support, life-saving cooperation].

In certain cases, emergency care begins even before emergency medical services arrive on the scene, as bystanders take immediate action. Dispatchers play a key role in supporting lay responders, yet their responsibility often remains unrecognized. The aim of this summary was to examine the cooperation between dispatchers and lay first aid providers from various perspectives. Advancements in technology are significantly improving prehospital care. The emergence of structured decision-support software, video-assisted resuscitation, and artificial intelligence-based systems can considerably enhan

Evidence of Campylobacter jejuni reduction in broilers with early synbiotic administration.

C. jejuni is considered a food safety concern to both public health authorities and consumers since it is the leading bacterial cause of food-borne gastroenteritis in humans. A high incidence of C. jejuni in broiler flocks is often correlated to pathogen recovery in retail poultry meat, which is the main source of human infection. In this work broiler chickens were fed with a synbiotic product mixed with conventional feed using two different administration strategies. The synbiotic was formulated with the microencapsulated probiotic Bifidobacterium longum PCB133 and a xylo-oligosaccharide (XOS

From womb to world: The development of learning, memory, and predictive adaptation from fetal life through the neonatal period.

BACKGROUND: The developing fetus is increasingly recognized as an active participant in its own development rather than a passive recipient of sensory stimulation. Advances in fetal behavioral assessment, neuroimaging, electrophysiology, and developmental neuroscience have substantially expanded understanding of learning and memory before and immediately after birth. OBJECTIVE: To provide a comprehensive review of current evidence regarding fetal and neonatal learning and to examine the neural, behavioral, and developmental mechanisms that support the emergence of learning during the prenatal

Regional vulnerability in Huntington's disease: fMRI-guided molecular analysis in patients and a mouse model of disease.

Although the huntingtin gene is expressed in brain throughout life, phenotypically Huntington's disease (HD) begins only in midlife and affects specific brain regions. Here, to investigate regional vulnerability in the disease, we used functional magnetic resonance imaging (fMRI) to translationally link studies in patients with a mouse model of disease. Using fMRI, we mapped cerebral blood volume (CBV) in three groups: HD patients, symptom-free carriers of the huntingtin genetic mutation, and age-matched controls. In contrast to a region in the anterior caudate, in which dysfunction was linked

Docosahexaenoic acid protects from amyloid and dendritic pathology in an Alzheimer's disease mouse model.

Genetic data argues that Alzheimer's disease (AD) can be initiated by aggregates of a 42 amino acid beta amyloid peptide (Abeta42). The Abeta aggregates, notably small oligomer species, cause a cascade of events including oxidative damage, inflammation, synaptic toxicity and accumulation of intraneuronal inclusions; notably neurofibrillary tangles. Cognitive deficits are likely to begin with a failure of synaptogenesis and synaptic plasticity with dendritic spine loss and dying back of dendritic arbor. This is followed by neuron loss in key areas involved in learning and memory. Significant pr

Growth analysis in clinical genetics.

The number of genetic and environmental determinants of human growth is very large and their interplay is responsible for the enormously complex phenomenon of prenatal and postnatal growth and simultaneous differentiation in function. Genetic factors participate substantially in determining size at birth, but become more and increasingly important in the realization of inherent potential after the age of two years. Thus, prenatal growth is determined by a greater number of environmental factors than postnatal growth, including parity, pregnancy spacing, maternal age, size, blood pressure, race

Increased renal expression of monocyte chemoattractant protein-1 and osteopontin in ADPKD in rats.

BACKGROUND: Human autosomal-dominant polycystic kidney disease (ADPKD) is variable in the rate of deterioration of renal function, with end-stage renal disease (ESRD) occurring in only approximately 50% of affected individuals. Evidence suggests that interstitial inflammation may be important in the development of ESRD in ADPKD. Han:SPRD rats manifest ADPKD that resembles the human disease. Homozygous cystic (Cy/Cy) rats develop rapidly progressive PKD and die near age 3 weeks. Heterozygous (Cy/+) females develop slowly progressive PKD without evidence of renal dysfunction until the second yea

The origin and dispersion of human parasitic diseases in the old world (Africa, Europe and Madagascar).

The ancestors of present-day man (Homo sapiens sapiens) appeared in East Africa some three and a half million years ago (Australopithecs), and then migrated to Europe, Asia, and later to the Americas, thus beginning the differentiation process. The passage from nomadic to sedentary life took place in the Middle East in around 8000 BC. Wars, spontaneous migrations and forced migrations (slave trade) led to enormous mixtures of populations in Europe and Africa and favoured the spread of numerous parasitic diseases with specific strains according to geographic area. The three human plasmodia (Pla

Exploring the Early-Life Microbiomes of Preterm Infants and Their Childhood Health Outcomes (The BLOOM Study): Protocol for a Prospective, Observational Cohort Study.

BACKGROUND: Establishment of the gut microbiome during the first years of life is critically important for long-term health and development. In preterm infants, microbial colonization is disrupted due to their developmental immaturity and the myriad of pre- and postnatal exposures. Unfortunately, this places them at an elevated risk for adverse health outcomes into childhood (eg, asthma and impaired neurodevelopment). Understanding the developmental trajectory of the microbiome in preterm infants, the factors predicting these microbial patterns, and the links to childhood health can offer oppo

Alzheimer's disease is an inherent, natural part of human brain aging: an integrated perspective.

Alzheimer disease is one of the most challenging demons in our society due to its very high prevalence and its clinical manifestations which cause deterioration of cognition, intelligence, and emotions - the very capacities that distinguish Homo sapiens from other animal species. Besides the personal, social, and economical costs, late stages of AD are vivid experiences for the family, relatives, friends, and general observers of the progressive ruin of an individual who turns into a being with lower mental and physical capacities than less evolved species. A human brain with healthy cognition

Hsp70 May Be a Molecular Regulator of Schistosome Host Invasion.

Schistosomiasis is a debilitating disease that affects over 240 million people worldwide and is considered the most important neglected tropical disease following malaria. Free-swimming freshwater cercariae, one of the six morphologically distinct schistosome life stages, infect humans by directly penetrating through the skin. Cercariae identify and seek the host by sensing chemicals released from human skin. When they reach the host, they burrow into the skin with the help of proteases and other contents released from their acetabular glands and transform into schistosomula, the subsequent la

Toward tissue-engineering of nasal cartilages.

Nasal cartilage pathologies are common; for example, up to 80% of people are afflicted by deviated nasal septum conditions. Because cartilage provides the supportive framework of the nose, afflicted patients suffer low quality of life. To correct pathologies, graft cartilage is often required. Grafts are currently sourced from the patient's septum, ear, or rib. However, their use yields donor site morbidity and is limited by tissue quantity and quality. Additionally, rhinoplasty revision rates exceed 15%, exacerbating the shortage of graft cartilage. Alternative grafts, such as irradiated allo

A timeline of structural and functional consequences to ipRGCs in a mouse model of Alzheimer's disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder that affects cognitive, sensory and motor systems, including the visual system and has a significant impact on autonomy and quality of life. Emerging evidence suggests that visual system abnormalities may enable early detection and monitoring for AD, appearing before cognitive symptoms. Intrinsically photosensitive retinal ganglion cells (ipRGCs or mRGCs) are among the first neurons affected in AD. This study investigates the structural and functional changes in ipRGCs during aging. ipRGC and retinal ganglion cell (RGC) degen

A comprehensive review on NUI, multi-sensory interfaces and UX design for applications and devices for visually impaired users.

In today's world, there has been a significant increase in the use of devices, gadgets, and mobile applications in our daily activities. Although this has had a significant impact on the lives of the general public, people who are Partially Visually Impaired SPVI, which includes a much broader range of vision loss that includes mild to severe impairments, and Completely Visually Impaired (CVI), who have no light perception, still face significant obstacles when trying to access and use these technologies. Exploring the impact of adolescent exposure to cannabinoids and nicotine on psychiatric risk: insights from translational animal models.

Adolescence represents a highly sensitive period of mammalian neurodevelopment wherein critical synaptic and structural changes are taking place in brain regions involved in cognition, self-regulation and emotional processing. Importantly, neural circuits such as the mesocorticolimbic pathway, comprising the prefrontal cortex, sub-cortical mesolimbic dopamine system and their associated input/output centres, are particularly vulnerable to drug-related insults. Human adolescence represents a life-period wherein many individuals first begin to experiment with recreational drugs such as nicotine

The Nurse's Part in the Recognition of Cancer of the Breast.

Editor's note: From its first issue in 1900 through to the present day, AJN has unparalleled archives detailing nurses' work and lives over more than a century. These articles not only chronicle nursing's growth as a profession within the context of the events of the day, but they also reveal prevailing societal attitudes about women, health care, and human rights. Today's nursing school curricula rarely include nursing's history, but it's a history worth knowing. To this end, From the AJN Archives highlights articles selected to fit today's topics and times. This April 1915 article was written

Clinical and Demographic Evaluation of a Holoprosencephaly Cohort From the Kyoto Collection of Human Embryos.

Holoprosencephaly (HPE) is a genetically and phenotypically heterogeneous disorder involving developmental defects. HPE is a rare condition (1/10,000-20,000 newborns) but can be found as frequently as 1/250 among conceptions, suggesting that most HPE embryos are incompatible with postnatal life and result in spontaneous abortions during the first trimester of gestation. Beginning in 1961, the Kyoto University in Japan collected over 44,000 human conceptuses in collaboration with several hundred domestic obstetricians. Over 200 cases of HPE have been identified in the Kyoto collection, which re
